Tissue-engineered conduit using bladder acellular matrix and bladder epithelial cells for urinary diversion in rabbits

Background For muscle invasive bladder cancer, radical cystectomy is the most effective treatment now and urinary diversion is often necessary. The use of intestinal tissue for urinary diversion is frequently associated with complications. In this study, we aimed to make a tissue-engineered conduit （TEC） using bladder epithelial cells and bladder acellular matrix （BAM） for urinary diversion in rabbits. Methods Bladder epithelial cells of rabbit were cultivated and expanded in vitro, then seeded on BAM, and cultured for 7 days. Then cell-seeded graft was used to make TEC. In the experimental group, most of bladder of the rabbit was removed while bladder trigone was retained. The proximal end of TEC was anastomosed with bladder trigone and the distal end was anastomosed with the abdominal stoma. In the control group, TEC was made using unseeded BAM. Haematoxylin and eosin staining was conducted, respectively, at 1, 2, 4, and 8 weeks postoperatively. Immunohistochemistry was performed 8 weeks postoperatively. Intravenous urography, retrograde pyelography, and cystoscopy of TEC were made at 12 weeks postoperatively. Results All animals were alive in the experimental group. Haematoxylin and eosin staining showed epithelial coverage in TEC. Immunohistochemistry showed anti-cytokeratin AEI/AE3 antibody and anti-ZO1 antibody positive, confirming there were mature and functional epithelial cells on the lumen of TEC. Retrograde pyelography and intravenous urography showed that TEC developed well and that there was no obstruction. In the control group, four rabbits were dead within 2 weeks and scar formation, atresia, and severe hydronephrosis were found. Conclusions We successfully made TEC using BAM and bladder epithelial cells for urinary diversion in rabbits. The lumen of this new TEC covered mature epithelial cells and could prevent urinary extravasation.

As ectotherms, fish are highly sensitive to temperature fluctuations, which can profoundly impact their reproductive cycles. In this study, we investigated the fertility and histological characteristics of zebrafish(Danio rerio) ovaries exposed to a temperature gradient ranging from the thermopreferendum temperature of the species,27℃, to lower temperatures of 22℃, 20℃, and 13℃ over a period of two weeks. Comparative metabolomic(six biological replicates for each temperature) and transcriptomic(four biological replicates for each temperature) analyses were conducted under the four temperature conditions. Results indicated that lower temperatures inhibited oocyte development and differential metabolites were involved in steroid hormone production,antioxidant function, and lipid and protein *** reproductive hormones, increased proteolysis,and lipid degradation significantly impeded oocyte development and egg maturation. Notably, a significant increase in bile acid content was noted in the ovaries of the cold-treated fish, indicating that bile acids play a critical role in ovarian failure. Overall, these findings provide valuable insights into the mechanisms governing the reproductive response of fish to cold stress.

