Repairing of rutin to the toxicity of combined F-53B and chromium pollution on the biofilm formed by Pseudomonas aeruginosa

The wastewater discharge from the process of chrome plating,which contains 6:2 chlorinated polyfluorinated ether sulfonate(F-53B)and chromium(Cr),may be toxic to biofilm.In this study we found that the biofilm formed by Pseudomonas aeruginosa PAO1 was inhibited by exposure to a combination of F-53B and Cr(VI).The combined pollution damaged the cell membranes and the structure of the biofilm,and inhibited the production of the Pseudomonas quinolone-based signal,which affected biofilm formation.Moreover,the secretion of extracellular polymeric substances decreased as a result of this combined exposure.Exposure to F-53B and Cr(VI)individually or in combination could induce the excessive accumulation of intracellular reactive oxygen species(ROS),and the ROS positive rate of the bacteria increased under the treatment with 0.2 mmol/L of Cr(VI)and 250 nmol/L of F-53B,respectively.In addition,the activities of superoxide dismutase(SOD)and catalase(CAT)were enhanced for scavenging ROS in the bacteria that were exposed to Cr(VI)and F-53B.As an antioxidant,rutin was used to repair the toxicity of Cr(VI)and F-53B towards the biofilm formed by the bacteria.When rutin was added to the bacteria medium,with either Cr(VI)or F-53B as pollutant,or with the combined pollutants,the extracellular protein content of the bacteria recovered to 0.84,0.94,and 0.85 times that of the control,respectively.Meanwhile,the accumulation of ROS and the activities of SOD and CAT decreased,which indicated that the addition of rutin can alleviate the oxidative stress and promote the antioxidant stress system.

Metal-free photocatalysts have attracted growing concern recently.Herein,the composites combining g-C_(3)N_(4)with wood pulp cellulose biochar(WPBC/g-C_(3)N_(4))were synthesized to effectively activate peroxymonosulfate(PMS)under visible light for the degradation of diclofenac(DCF).The incorporation of WPBC endowed g-C_(3)N_(4)with enhanced visible light absorption,improved charge separation capability,reduced electrical conductivity,and increased photocatalytic and PMS activation capability.Based on quenching tests,electron paramagnetic resonance(EPR),electrochemical analysis and solvent exchange experiments,both radical and nonradical mechanisms were proposed.Radical species including·OH,h^(+),·O_(2)^(-)were identified to contribute to DCF degradation.The 1O2 and electron transfer were the dominant nonradical pathways for DCF degradation.Moreover,the common influencing factors were examined,and DCF concentration was the most influential factor based on principal component analysis.Generally,the composites exhibited good reusability during consecutive runs.Based on HPLC/MS analysis,four intermediates were detected and the possible DCF degradation pathway was proposed.This work provided a potential strategy based on metal-free WPBC/g-C_(3)N_(4)for the photocatalytic activation of PMS to effectively degrade emerging contaminants in wastewater.

For the treatment of MRSA-infected wounds,the spatiotemporally sequential delivery of antibacterial and anti-inflammatory drugs is a promising strategy.In this study,ROS-responsive HA-PBA/PVA(HPA)hydrogel was prepared by phenylborate ester bond cross-linking between hyaluronic acid-grafted 3-amino phenylboronic acid(HA-PBA)and polyvinyl alcohol(PVA)to achieve spatiotemporally controlled release of two kinds of drug to treat MRSA-infected wound.The hydrophilic antibiotic moxifloxacin(M)was directly loaded in the hydrogel.And hydrophobic curcumin(Cur)with anti-inflammatory function was first mixed with Pluronic F127(PF)to form Cur-encapsulated PF micelles(Cur-PF),and then loaded into the HPA hydrogel.Due to the different hydrophilic and hydrophobic nature of moxifloxacin and Cur and their different existing forms in the HPA hydrogel,the final HPA/M&Cur-PF hydrogel can achieve different spatiotemporally sequential delivery of the two drugs.In addition,the swelling,degradation,self-healing,antibacterial,anti-inflammatory,antioxidant property,and biocompatibility of hydrogels were tested.Finally,in the MRSA-infected mouse skin wound,the hydrogel-treated group showed faster wound closure,less inflammation and more collagen deposition.Immunofluorescence experiments further confirmed that the hydrogel promoted better repair by reducing inflammation(TNF-a)and promoting vascular(VEGF)regeneration.In conclusion,this HPA/M&CurPF hydrogel that can spatiotemporally sequential deliver antibacterial and anti-inflammatory drugs showed great potential for the repair of MRSA-infected skin wounds.

A visible-light-assisted Pd/TiO_(2)gas sensor with carbon nanotubes electrodes for trace formaldehyde detection

Owing to the ppb-level detection standard toward the toxic and harmful gas,the detection of trace gases has become an important subject in the field of indoor environment management.However,the traditional resistive gas sensors hardly meet the requirement due to the weak signal generated by trace gas molecules that are difficult to capture.Herein,a visible-light-assisted Pd/TiO_(2)gas sensor is proposed to endow the effective detection of trace formaldehyde(HCHO)gas without heating temperature.Benefiting from the enhanced photocatalytic properties of TiO_(2)by Pd decoration,the visible-light-assisted Pd/TiO_(2)gas sensor can detect the HCHO gas as low as80×10^(–9)at room temperature.The successful preparation of nanoscale TiO_(2)sensing layer is facilitated by the ultrathin carbon nanotube interdigital electrode in the gas sensor,which avoids the discontinuity of the sensing layer caused by the excessive thickness of the traditional metal electrode.In addition,the whole preparation process of the Pd/TiO_(2)gas sensor with carbon nanotube electrodes is compatible with mainstream CMOS fabrication technology,which is expected to realize the batch fabrication and micro-integrated application of gas sensors.It is expected that our work can provide a new strategy for the batch preparation of high-performance trace HCHO gas sensors and their future applications in portable electronic devices such as smartphones.

