Background:Non-small cell lung cancer(NSCLC)patients with epidermal growth factor receptor(EGFR)mutations or anaplastic lymphoma kinase(ALK)fusions show dramatic responses to specific tyrosine kinase inhibitors(TKIs);however,after 10-12 months,secondary mutations arise that confer *** generated a murine xenograft model using patient-derived NSCLC cells isolated from the pleural fluid of two patients with NSCLC to investigate the mechanisms of resistance against the ALK-and EGFR-targeted TKIs crizotinib and osimertinib,***:Genotypes of patient biopsies and xenograft tumors were determined by whole exome sequencing(WES),and patients and xenograft-bearing mice received targeted treatment(crizotinib or osimertinib)*** mice were also treated for prolonged periods to identify whether the development of drug resistance and/or treatment responses were associated with tumor ***,the pathology of patients biopsies and xenograft tumors were compared ***:The histological characteristics and chemotherapy responses of xenograft tumors were similar to the actual *** showed that the genotypes of the xenograft and patient tumors were similar(an echinoderm microtu-bule-associated protein-like 4-ALK(EML4-ALK)gene fusion(patient/xenograft:CTC15035EML4-ALK)and EGFR L858R and T790M mutations(patient/xenograft:CTC15063EGFR L858R,T790M)).After continuous crizotinib or osimertinib treatment,WES data suggested that acquired ALK E1210K mutation conferred crizotinib resistance in the CTC15035EML4-ALK xenograft,while decreased frequencies of EGFR L858R and T790M mutations plus the appearance of v-RAF murine sarcoma viral oncogene homolog B(BRAF)G7V mutations and phosphatidylinositol-4-phosphate 3-kinase catalytic subunit type 2 alpha(PIK3C2A)A86fs frame shift mutations led to osimertinib resistance in the CTC15063EGFR L858R,T790M ***:We successfully developed a new method of generating drug resis

An Improved Method for Correction of Air Temperature Measured Using Different Radiation Shields

The variation of air temperature measurement errors using two different radiation shields （DTR502B Vaisala,Finland,and HYTFZ01,Huayun Tongda Satcom,China） was *** were collected in the field at the Daxing weather station in Beijing from June 2011 to May *** air temperature values obtained with these two commonly used radiation shields were lower than the reference records obtained with the new Fiber Reinforced Polymers （FRP） Stevenson *** most cases,the air temperature errors when using the two devices were smaller on overcast and rainy days than on sunny days; and smaller when using the imported rather than the Chinese *** measured errors changed sharply at sunrise and sunset,and reached maxima at *** diurnal variation characteristics were,naturally,related to changes in solar *** relationships between the record errors,global radiation,and wind speed were *** improved correction method was proposed based on the approach described by Nakamura and Mahrt （2005）（NM05）,in which the impact of the solar zenith angle （SZA） on the temperature error is considered and extreme errors due to changes in SZA can be corrected *** errors were reduced significantly after correction by either method for both *** error reduction rate using the improved correction method for the Chinese and imported shields were 3.3％ and 40.4％ higher than those using the NM05 method,respectively.

Alloying degree, particle size and the level of dispersion are the key structural parameters of Pt-Ru/C catalyst in fuel cells. Solvent（s） used in the preparation process can affect the particle size and alloying degree of the object substance, which lead to a great positive impact on its properties. In this work, three types of solvents and their mixtures were used in preparation of the Pt-Ru/C catalysts by chemical reduction of metal precursors with sodium borohydride at room temperature. The structure of the catalysts was characterized by X-ray diffraction （XRD） and Transmission electron microscopy （TEM）. The catalytic activity and stability for methanol electro-oxidation were studied by Cyclic Voltammetry （CV） and Chronoamperometry （CA）. Pt-Ru/C catalyst prepared in H2O or binary solvents of H2O and isopropanol had large particle size and low alloying degree leading to low catalytic activity and less stability in methanol electro-oxidation. When tetrahydrofuran was added to the above solvent systems, Pt-Ru/C catalyst prepared had smaller particle size and higher alloying degree which resulted in better catalytic activity, lower onset and peak potentials, compared with the above catalysts. Moreover, the catalyst prepared in ternary solvents of isopropanol, water and tetrahydrofuran had the smallest particle size, and the high alloying degree and the dispersion kept unchanged. Therefore, this kind of catalyst showed the highest catalytic activity and good stability for methanol electro-oxidation.

