Efficacy of Qifu lizhong enema prescription(芪附理中灌肠方)on intestinal mucosal tight junction function modulation of ulcerative colitis rat model

OBJECTIVE:To investigate the efficacy and mechanism of Qifu lizhong enema prescription(芪附理中灌肠方,QFlZ)on intervening ulcerative colitis(UC)rat model with TCM spleen and kidney Yang insufficiency syndrome METHODS:Seventy-two male Sprague-Dawley rats were randomly assigned to six groups:normal model,mesalazine,and QFlZ high,medium,and low dose groups,each with 12 rats.After 3 d of adaptation feeding,all groups except the normal group were induced using rhubarb decoction in combination with trinitrobenzene sulfonic acid(TNBS)/55%ethanol to establish a UC rat model.Following successful modeling,the normal and model groups received daily saline enema,while the Chinese medicine and Western medicine groups received daily QFlZ and Mesalazine enema for 2 weeks respectively.The disease activity index score,hematoxylin and eosin staining,immunohistochemistry,and Western blotting were used to determine the expression of claudin 1,claudin 2,zonula occludens-1protein(ZO-1),and F-actin proteins in each rat colon tissue following treatment.RESUlTS:QFlZ significantly alleviated the structural disorganization in the form of epithelial glands in the intestinal mucosa of rats with UC and retarded the progression of the disease.The intestinal mucosal epithelial cells of UC rats showed decreased expression of claudin 1,ZO-1,F-actin(Plaudin 2 appeared elevated(Plted in impaired TJ.Treatment with QFlZ resulted in elevated expression of claudin 1(Plaudin 2(Pllowed for repair of the intestinal mucosal TJ,which in turn served as a treatment for UC.CONClUSIONS:The mechanism of repairing TJ function and repairing the intestinal mucosal barrier by QFlZ may be associated with up-regulation of claudin 1,ZO-1,and F-actin levels,and down-regulation of claudin 2 expression level.

Since 2005,GlP-1 receptor(GlP-IR)agonists(GlP-IRAs)have been developed as therapeutic agents for type 2 diabetes(T2D).GlP-IR is not only expressed in pancreatic islets but also other organs,especially the lung.However,controversy on extra pancreatic GlP-IR expression still needs to be further resolved,utilizing different tools including the use of more reliable GlP-IR antibodies in immune-staining and co-immune-staining.Extra-pancreatic expression of GlP-IR has triggered extensive investigations on extra-pancreatic functions of GlP-1RAs,aiming to repurpose them into therapeutic agents for other disorders.Extensive studies have demonstrated promising anti-infammatory features of GlP-IRAs.Whether those features are directly mediated by GlP-IR expressed in immune cells also remains controversial.Following a brief review on GlP-1 as an incretin hormone and the development of GlP-IRAs as therapeutic agents for T2D,we have summarized our current understanding of the antiinflammatory features of GlP-IRAS and commented on the controversy on extra-pancreatic GlP-IR expression.The main part of this review is a literature discussion on GlP-IRA utilization in animal models with chronic airway diseases and acute lung injuries,including studies on the combined use of mesenchymal stem cell(MSC)based therapy.This is followed by a brief summary.

Eukaryotic cells are confined by membranes that create hydrophobic barriers for substance and information exchange between the inside and outside of the cell.These barriers are formed by assembly of lipids and protein in aqueous environments.lipids not only serve as building blocks for membrane construction,but also possess regulatory functions in cellular activities.These regulatory lipids are non-uniformly distributed in membrane systems;their temporal and spatial accumulation in specific membranes decodes environmental cues and changes cellular activity accordingly.Phosphoinositides(PIs)are phospholipids that exert regulatory effects.In recent years,research on PIs roles in regulating plant growth,development,and responses to environmental stress is increasing.Several reviews have been published on the composition of PIs,intermolecular transferring of PIs by lipid kinases(phosphatases)or PI-PlCs,subcellular localization,and specially their functions in plant developments.Herein,we review the crucial regulatory functions of PIs in plant stress responses,with a particular focus on PIs involved in membrane trafficking.

The Dong-Xi-Taijinaier Salt lake(DXTSl) of Qaidam Basin is known as one of rich lithium reserves in China.However,the source of lithium deposits in this region remains unclear.The hydrochemistry and lithium isotopes(δ7li) of water and solid samples along the Nalenggele River catchment were studied in combination with rock leaching experiments.The results showed the hot springs in the upstream contained high content of lithium and ratio of li × 1 000/total dissolved solids(1.5–3.5),but the lower δ7li values(4.33‰–11.59‰),meaning that it is one important lithium sources.The leaching experiments by the rich-lithium volcanic rocks(average content of 474.7 mg/kg) showed that under temperature of 20–80 ℃ and long time duration(24–96 h),the concentration of lithium released by leaching of the sampled volcanic rocks increased from 0.58 to 1.49 mg/l,with the leaching rate decreasing from 0.024 to 0.015 mg/(l·h) and the corresponding lithium release rate increasing from 29% to 74%.Calculation based on water-salt balance showed that the total amount of lithium contribution from hydrothermal fluids and volcanic rock weathering is about 4.99 and 0.29 Mt since 13.1 ka,respectively.Therefore,hot springs and leaching of volcanic rocks are main contributors to lithium deposit in DXTSl.

Alginate lyase mainly produces active alginate oligosaccharides(AOS)by degrading alginate viaβ-elimination process.In this study,the Pseudoalteromonas sp.Alg6B alginate lyase-encoding gene alg6B-7 from polysaccharide lyase(Pl)-7 family was successfully cloned,sequenced,expressed in Escherichia coli.Based on rational design and amino acid sequence alignment of the alginate lyase from various sources,four positive mutants were obtained.The specific enzyme activities of four mutants I62A,A99K,V132S,and l157T were 38.84%,42.85%,75.8%and 51.83%higher than that of the wild enzyme,respectively.The K_(cat)/K_(m) values of the four mutants were both increased,and the catalytic efficiency of V132S was 1.92-fold higher than that of the wild enzyme,especially.The rational design that was employed in this study achieved the dramatic improvement of catalytic activity,which may provide the application potential in industrial production.

Potato(Solanum tuberosum l.)is susceptible to enzymatic browning after fresh processing,resulting in color change and potential alteration in the nutritional quality.In this study,a popular potato cultivar,Feiwuruita,was used to profile the metabolites involved in color and nutritional quality changes in fresh shreds stored at 0 and 4 h at 25°C(designated CK and CK4H,respectively).The shreds turned brown within 4 h of storage.In all,723 metabolites consisting 12 classes of compounds were detected in the samples,largely lipids,phenolic acids,alkaloids,amino acids and derivatives,flavonoids,organic acids,nucleotides and derivatives.Of these,163 metabolites accumulated differentially between CK and CK4H shreds.Polyphenolic compounds(phenolic acids and flavonoids)mostly increased in the shreds after 4 h storage.Conversely,the short-term storage drastically reduced lipid compounds(25 lysoPC and 19 lysoPE),while essential alkaloids and terpenoid compounds that are beneficial to human health increased in accumulation.The findings present global metabolome and nutritional composition changes in short-term stored shreds of Feiwuruita.This study provides important foundation for future studies on browning prevention/reduction and for better utilization of Feiwuruita.
