Mapping theArabidopsis Metabolic Landscape by Untargeted Metabolomics at Different Environmental Conditions

Metabolic genome-wide association studies （mGWAS）, whereupon metabolite levels are regarded as traits, can help unravel the genetic basis of metabolic networks. A total of 309Arabidopsis accessions were grown under two independent environmental conditions （control and stress） and subjected to untargeted LC-MS- based metabolomic profiling; levels of the obtained hydrophilic metabolites were used in GWAS. Our two- condition-based GWAS for more than 3000 semi-polar metabolites resulted in the detection of 123 highly resolved metabolite quantitative trait loci （p ≤ 1.0E-08）, 24.39% of which were environment-specific. Interestingly, differently from natural variation in Arabidopsis primary metabolites, which tends to be controlled by a large number of small-effect loci, we found several major large-effect loci alongside a vast number of small-effect loci controlling variation of secondary metabolites. The two-condition-based GWAS was fol- lowed by integration with network-derived metabolite-transcript correlations using a time-course stress experiment. Through this integrative approach, we selected 70 key candidate associations between struc- tural genes and metabolites, and experimentally validated eight novel associations, two of them showing differential genetic regulation in the two environments studied. We demonstrate the power of combining large-scale untargeted metabolomics-based GWAS with time-course-derived networks both performed under different ablotic environments for identifying metabollte-gene associations, providing novel global insights into the metabolic landscape of Arabidopsis.

本文建立了完整的循环流化床流体动力学数学模型。内容包括顶部稀区和底部密区局部状态的模拟和径向分布的计算。并确定了两区共存和三种不同操作方式存在的条件。研究表明,能量最小在循环流化床的气固相互作用中起着重要的作用。顶部稀区和底部密区的气固相互作用机理完全不同。底部密区处于颗粒流体协调(FPC)状态,其稳定条件是悬浮输送能耗N_st等于最小值;而顶部稀区处于流体控制状态,其稳定条件是N_st=最大值或W_st=最小值。(W_st)_FPC=(W_st)_FD时,两区共存或过渡,系统处于饱和夹带状态。两个区域中的径向分布也都必须满足相应的整体稳定性条件。

Dimensioning of Punctiform Metal-Composite Joints: A Section-Force Related Failure Criterion

Reliable line production processes and simulation tools play a central role for the structural integration of thermoplastic composites in advanced lightweight constructions. Provided that material-adapted joining technologies are available, they can be applied in heavy-duty multi-material designs (MMD). A load-adapted approach was implemented into the new fully automatic and fault-tolerant thermo mechanical flow drill joining (FDJ) concept. With this method it is possible to manufacture reproducible high strength FRP/metal-joints within short cycle times and without use of extra joining elements for the first time. The analysis of FDJ joints requires a simplified model of the joint to enable efficient numerical simulations. The present work introduces a strategy in modeling a finite-element based analogous-approach for FDJ-joints with glass fiber reinforced polypropylene and high-strength steel. Combined with a newly developed section-force related failure criterion, it is possible to predict the fundamental failure behavior in multi-axial stress states. The functionality of the holistic approach is illustrated by a demonstrator that represents a part of a car body-in-white structure. The comparison of simulated and experimentally determined failure loads proves the applicability for several combined load cases.

Dear Editor, The tropical plant Madagascar periwinkle Catharanthus roseus （L.） *** is a rich source of plant-derived medicinal ter-penoid indole alkaloids （TIAs）, including the anti-hypertensive ajmalicine, the sedative compound serpentine, and the anti-cancer drugs vinblastine and vincristine. However, the latter two compounds are produced in C. roseus plants only in very low amounts. Elicitors such as hormones （e.g. jasmonates or salicylic acid） activate plant natural defense responses, includ-ing increased secondary metabolite production （EI-Sayed and Verpoorte, 2007; Lackman et al.. 2011）.

