KHK-A promotes fructose-dependent colorectal cancer liver metastasis by facilitating the phosphorylation and translocation of PKM2

Excessive fructose diet is closely associated with colorectal cancer(CRC)***,fructose’s specific function and precise mechanism in colorectal cancer liver metastasis(CRLM)is rarely ***,this study reported that the fructose absorbed by primary colorectal cancer could accelerate CRLM,and the expression of KHK-A,not KHK-C,in liver metastasis was higher than in paired primary ***,KHK-A facilitated fructose-dependent CRLM in vitro and in vivo by phosphorylating PKM2 at ***2 phosphorylated by KHK-A inhibited its tetramer formation and pyruvic acid kinase activity but promoted the nuclear accumulation of *** and aerobic glycolysis activated by nuclear PKM2 enhance CRC cells’migration ability and anoikis resistance during CRLM ***-46 treatment,targeting the phosphorylation of PKM2,inhibited the pro-metastatic effect of ***,c-myc activated by nuclear PKM2 promotes alternative splicing of KHK-A,forming a positive feedback loop.

Central nervous system(CNS)trauma,including traumatic brain injury and spinal cord injury,has a high rate of disability and mortality,and effective treatment is currently *** studies have revealed that neural inflammation plays a vital role in CNS *** the initial enzyme in neuroinflammation,cytosolic phospholipase A_(2)(cPLA2)can hydrolyze membranous phosphatides at the sn-2 position in a preferential way to release lysophospholipids andω3-polyunsaturated fatty acid dominated by arachidonic acid,thereby inducing secondary *** there is substantial fresh knowledge pertaining to cPLA2,in-depth comprehension of how cPLA2 participates in CNS trauma and the potential methods to amelio rate the clinical res ults after CNS trauma are still *** present review summarizes the latest understanding of how cPLA2 participates in CNS trauma,highlighting novel findings pertaining to how cPLA2 activation initiates the potential mechanisms specifically,neuroinflammation,lysosome membrane functions,and autophagy activity,that damage the CNS after ***,we focused on testing a variety of drugs capable of inhibiting cPLA2 or the upstream pathway,and we explored how those agents might be utilized as treatments to improve the results following CNS *** review aimed to effectively understand the mechanism of cPLA2 activation and its role in the pathophysiological processes of CNS trauma and provide clarification and a new referential framework for future research.

In radiation measurement and digital nuclear spectrum systems,traditional nuclear signal processing circuits in nuclear electronics have been gradually replaced by digital algorithm modules with the application of highperformance programmable hardware logic devices(such as FPGA or DSP).Referring to the digital realization method of inverse RC integral circuit systems,the function of the pole-zero cancellation(PZC)circuit was analyzed,a new modified cascade equivalent model of PZC was established,and the time-domain digital PZC(DPZC)recursive algorithm was derived in detail in this *** parameters kIand k_(D)are included in the new algorithm,where kIshould match the exponential decay time constant of the input signal to realize the pole-zero compensation,while the decay time constant of the output signal can be changed with the adjustable parameter k_(D)(which is larger than the decay time constant of the input signal).Based on the new DPZC algorithm module,two trapezoidal(triangular)shaping filters were designed and *** amplitude–frequency characteristics of the output signal of the proposed trapezoidal shaping algorithm and the convolution trapezoidal shaping algorithm were compared,with fixed peaking *** results show that the trapezoidal shaping algorithm based on DPZC can better suppress high-frequency ***,based on the Na I(Tl)scintillator(u75 mm×75 mm)detector and^(137)Cs source,the effect of the k_(D)value on the energy resolution of the DPZC trapezoidal(triangular)shaping algorithm was *** experimental results show that,with an increase in k_(D),the energy resolution of the system improved and reached the maximum when k_(D)was greater than 10,and the optimal energy resolution of the system was 7.72%.

Some Stemona alkaloids belonging to the tuberostemospironine group possess a spirolactone moiety with anti-configuration （C-9/C-9a）. In this paper, we describe two approaches to this structural unity. By using bromine atom as a traceless directing group, the SmI2-mediated reductive coupling of ketone 6 and fl-bromomethacrylate proceeded with complete anti-diastereoselectivity. In the absence of an a-directing （chelation） group, the one-pot reaction of the ketone derived from alcohol 15 with the organozinc reagent generated from bromomethacrylate af- forded spiro-a-methylene-y-lactone derivative 16 as a single diastereomer. These two highly diastereoselective methods would find application in the synthesis of stemona alkaloids containing anti-configured spiro-lactone/pyr- rolidine moieties. In addition, on the basis of our previous work, the total synthesis of （-）-9-epi-11-demethyl- sessilifoliamide J （11）, and an improved synthesis of （-）-9,11-di-epi-sessilifoliamide J （9） were accomplished.