In the past, the surgeons usually adopted ileal ureter plasty or ureter bladder flap plasty （Boari flap plasty） to restorelong-term ureteral mucosal avulsions and long or entire ureteral segment defects caused mostly by ureteroscope operations. But there are still certain difficulties in the restoration of long segment ureteral defects （〉20 cm） using traditional methods. In order to overcome traditional surgical approaches, we designed a new ureteroplasty operation using spiral pedunculated bladder muscle flap to restore long segment ureteral defects. METHODS Six patients who presented long segment ureteral defects caused in the course of ureteroscopic lithotripsy due to ureteropelvic junction stenosis and stones （length of defects： 21-25 cm, mean length： 22.5 cm）, were given general anesthesia, and made to lay in the horizontal position while indwelling triple lumen catheters. These patients had Gibson incision in the hypogastrium of the injured sides, and we could prolong the surgical incision up to the epigastrium or the flank abdomen moderately if necessary. We exposed the retroperitoneal space, transected the umbilical ligaments, peritoneal adhesions, spermaducts or the round ligaments to dissociate the bladder to the maximum extent possible. We could increase the mobility of the bladder by dissociating the contralateral superior vesical arteries along the anterior trunk of the internal iliac artery. We could also identify the stump of the distal ureter along the path between the ureter and bladder below the iliac vessels, and then we had to ligate or transfix the ureteral stump. We dissociated the ureteropelvic junction carefully, trimmed the stump to the inclined plane in order to facilitate the anastomose between the bladder muscle flap and the trimmed ureteral stump while locating and tracting it by the suture. The li-Mg-B-H composite(2liBH_(4)+MgH_(2))has a high capacity of 11.4 wt%as a hydrogen storage ***,the slow kinetics and poor cycling stability severely restrict its practical *** this work,a layered Nb_(2)C MXene was first synthesized and then introduced to tailor the kinetics and cycling stability of the li-Mg-B-H *** milled 2liH+MgB_(2)composites were initially hydrogenated to obtain the 2liBH_(4)+MgH_(2)*** 2liBH_(4)+MgH_(2)+5wt%Nb_(2)C can release 9.0 wt%H_(2)in 30 min at 400℃,while it is only 2.7 wt%for the undoped 2liBH_(4)+MgH_(2).The dehydrogenation activation energies of MgH_(2)and liBH_(4)are 123 and 154 kJ·mol^(-1)respectively for the 5 wt%Nb_(2)C-doped composite,lower than the undoped composite(164 and 165 kJ·mol^(-1)).The 2liBH_(4)+MgH_(2)+5 wt%Nb_(2)C possesses excellent cycling stability,with the reversible capacity only slightly reduced from 9.4 wt%for the 1st cycle to 9.3 wt%for the 20th ***_(2)C keeps stable in the composite and acts as an efficient catalyst for the li-Mg-B-H *** is believed that both the layered structure and the active Nb element contribu te to the enhanced hydrogen storage performances of the li-Mg-B-H *** work confirms that the Nb_(2)C MXene with layered stru cture has a significant enhancing impact on the li-Mg-B-H hydrogen storage materials,which is different from the bulk NbC.

Developing new polymeric semiconductors with excellent device performance is essential for organic electronics. Herein, we synthesized two new thiazoloisoindigo(Tz II)-based polymers, namely, P(Tz II-d Th-d Th) and P(Tz II-d Th-d Tz), by copolymerizing thiophene-flanked Tz II with bithiophene and bithiazole, respectively. Owing to the more electron-deficient nature of bithiazole than bithiophene, P(Tz II-d Th-d Tz)possesses deeper LUMO/HOMO levels of-3.45/-5.47 e V than P(Tz II-d Th-d Th)(-3.34/-5.32 e V). The organic field-effect transistor(OFET) devices based on P(Tz II-d Th-d Th) exhibited p-type behaviors with an average hole mobility value as high as 1.43 cm^(2)·V^(-1)·s^(-1), while P(Tz II-d Th-d Tz)showed typical ambipolar characteristics with average hole and electron mobilities of 0.38 and 0.56 cm^(2)·V^(-1)·s^(-1). In addition, we compared the performances of both polymers with other Tz II-based polymers reported in our previous work, and showed that the charge carrier polarity can be manipulated by adjusting the number of the thiophene units between the acceptor unit. As the increase of the number of thiophene rings,charge carrier polarity shifts from electron-dominated ambipolar transport to hole-dominated ambipolar transport and then to unipolar hole transport in OFETs, which provides an effective molecular design strategy for further optimization of polymer OFET performance.

In future optical transport networks,lightpath performance analysis is of great practical significance for fully automated *** general,the quality of transmission(QoT)of lightpaths,measured by optical quality factor or optical signal-to-noise ratio,has a complex time-varying process,along with the interactions of the other lightpath state parameters(LSPs),such as transmit power,chromatic dispersion,polarization mode *** studies are mostly focused on lightpath QoT estimation,but ignoring lightpath-level data *** this case,our article proposes a novel lightpath performance analysis method considering recurrence plot(RP)and cross recurrence plot(CRP).Firstly,we give a detailed interpretation on the recurrence patterns of LSPs via a qualitative 2-D RP representation and its quantitative *** can potentially enable the accurate and fast lightpath failure detection with a low computational *** the other hand,CRP is devoted to modeling the relationships between lightpath QoT and LSPs,and the correlation degree is determined by a geometric mean of multiple indexes of cross recurrence quantification *** the view of application,such CRP analysis can provide the effective knowledge sharing to guarantee more credible QoT *** experiments on a real-world optical network dataset have clearly demonstrated the effectiveness of our proposal.
