Bacillus velezensis JK-XZ8 prevents and controls crown gall disease on Prunus subhirtella by colonizing and inducing resistance

Crown gall of Prunus subhirtella Miq., flowering cherry, is a soil-borne bacterial disease caused by Agrobacterium tumefaciens Smith & Townsend and has caused serious damage to the species in recent years in China. In this study, the colonization of sapling roots, the biocontrol efficacy, the expression of defense-related genes and the enzyme activity in roots of P. subhirtella inoculated with the Bacillus velezensis JK-XZ8 against A. tumefaciens were determined under greenhouse conditions. The results showed that the JK-XZ8 strain colonized in the rhizosphere and root surfaces of the saplings. Sole application of the strain increased activities of polyphenol oxidase (PPO) and phenylalanine ammonia lyase (PAL) without affecting superoxide dismutase (SOD). SOD and PPO activities increased in the prevention group, and the activity of PAL increased in both the prevention and cure groups. The expression of the defense-related genes PAL and PR-1 of salicylic acid pathway in roots increased in both the prevention and cure groups. Simultaneously on day 3, the expression of both COI and MYC2 genes of the jasmonic acid pathway significantly increased in the two groups. The incidence of crown gall in the prevention and the cure groups were 48.1% and 66.7%, respectively, significantly lower compared to the pathogens alone (87.5%). This study showed that the application of the JK-XZ8 strain reduced the incidence of crown gall in P. subhirtella saplings in the two groups, and the prevention group had better control efficacy. In addition, the JK-XZ8 strain protects against crown gall by developing induced systemic resistance and systemic acquired resistance in the roots of the cherry saplings.

Combining ectomycorrhizal fungi and plant growth-promoting rhizobacteria to enhance salt tolerance of Metasequoia glyptostroboides

Plant growth and productivity are negatively affected by soil salinity.This study investigated the effects of the rhizosphere-promoting bacterium,Bacillus paramycoides JYZ-SD5,and the ectomycorrhizal fungus,Schizophyllum commune Be,on the growth of Metasequoia glyptostroboides under salt stress.Changes in biomass,root growth,root ion distribution and in vivo enzyme activities were determined under different treatments(Be,JYZ-SD5,and Be+JYZ-SD5).The results show that all inoculations increased chlorophyll content,shoot length and root diameter with or without salt stress,and the effect of Be+JYZ-SD5was the strongest.JYZ-SD5 and Be+JYZ-SD5 treatments significantly increased root length,surface area,bifurcation number,tip number,main root length and diameter under salt stress.Normal chloroplast structures developed under both single and double inoculations.Relative to the control,root activities of M.glyptostroboides in the Be,JYZSD5,and Be+JYZ-SD5 treatments increased by 31.3%,17.2%,and 33.7%.All treatments increased the activities of superoxide dismutase(SOD),peroxidase(POD),Na^(+)-K^(+)-ATPase and Ca^(2+)-Mg^(2+)-ATPase.The strongest effect was by Be+JYZ-SD5.Analysis of root ion distribution showed that,under salt stress,Na^(+)and K^(+)decreased and were concentrated in the epidermis or cortex.Na/K ratios also decreased.The Be+JYZ-SD5 treatment increased betaine by 130.3%and 97.9%under 50 mM and 100 mM salt stress,respectively.Together,these changes result in the activation of physiological and biochemical processes involved in the mitigation of salinity-induced stress in M.glyptostroboides.

Geometrical and electrical optimization of NS-SDBD streamwise plasma heat knife for aircraft anti-icing

Ice accretion on aircraft encountering supercooled water droplets in clouds poses great risks to flight performance and safety.With the aim of optimizing the newly developed streamwise plasma heat knife method for anti-icing,a parametric investigation is carried out in this work.The influence of the detailed voltage profile on the heating effects of a Surface Dielectric Barrier Discharge driven by Nanosecond Pulses(NS-SDBD)is investigated,and a comparison of the antiicing performance among different configurations of streamwise plasma heat knife is made.The results show that columnar high-temperature regions produced by a multi-streamer discharge appear at small pulse rise time,but become diffuse as the pulse rise time increases.An optimal pulse rise time exists to provide a wide range and high value of temperature,which is found to be 150 ns for the setup in the present study.The influence of the pulse fall time is much weaker than that of the rise time.The range and value of the temperature decrease with increasing pulse fall time.A greater pulse width is found to improve the heating effect by increasing the discharge power.When a spanwise electrode is placed connecting the streamwise electrodes of the streamwise plasma heat knife at the airfoil leading edge,the anti-icing performance becomes poorer,whereas good performance is achieved when the spanwise electrode is at the edge of the streamwise electrodes.Based on this,a three-level configuration of the plasma heat knife is proposed,and its anti-icing performance is found to be much better than that of the original configuration.

