Effects of dosimetric inadequacy on local control and toxicities in the patients with T4 nasopharyngeal carcinoma extending into the intracranial space and treated with *** radiotherapy plus chemotherapy

Background:To protect neurological tissues,underdosing occurs in most cases of T4 nasopharyngeal carcinoma(NPC) with intracranial *** this study,we aimed to evaluate the effect of dosimetric inadequacy on local control and late neurological toxicities for patients treated with intensity-modulated radiotherapy(IMRT) plus ***:We prospectively enrolled patients who had non-metastaticT4 NPC with intracranial extension treated between January 2009 and November *** prescribed dose was 66.0-70.4 Gy to the primary planning target volume(primary gross tumor volume [GTVp;i.e.,the nasopharyngeal tumor] +5.0 mm).Dose-volume histogram parameters were calculated,including minimum point dose(D_(min)) and dose to 95% of the target volume(D95).All patients received chemotherapy with the cisplatin,5-fluorouracil,and docetaxel *** were estimated using the Kaplan-Meier method and compared using the log-rank ***:In total,41 patients were *** local partial response rate was 87.8% after induction *** a median follow-up of 51 months,7 patients experienced failure in the nasopharynx;the 3-year local failure-free survival and overall survival rates of the 41 patients were 87.4% and 90.2%,*** actual mean D_(min) to the GTVp was 55.2 Gy(range 48.3-67.3 Gy),and D95 was 61.6 Gy(range 52.6-69.0 Gy).All doses received by neurological organs remained well within their dose *** patients developed temporal lobe necrosis or other neurological ***:With relative underdosed IMRT plus effective chemotherapy,the patients achieved satisfactory local control with few late toxicities of the central nervous *** the acceptable extent of dosimetric inadequacy requires further exploration.

Advanced forecasting of career choices for college students based on campus big data

Career indecision is a difficult obstacle confronting adolescents. Traditional vocational assessment research measures it by means of questionnaires and diagnoses the potential sources of career indecision. Based on the diagnostic outcomes, career counselors develop treatment plans tailored to students. However, because of personal motives and the architecture of the mind, it may be difficult for students to know themselves, and the outcome of questionnaires may not fully reflect their inner states and statuses. Selfperception theory suggests that students＇ behavior could be used as a clue for inference. Thus, we proposed a data-driven framework for forecasting student career choice upon graduation based on their behavior in and around the campus, thereby playing an important role in supporting career counseling and career guidance. By evaluating on 10M behavior data of over four thousand students, we show the potential of this framework for this functionality.

Overexpression of GhPFN2 enhances protection against Verticillium dahliae invasion in cotton

Growing evidence indicates that actin cytoskeleton is involved in plant innate immune responses, but the functional mechanism remains largely unknown. Here, we investigated the behavior of a cotton profilin gene （GhPFN2） in response to Verticillium dahliae invasion, and evaluated its contribution to plant defense against this soil-borne fungal pathogen. GhPFN2 expression was up-regulated when cotton root was inoculated with V. dahliae, and the actin architecture was reorganized in the infected root cells, with a clear increase in the density of filamentous actin and the extent of actin btmdling. Compared to the wild type, GhPFN2-overexpressing cotton plants showed enhanced protection against V. dahliae infection and the actin cytoskeleton organization in root epidermal cells was clearly altered, which phenocopied that of the wild-type （WT） root cells challenged with V. dahliae. Bactericidal ability of target acidic phospholipids and phagocytosis of CDC42 GTPase-mediated cytoskeletal rearrangement underlie functional conservation of CXCL12 in vertebrates

