Compression loading of osteoclasts attenuated micro RNA-146a-5p expression,which promotes angiogenesis by targeting adiponectin

Osteoclastogenesis in alveolar bone induced by compression stress triggers orthodontic tooth movement.Compression stress also stimulates angiogenesis,which is essential for osteoclastogenesis.However,the effects of osteoclastogenesis induced by compression on angiogenesis are poorly understood.In vivo,we found the markers of angiogenesis increased during orthodontic bone remodeling.In vitro,osteoclast-derived exosomes increased proliferation,migration,and tube formation of human umbilical vein endothelial cells(HUVECs),as well as expression of vascular endothelial growth factor and CD31.The promotive effects of exosomes derived from compressed osteoclasts were greater than those derived from osteoclasts without compression.Next,we analyzed changes in the micro RNA transcriptome after compression stress and focused on micro RNA146 a-5 p(mi R-146 a),which was significantly decreased by compression.Transfection of an inhibitor of mi R-146 a stimulated angiogenesis of HUVECs while mi R-146 a mimics repressed angiogenesis.Adiponectin(ADP)was confirmed to be a target of mi R-146 a by dual luciferase reporter assay.In HUVECs treated with exosomes,we detected increased ADP which promoted angiogenesis.Knockdown of ADP in HUVECs reduced the promotive effects of exosomes.Our results demonstrate that the decreased mi R-146 a observed in osteoclasts after compression promotes angiogenesis by targeting ADP,suggesting a novel method to interfere with bone remodeling induced by compression stress.

Placenta accreta spectrum(PAS)disorders are severe obstetric complications and can cause life-threatening hemorrhage,hysterectomy,or even death.PAS disorders in one of dichorionic diamniotic twins are rare to be seen in clinical practice and lack of standard management strategies.We reported a case of retained placenta creta after selective fetal reduction in one of dichorionic diamniotic twins.A 32-year-old woman with a history of cesarean section was diagnosed with placenta increta in one of dichorionic diamniotic twins at 16 weeks of gestation.After discussion,the couple required to be hospitalized for selective fetal reduction.She received selective termination by ultrasound-guided single fetal intracardiac injection of potassium chloride.The follow-up prenatal ultrasound examinations detected the placenta of the reduced fetus did not stop invade the myometrium of the uterus but developed from placenta increta to placenta percreta.The patient was diagnosed with placental percreta by prenatal ultrasound signs and surgical findings.The patient underwent scheduled cesarean delivery at 34^(+5)gestational weeks.The outcome of maternal and fetal was satisfactory for our elaborate operation and optimal management.PAS disorders are severe obstetric complications with an increasing incidence.Combined with twin pregnancy makes PAS disorders more intractable.We should learn that selective fetal reduction probably cannot solve it thoroughly.Intensive and standard management is essential to promote maternal and fetal outcomes.

Exosomes derived from osteoclasts under compression stress inhibit osteoblast differentiation

Orthodontic tooth movement is triggered by orthodontic force loading on the periodontal ligament and is achieved by alveolar bone remodeling,which is regulated by intimate crosstalk between osteoclastogenesis and osteoblast differentiation.Whether the communication between osteoclasts and osteoblasts is influenced by orthodontic compression stress requires further clarification.In this study,osteoclasts were differentiated for 10 days.On day 4 of differentiation,the number of pre-osteoclasts peaked,as determined by the increased expression of RANK and the number of multinucleated cells.After 24 h of compression stress loading,on day 4,the number of osteoclasts increased,and the optimal magnitude of stress to promote osteoclastogenesis was determined as 1 g/cm2.Moreover,the results of RNA-sequencing analysis showed that the miRNA expression profile changed markedly after compression loading and that many of the altered miRNAs were associated with cell communication functions.A series of indirect co-culture experiments showed an inhibitory effect of osteoclasts on osteoblast differentiation,especially after compression.Next,we added osteoclast-derived exosomes to hPDLSCs during osteoblast differentiation.Exosomes derived from osteoclasts under compression(cEXO)showed a greater inhibitory effect on osteoblast differentiation,compared to exosomes from osteoclasts without compression(EXO).Therefore,we analyzed differentially expressed miRNAs associated with bone development functions in exosomes:miR-223-5p and miR-181a-5p were downregulated,whereas miR-133a-3p,miR-203a-3p,miR-106a-5p,and miR-331-3p were upregulated;these altered expressions may explain the enhanced inhibitory effect of compression stress.

Diagnosis of placenta accreta spectrum(PAS)Before 2018,there were various terms used to describe PAS disorders,such as morbidly adherent placenta(US/Egypt/Israel),adhesive placenta(Italy/Argentina),and pernicious placenta(Chinese mainland).An inclusive standardized terminology PAS was proposed by the International Federation of Gynecology and Obstetrics(FIGO)in March 2018 and endorsed by the Society for Maternal-Fetal Medicine and American College of Obstetricians and Gynecologists,Royal College of Obstetricians and Gynecologists,and the Society of Obstetricians and Gynecologists of Canada.1 It describes the grade of PAS which are separated into three categories:placenta adherenta or creta(PC)when the villi adhere directly to the myometrium without a decidual interface,placenta increta(PI)when the villi invade the myometrium and placenta percreta(PP)when the villi invade the full thickness of the uterine wall including the serosa.