Minimizing airfoil drag at low angles of attack with DBD-based turbulent drag reduction methods

Dielectric Barrier Discharge(DBD)based turbulent drag reduction methods are used to reduce the total drag on a NACA 0012 airfoil at low angels of attack.The interaction of DBD with turbulent boundary layer was investigated,based on which the drag reduction experiments were conducted.The results show that unidirectional steady discharge is more effective than oscillating discharge in terms of drag reduction,while steady impinging discharge fails to finish the mission(i.***.drag increase).In the best scenario,a maximum relative drag reduction as high as 64%is achieved at the freestream velocity of 5 m/s,and a drag reduction of 13.7%keeps existing at the freestream velocity of 20 m/s.For unidirectional discharge,the jet velocity ratio and the dimensionless actuator spacing are the two key parameters affecting the effectiveness.The drag reduction magnitude varies inversely with the dimensionless spacing,and a threshold value of the dimensionless actuator spac-ing of 540(approximately five times of the low-speed streak spacing)exists,above which the drag increases.When the jet velocity ratio smaller than 0.05,marginal drag variation is observed.In con-trast,when the jet velocity ratio larger than 0.05,the experimental data bifurcates,one into the drag increase zone and the other into the drag reduction zone,depending on the value of dimensionless actuator spacing.In both zones,the drag variation magnitude increases with the jet velocity ratio.The total drag reduction can be divided into the reduction in pressure drag and turbulent friction drag,as well as the increase in friction drag brought by transition promotion.The reduction in tur-bulent friction drag plays an important role in the total drag reduction.

建筑设计参数对建筑运营过程中的能耗和成本有着重要的影响,而新疆冬季严寒且漫长,使得该地区建筑能耗及运营费用高于其他地区。因此,本文利用离差最大化法结合正交试验设计,以某教学建筑为例,考察了在新疆4个热工分区的典型城市气象条件下,包括中庭面积比、天窗面积比、不同立面的窗墙比、外围护保温层厚度、外窗类型等8个设计参数对建筑能耗和全局成本的影响,并通过综合考虑建筑能耗和全局成本目标,确定最佳的建筑设计方案。研究结果表明,在4个典型城市气象条件下,最佳设计方案可以实现建筑节能10.47%~13.48%,全局成本降低32.62~62.91元/m^(2)。研究有助于设计者选择适用于该地区的最佳设计参数,从而实现建筑的节能目标。

【目的】随着非常规油气理论的发展,沉积有机质富集机理再次成为热点研究,其中有机质富集主控因素成为争论的焦点。以往的研究对沉积过程的分析较少,这可能是存在争论的原因之一。【方法】结合沉积学与地球化学研究方法,在分析层序地层学和沉积环境的基础上,详细开展元素地球化学研究,讨论富有机岩沉积有机质富集机理。【结果】四川盆地北部和鄂西盆地上二叠统吴家坪组自下而上分为吴一段、吴二段和吴三段,大隆组自下而上分为大一段和大二段。吴家坪组至大隆组自下而上总共分为五个三级层序SQ1~SQ5。由层序地层学分析将广利海槽和鄂西海槽的发展分为四个阶段,分别为初始、快速发展、高潮稳定和萎缩阶段。初始阶段主要发育SQ2时期斜坡相暗色硅质灰岩,快速发展阶段主要发育SQ3时期陆棚相钙质页岩,高潮稳定阶段主要发育SQ4时期盆地相黑色硅质岩,萎缩阶段主要发育SQ5时期斜坡相石灰岩。含异常高有机质的甜点段主要形成于SQ3高位域至SQ5海侵域沉积期,在该时期Fe、Cu、Ni和Zn营养元素周期性富集,Mo、V、Fe/Al和S等氧化还原敏感元素均表现为较高的水平,热液强度参数Al/(Al+Fe+Mn)和Al-Fe-Mn三角图均指示频繁的热液活动特征。【结论】在甜点段,高营养元素含量和初级生产力水平的出现早于强还原条件,说明沉积水体的氧化还原条件主要受控于沉积有机质沉降埋藏过程中对氧的消耗,有机质的富集主要受较高初级生产力的控制。营养物质P、Si、Fe、Zn的供应主要与裂陷槽形成过程中深部热液活动和周围火山活动有关,这些构造活动周期性带来大量营养物质提高了初级生产力水平。因此海洋表层高的初级生产力水平是甜点段异常高有机质富集的启动和基础条件,而海洋底部水体较强的还原环境是有机质埋藏和保存的关键因素。

