AIM:To identify a maculopathy patient caused by new recessive compound heterozygous variants in ***:Comprehensive retinal morphological and functional examinations were evaluated for the patient with RP1L1 *** sequence capture array technique was used to screen potential pathologic *** chain reaction and Sanger sequencing were used to confirm the screening ***:Fundus examination showed round macular lesions appeared in both *** coherence tomography showed that the inner segment/outer segment continuity was disorganized and disruptive in the left eye,but it was uneven and slightly elevated in the right *** autofluorescence showed patchy hyper-autofluorescence in the *** field examination indicates central defects in both ***(ERG)and multifocal ERG showed no obvious *** fluorescein angiography in the macula showed obviously irregular hyper-fluorescence in the right eye and slightly hyper-fluorescence in the left *** found that the proband carried a missense variant(c.1972C>T)and a deletion variant(c.4717_4718del)of RP1L1,which were originated from the parents and formed compound heterozygous *** variants are likely pathogenic according to the ACMG *** imaging,ERG and detailed medical history are important diagnostic tools for differentiating between acquired and inherited retinal ***:A maculopathy case with detailed retinal phenotype and new recessive compound heterozygous variants of RP1L1 is identified in a Chinese family,which expands the understanding of phenotype and genotype in RP1L1 maculopathy.

Silencing transformer and transformer-2 in Zeugodacus cucurbitae causes defective sex determination with inviability of most pseudomales

transformer is a switch gene for sex determination in many insects, which cooperates with transformer-2 that is expressed in both sexes to regulate female differentiation, particularly in dipterans. Zeugodacus cucurbitae(Coquillett) is a very destructive pest worldwide, however, its sex determination pathway remains largely uncharacterized. Here, we show that the female sex ratio is sharply reduced with knockdown of either transformer or transformer-2 by RNA interference in early embryos of Z. cucurbitae. Most of the males grown from the embryos with transient transformer and transformer-2 suppression mated with wild-type females and produced mixed sex progeny, with one exception that produced only female progeny, and all of the few remaining males failed to mate with wild-type females and produced no progeny. The exceptional male and those males with mating failure were XX pseudomales as determined by the detection of Y chromosome-linked Maleness-on-the-Y, indicating that most XX pseudomales are not viable. The phenotypes of transformer and transformer-2 suggest that they play a key role in regulating sex determination and are required for female sexual development of Z. cucurbitae. Our results will be beneficial to the understanding of sex determination in Z. cucurbitae and can facilitate the development of genetic sexing strains for its biological control.

Adhesive Contact of Elastic Solids with Self-Affine Fractal Rough Surfaces

The elastic adhesive contact of self-affine fractal rough surfaces against a rigid flat is simulated using the finite element method. An array of nonlinear springs, of which the force-separation law obeys the Lennard–Jones potential, is introduced to account for the interfacial adhesion. For fractal rough surfaces, the interfacial interaction is generally attractive for large mean gaps, but turns repulsive as the gap continuously shrinks. The interfacial interactions at the turning point corresponding to the spontaneous contact are shown for various surfaces. For relatively smooth surfaces, the probability density distributions of repulsion and attraction are nearly symmetric. However, for rougher surfaces, the simulation results suggest a uniform distribution for attraction but a monotonously decreasing distribution with a long tail for repulsion. The pull-off force rises with increasing ratio of the work of adhesion to the equilibrium distance, whereas decreases for solids with a higher elastic modulus and a larger surface roughness. The current study will be helpful for understanding the adhesion of various types of rough solids.

Gradient sparsification for efficient wireless federated learning with differential privacy

Federated learning(FL) enables distributed clients to collaboratively train a machine learning model without sharing raw data with each other. However, it suffers from the leakage of private information from uploading models. In addition, as the model size grows, the training latency increases due to the limited transmission bandwidth and model performance degradation while using differential privacy(DP)protection. In this paper, we propose a gradient sparsification empowered FL framework with DP over wireless channels, to improve training efficiency without sacrificing convergence performance. Specifically, we first design a random sparsification algorithm to retain a fraction of the gradient elements in each client’s local model, thereby mitigating the performance degradation induced by DP and reducing the number of transmission parameters over wireless channels. Then, we analyze the convergence bound of the proposed algorithm, by modeling a non-convex FL problem. Next, we formulate a time-sequential stochastic optimization problem for minimizing the developed convergence bound, under the constraints of transmit power, the average transmitting delay, as well as the client’s DP requirement. Utilizing the Lyapunov drift-plus-penalty framework, we develop an analytical solution to the optimization problem. Extensive experiments have been implemented on three real-life datasets to demonstrate the effectiveness of our proposed algorithm. We show that our proposed algorithms can fully exploit the interworking between communication and computation to outperform the baselines, i.e., random scheduling, round robin, and delay-minimization algorithms.

