Characterization of SOX3 Gene in an Ovoviviparous Teleost, Black Rockfish(Sebastes schlegeli)

Evolutionary evidence suggests that Sox3, a member of the high-mobility-group(HMG) family of transcription factors, is an ancestral precursor of Sry and is involved in sex determination similar to Sry. However, there is limited information regarding the SOX3 gene of the black rockfish(Sebastes schlegeli). In this study, we first isolated SOX3 gene from the gonads of S. schlegeli by homology cloning. The full-length of S. schlegeli SOX3(SsSOX3) c DNA was 1386 bp, comprising a 906-bp open reading frame, which encodes a peptide showing 93.6% and 93.9% homology with the Sox3 proteins of Epinephelus coioides and Oryzias latipe, respectively. Comparison of the cDNA sequence of the Ss SOX3 gene with the corresponding genomic DNA fragment revealed that the SsSOX3 gene consists of a single exon. Phylogenetic analysis demonstrated the evolutionary relationship of Ss SOX3 with other known SOXB1 genes in fish and tetrapods. The promoter region contains binding sites of several transcriptional factors that might participate in the regulation of Ss SOX3 expression. Quantitative real-time PCR analysis indicated that SsSOX3 was expressed in all the investigated larval developmental stages from 1 to 35 days after birth and the level of expression gradually decreased as the development proceeded. SsSOX3 exhibited sexually dimorphic expression in adult gonads, with high expression in the ovary but low expression in the testis. In situ hybridization revealed that SsSOX3 was strongly expressed in oocytes and follicular cells of ovaries but slightly expressed in germ cells of testicular tissues. Therefore, this study suggests that Ss SOX3 plays an important role in oogenesis and ovary differentiation in S. schlegeli.

Purpose:To improve our understanding of macular schisis in highly myopic eyes. Methods:From 2002.6-2003.12, the highly myopic eyes(>6D) which has macular schisis found by OCT at our hospital were included in the study. Each eye then received further examinations including visual acuity,refractive error and ultra-sonography. Then the results were analyzed to find the characteristic of the entity. Results:During that period, OCT found macular schisis in 38 highly myopic eyes from 36 patients. Their vision was low, and their average axial length and refractive error were 29.6mm and -10.5D respectively. The appearance and the area of the schisis are different among the eyes. But the schisis can be largely divided into two types: inner and outer schisis. They can exit independently or concurrently in the same eye. And the schisis was often accompanied by other macular findings, like retinal detachment, macular hole. Conclusion:Macular schisis in highly myopic eyes is not uncommon in the clinic. OCT is useful in its diagnosis.

Molecular Cloning, Expression Pattern, and 3D Structural Prediction of the Cold Inducible RNA- Binding Protein(CIRP) in Japanese Flounder(Paralichthys olivaceus)

Cold-inducible RNA-binding protein(CIRP) is a kind of RNA binding proteins that plays important roles in many physiological processes. The CIRP has been widely studied in mammals and amphibians since it was first cloned from mammals. On the contrary, there are little reports in teleosts. In this study, the Po CIRP gene of the Japanese flounder was cloned and sequenced. The genomic sequence consists of seven exons and six introns. The putative Po CIRP protein of flounder was 198 amino acid residues long containing the RNA recognition motif(RRM). Phylogenetic analysis showed that the flounder Po CIRP is highly conserved with other teleost CIRPs. The 5' flanking sequence was cloned by genome walking and many transcription factor binding sites were identified. There is a Cp Gs region located in promoter and exon I region and the methylation state is low. Quantitative real-time PCR analysis uncovered that Po CIRP gene was widely expressed in adult tissues with the highest expression level in the ovary. The m RNA of the Po CIRP was maternally deposited and the expression level of the gene was regulated up during the gastrula and neurula stages. In order to gain the information how the protein interacts with m RNA, we performed the modeling of the 3D structure of the flounder Po CIRP. The results showed a cleft existing the surface of the molecular. Taken together, the results indicate that the CIRP is a multifunctional molecular in teleosts and the findings about the structure provide valuable information for understanding the basis of this protein's function.

Acute kidney injury(AKI)is a common clinical complication associated with high mortality in *** cells and cytokines have recently been described to play essential roles in AKI *** dendritic cells(pDCs)are a unique DC subset that specializes in type Ⅰ interferon(IFN)***,we showed that pDCs rapidly infiltrated the kidney in response to AKI and contributed to kidney damage by producing IFN-α.Deletion of pDCs using DTR^(BDCA2) transgenic(Tg)mice suppressed cisplatin-induced AKI,accompanied by marked reductions in proinflammatory cytokine production,immune cell infiltration and apoptosis in the *** contrast,adoptive transfer of pDCs during AKI exacerbated kidney *** further identified IFN-α as the key factor that mediated the functions of pDCs during AKI,as IFN-α neutralization significantly attenuated kidney ***,IFN-α produced by pDCs directly induced the apoptosis of renal tubular epithelial cells(TECs)in *** addition,our data demonstrated that apoptotic TECs induced the activation of pDCs,which was inhibited in the presence of an apoptosis ***,similar deleterious effects of pDCs were observed in an ischemia reperfusion(IR)-induced AKI ***,increased expression of IFN-α in kidney biopsies was observed in kidney transplants with *** together,the results of our study reveal that pDCs play a detrimental role in AKI via IFN-α.

Effects of hetero-multinuclear Pt-Ru metal complexes on the fibril foramtion of human islet amyloid polypeptide

Human islet amyloid polypeptide（hIAPP） is synthesized by islets beta cell of langerhans, and consists of 37 amino acids. Misfolding and fibrillization of hIAPP is related to type II diabetes mellitus（T2DM） [1]. Inhibitors of hIAPP aggregation include aromatic organic molecules, short peptides and metal complexes. Vanadium complexes have been used as diabetes drugs for more than 100 years[2]. However, the medicinal mechanism is not very clear. In this work, four Schiff base oxidovandium（IV） complexes were selected to suppress the fibril formation of hIAPP. The results indicated that four oxidovandium（IV） complexes effectively inhibited the fibrillization of hIAPP, changed the aggregates into monomers and oligomers, and decreased the cytotoxicity induced by hIAPP. The interaction mechanism between the complexes and the peptide was also investigated. Our research provides a new outlook in the design of vanadium complexes as potential inhibitors of hIAPP against T2DM.
