Molecular and functional characterization of three odorant-binding proteins from the wheat blossom midge,Sitodiplosis mosellana

Sitodiplosis mosellana,a periodic but devastating wheat pest,relies on wheat spike volatiles as a cue in sclecing hosts for *** odorant-binding proteins(OBPs)are thought to play essential roles in filtering,binding and transporting hydropho-bic odorant molecules to specific *** date,the molecular mechanisms underlying *** olfaction are poorly ***,three *** antenna-specific OBP genes,SmosOBPII,16 and 21,were cloned and bacterially *** properties of the recombinant proteins to 28 volatiles emitted from wheat spikes were in-vestigated using fluorescence competitive binding *** analysis suggested that these SmosOBPs belong to the Classic OBP ***-binding analysis showed that all three SmosOBPs preferentially bound alcohol,ester and ketone com-pounds,and SmosOBP11 and 16 also selectively bound terpenoid *** par-ticular,the three SmosOBPs had high binding affinities(Ki<20μmol/L)to 3-hexanol and cis-3-hexenylacetate that elicited strong electroantennogram(EAG)response fromfemale *** addition,SmosOBP11 displayed significantly higher binding(Ki<8μmo/L)than SmosOBP16 and 21 to l-octen-3-ol,D-panthenol,a-pinene and heptyl acetate which elicited significant EAG response,suggesting that SmosOBP11 plays a ma-jor role in recognition and transportation of these *** findings have provided important insight into the molecular mechanism by which *** specifically rec-ognizes plant volatiles for host selection,and have facilitated identification of effective volatile attractants that are potentially useful for pest monitoring and trapping.

Juvenile hormone(JH),a growth regulator,inhibits ecdysteroid-induced meta-morphosis and controls insect development and ***-tolerant(Met)and Krippel homolog I(Kr-h1)are two proteins involved in JH *** gain some insight into their function in development of Sitodiplosis mosellana,an insect pest undergoing obligatory larval diapause at the mature 3rd instar stage,we cloned full-length complemen-tary DNAs of Met and Kr-h1 from this *** encoded a putative protein,which contained three domains typical of the bHLH-PAS family and eight conserved amino acid residues important for JH ***-h1 encoded a protein showing high sequence homology to its counterparts in other specics,and contained all eight highly conserved Zn-finger motifs for *** patterns of SmMet and SmKr hl were de-velopmentally regulated and JH III responsive as *** mRNA abundance increased as larvae entered carly 3rd instar,pre-diapause and maintenance stages,and peaked during post-diapause quiescence,a pattern correlated with JH titers in this *** from reduced expression of SmMer,SmKr-h1 mRNA increased at mid-to-late period of post-diapause *** application of JH II on diapausing larvac also induced the two genes in a dose-dependent *** of SmuMer and SmKr-h1 clearly declined in the pre-pupal phase,and was significantly higher in female adults than male *** results suggest that JH-responsive SmMet and SmKr-h1 might play key roles in diapause induction and maintenance as well as in post-diapause quiescence and adult reproduction,whereas metamorphosis from larvae to pupac might be correlated with their reduced expression.

High basal defense gene expression determines sorghum resistance to the whorl-feeding insect southwestern corn borer

Southwestern corn borer （SWCB, Diatraea grandiosella） and fall armyworm （FAW, Spodopterafrugiperda） are major pests of sorghum in the southern United States. Host plant resistance is a desirable means for reducing plant damage and yield losses from both insects. In this study, we evaluated 12 sorghum lines for whorl-stage resistance to leaf-feeding SWCB and FAW in greenhouse and laboratory bioassays. Differential plant responses were detected against the two insects. Among 12 lines tested, CM1821, Della and PI196583 were resistant to both insects, while BTx2752 was largely susceptible. Line R.09110 was resistant to SWCB, but susceptible to FAW, whereas Redbine-60 was suscep- tible to SWCB, but not to FAW. In addition, we quantified various chemical components in the plants and determined their association with insect resistance. Tannin and chloro- phyll in leaves did not show any significant correlation with resistance to either insects, but contents of soluble protein in general were negatively correlated with resistance to both insects. Endogenous soluble sugar and dhurrin were only positively correlated with resistance to SWCB, but not with FAW resistance. To gain some molecular insight into resistance mechanism of sorghum to SWCB, we performed qPCR reactions for key genes encoding enzymes involved in dhurrin and jasmonic acid （JA） biosynthesis on selected resistant or susceptible lines. Although these genes were rapidly and strongly induced by insect feeding in all lines, the observed resistance is likely explained by higher constitutive dhurrin contents in some resistant lines and higher basal JA biosynthesis in others. Our results suggest that sorghum utilizes multiple strategies to defend itself against SWCB.

Synthetic chemistry plays an indispensable role in drug discovery,contributing to hit compounds identification,lead compounds optimization,candidate drugs preparation,and so *** Nobel Prize laureate James Black emphasized,“the most fruitful basis for the discovery of a new drug is to start with an old drug”***-stage modification or functionalization of drugs,natural products and bioactive compounds have garnered significant interest due to its ability to introduce diverse elements into bioactive compounds *** modifications alter the chemical space and physiochemical properties of these compounds,ultimately influencing their potency and *** enrich a toolbox of chemical modification methods for drug discovery,this review focuses on the incorporation of halogen,oxygen,and nitrogen—the ubiquitous elements in pharmacophore components of the marketed drugs—through late-stage modification in recent two decades,and discusses the state and challenges faced in these *** also emphasize that increasing cooperation between chemists and pharmacists may be conducive to the rapid discovery of new activities of the functionalized ***,we hope this review would serve as a valuable resource,facilitating the application of late-stage modification in the construction of novel molecules and inspiring innovative concepts for designing and building new drugs.

