Brain-targeting,acid-responsive antioxidant nanoparticles for stroke treatment and drug delivery

Stroke is the leading cause of death and ***,there is no effective pharmacological treatment for this disease,which can be partially attributed to the inability to efficiently deliver therapeutics to the *** we report the development of natural compound-derived nanoparticles(NPs),which function both as a potent therapeutic agent for stroke treatment and as an efficient carrier for drug delivery to the ischemic ***,we screened a collection of natural nanomaterials and identified betulinic acid(BA)as one of the most potent antioxidants for stroke ***,we engineered BA NPs for preferential drug release in acidic ischemic tissue through chemically converting BA to betulinic amine(BAM)and for targeted drug delivery through surface conjugation of AMD3100,a cXcR4 *** resulting AMD3100-conjugated BAM NPs,or A-BAM NPs,were then assessed as a therapeutic agent for stroke treatment and as a carrier for delivery of NA1,a neuroprotective *** show that intravenous administration of A-BAM NPs effectively improved recovery from stroke and its efficacy was further enhanced when NA1 was *** to their multifunctionality and significant efficacy,we anticipate that A-BAM NPs have the potential to be translated both as a therapeutic agent and as a drug carrier to improve the treatment of stroke.

Background:The period following pregnancy is a critical time window when future habits with respect to physical activity(PA) and sedentary behavior(SB) are established;therefore,it warrants *** purpose of this scoping review was to summarize public health-oriented country-specific postpartum PA and SB guidelines ***:To identity guidelines published since 2010,we performed a(a) systematic search of 4 databases(cINAHL,Global Health,PubMed,and SPORTDiscus),(b) structured repeatable web-based search separately for 194 countries,and(c) separate web-based *** the most recent guideline was included for each ***:We identified 22 countries with public health-oriented postpartum guidelines for PA and 11 countries with SB *** continents with guidelines included Europe(n=12),Asia(n=5),Oceania(n=2),Africa(n=1),North America(n=1),and South America(n=1).The most common benefits recorded for PA included weight control/management(n=10),reducing the risk of postpartum depression or depressive symptoms(n=9),and improving mood/well-being(n=8).Postpartum guidelines specified exercises to engage in,including pelvic floor exercises(n=17);muscle strengthening,weight training,or resistance exercises(n=13);aerobics/general aerobic activity(n=13);walking(n=11);cycling(n=9);and swimming(n=9).Eleven guidelines remarked on the interaction between PA and breastfeeding;several guidelines stated that PA did not impact breast milk quantity(n=7),breast milk quality(n=6),or infant growth(n=3).For SB,suggestions included limiting long-term sitting and interrupting sitting with ***:country-specific postpartum guidelines for PA and SB can help promote healthy behaviors using a culturally appropriate context while providing specific guidance to public health practitioners.

Attenuated retinoic acid signaling is among the early responses in mouse uterus approaching embryo attachment

The uterus is transiently receptive for embryo implantation. It remains to be understood why the uterus does not reject a semi-allogeneic embryo (to the biological mother) or an allogeneic embryo (to a surrogate) for implantation. To gain insights, we examined uterine early response genes approaching embryo attachment on day 3 post coitum (D3) at 22 hours when blue dye reaction, an indication of embryo attachment, had not manifested in mice. c57BL/6 pseudo-pregnant (control) and pregnant mouse uteri were collected on D3 at 22 hours for microarray analysis. The self-assembling-manifold (SAM) algorithm identified 21,858 unique probesets. Principal component analysis indicated a clear separation between the pseudo-pregnant and pregnant groups. There were 106 upregulated and five downregulated protein-coding genes in the pregnant uterus with fold change (fc) >1.5 andq value cant GO biological process (GOBP) terms (P ciated with immune responses, with a dominant natural killer (NK) cell activation signature. Among the top eight upregulated protein-coding genes,cyp26a1 inactivates retinoic acid (RA) whileLrat promotes vitamin A storage, both of which are expected to attenuate RA bioavailability;Atp6v0d2 andGjb2 play roles in ion transport and transmembrane transport;Gzmb,Gzmc, andIl2rb are involved in immune responses; andTdo2 is important for kynurenine pathway. Most of these genes or their related pathways have functions in immune regulations. RA signaling has been implicated in immune tolerance and immune homeostasis, and uterine NK cells have been implicated in immunotolerance at the maternal-fetal interface in the placenta. The mechanisms of immune responses approaching embryo attachment remain to be elucidated. The coordinated effects of the early response genes may hold the keys to the question of why the uterus does not reject an implanting embryo

Highly enhanced UV absorption and light emission of monolayer WS_(2)through hybridization with Ti_(2)N MXene quantum dots and g-c_(3)N_(4)quantum dots