Chemokine CXCL12 plays a crucial role in both direct bactericidal activity and phagocytosis in humans. However, the mechanisms and evolutionary functions of these processes in vertebrates remain largely unknown. In this study, we found that the direct bactericidal activity of CXCL12 is highly conserved across various vertebrate lineages, including Arctic lamprey(Lampetra japonica), Basking shark(Cetorhinus maximus), grass carp(Ctenopharyngodon idella), Western clawed frog(Xenopus tropicalis), Green anole(Anolis carolinensis), chicken(Gallus gallus), and human(Homo sapiens). CXCL12 also has been shown to promote phagocytosis in lower and higher vertebrates. We then employed C. idella CXCL12a(CiCXCL12a) as a model to further investigate its immune functions and underlying mechanisms. CiCXCL12a exerts direct broad-spectrum antibacterial activity by targeting bacterial acidic phospholipids, resulting in bacterial cell membrane perforation, and eventual lysis. Monocytes/macrophages are attracted to the infection sites for phagocytosis through the rapid production of CiCXCL12a during bacterial infection. CiCXCL12a induces CDC42 and CDC42 GTPase activation, which in turn mediates F-actin polymerization and cytoskeletal rearrangement. The interaction between F-actin and Aeromonas hydrophila facilitates bacterial internalization into monocytes/macrophages. Additionally, A. hydrophila is colocalized within early endosomes, late endosomes and lysosomes, ultimately degrading within phagolysosomes. CiCXCL12a also activates PI3K-AKT, JAK-STAT5 and MAPK-ERK signaling pathways. Notably, only the PI3K-AKT signaling pathway inhibits LPS-induced monocyte/macrophage apoptosis. Thus, CiCXCL12a plays key roles in reducing tissue bacterial loads, attenuating organ injury, and decreasing mortality rates. Altogether, our findings elucidate the conserved mechanisms underlying CXCL12-mediated bactericidal activity and phagocytosis, providing novel perspectives into the immune functions of CXC

New dual functional CYP450 gene involves in isoflavone biosynthesis in Glycine max L.

Glycine max *** a large amount of isoflavonoid compounds,which is beneficial for plant defense,plant-microbe symbiotic interactions,and human *** CYP450 subfamily genes are involved in the flavonoid biosynthetic pathway in *** the present study,we found 24 CYP82 subfamily genes were differentially expressed in various tissues of soybean,in Phytophthora sojae-infected soybean varieties and in soybean hairy roots treated with cell wall glucan *** of them(GmCYP82A2,GmCYP82A3,GmCYP82A4,GmCYP82A23,GmCYP82C20 and GmCYP82D26)were co-expressed with other known isoflavonoid pathway genes in *** enzymatic activity in yeast feeding assays showed that only GmCYP82D26 was able to convert naringenin to daidzein with both aryl migration and dehydration *** GmCYP82D26 was over-expressed in soybean hairy roots,the contents of the two major isoflavonoid aglycones in soybean(daidzein and genistein)were reduced,but total flavonoids were not *** GmCYP82D26 was suppressed by RNAi in the hairy roots,daidzein content was decreased but genistein content was increased,with unchanged total flavonoid ***82D26 was found to be localized in the endoplasmic reticulum at subcellular level when transiently expressed in tobacco leaf ***82D26 gene was preferentially expressed in roots,with low expression level in other tissues in *** modeling and molecular docking showed that GmCYP82D26 could form hydrogen bond with both HEM and naringenin at C5-OH and C4 *** these results indicated that GmCYP82D26 possesses new and dual enzymatic activity,which bridges the two branches(daidzein and genistein branch)of isoflavonoid pathway in soybean.

Pre-summer Persistent Heavy Rain over Southern China and Its Relationship with Intra-seasonal Oscillation of Tropical Atmosphere

Based on daily precipitation data supplied by the Chinese meteorological administration,hourly reanalysis datasets provided by the ECMWF and daily outgoing long wave radiation supplied by the NOAA,the evolution regularity of continuous heavy precipitation over Southern China(SC)from April to June in 1979-2020 was systematically *** interaction between specific humidity and circulation field at the background-scale,the intra-seasonal-scale and the synoptic-scale,and its influence on persistent heavy precipitation over the SC during the April-June rainy season were quantitatively diagnosed and *** results are as *** heavy rainfall events(PHREs)over the SC during the April-June rainy season occur frequently from mid-May to mid-and late-June,exhibiting significant intra-seasonal oscillation(10-30-day)*** integrated moisture flux convergence(VIMFC)can well represent the variation of the PHREs.A multiscale quantitative diagnosis of the VIMFC shows that the pre-summer PHREs over the SC are mainly affected by the background water vapor(greater than 30 days),intraseasonal circulation disturbance(10-30-day)and background circulation(greater than 30 days),and water vapor convergences are the main *** SC is under the control of a warm and humid background and a strong intraseasonal cyclonic circulation,with strong convergence and ascending movements and abundant water vapor conditions during the period of the ***,the westward inter-seasonal oscillation of tropical atmosphere keeps the precipitation system over the SC for several consecutive days,eventually leading to the occurrence,development and persistence of heavy precipitation.

