Identifying disease-associated RNAs is crucial in revealing the pathogenic mechanisms of diseases [1], and biologists have made notable progress in this field [2]. Available online Immunoglobulins G(IgGs)are Y-shaped globular proteins,however,their high flexibility and heterogeneity pose great challenges to their structure and conformation *** structure of IgG closely correlates to its biofunctions,such as the antibody escape of human immunodeficiency virus(HIV)could attribute to the distance mismatch between the ends of two Fab arms(antigen-binding sites)and envelope glycoprotein spikes on virion ***,we report the first use of mobility capillary electrophoresis(MCE)and native mass spectrometry(nMS)to resolve the internal geometric structure and conformation of an IgG(trastuzumab)in solution *** proteolysis,the ellipsoid dimensions of IgG and its subunits were measured by MCE-nMS *** was then reconstructed,in which the sizes and relative positions of these three subunits in three-dimensional space were *** was found that the two Fab arms have an angle of~102.1°and a distance of~11.0 nm between the two antigen-binding sites under native condition,and the Fc arm was tilted~16.0°towards one of the Fab *** was not on the plane of Fab-Fab,but has an angle of no larger than 103.1°.Under acidic environment(pH 3.0),each subunit of the IgG would unfold into larger dimensions,and the angles between these subunits also *** great potential for tumor imaging and therapy,the structure of F(ab')_(2)fragments was also measured and validated by molecular dynamic *** was found that the electrostatic force among these three subunits and steric hindrance stemming from Fc help maintaining the angle between two Fab arms.

Increasing evidence suggests that deregulated RNA splicing factors play critical roles in tumorigenesis; however, their specific involvement in colon cancer remains largely unknown. Here we report that the splicing factor RBM25 is overexpressed in colon cancer, and this increased expression correlates with a poor prognosis of patients with colon cancer. Functionally, RBM25 ablation suppresses the growth of colon cancer cells both in vitro and in vivo. Mechanistically, our transcriptome-wide analysis of splicing events revealed that RBM25 regulates a large number of cancer-related alternative splicing events across the human genome in colon cancer. Particularly, RBM25 regulates the splicing of MNK2 by interacting with the poly G rich region in exon 14a, thereby inhibiting the selection of the proximal 3'splice site(ss), resulting in the production of the oncogenic short isoform, MNK2b. Knockdown of RBM25 leads to an increase in the MNK2a isoform and a decrease in the MNK2b isoform. Importantly, re-expression of MNK2b or blocking the 3′ ss of the alternative exon 14a with ASO partially reverses the RBM25 knockdown mediated tumor suppression. Moreover, MNK2b levels were significantly increased in colon cancer tissues, which is positively correlated with the expression level of RBM25. Collectively, our findings uncover the critical role of RBM25 as a key splicing factor in colon cancer, suggesting its potential as a prognostic marker and therapeutic target.

Extensive efforts have been made to pursue a low-friction state with promising applications in many fields,such as mechanical and biomedical *** which,the load capacity of the low-friction state has been considered to be crucial for industrial ***,we report a low friction under ultrahigh contact pressure by building a novel self-assembled fluorinated azobenzene layer on an atomically smooth highly-oriented pyrolytic graphite(HOPG)*** friction coefficients could be as low as 0.0005 or even lower under a contact pressure of up to 4 *** demonstrates that the low friction under ultrahigh contact pressure is attributed to molecular *** fluorination leads to effective and robust lubrication between the tip and the self-assembled layer and enhances tighter rigidity which can reduce the stress concentration in the substrate,which was verified by density functional theory(DFT)and molecular dynamics(MD)*** work provides a new approach to avoid the failure of ultralow friction coefficient under relatively high contact pressure,which has promising potential application value in the future.

