Effect of various aromatic compounds with different functional groups on enzymatic hydrolysis of microcrystalline cellulose and alkaline pretreated wheat straw

Low molecular aromatic compounds are detrimental to the enzymatic hydrolysis of lignocellu-lose.However,the specific role of their functional groups remains unclear.Here,a series of nine aromatic compounds as additives were tested to understand their effect on the hydrolysis yield of microcrystalline cellulose(MCC)and alkaline pretreated wheat straw.Based on the results,the inhibition of aldehyde groups on MCC was greater than that of carboxyl groups,whereas for the alkaline pretreated wheat straw case,the inhibitory effect of aldehyde groups was lower than that of carboxyl groups.Increased methoxyl groups of aromatic compounds reduced the inhibitory ef-fect on enzymatic hydrolysis of both substrates.Stronger inhibition of aromatic compounds on MCC hydrolysis was detected in comparison with the alkaline pretreated wheat straw,indicating that the substrate lignin can offset the inhibition to a certain extent.Among all aromatic com-pounds,syringaldehyde with one aldehyde group and two methoxyl groups improved the glucan conversion of the alkaline pretreated wheat straw.

CDC42 controls intestinal epithelial(IEC)stem cell(IESC)division.How aberrant CDC42 initiates intestinal inflammation or neoplasia is unclear.We utilized models of inflam-matory bowel diseases(IBD),colorectal cancer,aging,and IESC injury to determine the loss of intestinal Cdc42 upon inflammation and neoplasia.Intestinal specimens were collected to determine the levels of CDC42 in IBD or colorectal cancer.Cdc42 floxed mice were crossed with Villin-Cre,Villin-CreERT2 and/or Lgr5-eGFP-IRES-CreERT2,or Bmi1-CreERT2 mice to generate Cdc42 deficient mice.Irradiation,colitis,aging,and intestinal organoid were used to evaluate CDC42 upon mucosal inflammation,IESC/progenitor regenerative capacity,and IEC repair.Our studies revealed that increased CDC42 in colorectal cancer correlated with lower survival;in contrast,lower levels of CDC42 were found in the inflamed IBD colon.Colonic Cdc42 depletion significantly reduced Lgr5+IEsCs,increased progenitors'hyperplasia,and induced mucosal inflammation,which led to crypt dysplasia.Colonic Cdc42 depletion markedly enhanced irra-diation-or chemical-induced colitis.Depletion or inhibition of Cdc42 reduced colonic Lgr5+IESC regeneration.In conclusion,depletion of Cdc42 reduces the IESC regeneration and IEC repair,leading to prolonged mucosal inflammation.Constitutive monogenic loss of Cdc42 in-duces mucosal inflammation,which could result in intestinal neoplasia in the context of aging.

Cellular metabolism is the fundamental process by which cells maintain growth and self-renewal.It produces energy,furnishes raw materials,and intermedi-ates for biomolecule synthesis,and modulates enzyme activity to sustain normal cellular functions.Cellular metabolism is the foundation of cellular life processes and plays a regulatory role in various biological functions,including pro-grammed cell death.Ferroptosis is a recently discovered form of iron-dependent programmed cell death.The inhibition of ferroptosis plays a crucial role in tumorigenesis and tumor progression.However,the role of cellular metabolism,particularly glucose and amino acid metabolism,in cancer ferroptosis is not well understood.Here,we reviewed glucose,lipid,amino acid,iron and sele-nium metabolism involvement in cancer cell ferroptosis to elucidate the impact of different metabolic pathways on this process.Additionally,we provided a detailed overview of agents used to induce cancer ferroptosis.We explained that the metabolism of tumor cells plays a crucial role in maintaining intracellu-lar redox homeostasis and that disrupting the normal metabolic processes in these cells renders them more susceptible to iron-induced cell death,resulting in enhanced tumor cell killing.The combination of ferroptosis inducers and cel-lular metabolism inhibitors may be a novel approach to future cancer therapy and an important strategy to advance the development of treatments.

Media convergence is a media change led by technological innovation.Applying media convergence technology to the study of clustering in Chinese medicine can significantly exploit the advantages of media fusion.Obtaining consistent and complementary information among multiple modalities through media convergence can provide technical support for clustering.This article presents an approach based on Media Convergence and Graph convolution Encoder Clustering(MCGEC)for traditonal Chinese medicine(TCM)clinical data.It feeds modal information and graph structure from media information into a multi-modal graph convolution encoder to obtain the media feature representation learnt from multiple modalities.MCGEC captures latent information from various modalities by fusion and optimises the feature representations and network architecture with learnt clustering labels.The experiment is conducted on real-world multimodal TCM clinical data,including information like images and text.MCGEC has improved clustering results compared to the generic single-modal clustering methods and the current more advanced multi-modal clustering methods.MCGEC applied to TCM clinical datasets can achieve better results.Integrating multimedia features into clustering algorithms offers significant benefits compared to single-modal clustering approaches that simply concatenate features from different modalities.It provides practical technical support for multi-modal clustering in the TCM field incorporating multimedia features.