Overexpression of GhFIM2 propels cotton fiber development by enhancing actin bundle formation

Cell elongation and secondary wall deposition are two consecutive stages during cotton fiber development. The mechanisms controlling the progression of these two developmental phases remain largely unknown. Here, we report the functional characterization of the actin-bundling protein GhFIM2 in cotton fiber. Overexpression of GhFIM2 increased the abundance of actin bundles, which was accompanied with accelerated fiber growth at the fastelongating stage. Meanwhile, overexpression of GhFIM2 could propel the onset of secondary cell wall biogenesis. These results indicate that the dynamic rearrangement of actin higher structures involving GhFIM2 plays an important role in the development of cotton fiber cells.

Contribution of additives Cu to its accumulation in pig feces: study in Beijing and Fuxin of China

Massive amounts of pig manure are produced by intensive pig farm in China, and the composition of pig manure has changed much due to the use of feed additives. However, little is known about the exact Cu （copper） feed as additives or present as contaminants in pig feed and the residues in feces. One hundred and thirty-seven feeds and one hundred and forty-two fecal samples from 48 pig farms were collected in Beijing and Fuxin cities in 1999 and 2005, respectively. The concentrations of Cu were in the range of 6.86-395.19 mg/kg in the feed samples, and the mean values were in the order of weaner〉 grower-finisher〉 sow＇s feeds. The high concentrations over EU recommendations implied that excessive levels of Cu are fed on many pig farms in Beijing and Fuxin. Cu was also present in high concentrations in feces, and concentrations were highly variable. Cu concentrations in the feces from grower-finisher and weaner pigs were significantly greater than feces of sows. The super-intensive and small-scale farms had higher levels of Cu in feces than the middle farms. Cu concentrations in pig feces were approximately 5-times greater than in pig feeds. Feed management in grower-finisher pigs on super-intensive and small-scale pig farms is needed to reduce high Cu concentrations in feces and risks to soil contamination while feces are land-applied.

A molecular dynamics simulation method is used to study the growth of narrow single-wall carbon *** is found that the growth temperature and the species of small carbon clusters in the feedstock are important for the quality of the nanotubes *** is a temperature range of 1000-1500 K in which the narrow armchair single-wall carbon nanotubes can grow rapidly via adduction of C2 dimers,even without the existence of *** narrow zigzag tubes cannot keep open-ended *** the feedstock consists of various species of carbon clusters,the tubes cannot keep open-ended *** higher temperatures,the narrow nanotubes close rapidly in the noncatalytic environment,and the products grown are fullerene-like capsules.

Chiral su(3) Quark Model Study of Tetraquark States: cn■/cs

The new members of the charm-strange family Dsj^＊（2317）, Dsj（2460）, and Ds（2632）, which have the surprising properties, are challenging the present models. Many theoretical interpretations have been devoted to this issue. Most authors suggest that they are not the conventional cs^- quark model states, but possibly are four-quark states, molecule states, or mixtures of a P-wave cs^- and a four-quark state. In this work, we follow the four-quark-state picture, and study the masses of cnn^-s^-/css^-s^- states （n is u or d quark） in the chiral su（3） quark model. The numerical results show that the mass of the mixed four-quark state （cnn^-s^-/css^-s^-） with spin parity j^P ： 0^＋ might not be Ds （2632）. At the same time, we also conclude that Dsj^＊（2317） and Dsj（2460） cannot be explained as the pure four-quark state.