Global climate change is characterized by asymmetric warming,i.e.,greater temperature increases in winter,spring,and nighttime than in summer,autumn,and *** experiments were conducted using four wheat cultivars,namely‘Yangmai 18’(YM18),‘Sumai 188’(SM188),‘Yannong 19’(YN19),and‘Annong 0711’(AN0711),in the two growing seasons of 2019-2020 and 2020-2021,with passive night warming during different periods in the early growth *** treatments were night warming during the tillering-jointing(NW_(T-J)),jointing-booting(NWJ-B),and booting-anthesis(NWB-A)stages,with ambient temperature(NN)as the *** effects of night warming during different stages on wheat yield formation were investigated by determining the characteristics of dry matter accumulation and translocation,as well as sucrose and starch accumulation in wheat *** wheat yields of all four cultivars were significantly higher in NW_(T-J)than in NN in the 2-year *** yield increases of semi-winter cultivars YN19 and AN0711 were greater than those of spring cultivars YM18 and *** NW_(T-J)increased wheat yield mainly by increasing the 1,000-grain weight and the number of fertile spikelets,and it increased dry matter accumulation in various organs of wheat at the anthesis and maturity stages by increasing the growth rate at the vegetative growth *** flag leaf and spike showed the largest increases in dry matter ***_(T-J)also increased the grain sucrose and starch contents in the early and middle grain-filling stages,promoting yield ***,night warming between the tillering and jointing stages increased the pre-anthesis growth rate,and thus,wheat dry matter production,which contributed to an increase in wheat yield.

Objective To investigate whether melatonin(MT)secretion in different parts of the gastrointestinal tract(GIT)exhibits seasonal variations and its correlation with immune *** Sixty Sprague-Dawley rats were divided into control and model groups,and the pineal gland was removed in the model ***,jejunum,ileum,and colon tissues were obtained during the spring equinox,summer solstice,beginning of autumn,autumn equinox,and winter *** levels of MT,MT receptors(MR),arylalkylamine N-acetyltransferase(AANAT),hydroxyindole-O-methyltransferase(HIOMT),interleukin-2(IL-2),and interleukin-10(IL-10)in the GIT were measured using enzyme-linked immunosorbent *** Except for the stomach,the jejunum,ileum,and the colon showed seasonal tendencies in MT *** the control group,MT secretion in the jejunum and ileum was the highest in the long summer,and colonic MT secretion was the highest in *** the model group,MT levels in the colon were highest in the *** seasonal rhythms of the MR,AANAT,HIOMT,IL-2,and IL-10 in the colon were roughly similar to those of MT,and changed accordingly after *** Gastrointestinal MT secretion is related to seasonal changes,and MT secretion in each intestinal segment is influenced by different *** biological effects of MT in the gut are inextricably linked to the mediation of MR,and a hormone-receptor linkage exists between MT and *** effect of seasonal changes on the gastrointestinal immune system may be mediated through the regulation of seasonal secretion of MT.

Correlation between Neutrophil Percentage-to-albumin Ratio and Coronary Heart Disease Complicated with Diabetes Mellitus

[Objectives]This study was conducted to explore the relationship between neutrophil percentage-to-albumin ratio(NPAR)and coronary heart disease complicated with diabetes.[Methods]A total of 603 patients with coronary heart disease who underwent coronary angiography in Pingquan County Hospital from January,2023 to December,2023 and met the inclusion criteria were included as the research *** the patients were divided into a coronary heart disease complicated with diabetes group(CAD+T2DM group)(n=298 cases)and a control group(CAD group)(n=305 cases),according to patients medical history,heart color ultrasound and biochemical test *** clinical data,biochemical test results and coronary artery imaging data of patients were recorded,and the Gensini score was *** neutrophil percentage(NEUT%)and albumin count were determined to calculate NPAR.[Results]The NPAR value of the coronary heart disease complicated with diabetes mellitus group was(1.6±0.42),which was significantly higher than that of the control group(1.47±0.49),and the difference was statistically significant(P<0.05).The area under the ROC curve was 0.619(95%CI:0.591-0.675,P<0.05),and the prediction of coronary heart disease complicated with diabetes using NPAR showed a Youden index of 0.31,a sensitivity of 60.4%,a specificity of 40.3%,and a best cut-off score of 1.4506.[Conclusions]The neutrophil percentage-to-albumin ratio(NPAR)is closely related to coronary heart disease complicated with diabetes mellitus,and NPAR has clinical application value in the diagnosis of coronary heart disease complicated with diabetes mellitus.

《周礼》所构建的赋税体系中,有一条现今并未发现在先秦两汉时期实际实施过的征课政策,即“唯其漆林之征二十而五”,对漆林进行高税率课税。后世学者对此非常科税收政策,遵循两条解释理路:一为“抑末”、二为“返朴”。不过,这两种解释与《周礼》文本都不够契合。本文通过考察古代中国经济作物的征税政策,结合《周礼》文本与唐代茶税出现的历史背景,对漆林征课做出解释。
