Freeze‒thaw induced landslides(FTILs)in grasslands on the Tibetan Plateau are a geological disaster leading to soil *** landslides reduce biodiversity and intensify landscape fragmentation,which in turn are strengthen by the persistent climate change and increased anthropogenic ***,conventional techniques for mapping FTILs on a regional scale are impractical due to their labor-intensive,costly,and time-consuming *** study focuses on improving FTILs detection by implementing image fusion-based Google Earth Engine(GEE)and a random forest *** of multiple data sources,including texture features,index features,spectral features,slope,and vertical‒vertical polarization data,allow automatic detection of the spatial distribution characteristics of FTILs in Zhidoi county,which is located within the Qinghai‒Tibet Engineering Corridor(QTEC).We employed statistical techniques to elucidate the mechanisms influencing FTILs *** enhanced method identifies two schemes that achieve high accuracy using a smaller training sample(scheme A:94.1%;scheme D:94.5%)compared to other methods(scheme B:50.0%;scheme C:95.8%).This methodology is effective in generating accurate results using only~10%of the training sample size necessitated by other *** spatial distribution patterns of FTILs generated for 2021 are similar to those obtained using various other training sample sources,with a primary concentration observed along the central region traversed by the *** results highlight the slope as the most crucial feature in the fusion images,accounting for 93%of FTILs occurring on gentle slopes ranging from 0°to 14°.This study provides a theoretical framework and technological reference for the identification,monitoring,prevention and control of FTILs in *** developments hold the potential to benefit the management of grassland ecosystem,reduce economic losses,and promote grassland sustainability.

Brain-derived neurotrophic factor inducing angiogenesis through modulation of matrix-degrading proteases

Background Recent studies have proved that brain-derived neurotrophic factor （BDNF） possesses angiogenic activity in vitro and in vivo. However, the proangiogenic mechanism of BDNF has not yet been provided with enough information. To explore the proangiogenic mechanism of BDNF, we investigated the effects of BDNF on extracellular proteolytic enzymes, including matrix metalloproteinases （MMPs） and serine proteases, particularly the urokinase-type plasminogen activator （uPA）-plasmin system in human umbilical vein endothelial cells （HUVECs） model.Methods Tube formation assay was performed in vitro to evaluate the effects of BDNF on angiogenesis. The HUVECs were treated with various concentrations of BDNF （25-400 ng/ml） for different （6-48 hours）, reverse transcriptase-polymerase chain reaction （RT-PCR） was used to assay MMP-2, MMP-9, TIMP-1, and TIMP-2 mRNA in HUVECs, and the conditioned medium was analyzed for MMP and uPA activity by gelatin zymography and fibrin zymography, respectively, uPA, plasminogen activator inhibitor （PAI）-1, tissue inhibitors of metalloproteinase （TIMP）-1, and TIMP-2 were quantified by western blotting analysis.Results BDNF elicited robust and elongated angiogeneis in two-dimensional cultures of HUVECs in comparison with control. The stimulation of serum-starved HUVECs with BDNF caused obvious increase in MMP-2 and MMP-9 mRNA expression and induced the pro-MMP-2 and pro-MMP-9 activation without significant differences in proliferation. However, BDNF had no effect on TIMP-1 and TIMP-2 production. BDNF increased uPA and PAI-1 production in a dose-dependent manner. Maximal activation of uPA and PAI-1 expression in HUVECs was induced by 100 ng/ml BDNF, while effects of 200 ng/ml and 400 ng/ml BDNF were slightly reduced in comparison with with those of 100 ng/ml. Value of T cell receptor gamma alternate reading frame protein and keratin 5 in endometrial carcinoma

Background Tumors with different gene expression develop and progress in different ways. To deepen our understanding of the progression in endometrial cancer, and provide a useful tool for accurate diagnosis and prognosis assessment, we identified the new molecular prognostic markers in endometrial carcinoma and analyzed the relationship of them with clinical and pathological features of endometrial carcinoma. Methods Ninety-four cases of endometrial endometrioid adenocarcinoma with complete data from the Peking University People＇s Hospital from 2000 to 2008 and 40 cases of normal endometrium were enrolled. Among these, 30 endometrial endometrioid adenocarcinoma samples of different International Federation of Gynecology and Obstetrics （FIGO） stage were selected for further Agilent genome-wide microarray analysis. Significance analysis of microarrays （SAM） was used to identify genes that are significantly associated with tumor progress. Immunohistochemistry was utilized to identify the genes of interest in endometrial carcinoma and normal endometrium. The relationship between the genes and the age, clinical stage, histological grade, myometrium invaded depth, lymph node metastasis status, and the expression of ER, PR, P53, and PTEN were analyzed by X2 test. Results Analysis between FIGO 1988 stage I and stage III identified a 362-gene ＂progress signature＂; 171 downregulated and 191 up-regulated genes. Among the alterative genes, TARP （T cell receptor gamma alternate reading frame protein） and KRT5 （keratin 5） decreased 3.57 fold and 5.8 fold in FIGO stage III patients. The expression of TARP in endometrial carcinoma increased compared to normal endometrium, while that of KRT5 decreased （P〈0.05）. The expression of TARP and KRT5 decreased when stage, histological grading, myometrium invaded depth increased （P〈0.05）. Effects of insulin, insulin-like growth factor-I and -II on proliferation and intracellular signaling in endometrial carcinoma cells with different expression levels of insulin receptor isoform A

