Integrative transcriptome analysis identifies MYBL2 as a poor prognosis marker for osteosarcoma and a pan-cancer marker of immune infiltration

MYBL2(MYB proto-oncogene like 2)is an emerging prognostic marker for malignant tumors,and its potential role in osteosarcoma and its relationship with immune infiltration in pan-cancer is yet to be elucidated.We constructed a transcription factor activity profile of os-teosarcoma using the single-cell regulatory network inference algorithm based on single-cell RNA sequencing data obtained from the Gene Expression Omnibus.Subsequently,we calcu-lated the extent of MYBL2 activation in malignant proliferative osteoblasts.We also explored the association between MYBL2 and chemotherapy resistance in osteosarcoma.Furthermore,we systematically correlated MYBL2 with immunological signatures in the tumor microenviron-ment in pan-cancer,including immune cell infiltration,immune checkpoints,and tumor immu-notherapy prognosis.Finally,we developed and validated a risk score(MRGS),derived an osteosarcoma risk score nomogram based on MRGS,and tested its ability to predict prognosis.MYBL2 and gene enrichment analyses in osteosarcoma and pan-cancer revealed that MYBL2 was positively correlated with cell proliferation and tumor immune pathways.MYBL2 expres-sion positively correlated with SLC19A1 in pan-cancer and osteosarcoma cell lines.Pan-cancer immune infiltration analysis revealed that MYBL2 was correlated with myeloid-derived sup-pressor cells,Th2 cell infiltration,CD276,RELT gene expression,and tumor mutation burden.

A bounding surface visco-plasticity model considering generalized spacing ratio of soils

The non-unique critical state of soils with time-dependent behaviors is a significant issue in geotechnical engineering problems.However,previous bounding surface plasticity models cannot predict accurately the non-unique critical state of soils,because the distance between the compression line and critical state line charged by strain-rate effect is basically neglected.To fill this gap,a generalized spacing ratio of soils is defined in the elasto-viscoplastic framework,and a bounding surface visco-plasticity model is formulated and verified,which can consider the generalized spacing ratio.Specifically,the generalized spacing ratio of soils reflects the distance between the compression line and the critical state line of soils with time-dependent behaviors.Then,the generalized spacing ratio is introduced into an improved anisotropic bounding surface.A new expression of the visco-plastic multiplier is derived by solving the consistency equation of an anisotropic bounding surface.In the expression,a strain rate index is proposed to account for the strain-rate effect on visco-plastic strain increment,and a visco-plastic hardening modulus is derived to predict the visco-plastic response of soils in overconsolidation conditions.The model is then verified through constant strain rate tests and creep tests.Notably,it can capture the non-unique critical states of soils with time-dependent behaviors due to the generalized spacing ratio and the creep rupture of soils due to the visco-plastic multiplier that considers the stress ratio and visco-plastic strain rate.

Pd-catalyzed asymmetric carbonyl alkynylation:Synthesis of axial chiral ynones

Ynones are important skeletons in bioactive molecules and valuable building blocks for organic synthesis,thus great efforts have been devoted to their preparation.While,introducing prochiral substrates to construct ynones bearing a chiral framework is unrealized to date.Herein,we reported the first example of Pd/SOP-catalyzed asymmetric carbonylative alkynylation via a non-classical carbonylative Sonogashiratype approach(acyl-Pd(Ⅱ)species generated from nucleophiles).By using cyclic diaryliodonium salts as prochiral substrates,various axial chiral ynones with good functional group tolerance(39 examples),satisfied yields(71%-96%)and excellent enantioselectivities(generally 94%-99%ee)were produced.Synthesis of bioactive compounds,scale-up experiment and useful transformations were also conducted to demonstrate the utility of this process.

The co-transport of Cd(Ⅱ) and nZnO in saturated soil packed column:effects of ionic strength and pH

The rapid development and widespread use of ZnO nanoparticles(nZnO) in various industries have raised concerns about their potential environmental impact.Therefore,understanding the fate and role of nZnO in the natural environment is crucial for mitigating their hazardous effects on the environment and human safety.The purpose of the present study was to provide scientific support for understanding and eliminating the joint risk of nanoparticle and heavy metal pollution in the soil environment by revealing the co-transport characteristics of Cd(Ⅱ) and ZnO nanoparticles(nZnO) in soil under different ionic strength(IS) and pH.The impacts of different IS and pH on the co-transport of Cd(Ⅱ) and nZnO in a20 cm long with an inner diameter of 2.5 cm acrylic column packed with 10 cm high soil samples were investigated in the present study.In the above system,a500 μg L^(-1) Cd(Ⅱ) loaded nZnO suspension pulse with varying IS or pH was introduced into the soil column for leaching over 5 PVs,followed up by 5 PVs background solutions without nZnO.The IS was 1,10,or 50 mM NaCl,with pH6,or the pH was 6,7 or 8 with 1 mM NaCl.Meanwhile,Sedimentation experiments for nZnO,adsorption of Cd(Ⅱ) on soil,and nZnO,DLVO theory calculation for the same background condition were conducted.The presence of nZnO significantly increased the mobility of Cd(Ⅱ) as a result of its strong adsorption capacity for nZnO-associated Cd(Ⅱ).However,with the increase of IS,the co-transport of nZnO and Cd(Ⅱ) was decreased and the retention of nZnO in the soil column due to more nZnO attended to aggregate and sediment during the transport and the decrease in the adsorption capacity of nZnO for Cd(Ⅱ) by competition of Na^(+).When pH was 6,7,and 8,the co-transport of nZnO and Cd(Ⅱ) increased with higher pH due to the lower electrostatic attraction between nZnO and soil under higher pH.Meanwhile,the DLVO theory was fitted to describe the above co-transport process of nZnO and Cd(Ⅱ).More attention should be paid to the presence of nZnO on the migration of Cd(Ⅱ) i