Background:Massive bleeding is the main concern for the management of placenta percreta(PP).Intra-abdominal aortic balloon occlusion(IABO)is one method for pelvic devascularization,but the efficacy of IABO is uncertain.This study aims to investigate the outcomes of IABO in PP patients.Methods:We retrospectively reviewed the clinical data of PP cases from six tertiary centers in China between January 2011 and December 2015.PP cases with/without the use of IABO were analyzed.Propensity score matching analysis was performed to reduce the effect of selection bias.Postpartum hemorrhage(PPH)and the rate of hysterectomy,as well as neonatal outcomes,were analyzed.Results:One hundred and thirty-two matched pairs of patients were included in the final analysis.Compared with the control group,maternal outcomes,including PPH(68.9%vs.87.9%,χ^(2)=13.984,P<0.001),hysterectomy(8.3%vs.65.2%,χ^(2)=91.672,P<0.001),and repeated surgery(1.5%vs.12.1%,χ^(2)=11.686,P=0.001)were significantly reduced in the IABO group.For neonatal outcomes,Apgar scores at 1 minute(8.67±1.79vs.8.53±1.68,t=-0.638,P=0.947)and 5 minutes(9.43±1.55vs.9.53±1.26,t=0.566,P=0.293)were not significantly different between the two groups.Conclusions:IABO can significantly reduce blood loss,hysterectomies,and repeated surgeries.This procedure has not shown harmful effects on neonatal outcomes.

Genetic information can be critical in identifying conservation priorities and developing conservation strategies.There is an urgent need for noninvasive genetic tools to study the wild populations of Asian colobine monkeys.The majority of these species are threatened with habitat destruction,population reduction and even extinction,but generally lack information on their genetic diversity and population structure.Genetic sampling and tissue collection have been scarce in these species owing to strict regulations on manipulation of endangered species,and the difficulties and risks associated with capturing these arboreal and fast-moving monkeys in the challenging environments that they inhabit.These difficulties have hindered the development of molecular genetic markers,which are usually derived from tissues or blood.In this study,we present a method for de novo microsatellite isolation and genotyping using DNA from noninvasive origins of a critically endangered Asian colobine,the white-headed langur(Trachypithecus leucocephalus).Genomic DNA isolated from hair was shown to be sufficient for microsatellite enrichment and isolation,with similar isolation efficiencies as from tissue DNA.We identified and characterized 20 polymorphic microsatellite loci,and evaluated their amplification success and genotyping reliability with 86 field-collected fecal samples.These results show that this panel of loci can produce reliable genotypes from fecal samples,and represent a useful tool for noninvasive investigation of genetic structure,individual identification and kinship assessment in this highly endangered species.Our approach can be applied to conservation genetic studies of other wild species that lack sequence information and tissue samples.

Background:Single-cell RNA sequencing(scRNA-seq)technology is now becoming a widely applied method of transcriptome exploration that helps to reveal cell-type composition as well as cell-state heterogeneity for specific biological processes.Distinct sequencing platforms and processing pipelines may contribute to various results even for the same sequencing samples.Therefore,benchmarking sequencing platforms and processing pipelines was considered as a necessary step to interpret scRNA-seq data.However,recent comparing efforts were constrained in sequencing platforms or analyzing pipelines.There is still a lack of knowledge of analyzing pipelines matched with specific sequencing platforms in aspects of sensitivity,precision,and so on.Methods:We downloaded public scRNA-seq data that was generated by two distinct sequencers,NovaSeq 6000 and MGISEQ 2000.Then data was processed through the Drop-seq-tools,UMI-tools and Cell Ranger pipeline respectively.We calculated multiple measurements based on the expression profiles of the six platform-pipeline combinations.Results:We found that all three pipelines had comparable performance,the Cell Ranger pipeline achieved the best performance in precision while UMI-tools prevailed in terms of sensitivity and marker calling.Conclusions:Our work provided an insight into the selection of scRNA-seq data processing tools for two sequencing platforms as well as a framework to evaluate platform-pipeline combinations.

Histone H2A monoubiquitination is associated with transcriptional repression and needs to be removed by deubiquitinases to facilitate gene transcription in eukaryotes.However,the deubiquitinase responsible for genome-wide H2A deubiquitination in plants has yet to be identified.In this study,we found that the previously identified PWWP-EPCR-ARID-TRB(PEAT)complex components interact with both the ubiquitin-specific protease UBP5 and the redundant histone acetyltransferases HAM1 and HAM2(HAM1/2)to form a larger version of PEAT complex in Arabidopsis thaliana.UBP5 functions as an H2A deubiquitinase in a nucleosome substrate-dependent manner in vitro and mediates H2A deubiquitination at the whole-genome level in vivo.HAM1/2 are shared subunits of the PEAT complex and the conserved NuA4 histone acetyltransferase com-plex,and are responsible for histone H4K5 acetylation.Within the PEAT complex,the PWWP components(PWWP1,PWWP2,and PWWP3)directly interact with UBP5 and are necessary for UBP5-mediated H2A deu-biquitination,while the EPCR components(EPCR1 and EPCR2)directly interact with HAM1/2 and are required for HAM1/2-mediated H4K5acetylation.Collectively,our study not onlyidentifies dual roles of thePEAT com-plex in H2A deubiquitination and H4K5 acetylation but also illustrates how these processes collaborate at the whole-genome level to regulate the transcription and development in plants.
