Gap junction(GJ)has been indicated to have an intimate correlation with adhesion junction.However,the direct interaction between them partially remains elusive.In the current study,we aimed to elucidate the role of N-cadherin,one of the core components in adhesion junction,in mediating connexin 43,one of the functional constituents in gap junction,via transforming growth factor-β1(TGF-β1)induction in osteoblasts.We first elucidated the expressions of N-cadherin induced by TGF-β1 and also confirmed the upregulation of Cx43,and the enhancement of functional gap junctional intercellular communication(GJIC)triggered by TGF-β1 in both primary osteoblasts and MC3T3 cell line.Colocalization analysis and Co-IP experimentation showed that N-cadherin interacts with Cx43 at the site of cell–cell contact.Knockdown of N-cadherin by siRNA interference decreased the Cx43 expression and abolished the promoting effect of TGF-β1 on Cx43.Functional GJICs in living primary osteoblasts and MC3T3 cell line were also reduced.TGF-β1-induced increase in N-cadherin and Cx43 was via Smad3 activation,whereas knockdown of Smad3 signaling by using siRNA decreased the expressions of both N-cadherin and Cx43.Overall,these data indicate the direct interactions between N-cadherin and Cx43,and reveal the intervention of adhesion junction in functional gap junction in living osteoblasts.

The automation of traditional Chinese medicine(TCM)pharmaceuticals has driven the development of process analysis from offline to online.Most of common online process analytical technologies are based on spectroscopy,making the identification and quantification of specific ingredients still a challenge.Herein,we developed a quality control(QC)system for monitoring TCM pharmaceuticals based on paper spray ionization miniature mass spectrometry(mini-MS).It enabled real-time online qualitative and quantitative detection of target ingredients in herbal extracts using mini-MS without chromatographic separation for the first time.Dynamic changes of alkaloids in Aconiti Lateralis Radix Praeparata(Fuzi)during decoction were used as examples,and the scientific principle of Fuzi compatibility was also investigated.Finally,the system was verified to work stably at the hourly level for pilot-scale extraction.This mini-MS based online analytical system is expected to be further developed for QC applications in a wider range of pharmaceutical processes.

Community composition,co-occurrence,and environmental drivers of bacterioplankton community in surface and 50-m water layers in the subarctic North Pacific

The Western Subarctic Gyre(WSG)is one of the two gyre-systems in the subarctic North Pacific known for high nutrient and low-chlorophyll waters.However,the bacterioplankton in marine water of this area,either in terms of the taxonomic composition or functional structure,remains relatively unexplored.A total of 22 sampling sites from two water layers(surface water,SW and 50-m layer water,FW)were collected in this area.The physiochemical parameters of waters,Synechococcus,and bacterial density,as well as the bacterioplankton community composition and distribution pattern,were analyzed.The nutrient concentrations of DIN,DIP,and DSi,Chl-a concentration,and the average abundance of heterobacteria in FW were higher than those in SW.However,temperature and the average abundance of Synechococcus and pico-eukaryotes were higher in SW.A total of 3269 OTUs were assigned,and 2123OTUs were commonly shared;moreover,similar alpha diversity patterns were observed in both SW and FW.The bacterioplankton community showed significantly obvious correlation with salinity,DIP,DIN,and Chl a in both SW and FW.Proteobacteria,Cyanobacteria,Bacteroidota,Actinobacteriota,and Firmicutes were the main phyla while Synechococcus_CC9902,Psychrobacter,and Sulfitobacter were the dominant genera in each sampling site.Most correlations that happened between the OTUs in the cooccurrence network were positive and inter-module.Higher edges and graph density were found in SW,indicating that more correlations occurred,and the community was more complex in SW.This study provided novel knowledge on the bacterioplankton community structure and the correlation characteristics in WSG.

Graphene oxide(GO)exhibits good mechanical and physicochemical characteristics and has extensive application prospects in bone tissue engineering.However,its effect on angiogenesis is unclear,and its potential toxic effects are heavily disputed.Herein,we found that nanographene oxide(NGO)synthesized by one-step water electrolytic oxidation is smaller and shows superior biocompatibility.Moreover,NGO significantly enhanced angiogenesis in calvarial bone defect areas in vivo,providing a good microenvironment for bone regeneration.Endothelial tip cell differentiation is an important step in the initiation of angiogenesis.We verified that NGO activates endothelial tip cells by coupling with lysophosphatidic acid(LPA)in serum via strong hydrogen bonding interactions,which has not been reported.In addition,the mechanism by which NGO promotes angiogenesis was systematically studied.NGO-coupled LPA activates LPAR6 and facilitates the formation of migratory tip cells via Hippo/Yes-associated protein(YAP)independent of reactive oxygen species(ROS)stimulation or additional complex modifications.These results provide an effective strategy for the application of electrochemically derived NGO and more insight into NGO-mediated angiogenesis.