Heterotopic gastric mucosa(HGM) in the rectum is an extremely rare clinical entity which may be missed or misdiagnosed due to a lack of *** the present study,a 14-year-old girl visited our hospital due to a 5-year history of repeated *** showed a solitary superficial depressed lesion approximately 5 cm in size and a concomitant 1.5 cm deep diverticulum in the *** examination of the endoscopic biopsy showed typical ectopic gastric mucosa in the depressed lesion and inside the *** band imaging further confirmed the histological *** ultrasound indicated that the lesion originated from the mucosal layer,and partially involved the submucosal *** submucosal dissection was performed in this patient due to the large size and shape of the *** bleeding,perforation or other adverse events were *** presence of HGM in the diverticular cavity greatly increased the surgical difficulty.A literature review was also carried out in our study.

Kinetics of hot metal desulfurization were studied using CaO-SiO2-Al2O3-Na2O-TiO2 slag in the range of 1400-1500℃on a laboratory *** results of kinetic experiments indicate that the desulfurization rate increases as the temperature,Al2O3 content,Na2O content,and TiO2 content increase and basicity increases from 1.01 to 1.75,but decreases when basicity increases from 1.75 to *** melting effect of slag is promoted as the temperature,Na2O content,and TiO2 content increase and Al2O3 content increases from 12.13 to 17.17 mass%,but worsened as basicity increases and Al2O3 content increases from 17.17 to 22.27 mass%.A kinetic model of hot metal desulfurization has been developed to calculate the mass transfer coefficient and the mass transfer resistance of sulfur in *** mass transfer coefficient of sulfur increases as the temperature,Al2O3 content,Na2O content,and TiO2 content increase and basicity *** mass transfer coefficients of sulfur were in the range of(5.02-23.78)×10^-7 m s^-*** activation energy was estimated to be 464.06 kJ mo1^-1 at the temperature from 1400 to 1450℃and 176.35 kJ mol-1 at the temperature from 1450 to 1500℃.The sulfur distribution at the slag-metal interface was observed using a mineral liberation *** result shows that the mass transfer of sulfur in slag is the controlling step at high temperature during the desulfurization process.

In order to explore the total-pressure distortion test assessment method for a turbofan engine, a Controlled Variable Double-Baffle Distortion Generator(CVDBDG) with a horizontal symmetry moving form was developed, which can adjust the steady-state and time–variant distortion separately in real time. The inlet total-pressure distortion test was conducted on an afterburner turbofan engine. The distortion parameters of CVDBDG and the instability characteristics of the engine were measured. The experimental data were modeled and analyzed by using back propagation artificial neural networks, and the work envelope of CVDBDG was obtained. Based on the analysis of the data on the engine’s instability, the properties of CVDBDG used for the stability assessment were preliminarily evaluated. The results show that CVDBDG can simulate both steady-state and time–variant distortions simultaneously in a range determined by three *** the condition of symmetric double baffles, a critical depth of insertion exists, beyond which the symmetric baffles will generate an asymmetric flow field. In the case of double baffles, compared to a single baffle, the engine exhibited different instability characteristics. Based on CVDBDG, it is expected that more efficient engine stability and durability assessment methods can be developed.

Atmospheric humidity is a sustainable low-value energy widely existing in natural environment,which is a promising candidate to solve the noncontinuous and low efficiency of low-value energy power *** the mono-substituted Dawson-type polyoxometalates are constructed to be highly dispersed organic ammonium-polyoxoanion clusters and are assembled into thin films power generators with micropores,working in atmospheric *** optimal polyoxometalates generator with the thickness of 7.2μm and the area of 0.36 cm^(2) produces a voltage of 0.68 V and a current density of 19.5μA·cm^(-2) under simulated natural environment,and works continuously and stably under almost all-natural environments(humidity 10%–90%).The highly dispersed polyoxometalate nanoclusters can form microporous in polyoxometalate films to effectively absorb atmospheric humidity and spontaneously form distribution gradient of water,which is the structural basis of power *** continuous power generation may be maintained by the effective adsorption and utilization of H_(2)O,the huge electrostatic field of organic ammonium-polyoxoanion clusters,and the reasonably designed polyoxometalates containing inorganic small ions with high *** is the first humidity generator designed with polyoxometalates,which may provide a new research direction for polyoxometalates in sustainable utilization of low-value energy.

The basic structure of aromatic compounds that are abundant in coal is the carbonaceous precursor derived from carbon ***,it remains to be a huge challenge to prepare carbon microspheres using coal due to the complex construction and composition of ***,a simple and viable way to obtain coal-based microporous carbon microspheres was developed by means of ethanol pyrolysis and a sequential extraction *** as-prepared carbon microsphere featured aspherical micron particles of a uniform size(0.6-1.6㎛),abundant O-functional groups,excellent thermal stability,high SBET(415.5-983.2 m^(2)/g),and plentiful ultra-micropores(63.15-72.72%).The coal-based carbon microsphere exhibited a noteworthy CO_(2)uptake(3.19-4.97 mmol/g at 273 K and 1.0 bar),acceptable CO_(2)/N_(2)selectivity(IAST:23-46)and moderate isosteric heats(20-32 kJ/mol).This synthetic strategy is important for the preparation of ultramicroporous carbon microspheres using coal,and the synthetic carbon microspheres have promising prospects for highly efficient CO_(2)capture.
