为了充分发挥本课题组前期设计的基于硝基化T表位的靶向PD-L1的肿瘤疫苗(PD-L1-NitraTh)的抑瘤活性,选择了作用机制不同的几种佐剂进行比较,以期筛选出最适用于此类疫苗的佐剂。研究结果显示,Poly(I:C)、CPG1018、肿节风多糖SGP2及GM-CSF等佐剂均可以提高PD-L1-NitraTh疫苗的免疫原性,其中,Poly(I:C)组诱导产生的抗体滴度最高。对T细胞分化相关转录因子的qPCR检测结果显示,Poly(I:C)减少了GATA3和FoxP3的表达,提示对CD4^(+)T细胞分化有较强的影响。同时,相比其他佐剂,Poly(I:C)可以辅助PD-L1-NitraTh增加肿瘤内T淋巴细胞以及CD11b^(+)细胞浸润,提示Poly(I:C)佐剂可能适用于以硝基化T表位为基础的肿瘤疫苗。

Hepatocellular carcinoma(HCC)is the fourth leading cause of cancer-associated death worldwide.Angiogenesis,the process of formation of new blood vessels,is required for cancer cells to obtain nutrients and oxygen.HCC is a typical hypervascular solid tumor with an aberrant vascular network and angiogenesis that contribute to its growth,progression,invasion,and metastasis.Current anti-angiogenic therapies target mainly tyrosine kinases,vascular endothelial growth factor receptor(VEGFR),and plateletderived growth factor receptor(PDGFR),and are considered effective strategies for HCC,particularly advanced HCC.However,because the survival benefits conferred by these anti-angiogenic therapies are modest,new anti-angiogenic targets must be identified.Several recent studies have determined the underlying molecular mechanisms,including pro-angiogenic factors secreted by HCC cells,the tumor microenvironment,and cancer stem cells.In this review,we summarize the roles of pro-angiogenic factors;the involvement of endothelial cells,hepatic stellate cells,tumor-associated macrophages,and tumor-associated neutrophils present in the tumor microenvironment;and the regulatory influence of cancer stem cells on angiogenesis in HCC.Furthermore,we discuss some of the clinically approved anti-angiogenic therapies and potential novel therapeutic targets for angiogenesis in HCC.A better understanding of the mechanisms underlying angiogenesis may lead to the development of more optimized anti-angiogenic treatment modalities for HCC.

Photothermal(PTT) and photodynamic(PDT) combined therapy has been hindered to clinical translation, due to the lack of available biomaterials, difficult designs of functions,and complex chemical synthetic or preparation procedures. To actualize a high-efficiency combination therapy for cancer via a feasible approach, three readily available materials are simply associated together in one-pot, namely the single-walled carbon nanohorns(SWCNH), zinc phthalocyanine(ZnPc), and surfactant TPGS. The established nanodispersion is recorded as PCT. The association of SWCNH/ZnPc/TPGS was confirmed by energy dispersive spectrum, Raman spectrum and thermogravimetric analysis. Under lighting, PCT induced a temperature rising up to about 60 ℃ due to the presence of SWCNH, production a 7-folds of singlet oxygen level elevation because of ZnPc, which destroyed almost all4T1 tumor cells in vitro. The photothermal effect of PCT depended on both laser intensity and nanodispersion concentration in a linear and nonlinear manner, respectively. After a single peritumoral injection in mice and laser treatment, PCT exhibited the highest tumor temperature rise(to 65 ℃) among all test groups, completely destroyed primary tumor without obvious toxicity, and inhibited distant site tumor. Generally, this study demonstrated the high potential of PCT nanodispersion in tumor combined therapy.

基于IAHP-IRMO法的既有路基挡土墙评估指标权重确定

An acidic polysaccharide was isolated from the residue of Sarcandra glabra(Thunb.) Nakai(SERP1) by water extraction and purified by decoloration and ion exchange chromatography.The structure of SERPl was determined by HPLC,HPSEC-MALLS,FT-IR,NMR,and AFM analysis.The results suggested that SERPl was a homogeneous heteropolysaccharide with absolute molecular weight of 4.208×10 Da in aqueous phase.1,4-linked α-D-galacturonic acid,methylesterified 1,4-linked α-D-galacturonic acid,1,4-linked α-D-glucuronic acid,1,5-linked α-Larabinose,1,3-linked β-D-galactose,1,4-linked α-D-glucose,1,4,6-linked β-D-glucose,1,6-linkedβ-D-glucose,and 1,2-linked rhamnose were exist in SERP1.In vitro α-glucosidase inhibition assay indicated that SERP1 had a low IC value of 49.01 μg/mL,which exhibited stronger a-glucosidase enzyme inhibitory activity than that of acarbose at the same concentration.The treatment of SERP1 to type 2 diabetes mellitus mice alleviated the hyperglycemia,increased glucose utilization of peripheral tissues of liver and inhibited the liver injury.