Toxic effect of perfluorooctane sulfonate on plants in vertical-flow constructed wetlands

Per-and polyfluoroalkyl substances(PFASs)can be taken up and bioaccumulated in plants,but the toxic mechanisms of PFASs on wetland plants are still unclear.In present study,the toxic influences of perfluorooctane sulfonate(PFOS)on Eichhornia crassipes(E.crassipes)and Cy perus alternifolius(C.alternifolius)in a vertical-sub surface-flow constructed wetland were evaluated.The results showed that E.crassipes was more tolerant to PFOS stress than C.alternifolius,and the growth and chlorophyll synthesis of the two plants were promoted by low concentration(<0.1 mg/L)of PFOS,and the chlorophyll synthesis was inhibited by high concentration(10 mg/L)of PFOS but the growth did not change obviously.The catalase activity and malondialdehyde content in the leaves of the two plants increased,peroxidase activity decreased under exposure to high concentrations of PFOS,and superoxide dismutase activity did not change.Under PFOS stress,the membrane of plant leaves and the cell structure of the two wetland plants were destroyed,and the mitochondrial contour of root cells became incomplete.Tanscriptomic analysis showed that the expression levels of genes related to cell wall formation,the cell apoptosis pathway,material synthesis,and metabolism in the plants were changed by PFOS.Analysis in fluorogenic quantitative real time polymerase chain reaction(RT-qPCR)also confirmed that the photosynthesis system of E.crassipes was inhibited,while that of C.alternifolius was promoted.

Potential role of hippocampal neurogenesis in spinal cord injury induced post-trauma depression

It has been reported both in clinic and rodent models that beyond spinal cord injury directly induced symptoms, such as paralysis, neuropathic pain, bladder/bowel dysfunction, and loss of sexual function, there are a variety of secondary complications, including memory loss, cognitive decline, depression, and Alzheimer's disease. The largescale longitudinal population-based studies indicate that post-trauma depression is highly prevalent in spinal cord injury patients. Yet, few basic studies have been conducted to address the potential molecular mechanisms. One of possible factors underlying the depression is the reduction of adult hippocampal neurogenesis which may come from less physical activity, social isolation, chronic pain, and elevated neuroinflammation after spinal cord injury. However, there is no clear consensus yet. In this review, we will first summarize the alteration of hippocampal neurogenesis post-spinal cord injury. Then, we will discuss possible mechanisms underlie this important spinal cord injury consequence. Finally, we will outline the potential therapeutic options aimed at enhancing hippocampal neurogenesis to ameliorate depression.

Cryptic diversity and rampant hybridization in annual gentians on the Qinghai-Tibet Plateau revealed by population genomic analysis

Understanding the evolutionary and ecological processes involved in population differentiation and speciation provides critical insights into biodiversity formation. In this study, we employed 29,865 single nucleotide polymorphisms(SNPs) and complete plastomes to examine genomic divergence and hybridization in Gentiana aristata, which is endemic to the Qinghai-Tibet Plateau(QTP) region. Genetic clustering revealed that G. aristata is characterized by geographic genetic structures with five clusters(West, East, Central, South and North). The West cluster has a specific morphological character(i.e., blue corolla) and higher values of FSTcompared to the remaining clusters, likely the result of the geological barrier formed by the Yangtze River. The West cluster diverged from the other clusters in the Early Pliocene;these remaining clusters diverged from one another in the Early Quaternary. Phylogenetic reconstructions based on SNPs and plastid data revealed substantial cyto-nuclear conflicts. Genetic clustering and D-statistics demonstrated rampant hybridization between the Central and North clusters,along the Bayankala Mountains, which form the geological barrier between the Central and North clusters. Species distribution modeling demonstrated the range of G. aristata expanded since the Last Interglacial period. Our findings provide genetic and morphological evidence of cryptic diversity in G. aristata, and identified rampant hybridization between genetic clusters along a geological barrier.These findings suggest that geological barriers and climatic fluctuations have an important role in triggering diversification as well as hybridization, indicating that cryptic diversity and hybridization are essential factors in biodiversity formation within the QTP region.

Unnatural amino acids(UAAs)have gained significant attention in protein engineering and drug development owing to their ability to introduce new chemical functionalities to proteins.In eukaryotes,genetic code expansion(GCE)enables the incorporation of UAAs and facilitates posttranscriptional modification(PTM),which is not feasible in prokaryotic systems.GCE is also a powerful tool for cell or animal imaging,the monitoring of protein interactions in target cells,drug development,and switch regulation.Therefore,there is keen interest in utilizing GCE in eukaryotic systems.This review provides an overview of the application of GCE in eukaryotic systems and discusses current challenges that need to be addressed.

The spectrum and stability of travelling pulses in a coupled FitzHugh-Nagumo equation

For a coupled slow-fast FitzHugh-Nagumo(FHN)equation derived from a reaction-diffusionmechanics(RDM)model,Holzer et al.(2013)studied the existence and stability of the travelling pulse,which consists of two fast orbit arcs and two slow ones,where one fast segment passes the unique fold point with algebraic decreasing and two slow ones follow normally hyperbolic critical curve segments.Shen and Zhang(2021)obtained the existence of the travelling pulse,whose two fast orbit arcs both exponentially decrease,and one of the slow orbit arcs could be normally hyperbolic or not at the origin.Here,we characterize both the nonlinear and spectral stability of this travelling pulse.