A Reliable Iteration Algorithm for One-Bit Compressive Sensing on the Unit Sphere

The one-bit compressed sensing problem is of fundamental importance in many areas,such as wireless communication,statistics,and so ***,the optimization of one-bit problem coustrained on the unit sphere lacks an algorithm with rigorous mathematical proof of convergence and *** this paper,an iteration algorithm is established based on difference-of-convex algorithm for the one-bit compressed sensing problem constrained on the unit sphere,with iterating formula■,where C is the convex cone generated by the one-bit measurements andη_(1)>η_(2)>1/*** new algorithm is proved to converge as long as the initial point is on the unit sphere and accords with the measurements,and the convergence to the global minimum point of the l_(1)norm is discussed.

Theoretical Exploration of Alloying Effects on Stabilities and Mechanical Properties ofγʹPhase in Novel Co–Al–Nb-Base Superalloys

The new developedγ/γʹCo–Al–Nb-base alloys show great potentials as high-temperature ***,finding appropriate compositions to improve performance of alloys still poses a great challenge to the development of Co–Al–Nb-base *** by the lack of alloying effects on fundamental properties of criticalγʹphase,we systematically performed a theoretical investigation on the effect of alloying elements TM(TM:Ti,V,Cr,Zr,Mo,Ta,W,Re,and Ru)on phase stabilities and mechanical properties of L1_(2)-typeγʹ(Co,Ni)_(3)(Al,Nb).By analyzing the stability ofγʹphase with respect to its competitive B2 and D0_(19) phases,the results shown that Ti,V,and Cr enhance the L1_(2) stability and widen the L1_(2)–D0_(19) energy barrier,in which V yields the maximum *** analysis of electronic structure indicated that the alternation of valence electrons at fermi level would be the atomic origin for doping TM inγʹ*** calculated results of mechanical properties shown that V and Cr are expected to be optimal dopant for enhancing the strength and the ductility ofγʹ*** addition of Ta is also beneficial for enhancing the strength at the slight expense of ductility ofγʹ*** drawing the mechanical maps,the preferred composition range for the phases with desired properties is roughly demarcated in theory for the multi-addition of V/Cr and V/Ta inγʹ*** findings would be useful for optimizing the performance of novelγ/γʹCo–Al–Nb-base superalloys.

Nutrient transport following water transfer through the world's largest water diversion channel

Nutrient levels in the artificial channel constructed for the Middle Route Project are significant indicators of water quality safety and aquatic ecological integrity for this large,interbasin ***,the distribution and transport of nutrients along the channel were poorly *** on a time-series dataset as well as mass balance and material flow analysis methods,the water and nutrient transport fluxes in the Middle Route of the South-to-North Water Diversion Project were identified in this *** results indicate that the nutrient concentrations varied considerably with time,but there was no significant difference among the 30 stations of the main *** temperature difference was the major factor in the large fluctuations of water quality indicators over *** nutrient loadings varied with the water volume outputs from the main channel to the waterreceiving *** deposition was an important source of nutrients in the main channel,accounting for 9.13%,20.6%,and 0.635%of the nitrogen,phosphorus,and sulfur input from the Danjiangkou Reservoir,*** 2021,a net accumulation of 988 tons of N,29 tons of P,and 2,540 tons of S,respectively,were present in the main *** increase of these external and internal nutrient loadings would cause water quality fluctuation and deterioration in some local sections of the main *** study quantified the spatial and temporal patterns of nutrient transport in the Middle Route and revealed the ecological effects on the aquatic environment,assisting authorities on the project to develop effective water conservation strategies.

Collectively induced transparency and absorption in waveguide quantum electrodynamics with Bragg atom arrays

Collective quantum states, such as subradiant and superradiant states, are useful for controlling optical responses in many-body quantum systems. In this work, we study novel collective quantum phenomena in waveguide-coupled Bragg atom arrays with inhomogeneous *** atoms without free-space dissipation, collectively induced transparency is produced by destructive quantum interference between subradiant and superradiant states. In a large Bragg atom array, multi-frequency photon transparency can be obtained by considering atoms with different frequencies. Interestingly, we find collectively induced absorption(CIA) by studying the influence of free-space dissipation on photon transport. Tunable atomic frequencies nontrivially modify decay rates of subradiant states. When the decay rate of a subradiant state equals to the free-space dissipation, photon absorption can reach a limit at a certain frequency. In other words, photon absorption is enhanced with low free-space dissipation, distinct from previous photon detection schemes. We also show multi-frequency CIA by properly adjusting atomic frequencies. Our work presents a way to manipulate collective quantum states and exotic optical properties in waveguide quantum electrodynamics(QED) systems.

Developing an energy supply-chain based on renewable biomass holds great potential to build a low carbon ***-energy-density(HED)jet fuel,featuring unique fused/strained cycloalkanes,is of great significance for volume-limited military aircrafts,as their high density and combustion heat can extend flight duration and increase the ***,the exploration of biomass-based routes towards HED fuel has drawn much attention over the past *** reaction features rapid construction of various carbocycles in an atom-and step-economical *** elegant strategy has been widely applied in the manufacture of sustainable HED *** we carefully summarize the progress achieved in this fascinating area and the review is categorized by the cycloaddition patterns including[4+2],[2+2],[4+4],and[2+1]***,the energy densities of the as-prepared biofuels and petroleumbased fuels(conventional Jet-A and advanced JP-10)are also *** review will provide important insights into rational design of new HED fuel with different ring-types/sizes and inspire the chemists to turn those literature studies into practical applications in military field.