Learning to Branch in Combinatorial Optimization With Graph Pointer Networks

Traditional expert-designed branching rules in branch-and-bound(B&B) are static, often failing to adapt to diverse and evolving problem instances. Crafting these rules is labor-intensive, and may not scale well with complex problems.Given the frequent need to solve varied combinatorial optimization problems, leveraging statistical learning to auto-tune B&B algorithms for specific problem classes becomes attractive. This paper proposes a graph pointer network model to learn the branch rules. Graph features, global features and historical features are designated to represent the solver state. The graph neural network processes graph features, while the pointer mechanism assimilates the global and historical features to finally determine the variable on which to branch. The model is trained to imitate the expert strong branching rule by a tailored top-k Kullback-Leibler divergence loss function. Experiments on a series of benchmark problems demonstrate that the proposed approach significantly outperforms the widely used expert-designed branching rules. It also outperforms state-of-the-art machine-learning-based branch-and-bound methods in terms of solving speed and search tree size on all the test instances. In addition, the model can generalize to unseen instances and scale to larger instances.

Characterization, preparation, and reuse of metallic powders for laser powder bed fusion: a review

Laser powder bed fusion(L-PBF) has attracted significant attention in both the industry and academic fields since its inception, providing unprecedented advantages to fabricate complex-shaped metallic components. The printing quality and performance of L-PBF alloys are infuenced by numerous variables consisting of feedstock powders, manufacturing process,and post-treatment. As the starting materials, metallic powders play a critical role in infuencing the fabrication cost, printing consistency, and properties. Given their deterministic roles, the present review aims to retrospect the recent progress on metallic powders for L-PBF including characterization, preparation, and reuse. The powder characterization mainly serves for printing consistency while powder preparation and reuse are introduced to reduce the fabrication costs.Various powder characterization and preparation methods are presented in the beginning by analyzing the measurement principles, advantages, and limitations. Subsequently, the effect of powder reuse on the powder characteristics and mechanical performance of L-PBF parts is analyzed, focusing on steels, nickel-based superalloys, titanium and titanium alloys, and aluminum alloys. The evolution trends of powders and L-PBF parts vary depending on specific alloy systems, which makes the proposal of a unified reuse protocol infeasible. Finally,perspectives are presented to cater to the increased applications of L-PBF technologies for future investigations. The present state-of-the-art work can pave the way for the broad industrial applications of L-PBF by enhancing printing consistency and reducing the total costs from the perspective of powders.

Due to their excellent fluorescence properties and biological function,cyanine dyes have been widely applied in biological imaging.Heptamethine cyanine(Cy7)dyes,as a type of classic near-infrared(NIR)fluorescent dyes,are considered as one of the effective fluorescent tools in the living organisms due to their good biocompatibility and very low background interference.Bioorthogonal reactions performed in living cells and tissues have developed by leaps and bounds in recent years.The NIR fluorescent labeling technique involving cyanine has attracted widespread attention.This review summarizes their recent application in the field of bioorthogonal imaging,mainly concluding Cy7-type dyes,labeling strategy,bioimaging application,etc.We expect this work can provide some helps for the studies of NIR bioorthogonal reaction in vivo.

Benefiting from the great advances of the femtosecond laser two-photon polymerization(TPP)technology,customized microcantilever probes can be accurately 3-dimensional(3D)manufactured at the nanoscale size and thus have exhibited considerable potentials in the fields of microforce,micro-vibration,and microforce sensors.In this work,a controllable microstructured cantilever probe on an optical fiber tip for microforce detection is demonstrated both theoretically and experimentally.The static performances of the probe are firstly investigated based on the finite element method(FEM),which provides the basis for the structural design.The proposed cantilever probe is then 3D printed by means of the TPP technology.The experimental results show that the elastic constant k of the proposed cantilever probe can be actively tuned from 2.46N/m to 62.35N/m.The force sensitivity is 2.5nm/μN,the Q-factor is 368.93,and the detection limit is 57.43nN.Moreover,the mechanical properties of the cantilever probe can be flexibly adjusted by the geometric configuration of the cantilever.Thus,it has an enormous potential for matching the mechanical properties of biological samples in the direct contact mode.

CNN-LSTM based incremental attention mechanism enabled phase-space reconstruction for chaotic time series prediction

To improve the prediction accuracy of chaotic time series and reconstruct a more reasonable phase space structure of the prediction network,we propose a convolutional neural network-long short-term memory(CNN-LSTM)prediction model based on the incremental attention mechanism.Firstly,a traversal search is conducted through the traversal layer for finite parameters in the phase space.Then,an incremental attention layer is utilized for parameter judgment based on the dimension weight criteria(DWC).The phase space parameters that best meet DWC are selected and fed into the input layer.Finally,the constructed CNN-LSTM network extracts spatio-temporal features and provides the final prediction results.The model is verified using Logistic,Lorenz,and sunspot chaotic time series,and the performance is compared from the two dimensions of prediction accuracy and network phase space structure.Additionally,the CNN-LSTM network based on incremental attention is compared with long short-term memory(LSTM),convolutional neural network(CNN),recurrent neural network(RNN),and support vector regression(SVR)for prediction accuracy.The experiment results indicate that the proposed composite network model possesses enhanced capability in extracting temporal features and achieves higher prediction accuracy.Also,the algorithm to estimate the phase space parameter is compared with the traditional CAO,false nearest neighbor,and C-C,three typical methods for determining the chaotic phase space parameters.The experiments reveal that the phase space parameter estimation algorithm based on the incremental attention mechanism is superior in prediction accuracy compared with the traditional phase space reconstruction method in five networks,including CNN-LSTM,LSTM,CNN,RNN,and SVR.
