Harnessing bacteria for tumor therapy:Current advances and challenges

After a century of standstill,bacteria-based tumor therapy has resurged recently benefiting from the revolution of tumor immunotherapy,which provides unique solutions to tackle the obstacles of traditional tumor treatments.Obligate and facultative anaerobes with active tropism can selectively colonize at tumor sites and suppress tumor growth via different mechanisms,serving as attractive tools for tumor treatment either as a monotherapy or combining with other therapies for synergistic anti-tumor effects.In this critical review,we introduce the recent advances of bacteria-based tumor therapy from the following aspects.First,the general properties of bacteria are reviewed emphasizing on their structural components related to tumor immunotherapy,and the main bacteria that have been used in tumor therapy are listed.Then,the benefits of bacteria for tumor therapy are illustrated,such as tumor targetability,deep penetration,and facile genetic engineering for attenuation,enhanced efficacy,as well as bioimaging.Next,anti-tumor mechanisms of bacteria are summarized,which refer to intrinsic tumoricidal activities,immune activation,bacteria metabolism,and their capability to regulate gut microbiota homeostasis.Moreover,bacteria could act as carriers to deliver various types of therapeutics to achieve combination therapy with improved efficacy.In addition,several challenges for anti-tumor applications of bacteria are discussed regarding the delivery,efficacy and safety issues,and potential solutions are also provided.Finally,the possible improvements and perspectives are discussed in the end,which provide a guideline for the design of advanced bacteria-based tumor therapeutics in the future.

Therapeutic cancer vaccines have undergone a resurgence in the past decade.Because of the high level of immune cell accumulation and abundant capillary lymphatic system in the dermis,percutaneous vaccination is considered to be an ideal treatment route.For convenient administration,the recent development of microneedles(MNs)provides a safe,painless,and low-cost transdermal delivery strategy,which could bypass the first-pass metabolism of vaccines for enhanced stability and bioavailability.However,the therapeutic effect of MNs-based cancer vaccines is not optimal,which is limited by the complex set of host,tumor,and environmental factors,as well as the limited vaccine loading capacity.Therefore,further improvements are still required to push their clinical translation.In this critical review,we deliberate on how to improve the therapeutic effect of MNs-based vaccines for cancer immunotherapy,summarize the recent advances in MNs-based cancer vaccination,and provide an overview of various design strategies and mechanisms for active or passive targeting delivery,aiming to develop safer,more effective,and more stable MNs-based cancer vaccines.Finally,we briefly describe the potential of vaccine platforms in combination with other therapies,suggest the need to design vaccines according to specific circumstances,and discuss the biosafety of repeated administration for enhancing clinical efficacy.

Utilization of recycled solid waste as ecological reclamation materials based on plant growth experiments and soil quality analysis

During the highway construction,a large number of spoil areas will be generated while a large amount of waste slag and soil could not be rationally utilized.Besides,the vegetation recovery is slow in the spoil areas due to the soil depletion.Aiming at recycling the solid waste,the sieved engineering waste slag with local red clay and corn straw biochar was supplied to solve the problem of insufficient nutrients in engineering waste slag and soil.In addition,planting experiments of alfalfa(Medicago sativa L.)and Amorpha fruticosa L.combined with physical and chemical experiments were carried out to prove the feasibility of the novel improved substrate for the reclamation of spoil areas.The results show that the substrate's improvement effect is mainly affected by the soil to slag ratio and the biochar content.The improvement effect of soil matrix in highway spoil area decreases with the increase of the waste slag content,especially when the soil-slag ratio is less than 3,and the promotion of plants is limited.On the contrary,the improvement effect is proportional to the biochar content(3%-8%).But it is noted that the Cu and Pb in the soil will exceed the clean limit corresponding to the Nemero soil pollution index level when the biochar content is 8%.Therefore,it is recommended that the soil-slag ratio should be≥3,and the biochar content should reach 3%-5%.This research provides experimental basis and technical support for utilizing solid waste resources in the reclamation of highway spoil areas.

Ultrasonic vibration-assisted drilling(UVAD)has recently been successfully applied in the drilling of carbon fiber reinforced polymer/plastic(CFRP)due to its high reliability.Multiple defects have been observed in the CFRP drilling process which negatively affects the quality of the hole.The carbon fiber/bismaleimide(BMI)composites is an advanced kind of CFRPs with greater strength and heat resistance,having been rapidly applied in lightweight and high temperature resistant structures in the aerospace field.To suppress the defect during the drilling of carbon fiber/BMI composites,it is necessary to comprehensively understand the defect formation and suppression mechanism at different positions.In this study,the defects formation in both conventional drilling(CD)and UVAD were observed and analyzed.The variation trend in the defect factor and thrust force with the spindle speed and feed rate were acquired.The results revealed that the UVAD could significantly enhance the hole’s quality with no delamination and burr.Meanwhile,the defect suppression mechanism and thrust force in UVAD were analyzed and verified,where the method of rod chip removal affected the exit defect formation.In summary,UVAD can be considered a promising and competitive technique for drilling carbon fiber/BMI composites.

