Genome-Wide Screening of Aberrant Methylation Loci for Nonsyndromic Cleft Lip

Background： The pathogenicity of cleft lip （CL） is pretty complicated since it is influenced by the interaction of environment and genetic factors. The purpose of this study was to conduct a genome-wide screening of aberrant methylation loci in partial lesion tissues of patients with nonsyndromic CL （NSCL） and preliminarily validate candidate dysmethylated genes associated with ***： Fifteen healthy and sixteen NSCL fetal lip tissue samples were collected. The Infinium HumanMethylation450 BeadChip was used to screen aberrant methylation loci in three NSCL and three healthy lip tissues. The differential methylation sites and functions of the annotated genes between NSCL and healthy lip tissues were analyzed using minfi package of R software, cluster analysis, Gene Ontology （GO） annotation, and metabolic pathway annotation. Gene expression was assessed in nine differentially methylated genes by real-time polymerase chain reaction （PCR）. The transcriptions mRNA levels of three out of nine candidate genes were downregulated remarkably in NSCL lip tissues, and these three genes＇ abnormal methylation loci were validated by pyrosequencing in 16 NSCL cases and 15 healthy ***： In total, 4879 sites in the genes of NSCL odinopoeia fetuses showed aberrant methylation when compared with normal lip tissue genome. Among these, 3661 sites were hypermethylated and 1218 sites were hypomethylated as compared to methylation levels in healthy specimens. These aberrant methylation sites involved 2849 genes and were widely distributed among the chromosomes. Most differentially methylated sites were located in cytosine-phosphoric acid-guanine islands. Based on GO analysis, aberrantly methylated genes were involved in 11 cellular components, 13 molecular functions, and a variety of biological processes. Notably, the transcription of DAB1, REELIN, and FYN was significantly downregulated in lesion tissues of NSCL fetus （P 〈 0.05）. Pyrosequencing results validated that there

The presence of wrinkles in a membrane is the main factor that induces surface errors on space planar film reflect-arrays. Based on the commercial finite element (FE) package ABAQUS, a numerical procedure for membrane wrinkle analysis was set up, and used to analyze a square planar film reflect-array under pure shear force to evaluate its induced wrinkle characteristics. First, the effect of shear force on the wrinkle pattern of the array was studied and validated by experiment. Second, the effect of prestress was studied. When the prestress increases, the quantity of the wrinkles increases, and the amplitude of the wrinkles decreases. Third, the influence of the boundary conditions was investigated. A frame side edge structure has a relatively smooth surface, but also relatively high stress. Finally, the behavior of a joint seam was analyzed. The results indicate that a joint band has a significant influence on the wrinkle pattern of the membrane.

In the post-genomic era, identification of specific regulatory motifs or transcription factor binding sites （TFBSs） in non-coding DNA sequences, which is essential to elucidate transcriptional regulatory networks, has emerged as an obstacle that frustrates many researchers. Consequently, numerous motif discovery tools and correlated databases have been applied to solving this problem. However, these existing methods, based on different computational algorithms, show diverse motif prediction efficiency in non-coding DNA sequences. Therefore, understanding the similarities and differences of computational algorithms and enriching the motif discovery literatures are important for users to choose the most appropriate one among the online available tools. Moreover, there still lacks credible criterion to assess motif discovery tools and instructions for researchers to choose the best according to their own projects. Thus integration of the related resources might be a good approach to improve accuracy of the application. Recent studies integrate regulatory motif discovery tools with experimental methods to offer a complementary approach for researchers, and also provide a much-needed model for current researches on transcriptional regulatory networks. Here we present a comparative analysis of regulatory motif discovery tools for TFBSs.

BACKGROUND Thyroxine-binding globulin(TBG;the gene product of SERPINA7)is the main transporter of thyroid hormones in *** in the TBG gene may lead to inherited TBG *** have been 28 reported mutations that associate with complete TBG deficiency(TBG-CD).Here we identified a novel frameshift mutation causing early termination of the TBG protein and TBG-CD in a Chinese *** SUMMARY A 46-year-old Chinese man was referred to our hospital with normal free thyroxine,free triiodothyronine,thyrotropin,but lower total thyroxine and total triiodothyronine,and undetectable serum TBG,indicative of *** samples were obtained from the patient’s family members and thyroid function and serum TBG were *** DNA from peripheral blood was sequenced to detect possible TBG mutation(s).Quantitative PCR high-resolution melting curve analysis was used to screen TBG-Poly(L283F)among 117 Chinese men.A novel mutation of TBG(***135Alafs*21),a 19-nucleotide insertion in exon 1,was identified,which resulted in a truncated TBG protein product and caused *** other mutation,identified in the proband’s father,is a known polymorphism,TBG-Poly(L283F).The frequency of the TBG-Poly allele among 117 unrelated Han Chinese men from northeast China was 21.37%.CONCLUSION A novel mutation in the TBG gene associated with the TBG-CD phenotype was identified in a Chinese ***,it was found that 21.37%of Chinese males had TBG-Poly(L283F).

Using transfer matrix method,the optical transmission properties of 1-D photonic crystals composed partially of negative refraction media are analyzed. The transmission spectra of periodic photonic crystal,chirped photonic crystal and apodized photonic crystal are numerically simulated respectively. By contrast with optical transmission properties of ordinary photo-nic crystals made of positive refraction media,the transmission spectra of apodized photonic crystal become unregular,the Bragg flat-headed area recurs but the peak of transmission does not change significantly. Futhermore,the band gap range of chirped photonic crystal diminishes gradually.

Study of Coping-competence among Unmarried Pregnant Young Women in Three Big Cites in China

Objective To identify the coping-competence among unmarried pregnant young women. Methods A cross-sectional study was conducted in the setting of clinics-based. A total of 1 391 unmarried young women were recruited as the sample in Youth Clinics of 3 maternal care hospitals in Beijing, Jinan, and Guangzhou respectively in China. ＂Behavioral Attributes of Psychosocial Competence Scale-Condensed Form＂ was administered to identify the coping-competence of these women. All these women were aged 10-24 years old and were divided into three groups based on whether or not they had sex and pregnancy. The three groups were named as follows： pregnancy group with young women having had both sex and pregnancy, sex group with young women having had only sex but not pregnancy, and non-sex group with young women having no sex experience. Results Among the adolescents aged 10-19 years old, the coping-competence was different among the three groups （P=O.050）. Compared with the pregnancy group, the non-sex group were more inclined to active coping （P=0.026）. Among all the pregnant women aged 10-24 years old, the coping-competence was various by region （P〈0.001）： the women in Jinan were more inclined to active coping than the women in another two cites （P=0.009, P〈0.001）, and there was no difference between the women from Beijing and Guangzhou （P=0.324）. Conclusion This is the first study of coping among unmarried pregnant young women in China. The results supported the point of view that the pregnant adolescents were more inclined to passive coping, and the coping had regional differences.