Using Copula functions to predict climatic change impacts on floods in river source regions

Flood frequency in river source regions is significantly affected by rainfall and snowmelt as part of climatic changes.A traditional univariate flood frequency analysis cannot reflect the complexity of floods,and when used in isolation,it can only underestimate flood *** effective flood prevention and mitigation,it is essential to consider the combined effects of precipitation and *** functions can effectively quantify the joint distribution relationship between floods and their associated variables without restrictions on their distribution *** study uses copula functions to consider a multivariate probability distribution model of flood peak flow(Q)with cumulative snowmelt(CSm)and cumulative precipitation(CPr)for the Hutubi River basin located in northern Xinjiang,*** joint frequencies of rainfall and snowmelt floods are predicted using copula models based on the Coupled Model Intercomparison Project Phase 6 *** results show that Q has a significant positive correlation with 24-d CSm(r=0.559,p=0.002)and 23-d CPr(r=0.965,p<0.05).Flood frequency will increase in the future,and mid-(2050e2074)and long-term(2075e2099)floods will be more severe than those in the near-term(2025e2049).The probability of flood occurrence is higher under the SSP2-4.5 and SSP1-2.6 scenarios than under *** during the historical period(1990e2014)led to extreme floods,and increasing future precipitation trends are found to be *** increases with rising temperatures and occurs earlier than estimated,leading to an earlier flood period in the basin and more frequent snowmelt *** Q under the joint return period is larger than that during the same univariate return *** difference indicates that neglecting the interaction between precipitation and snowmelt for floods leads to an underestimation of the flood risk(with underestimations ranging from 0.3%to 22%).The underestimations decrease with an increase

Hydride ion(H-)conductors have drawn much attention due to their potential applications in hydrideion-based *** earth metal hydrides(REH_(x))have fast H-conduction which,unfortunately,is accompanied by detrimental electron conduction preventing their application as ion ***,REH_(x)(RE=Nd,Ce,and Pr)with varied grain sizes,rich grain boundaries,and defects have been prepared by ball milling and subsequent *** electronic conductivity of the ball-milled REH_(x)samples can be reduced by 2-4 orders of magnitude compared with the non-ball-milled *** relationship of electron conduction and miscrostructures in REH_(x)is studied and discussed based on experimental data and previously-proposed classical and quantum *** H-conductivity of all REH_(x)is about 10^(-4)to 10^(-3)S cm^(-1)at room temperature,showing promise for the development of H-conductors and their applications in clean energy storage and conversion.

Dear Editor,Protein histidine methylation has been found to be a widespread post-translational modification(PTM)in mammalian cells and represents more than 13%of all protein methylation events in the human methylome(Ning et al.,2016;Kapell and Jakobsson,2021).Recently,we and other groups independently identified METTL9 as the first broad-specificity histidine N1 methyltransferase in mammals(Davydova et al.,2021;Lv et al.,2021).METTL9 methylates a variety of substrates with a common x-His-x-His motif(His is for histidine;the second x is preferably small amino acids:A/C/G/S)and catalyzes methyl transfer to the last histidine.

Elemental doping is an effective strategy to enhance photocatalytic activity and extend the light absorption range of single-component *** this work,a series of La-doped CeO_(2) nanorods(La-CeO_(2)-x) with La content of 1 wt%-15 wt% are synthesized by a simple hydrothermal method and further used as photocatalyst for sulfamerazine(SMR) *** prepared La-CeO_(2)-x nanorods exhibit a great improvement in electron-hole pair migration and visible-light response due to the synergistic effect of abundant oxygen vacancies and heterogeneous elements(La).Consequently,La-CeO_(2)-x exhibited excellent visible-light photocatalytic performances and chemical stability for SMR degradation,the La-CeO_(2)-5 sample achieved the highest SMR degradation rate of 81%,which was 3.4 times higher than that of the original CeO_(2).Furthermore,three possible degradation pathways of SMR in La-CeO_(2) photocatalytic reactions were proposed by liquid chromatography-mass spectrometry ***,density functional theory calculations were carried out to provide an in-depth understanding of the structure-performance *** its excellent properties and better photocatalytic performance,this study demonstrates that La doping in CeO_(2) is an effective way to increase oxygen vacancy and improve the photochemical properties of photocatalysts.