近年来,中国页岩气聚焦“三新”(尚未开展工作或工作程度低的新区带、新层系及新类型)领域,加大理论攻关与勘探力度,页岩气增储上产再上新台阶。基于中国页岩气“三新”领域理论与勘探新进展,分析其勘探特点、变化趋势和发展前景,探索中国特殊页岩气地质理论,明确页岩气发展挑战与对策。结果表明:中国在四川盆地及邻区创新建立了五峰组-龙马溪组特色的高-过成熟度海相页岩气富集理论,累计发现页岩气田9个,探明页岩气地质储量近3×10^(12)m^(3),形成了450×10^(8)m^(3)/a的页岩气产能,年产页岩气250×10^(8)m^(3)。明确了近年来中国页岩气“三新”领域勘探具三大特点:①四川盆地及邻区五峰组-龙马溪组在深层、极浅层勘探取得重大新发展;②四川盆地筇竹寺组和吴家坪组等多个新层系实现新突破;③鄂尔多斯盆地西缘乌拉力克组获得新发现,东缘山西组取得新突破。页岩气“三新”领域的勘探实现了三大战略性转变:①目标由单一类型、单一盆地和单一层系转变为多类型、多盆地和多层系;②选区由盆内或盆缘为主转变为盆内、盆缘兼顾盆外弱改造区;③思路由纯粹富有机质页岩转变为富有机质页岩系统。通过梳理中国页岩气“三新”领域勘探挑战,指出“三新”领域是中国页岩气可持续发展的方向,并提出了下一步发展对策。

冻融循环作用下纤维混凝土动态抗压性能

为研究冻区混凝土动态抗压性能,对不同掺料的混凝土试件进行了先冻融循环后单轴动态抗压试验。研究了三组混凝土(PVA纤维、纳米SiO_(2)和PVA-纳米SiO_(2))在不同应变速率(10-5s^(-1)、10-4s^(-1)和10^(-3)s^(-1))下的强度变化率、应变损失率,分析得到三种混凝土动态抗压性能的优劣关系。通过试件切片的SEM电镜扫描试验,研究了掺料改善混凝土性能的微观作用机理。研究结果表明:单掺PVA纤维或纳米SiO2(P组、S组),对于混凝土受冻后的动态抗压性能无明显改善。双掺(SP组)时,纳米SiO2与水化产物Ca(OH)2反应生成的C-S-H凝胶填充了混凝土的孔隙和ITZ,增强了混凝土的密实性,为受冻后混凝土中PVA纤维更完整地发挥桥连作用提供了条件。冻融循环100次后的SP组混凝土在应变速率从10^(-4)s^(-1)到10^(-3)s^(-1)的情况下,其相较于P组、S组的强度提高约10%,PVA纤维和纳米SiO2的协同作用使混凝土受冻后的动态抗压性能改善明显。

对学习者自我调节学习过程的准确评价,是实现教学干预的必要前提。然而,已有评价方法多为总结性评价,实时性不足。为此,本研究提出了数据驱动的自我调节学习动态评价模型。以学习任务为单位对学习过程进行时序化处理,并构建学习过程中生成性数据到自我调节学习状态的映射关系。研究结果表明:(1)生成性数据是评价学习者自我调节学习状态的有力因素,模型具有较高的有效性。(2)学习者的自我调节学习状态在不同评价维度呈现出差异性变化趋势,其中任务分析、自我激励的信念和自我观察维度趋于稳定,而自我控制、自我判断和自我反应3个维度呈现明显波动性变化。(3)高低绩效群体在自我激励的信念、自我控制和自我反应3个评价维度上表现出显著差异,而在任务分析、自我观察和自我判断有3个维度上没有统计学差异。