Flexible magnetic materials with robust and controllable perpendicular magnetic anisotropy(PMA)are highly desirable for developing flexible high-performance spintronic ***,it is still challenge to fabricate PMA films on polymers ***,we report a facile method for synthesizing single-crystal freestanding SrRuO_(3) membranes with controlled crystal structure and orientation using water-soluble Ca_(3-x)Sr_(x)Al_(2)O_(3)6 sacrificial *** cooperative effect of crystal structure and orientation,flexible membranes reveal highly tunable magnetic anisotropy from in-plane to out-of-plane with a remarkable PMA energy of 7×10^(6) erg·cm^(−3).First-principle calculations reveal that the underlying mechanism of PMA modulation is intimately correlated with structure-controlled Ru 4d-orbital occupation,as well as spin-orbital matrix element differences,dependent on the crystal *** addition,even after 10,000 bending cycles,the PMA keeps stable,indicating a robust magnetism reliability in the prepared *** work provides a feasible approach to prepare the flexible oxide films with strong and controllable PMA.

High-entropy oxides(HEOs)have received significant attention because of their tunable mechanical properties and wide range of functional ***,the conventional method used for sintering HEOs requires prolonged processing time,which results in excessive grain growth,thereby compromising their ***,an ultrafast high-temperature sintering(UHS)strategy was adopted,and rock-salt composite(Mg_(0.2)Co_(0.2)Ni_(0.2)Cu_(0.2)Zn_(0.2))O was selected as model *** parameters were tuned to illustrate the influence of applied current and soaking time on the densification process and resulting grain ***,the electrochemical performance of UHS-synthesized microparticles as anode materials in lithium-ion batteries was *** results show that the ultrafast heating rate results in fine grains with a diameter of~6–8μm and density of 95%,which are much smaller and similar to those obtained using the conventional sintering method(25μm and 96%).Moreover,the high surface area and reactivity of the microparticles,as well as their sluggish diffusion effect and structural stability,contribute to outstanding performance with high capacity(336 mA·h/g at 1 A/g)and ultralong cyclability(1000 cycles).This novel technique offers valuable insights into the densification process of HEOs and other materials and can thus broaden their application range.

Eight new annonaceous acetogenins, squamotin A–D(1–4), annosquatin IV–V(5 and 6), muricin O(7) and squamosten B(8), together with four known ones(9–12) were isolated from the seeds of Annona squamosa. Their structures were elucidated by chemical methods and spectral data. The inhibitory activities of compound 1–9 against three multidrug resistance cell lines were evaluated. All tested compounds showed strong cytotoxicity.

Structural insights into the catalytic mechanism of aldehyde-deformylating oxygenases

The fatty alk（a/e）ne biosynthesis pathway found in cyanobacteria gained tremendous attention in recent years as a promising alternative approach for biofuel production. Cyanobacterial aldehyde-deformylating oxygenase （cADO）, which catalyzes the conversion of Cn fatty aldehyde to its corresponding Cn-1 alk（ale）ne, is a key enzyme in that pathway. Due to its low activity, alk（a/e）ne production by cADO is an inefficient process. Previous biochemical and structural investi. gations of cADO have provided some information on its catalytic reaction. However, the details of its cata- lytic processes remain unclear. Here we report five crystal structures of cADO from the Synechococcus elongates strain PCC7942 in both its iron-free and iron-bound forms, representing different states during its catalytic process. Structural comparisons and functional enzyme assays indicate that Giu144, one of the iron-coordinating residues, plays a vital role in the catalytic reaction of cADO. Moreover, the helix where Glu144 resides exhibits two distinct conformations that correlates with the different binding states of the di-iron center in cADO structures. Therefore, our results provide a structural explanation for the highly labile feature of cADO di-iron center, which we pro- posed to be related to its low enzymatic activity. On the basis of our structural and biochemical data, a possible catalytic process of cADO was proposed, which could aid the design of cADO with improved activity.