Background Hyperinsulinemia, insulin-like growth factor （IGF）-I and -Ⅱ （IGF-Ⅱ） are associated with increased risk of endometrial carcinoma. Insulin receptor isoform A （IR-A） is more frequently expressed in endombtrial carcinoma than in normal endometrial tissues. To better understand their roles in endometrial carcinoma, we investigated the effects of insulin, IGF-I, and IGF-II in endometrial carcinomas cells with different IR-A expression levels. Methods To explore the role of IR-A in mediating the activity of IGF-I, IGF-II, and insulin, we investigate the cellular proliferation of endometrial carcinoma cell lines RL95-2 and RL95-2-1R-A by MTS assays. Then we examined the protein kinase Akt phosphorylation and extracellular signal-regulated kinase （ERK） 1/2 phosphorylation in both cell lines by Western blotting. The effect of IGF-II and AG1024 on cell cycle progression and apoptosis was assessed by fiowcytometry. To examine whether the effects of IGFs were mediated by IR-A, we blocked IGF-I receptor （IGF-IR） in both cell lines using AG1024, an IGF-IR-specific inhibitor. Results IGF-I and IGF-II significantly enhanced proliferation of both cell lines （P 〈0.05）. By contrast, insulin significantly increased proliferation of RL95-2-1R-A cells only （P 〈0.05）. IGF-I and IGF-II significantly increased pAkt levels in RL95-2 cells and pERK1/2 levels in RL95-2-1R-A cells （all, P 〈0.05）. Insulin increased pERK1/2 levels in RL95-2-1R-A cells only （P 〈0.05）. LY294002 and PD98059 inhibited the specific signaling activities and cellular proliferation. After AG1024 pretreatment, neither IGF-I nor IGF-II affected pAkt levels in RL95-2 cells. IGF-II, but not IGF-I, increased pERK1/2 levels in RL95-2-1R-A cells. After AG1024 pretreatment, the proliferation rate and DNA content corresponding to the S phase increased and apoptosis decreased significantly in IGF-II-treated RL95-2-1R-A cells only （P 〈0.05）. Effect of traditional Chinese medicine compounds on rumen fermentation, methanogenesis and microbial flora in vitro

This study was conducted to investigate the effects of traditional Chinese medicine compounds(TCMC)on rumen fermentation, methane emission and populations of ruminal microbes using an in vitro gas production technique. Cablin patchouli herb(CPH), Atractylodes rhizome(AR), Amur Cork-tree(AC) and Cypsum were mixed with the weight ratios of 1:1:1:0.5 and 1:1:1:1 to make up TCMC1 and TCMC2,respectively. Both TCMC were added at level of 25 g/kg of substrate dry matter. In vitro gas production was recorded and methane concentration was determined at 12 and 24 h of incubation. After 24 h, the incubation was terminated and the inoculants were measured for pH, ammonia nitrogen, volatile fatty acids(VFA). Total deoxyribonucleic acid of ruminal microbes was extracted from the inocula, and populations were determined by a real-time quantitative polymerase chain reaction. Populations of total rumen methanogens, protozoa, total fungi, Ruminococcus albus, Fibrobacter succinogenes and Ruminococcus flavefaciens were expressed as a proportion of total rumen bacterial 16 S ribosomal deoxyribonucleic acid. Compared with the control, the 2 TCMC decreased(P < 0.05) total VFA concentration,acetate molar proportion, acetate to propionate ratio, gas and methane productions at 12 and 24 h,hydrogen(H) produced and consumed, and methanogens and total fungi populations, while the 2 TCMC increased(P < 0.05) propionate molar proportion. Traditional Chinese medicine compound 1 also decreased(P ≤ 0.05) R. flavefaciens population. From the present study, it is inferred that there is an effect of the TCMC in suppressing methanogenesis, probably mediated via indirect mode by channeling H2 utilized for methanogenesis to synthesis of propionate and direct action against the rumen microbes involved in methane formation. In addition, the relative methane reduction potential(RMRP) of TCMC2 was superior to that of TCMC1.

We develop a novel coumarin-all〈yne derivative （NC7-AL）, which can specifically react with Au^3＋ and give a colorimetric and fluorescent ＂turn-on＂ response toward Au3~, Notably, other alkynophilic metal species such as Au^＋, Ag^＋, Pd^2＋, Ni^2＋, Cu^2＋, and Hg^2＋ do not produce an interfering signal. A good linear relationship between emission intensity at 420 nm and Au^3＋ concentration from 0 to 2 equivalent is observed, and the detection limit （3σ/k） is estimated to be ca. 3.58 nmol/L. Background This retrospective study assessed the efficacy and safety of ketogenic diet therapies in children with epilepsy caused by SLC2A1 genetic mutations and glucose transporter type 1 deficiency *** Pediatric patients with epilepsy symptoms admitted to our medical center between January 2017 and October 2021 were included if they presented with an SLC2A1 genetic mutation on whole-exome *** analyzed the patients’convulsions and treatment with antiepileptic *** patients were followed up at different time periods after ketogenic diet *** Six patients with SLC2A1 mutations were included in this *** patients had seizures of different types and frequencies,and they took antiepileptic drugs to relieve their *** were then treated with a ketogenic diet for at least four *** analyzed epilepsy control rates at 1,2,3,6,and 12 months after ketogenic diet *** patients were seizure-free within a month of receiving the diet *** patients were followed up for six months,three were followed up for 12 months after the treatment,and there was no recurrence of epilepsy during this *** antiepileptic drug withdrawal,none of the patients experienced seizure relapse when receiving ketogenic diet treatment *** severe adverse events occurred during the *** Ketogenic diet therapy is very effective and safe for the treatment of epilepsy caused by SLC2A1 ***,patients with glucose transporter type 1 deficiency syndrome caused by SLC2A1 mutations should begin ketogenic diet treatment as soon as possible.