Two-dimensional(2D)transition metal dichalcogenides(TMD)are atomically thin semiconductors with promising optoelectronic applications across the visible ***,their intrinsically weak light absorption and the low photoluminescence quantum yield(PLQY)restrict their performance and potential use,especially in ultraviolet(UV)wavelength light *** dots(QD)derived from 2D materials(2D/QD)provide efficient light absorption and emission of which energy can be tuned for desirable light *** this study,we greatly enhanced the photon absorption and PLQY of monolayer(1L)tungsten disulfide(WS_(2))in the UV range via hybridization with 2D/QD,particularly titanium nitride MXene QD(Ti_(2)N MQD)and graphitic carbon nitride QD(GcNQD).With the hybridization of MQD or GcNQD,1LWS_(2)showed a maximum PL enhancement by 15 times with 300 nm wavelength excitation,while no noticeable enhancement was observed when the excitation photon energy was less than the bandgap of the QD,indicating that UV absorption by the QD played a crucial role in enhancing the light emission of 1L-WS_(2)in our 0D/2D hybrid *** findings present a convenient method for enhancing the photo-response of 1L-WS_(2)to UV light and offer exciting possibilities for harvesting UV energy using 1L-TMD.

The SARS-coV-2 Omicron variants are notorious for their transmissibility,but little is known about their subgenomic RNA(sgRNA)*** study applied RNA-seq to delineate the quantitative and qualitative profiles of canonical sgRNA of 118 respiratory samples collected from patients infected with Omicron BA.2 and compared with 338 patients infected with non-variant of concern(non-VOc)-D614G.A unique characteristic profile depicted by the relative abundance of 9 canonical sgRNAs was reproduced by both BA.2 and non-VOcD614G regardless of host gender,age and presence of ***,such profile was lost in samples with low viral load,suggesting a potential application of sgRNA pattern to indicate viral activity of individual patient at a specific time point.A characteristic qualitative profile of canonical sgRNAs was also reproduced by both BA.2 and *** presence of a full set of canonical sgRNAs carried a coherent correlation with crude viral load(AUc¼0.91,95%cI 0.88–0.94),and sgRNA ORF7b was identified to be the best surrogate marker allowing feasible routine application in characterizing the infection status of individual *** potentials in using sgRNA as a target for vaccine and antiviral development are worth pursuing.

Objective: To report the clinical maternal and fetal outcomes of pregnant women with coronavirus disease 2019 (cOVID-19), along with any associated pregnancy complications, in Hong Kong, china, and to assess the impact of cOVID-19 vaccination on these ***: This prospective registry-based observational study included pregnant women who were recruited through convenient sampling and had a laboratory-confirmed severe acute respiratory syndrome coronavirus 2 infection with a cycle threshold (ct) value result available on admission to eight local hospitals in Hong Kong, china. Data on clinical symptoms, laboratory results, medical treatments, delivery timing and mode, and pregnancy complications were extracted from the Hospital Authority’s electronic medical record system. Maternal, fetal, and pregnancy outcomes were compared between unvaccinated pregnant women with cOVID-19 and those who had received at least one dose of cOVID-19 vaccine before diagnosis. Nonparametric continuous variables and categorical variables were analyzed using the Mann-WhitneyU test and the Pearson’s chi-squared test respectively. AP value less than 0.05 was considered statistically ***: A total of 164 pregnant women were included, of whom 78 (47.56%) were nulliparous. cOVID-19 was diagnosed before 28 weeks’ gestation in 30 (18.29%), while 134 (81.71 %) were diagnosed at or after 28 weeks’ gestation. Sixty-two (37.80%) women received at least one dose of cOVID-19 vaccine. There were no significant differences between vaccinated and unvaccinated groups in the time interval between cOVID-19 diagnosis and delivery, the ct value, and the gestational age at infection onset or delivery (P > 0.05). The majority of women were symptomatic at diagnosis regardless of vaccination status (55 (88.71%) in vaccinated groupvs. 78 (76.47%) in unvaccinated group (P = 0.052). An efficient semi-blind source extraction algorithm and its applications to biomedical signal extraction

In many applications, such as biomedical engineering, it is often required to extract a desired signal instead of all source signals. This can be achieved by blind source extraction （BSE） or semi-blind source extraction, which is a powerful technique emerging from the neural network field. In this paper, we propose an efficient semi-blind source extraction algorithm to extract a desired source signal as its first output signal by using a priori information about its kurtosis range. The algorithm is robust to outliers and spiky noise because of adopting a classical robust contrast function. And it is also robust to the estimation errors of the kurtosis range of the desired signal providing the estimation errors are not large. The algorithm has good extraction performance, even in some poor situations when the kurtosis values of some source signals are very close to each other. Its convergence stability and robustness are theoretically analyzed. Simulations and experiments on artificial generated data and real-world data have confirmed these results.