The milestones in continuous casting were set by the SMS group and its predecessor companies（Schloemann, Concast and Mannesmann Demag） and testify to 50 years of leadership in continuous casting. The thin, medium and thick slabs produced on continuous casters from SMS Siemag form the basis for superior steel products. Today, approx. 95% of steel is produced by means of continuous casting. Being a frontrunner in this technology, SMS Siemag today offers the world’s most extensive portfolio of continuous casting machines meeting the respective demands of the market as well as the customers’ requirements. The great numbers of continuous casters which we have already successfully commissioned, or will be commissioning are eloquent proof of this successful and trustworthy cooperation with our customers. Continuous new developments are characteristic for the history of SMS Siemag. Interdisciplinary cooperation within knowledge networks, with a bearing on the tasks in hand, leads to the necessary innovations in plant engineering, automation and process technology and thereby to the solutions demanded by our customers. In this paper we are presenting several references which show that our customers manufacture high-quality products in their plants. We are also reporting some new developed technologies increasing production, reducing production costs, enhancing quality and saving energy.

22-gauge core vs 22-gauge aspiration needle for endoscopic ultrasound-guided sampling of abdominal masses

AIM To compare the aspiration needle(AN) and core biopsy needle(PC) in endoscopic ultrasound-guided fine needle aspiration(EUS-FNA) of abdominal *** Consecutive patients referred for EUS-FNA were included in this prospective single-center trial. Each patient underwent a puncture of the lesion with both standard 22-gauge(G) AN(Echo Tip Ultra; Cook Medical, Bloomington, Indiana, United States) and the novel 22 G PC(Echo Tip Pro Core; Cook Medical, Bloomington, Indiana, United States) in a randomized fashion; histology was attempted in the PC group only. The main study endpoint was the overall diagnostic accuracy, including the contribution of histology to the final diagnosis. Secondary outcome measures included material adequacy, number of needle passes, and *** Fifty six consecutive patients(29 men; mean age 68 years) with pancreatic lesions(n = 38), lymphadenopathy(n = 13), submucosal tumors(n = 4), or others lesions(n = 1) underwent EUS-FNA using both of the needles in a randomized order. AN and PC reached similar overall results for diagnostic accuracy(AN: 88.9 vs PC: 96.1, P = 0.25), specimen adequacy(AN: 96.4% vs PC: 91.1%, P = 0.38), mean number of passes(AN: 1.5 vs PC: 1.7, P = 0.14), mean cellularity score(AN: 1.7 vs PC: 1.1, P = 0.058), and complications(none). A diagnosis on the basis of histology was achieved in the PC group in 36(64.3%) patients, and in 2 of those as the sole modality. In patients with available histology the mean cellularity score was higher for AN(AN: 1.7 vs PC: 1.0, P = 0.034); no other differences were of statistical *** Both needles achieved high overall diagnostic yields and similar performance characteristics for cytological diagnosis; histological analysis was only possible in 2/3 of cases with the new needle.

结构牢固的集成变频电机曼内斯曼德马泰克将变频器同齿轮电机结合成一体给用户带来更大的利益。首先变频器安装于齿轮电机内大大减少了安装费用,其次,整个系统所需的只是供电和数据线,其结构极为紧凑,尺寸为80～90mm,输出功率为1.9kW,可适用于高达40℃的环境温度。这种装置通过总线系统控制,为IP54保护。除了“Interbus—S”传感器/换向器总线之外,曼内斯曼德马泰克还将提供现场总线系统,如“Profibus”,通过配备一个小型备选装置,就可通过控制开关或者模拟定点值来控制驱动器。电机与变频器相结合是真正的四象限装置,用于开始与完成操作。其应用范围可扩展至380V至500V,主频率从50Hz至60Hz。此种连接的研制成功为世界市场开辟了良好的前景。此外将变频器用于齿轮电机还可防止电磁干扰。集成变频电机安装费用特别小,这就使它能以较大的优势进入目前还主要使用变极控制式电机和机械驱动装置的应用领域。