开环模式谐振理论是阐释风电并网引起的电力系统振荡机理的研究成果,其中基于平均模型的离线时域仿真结果可信度存疑。为提升开环模式谐振的工程指导可靠度,首先基于硬件在环仿真,组建了用于验证开环模式谐振的半实物实验平台;其次建立双馈风电场并网系统的小信号模型,提出开环模式分析通用方法评估系统振荡风险;最后以两个算例进行了开环模式谐振现象的分析与实验。得到结论:硬件在环仿真的结果符合开环模式分析的结果,在开环模式谐振条件下,系统稳定性下降。留数法可以较为准确地预测系统闭环模式,系统参数的合理整定可以减小开环模式谐振带来的影响。

目的探讨伴导管分化的食管癌临床病理学特征及分子遗传学特征,为其诊治积累经验。方法收集宁波市临床病理诊断中心2011年6月至2022年12月确诊的伴导管分化的食管癌17例,整理临床及病理诊断信息,通过HE切片观察其组织学特征,选择肿瘤组织区域获取DNA,采用二代测序技术检测并分析肿瘤标本中的基因突变情况。结果17例病例,男性16例,女性1例;患者年龄54~77岁,中位年龄66岁。其中9例以伴导管分化的癌为主,肿瘤表面鳞状上皮均伴有高级别上皮内瘤变,肿瘤与异型鳞状上皮有移行,局灶伴有不同程度的鳞状细胞癌成分(<10%);另外8例肿瘤主体为不同分化程度的鳞状细胞癌或基底样鳞状细胞癌或肉瘤样癌,局灶区域为伴导管分化的癌(<10%);伴导管分化区域的肿瘤细胞主要呈小管状排列,小管呈双层结构,即管腔内层细胞和外层细胞。免疫组织化学结果显示肿瘤性小管外层细胞表达p63、p40、细胞角蛋白(CK)5/6、CK34βE12,内层细胞表达CK7。与文献报道的食管鳞状细胞癌相比较,本组病例中MYC(P=0.002)、TP63(P=0.002)、CDKN1C(P=0.002)和NFE2L2(P=0.045)等基因突变发生频率更低;信号通路层面,NOTCH信号通路(P=0.041)突变发生频率更高,而NRF2通路(P=0.013)和PI3K通路(P=0.009)突变相对于食管鳞状细胞癌发生频率更低。结论伴导管分化的食管癌是一种形态学特殊的食管癌,分子改变与普通的食管鳞状细胞癌也存在明显差异。

目的探讨肾上腺腺瘤样瘤的临床病理学特征,为其诊治积累经验。方法收集宁波市临床病理诊断中心2013年1月至2022年12月诊断的肾上腺腺瘤样瘤7例以及上海交通大学医学院附属瑞金医院病理科2014年1月至2022年10月诊断的肾上腺腺瘤样瘤6例,分析研究其临床表现、病理学特点以及预后,并复习相关文献。结果患者男性12例,女性1例。肾上腺腺瘤样瘤几乎均为单侧单灶发病,有明显男性倾向,平均年龄和中位年龄均为42岁,内分泌激素水平基本正常,CT显示低密度结节影,可囊性变或伴点状钙化,多数边界清,术前多考虑无功能皮质腺瘤或不能排除恶性,肿瘤大体实性或囊实性,平均最大径3.4 cm,镜下呈浸润性生长,排列成不规则腔隙样、囊状或实性,衬覆扁平或立方状细胞,实性区细胞上皮样、印戒样或梭形,无异型性,绝大多数病例间质伴有淋巴细胞浸润;免疫组织化学表达间皮和上皮的标志物,多数伴有L1CAM蛋白高表达。结论肾上腺腺瘤样瘤是一种罕见的间皮来源的良性肿瘤,术前难以明确诊断,病理学及免疫组织化学帮助确诊,预后好。

以2066名中小学生为研究对象,采用问卷调查法考察校园欺凌中学生旁观行为的性别和年级差异,并深入探讨其背后的道德推脱机制。结果发现:(1)男生的协同欺凌、煽风点火和保护行为水平高于女生。(2)随着年级的升高,协同欺凌行为在小学和初中分别表现出下降和增长趋势,保护行为在小学和初中分别表现出增长和下降趋势,置身事外行为在小学和初中均表现出增长趋势,煽风点火行为从小学到初中均表现出增长趋势。(3)协同欺凌和煽风点火行为的道德推脱机制为认知重建,委婉标签、有利比较和道德辩护均能显著正向预测协同欺凌和煽风点火行为。(4)除扭曲结果外,保护行为和置身事外行为的其他道德推脱机制作用不同,具体表现为有利比较、责任转移能显著负向预测保护行为,责备归因能显著正向预测置身事外行为,委婉标签和非人性化能显著负向预测保护、正向预测置身事外行为。因此,树立正确的道德认知是校园欺凌干预方案的关键内容,且干预的旁观行为不同,学生道德推脱认知转变的重点也不同。

为了实现物流分拣多目标精准跟踪,提出一种激光雷达融合机器视觉的物流分拣多目标视频跟踪方法。通过激光雷达采集检测环境内的点云数据,并对其滤波处理。将机器视觉采集的物流分拣视频去噪,采用Sobel边缘检测算子检测去噪后的图像边缘,引入分裂合并算法对去噪图像展开多区域分割处理,获取比较明显的多目标视频区域。利用透视变换原理对图像数据融合处理,实时确定目标位置,实现物流分拣多目标视频跟踪。经过实验测试证明,所提方法的物流分拣多目标视频跟踪结果准确率超过95%,且延误时间控制在13 ms以内。
