Tianxiangdan(TXD) alleviates myocardial ischemia reperfusion-induced ferroptosis through the activation of estrogen receptor alpha(ERα)

Tianxiangdan（TXD）, a traditional Chinese herbal remedy, demonstrates efficacy in mitigating myocardial ischemia-reperfusion（I/R）-induced damage. This study employed network pharmacology to evaluate the therapeutic targets and mechanisms of TXD in treating I/R. Highperformance liquid chromatography-mass spectrometry（HPLC-MS） identified 86 compounds in TXD. Network pharmacological analysis predicted potential target genes and their modes of action. Cardiac function, ischaemic ST changes, lactate dehydrogenase（LDH）,malondialdehyde（MDA）, superoxide dismutase（SOD） activity, myocardial fiber, and infarct size were assessed using in vivo and in vitro I/R injury models. Estrogen receptor alpha（ERα）protein expression and estradiol（E2） levels were measured to confirm TXD's impact on estrogen levels and ERα expression. To examine if TXD reduces I/R injury through ERα, an AZD group(300 nmol·L-1AZD9496 and 15% TXD serum) was compared to a TXD group（15% TXD serum）. The study hypothesized that TXD upregulates the ERα-mediated iron metamorphosis pathway. I/R injury-induced ferroptosis was identified using a Fer-1 group(1.0 μmol·L-1Fer-1 and 15% TXD serum) to elucidate the potential association between ferroptosis and ERαproteins. A DCFH-DA probe detected reactive oxygen species（ROS） and Fe2+, while Western blotting assessed target protein expression. Both in vitro and in vivo experiments demonstrated that TXD attenuated I/R injury by reducing elevated ST-segment levels, improving cardiac injury biomarkers（LDH, MDA, and SOD）, alleviating pathological features, and preventing I/R-induced loss of cell viability in vitro. The effects and mechanisms of TXD on I/R injury-associated ferroptosis were investigated using I/R-induced H9c2 cells. The TXD group showed significantly decreased ROS and Fe2+levels, while the AZ group（treated with AZD9496） exhibited increased levels. Enhancement of motor functional recovery in thoracic spinal cord injury: voluntary wheel running versus forced treadmill exercise

Spinal cord injury necessitates effective rehabilitation strategies, with exercise therapies showing promise in promoting recovery. This study investigated the impact of rehabilitation exercise on functional recovery and morphological changes following thoracic contusive spinal cord injury. After a 7-day recovery period after spinal cord injury, mice were assigned to either a trained group(10 weeks of voluntary running wheel or forced treadmill exercise) or an untrained group. Bi-weekly assessments revealed that the exercise-trained group, particularly the voluntary wheel exercise subgroup, displayed significantly improved locomotor recovery, more plasticity of dopaminergic and serotonin modulation compared with the untrained group. Additionally, exercise interventions led to gait pattern restoration and enhanced transcranial magnetic motor-evoked potentials. Despite consistent injury areas across groups, exercise training promoted terminal innervation of descending axons. In summary, voluntary wheel exercise shows promise for enhancing outcomes after thoracic contusive spinal cord injury, emphasizing the role of exercise modality in promoting recovery and morphological changes in spinal cord injuries. Our findings will influence future strategies for rehabilitation exercises, restoring functional movement after spinal cord injury.

Value Assessment Method for the Grid-Alternative Energy Storage Based on Coordinated Planning Framework

Impacts of near-M_(s)austempering treatment on microstructure evolution and bainitic transformation kinetics of a medium Mn steel

The microstructure evolution and bainitic transformation of an Fe-0.19C-4.03Mn-1.48Si steel subjected to near-M_(s)austempering treatment were systematically investigated by combining dilatometer,X-ray diffraction,and electron *** additional austempering treatments with isothermal temperatures above M_(s)were used as *** show that the incubation period for the bainitic transformation occurs when the medium Mn steel is treated with the austempering temperature above M_(s).However,when subjected to near-M_(s)isothermal treatment,the medium Mn steel does not show an incubation period and has the fastest bainitic transformation ***,the largest volume fraction of bainite with a value of 74.7%is obtained on the condition of near-M_(s)austempering treatment after cooling to room *** and microstructure evolution analysis indicates that the elimination of the incubation period and the fastest rate of bainitic transformation are related to the preformed *** advent of preformed martensite allows the specimen to generate more bainite in a limited *** bainitic ferrite nucleation at austenite grain boundaries and through autocatalysis at ferrite/austenite interfaces,a model is established to understand the kinetics of bainite formation and it can describe the nucleation rate of bainitic transformation well when compared to the experimental results.

From waste to wealth:Coal tar residue derived carbon materials as low-cost anodes for potassium-ion batteries

The depression mechanism of sulfite ions on sphalerite and Pb^(2+)activated sphalerite in the flotation separation of galena from sphalerite still lacked in-depth ***,the depression mechanism of sulfite ions on sphalerite and Pb^(2+)activated sphalerite in the flotation separation of galena from sphalerite was further systematically investigated with experiments and density functional theory(DFT)*** X-ray photoelectric spectroscopy(XPS)results,DFT calculation results,and frontier molecular orbital analysis indicated that sulfite ions were difficult to be adsorbed on sphalerite surface,suggesting that sulfite ions achieved depression effects on sphalerite through other non-adsorption ***,the oxygen content in the surface of sphalerite treated with sulfite ions in creased,which enhanced the hydrophilicity of the sphalerite and further increased the difference in hydrophilicity between sphalerite and ***,sulfite ions were chelated with lead ions to form PbSO_(3)in *** hydrophilic PbSO_(3)was more easily adsorbed on sphalerite than *** interaction between sulfite ions and lead ions could effectively inhibit the activation of *** addition the UV spectrum showed that after adding sulfite ions,the peak of perxanthate in the sphalerite treated xanthate solution was significantly stronger than that in the galena with xanthate solution,indicating that xanthate interacted more readily with sulfite ions and oxygen mo lecules within the sphalerite system,leading to the formation of ***,sulfite ions hardly depressed the flotation of ga lena and could promote the flotation of galena to some *** study deepened the understanding of the depression mechanism o sulfite ions on sphalerite and Pb^(2+)activated sphalerite.

Design,Synthesis and Pharmacological Evaluation of Novel 4-Phenoxyquinoline Derivatives as VEGFR2 Kinase Inhibitors for Tumor Treatment

Vascular endothelial growth factor 2(VEGFR2)plays a vital role in regulating of tumor metastasis and angiogenesis,which has emerged as one of the effective targets for clinical tumor ***,a series of novel facilely accessible 4-phenoxyquinoline derivatives was prepared and assessed for their antitumor activity against three human tumor cell lines(SGC-7901,HepG2 and A549).Among these compounds,6a,6b and 6c show strong antitumor activity on HepG2 cells[the drug concentration of eliminating half of tumor cells(IC_(50))=9.33,1.84,8.54μmol/L].Notably,compound 6b shows potent selective inhibitory activity against VEGFR2 kinase with an IC_(50) value of 4.66 nmol/*** excellent anti-angiogenesis capability of compound 6b was confirmed by tube formation and chick chorioallantoic membrane(CAM)*** vivo studies confirmed that compound 6b was able to inhibit tumor growth in HepG2 xenografts of BALB/c nude mice without obvious side or toxic *** results demonstrated that compound 6b exhibited remarkable anti-angiogenesis and tumor growth inhibitory effects with less toxicity in vitro and in vivo *** findings highlighted the potential of compound 6b as a promising VEGFR2 kinase inhibitor for the development of antitumor drugs.