在既有路基挡土墙安全评估中,评估指标权重的确定非常重要。目前,挡土墙评估指标权重的确定大多采用确定型的层次分析法,忽略了评估指标不确定性和模糊性的影响。为了准确反映评估指标权重的不确定性和模糊性,通过分析评估指标权重计算方法,采用区间层次分析法(IAHP)构造区间数判断矩阵来确定既有路基挡土墙评估指标的权重。针对区间数判断矩阵中求解权重方法的不足,引入相离度的概念,建立单目标优化模型。将模型中的目标函数作为适应度函数,运用改进径向移动算法(IRMO)进行全局优化搜索,并对IRMO算法中的惯性权重递减模型wk进行改进。采用的曲线递减模型能提高全局优化搜索能力,从而获得评估指标的最优权重。对2个不同算例与其他方法进行对比分析,并结合工程实例进行验证计算。结果表明:IRMO算法运用于区间数判断矩阵权重优化搜索具有稳定性和高效性,能够有效减小不确定性带来的影响,搜索速度快、收敛效率高、搜索结果更为稳定、客观、合理。IAHP−IRMO法应用于既有路基挡土墙评估指标权重计算方面具有可行性、有效性与准确性。工程实例得出的安全评估结果与现场实际情况较为符合,该方法能够广泛应用于铁路、公路等既有路基挡土墙的安全评估指标权重确定,为挡土墙安全评估提供依据。

肠促胰岛素分泌肽胰高血糖素样肽-1(GLP-1)和葡萄糖依赖性促胰岛素释放多肽(GIP)能通过血糖依赖机制促进胰岛素分泌,其特异性结合受体GLP-1R和GIPR是治疗2型糖尿病的良好靶点。以本实验室前期设计的口服降糖多肽OHP2为基础,设计了可口服的新型降糖多肽——ODA。ODA较OHP2的亲脂性提高,在Caco-2细胞中的胞吞能力及跨细胞转运能力更强。ODA保留了OHP2对GLP-1R的激活能力,增强了与GIPR的结合能力。口服低剂量ODA(0.53 mg/kg)即可达到与口服OHP2(1.06 mg/kg)相当的降糖水平。研究结果显示,ODA是治疗2型糖尿病极具潜力的口服药物。

A study was conducted to evaluate the effects of dietary carbohydrates on triglyceride, cholesterol and fatty acid concentrations in abalone, Haliotis discus hannai Ino. Six semi-purified diets with different carbohydrates (dextrin, heat-treated wheat starch, wheat starch, corn starch, tapioca starch and potato starch, respectively), all containing a carbohydrate level of 33.5%, were fed to abalone (initial shell length: 29.98mm±0.09mm; initial weight: 3.42g±0.02g) for 24 weeks in a recirculation system. The results indicate that serum triglyceride concentrations were significantly (P <0.05) higher in the abalone fed with dextrin, heat-treated wheat starch and wheat starch than those fed with corn starch, and serum cholesterol concentrations were significantly (P <0.05) higher in the abalone fed with dextrin, heat-treated wheat starch than those fed with corn starch. Fatty acid C20:4n-6 in the foot muscles were significantly (P<0.05) lower in the abalone fed with dextrin than those fed with wheat starch, corn starch, tapioca starch and potato starch. Fatty acid C20:4n-6 in hepatopancreas was significantly (P<0.05) lower in abalone fed with heat-treated wheat starch than those fed with corn starch, tapioca starch and potato starch. Fatty acid C22:6n-3 in the foot muscles were significantly (P<0.05) lower in the abalone fed with dextrin and heat-treated wheat starch than those fed with wheat starch and potato starch.

A novel method on designing bispectral windows is proposed in this letter. Compound functions based on a function with binary quadric form, starting from the symmetry characteristic of three-order moments are used as the 2-D windowfunctions. Two approaches on how to find the expressions of the compound functions are discussed in detail. One is to approximate the compound function after being Taylor expanded under the Minimum Mean Square Error (MMSE) criteria. Another is to compound the hyperbolic secant function and the binary quadric function directly. According to theoretical analysis, the first type new windows have been proved as slightly better than the conventional ones, the second type new windows are much better than the conventional ones, and the bispectral estimation mean square error approximates to 0.
