Inhibition of microRNA-29b suppresses oxidative stress and reduces apoptosis in ischemic stroke

MicroRNAs(miRNAs) regulate protein expression by antagonizing the translation of mRNAs and are effective regulators of normal nervous system development, function, and disease. Micro RNA-29 b(mi R-29 b) plays a broad and critical role in brain homeostasis. In this study, we tested the function of mi R-29 b in animal and cell models by inhibiting mi R-29 b expression. Mouse models of middle cerebral artery occlusion were established using the modified Zea-Longa suture method. Prior to modeling, 50 nmol/kg mi R-29 b antagomir was injected via the tail vein. Mi R-29 b expression was found to be abnormally increased in ischemic brain tissue. The inhibition of mi R-29 b expression decreased the neurological function score and reduced the cerebral infarction volume and cell apoptosis. In addition, the inhibition of mi R-29 b significantly decreased the malondialdehyde level, increased superoxide dismutase activity, and Bcl-2 expression, and inhibited Bax and Caspase3 expression. PC12 cells were treated with glutamate for 12 hours to establish in vitro cell models of ischemic stroke and then treated with the mi R-29 antagomir for 48 hours. The results revealed that mi R-29 b inhibition in PC12 cells increased Bcl-2 expression and inhibited cell apoptosis and oxidative damage. These findings suggest that the inhibition of mi R-29 b inhibits oxidative stress and cell apoptosis in ischemic stroke, producing therapeutic effects in ischemic stroke. Ethical considerations of cellular immunotherapy for cancer

With the rapid development of immunology, molecular biology, and associated technologies such as next-generation sequencing, cellular immunotherapy has recently become the fourth major cancer treatment. Immunotherapies based on T cells, natural killer cells, and dendritic cells play key roles in cancer immunotherapy. However, their application in clinical practice raises several ethical issues. Thus, studies should focus on proper adherence to basic ethical principles that can effectively guide and solve related clinical problems in the course of treatment, improve treatment effects, and protect the rights and interests of patients. In this review, we discuss cellular immunotherapy-related ethical issues and highlight the ethical practices and current status of cellular immunotherapy in China. These considerations may supplement existing ethical standards in cancer immunotherapy.

Effects of aggregate size distribution and carbon nanotubes on the mechanical properties of cemented gangue backfill samples under true triaxial compression

The mechanical behavior of cemented gangue backfill materials(CGBMs)is closely related to particle size distribution(PSD)of aggregates and properties of cementitious ***,the true triaxial compression tests,CT scanning,SEM,and EDS tests were conducted on cemented gangue backfill samples(CGBSs)with various carbon nanotube concentrations(P_(CNT))that satisfied fractal theory for the PSD of *** mechanical properties,energy dissipations,and failure mechanisms of the CGBSs under true triaxial compression were systematically *** results indicate that appropriate carbon nanotubes(CNTs)effectively enhance the mechanical properties and energy dissipations of CGBSs through micropore filling and microcrack bridging,and the optimal effect appears at P_(CNT)of 0.08wt%.Taking PSD fractal dimension(D)of 2.500 as an example,compared to that of CGBS without CNT,the peak strength(σ_(p)),axial peak strain(ε_(1,p)),elastic strain energy(Ue),and dissipated energy(U_(d))increased by 12.76%,29.60%,19.05%,and90.39%,***,excessive CNTs can reduce the mechanical properties of CGBSs due to CNT agglomeration,manifesting a decrease inρ_(p),ε_(1,p),and the volumetric strain increment(Δε_(v))when P_(CNT)increases from 0.08wt%to 0.12wt%.Moreover,the addition of CNTs improved the integrity of CGBS after macroscopic failure,and crack extension in CGBSs appeared in two modes:detour and pass through the ***σ_(p)and U_(d)firstly increase and then decrease with increasing D,and porosity shows the opposite ***ε_(1,p)andΔε_(v)are negatively correlated with D,and CGBS with D=2.150 has the maximum deformation parameters(ε_(1,p)=0.05079,Δε_(v)=0.01990)due to the frictional slip effect caused by coarse *** increasing D,the failure modes of CGBSs are sequentially manifested as oblique shear failure,"Y-shaped"shear failure,and conjugate shear failure.

Factors associated with a SARS-CoV-2 recurrence after hospital discharge among patients with COVID-19:systematic review and meta-analysis

Background:The proportion of recurrences after discharge among patients with coronavirus disease 2019(COVID-19)was reported to be between 9.1%and 31.0%.Little is known about this issue,however,so we performed a meta-analysis to summarize the demographical,clinical,and laboratorial characteristics of non-recurrence and recurrence ***:Comprehensive searches were conducted using eight electronic *** re-garding the demographic,clinical,and laboratorial characteristics of both recurrence and non-recurrence groups were extracted,and quantitative and qualitative analyses were ***:Ten studies involving 2071 COVID-19 cases were included in this *** proportion of recurrence cases involving patients with COVID-19 was 17.65%(between 12.38%and 25.16%)while older patients were more likely to experience recurrence(weighted mean difference(WMD)=1.67,range between 0.08 and 3.26).The time from discharge to recurrence was 13.38 d(between 12.08 and 14.69 d).Patients were categorized as having moderate severity(odds ratio(OR)=2.69,range between 1.30 and 5.58),while those with clinical symptoms including cough(OR=5.52,range between 3.18 and 9.60),sputum production(OR=5.10,range between 2.60 and 9.97),headache(OR=3.57,range between 1.36 and 9.35),and diz-ziness(OR=3.17,range between 1.12 and 8.96)were more likely to be associated with *** pre-senting with bilateral pulmonary infiltration and decreased leucocyte,platelet,and CD4+T counts were at risk of COVID-19 recurrence(OR=1.71,range between 1.07 and 2.75;WMD=?1.06,range between?1.55 and?0.57,WMD=?40.39,range between?80.20 and?0.48,and WMD=?55.26,range between?105.92 and?4.60,respectively).Conclusions:The main factors associated with the recurrence of severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)after hospital discharge were older age,moderate severity,bilateral pulmonary infiltration,laboratory findings including decreased leucocytes,platelets,and CD4+T counts,a

An experimental investigation on static directiona stability

A generic aircraft usually loses its static directional stability at moderate angle of attack(typically 20–30°). In this research, wind tunnel studies were performed using an aircraft model with moderate swept wing and a conventional vertical tail. The purpose of this study was to investigate flow mechanisms responsible for static directional stability. Measurements of force, surface pressure and spatial flow field were carried out for angles of attack from 0° to 46° and sideslip angles from-8° to 8°. Results of the wind tunnel experiments show that the vertical tail is the main contributor to static directional stability, while the fuselage is the main contributor to static directional instability of the model. In the sideslip attitude for moderate angles of attack, the fuselage vortex and the wing vortex merged together and changed asymmetrically as angle of attack increased on the windward side and leeward side of the vertical tail. The separated asymmetrical vortex flow around the vertical tail is the main reason for reduction in the static directional stability. Compared with the wing vortices, the fuselage vortices are more concentrated and closer to the vertical tail, so the yawing moment of vertical tail is more unstable than that when the wings are absent. On the other hand,the attached asymmetrical flow over the fuselage in sideslip leads to the static directional instability of the fuselage being exacerbated. It is mainly due to the predominant model contour blockage effect on the windward side flow over the model in sideslip, which is strongly affected by angle of attack.