Diffusion mechanism of immiscible Fe-Mg system induced by high-density defects at the steel/Mg composite interface

Due to positive mixing heat between Fe and Mg,it is difficult to diffuse for Fe-Mg at the interface of steel/Mg laminated composites,resulting in the inability to achieve high-strength metallurgical bonding.In this paper,20#steel/Mg laminated composites were prepared by large deformation rolling and subse-quent diffusion heat treatment process.The interfacial bonding strength was improved by constructing high-density crystal defects at the interface to promote element diffusion.The mechanisms of interface morphology evolution and element diffusion were analyzed by finite element simulation and theoretical calculation.The results show after diffusion heat treatment,the bond strength of the large deformation rolled interface was increased from 14 to 30 MPa.Fe-Mg transition layer with about 80 nm thickness as well as high-density vacancies,dislocations and grain boundaries were formed in the large deforma-tion rolled interface region.During diffusion heat treatment,Mg elements diffused into grain interior and grain boundary regions of 20#steel under the effect of heat-force coupling,and the thickness of Fe-Mg transition layer increased to 150 nm,forming an Fe-based supersaturated solid solution.The in-terface with high-density defects constituted a non-equilibrium interface.The 20#steel internal energy in the non-equilibrium interface is able to overcome positive mixing heat of immiscible Fe-Mg system and provide the driving force for Mg elements diffusion.Promoting elemental diffusion by constructing high-density defects can be a new concept to achieve metallurgical bonding at the interface of immiscible metal laminated composites.

The use of tree shrews as experimental animals for biomedical research is a new practice.Several recent studies suggest that tree shrews are suitable for studying cancers,including breast cancer,glioblastoma,lung cancer,and hepatocellular carcinoma.However,the telomeres and the telomerase of tree shrews have not been studied to date.Here,we characterize telomeres and telomerase in tree shrews.The telomere length of tree shrews is approximately 23 kb,which is longer than that of primates and shorter than that of mice,and it is extended in breast tumor tissues according to Southern blot and flow-fluorescence in situ hybridization(FISH)analyses.Tree shrew spleen,bone marrow,testis,ovary,and uterus show high telomerase activities,which are increased in breast tumor tissues by telomeric repeat amplification protocol assays.The telomere length becomes shorter,and telomerase activity decreases with age.The tree shrew TERT and TERC are more highly similar to primates than to rodents.These findings lay a solid foundation for using tree shrews to study aging and cancers.

Inflammatory bowel disease(IBD)has become a global disease with accelerating incidence worldwide in the 21st century while its accurate etiology remains unclear.In the past decade,gut microbiota dysbiosis has consistently been associated with IBD.Although many IBDassociated dysbiosis have not been proven to be a cause or an effect of IBD,it is often hypothesized that at least some of alteration in microbiome is protective or causative.In this article,we selectively reviewed the hypothesis supported by both association studies in human and pathogenesis studies in biological models.Specifically,we reviewed the potential protective bacterial pathways and species against IBD,as well as the potential causative bacterial pathways and species of IBD.We also reviewed the potential roles of some members of mycobiome and virome in IBD.Lastly,we covered the current status of therapeutic approaches targeting microbiome,which is a promising strategy to alleviate and cure this inflammatory disease.

Over the last few years,great advances have been achieved in exploration of high-mobility two-dimensional(2D)semiconductors such as metal chalcogenide InSe and noble-transition-metal dichal-cogenide PdSe_(2).These materials are competitive candidates for constructing next-generation optoelec-tronic devices owing to their unique crystalline and electronic structures.Moreover,the optical and electronic properties of 2D materials can be efficiently modified via precisely engineering their band structures,which is critical for widening specific applications ranging from high-performance opto-electronics to catalysis and energy harvesting.In this review,we focus on the progress in bandgaps engineering of newly emerging high-mobility 2D semiconductors and their applications in optoelec-tronic devices,incorporating our recent study in the InSe and PdSe_(2)systems.First of all,we discuss the structure-property relationship of typical high-mobility 2D semiconductors(InSe and PdSe 2).Next,we analyze several viable strategies for bandgap engineering,including thickness,strain or pressure,alloying,heterostructure,surface modification,intercalation,and so on.Furthermore,we summarize the optoelectronic devices fabricated with such high-mobility 2D semiconductors.The conclusion and outlook in this topic are finally presented.This review aims to provide valuable insights in bandgap engineering of newly emerging 2D semiconductors and explore their potential in future optoelectronic applications.

Fault Attribute Reduction of Oil Immersed Transformer Based on Improved Imperialist Competitive Algorithm

The original fault data of oil immersed transformer often contains a large number of unnecessary attributes,which greatly increases the elapsed time of the algorithm and reduces the classification accuracy,leading to the rise of the diagnosis error rate.Therefore,in order to obtain high quality oil immersed transformer fault attribute data sets,an improved imperialist competitive algorithm was proposed to optimize the rough set to discretize the original fault data set and the attribute reduction.The feasibility of the proposed algorithm was verified by experiments and compared with other intelligent algorithms.Results show that the algorithm was stable at the 27th iteration with a reduction rate of 56.25%and a reduction accuracy of 98%.By using BP neural network to classify the reduction results,the accuracy was 86.25%,and the overall effect was better than those of the original data and other algorithms.Hence,the proposed method is effective for fault attribute reduction of oil immersed transformer.

Circular RNAs (circRNAs) are a special class of single-stranded RNA molecules with covalently closed loops widely expressed in eukaryotic organisms. CircRNAs have long been considered to play important roles in various physiological and pathological processes as non-coding RNAs. However, circRNAs have recently garnered considerable attention due to their ability to be translated into peptides/proteins via internal ribosome entry site- or N6-methyladenosine-mediated pathways or rolling translation mechanisms. Furthermore, dysregulation of translatable circRNAs and their encoded proteins has been associated with developing and progressing diseases such as cancer. This review aims to summarize the driving mechanisms of circRNA translation and the available strategies in circRNA translation research. The main focus is on the emerging biological functions of translatable circRNAs, their regulatory mechanisms, and potential clinical applications in human diseases to provide new perspectives on disease diagnosis, prognosis, and targeted therapy.