One of the most recent developments in the field of graph theory is the analysis of networks such as Butterfly networks,Benes networks,Interconnection networks,and David-derived networks using graph theoretic parameters.The topological indices(TIs)have been widely used as graph invariants among various graph theoretic tools.Quantitative structure activity relationships(QSAR)and quantitative structure property relationships(QSPR)need the use of TIs.Different structure-based parameters,such as the degree and distance of vertices in graphs,contribute to the determination of the values of TIs.Among other recently introduced novelties,the classes of ev-degree and ve-degree dependent TIs have been extensively explored for various graph families.The current research focuses on the development of formulae for different ev-degree and ve-degree dependent TIs for s−dimensional Benes network and certain networks derived from it.In the end,a comparison between the values of the TIs for these networks has been presented through graphical tools.

Network Embedding Algorithm for Vulnerability Assessment of Power Transmission Lines Using Integrated Structure and Attribute Information

In power systems,failures of vulnerable lines can trigger large-scale cascading failures,and vulnerability assessment is dedicated to locating these lines and reducing the risks of such failures.Based on a structure and attribute network embedding(SANE)algorithm,a novel quantitative vulnerability analysis method is proposed to identify vulnerable lines in this research.First,a two-layered random walk network with topological and electrical properties of transmission lines is established.Subsequently,based on the weighted degree of nodes in the two-layered network,the inter-layer and intra-layer walking transition probabilities are developed to obtain walk sequences.Then,a Word2Vec algorithm is applied to obtain lowdimension vectors representing transmission lines,according to obtained walk sequences for calculating the vulnerability index of transmissions lines.Finally,the proposed method is compared with three widely used methods in two test systems.Results show the network embedding based method is superior to those comparison methods and can provide guidance for identifying vulnerable lines.

A wall-thickness compensation strategy for wax pattern of hollow turbine blade

As an important index affecting the aerodynamic performance and the structural strength of hollow turbine blades, the wall-thickness precision of the blade is mainly inherited from the positional relationship between the corresponding wax pattern and the internal ceramic core.However, due to locating errors, the actual position of ceramic core is always deviated from the ideal position, which makes it difficult to guarantee the wall-thickness precision of the wax pattern.To solve this problem, a wall-thickness compensation strategy is proposed in this paper. Firstly,based on the industrial computed tomography(ICT) technique and curve matching algorithms, a model reconstruction method is developed, with which the 3D model of a trial wax pattern can be easily constructed. After that, focusing on eliminating the wall-thickness errors of the trial wax pattern, an optimization method for the pose of the ceramic core in the wax pattern is proposed. Then, by mapping the optimal pose of the ceramic core to length adjustments of the locating rods, the wall-thickness errors of the wax pattern can be greatly reduced. A case study is also given to illustrate the effectiveness of the proposed compensation strategy.

Oncogenes are critical factors in tumorigenesis of diverse cancer types and play essential roles in tumor immune escape.Mutations in Kirsten rat sarcoma viral oncogene homolog(KRAS)and epidermal growth factor receptor(EGFR)are among the most frequent gain-of-function alterations[1].After many years of in-depth research,inhibitors targeting EGFR or KRAS mutations have been successfully developed,however,their clinical benefit is relatively limited,and they will inevitably encounter the challenge of drug resistance.The emergence of resistance is attributed to secondary mutations in driver genes and other complicated factors.

Influence of TiB2 particles on machinability and machining parameter optimization of TiB2/Al MMCs

In situ formed TiB2 particle reinforced aluminum matrix composites （TiB2/Al MMCs） have some extraordinary properties which make them be a promising material for high performance aero-engine blade. Due to the influence of TiB2 particles, the machinability is still a problem which restricts the application of TiB2/Al MMCs. In order to meet the industrial requirements, the influence of TiB2 particles on the machinability of TiB2/Al MMCs was investigated experimentally. Moreover, the optimal machining conditions for this kind of MMCs were investigated in this study. The major conclusions are：（1） the machining force of TiB2/Al MMCs is bigger than that of non- reinforced alloy and mainly controlled by feed rate; （2） the residual stress of TiB2/AI MMCs is compressive while that of non-reinforced alloy is nearly neutral; （3） the surface roughness of TiB2/Al MMCs is smaller than that of non-reinforced alloy under the same cutting speed, but reverse result was observed when the feed rate increased; （4） a multi-objective optimization model for surface roughness and material removal rate （MRR） was established, and a set of optimal parameter combinations of the machining was obtained. The results show a great difference from SiC particle reinforced MMCs and provide a useful guide for a better control of machining process of this material.
