Exploring the therapeutic potential of Qi Teng Mai Ning recipe in ischemic stroke and vascular cognitive impairment

Background:This study aims to explore the therapeutic effects of the Qi Teng Mai Ning recipe on ischemic stroke and vascular cognitive impairment through its potential to modulate cellular autophagy,with a focus on identifying its active ingredients and their target ***:The study began with the identification of active ingredients in the Qi Teng Mai Ning *** proceeded to screen the gene expression omnibus database for ischemic stroke and vascular cognitive impairment-associated differentially expressed mRNAs and to identify cellular autophagy-related proteins via the Human Autophagy *** proteins were annotated with Gene Ontology and Kyoto Encyclopedia of Genes and Genomes functions and subjected to molecular docking with the recipe’s core active *** vitro cell experiments were conducted on hypoxic HT22 cells,involving CCK8 assay,lentiviral transfection to silence autophagy related 9B(ATG9B),immunofluorescence staining,and qPCR validation to investigate the effects of the recipe on ***:The analysis identified 104 active ingredients targeting 408 proteins and forming a complex ingredient-target *** 55 ischemic stroke-related and 909 vascular cognitive impairment-related differentially expressed mRNAs revealed 14 co-expressed *** docking showed quercetin,kaempferol,myrcene,and conferone as key ingredients targeting autophagy-related *** experiments indicated that the recipe significantly enhanced cell viability under hypoxic conditions,reduced apoptosis,and modulated the expression of autophagy-related factors,thereby decreasing apoptosis rates in HT22 ***:The Qi Teng Mai Ning recipe offers protective effects against ischemic stroke and vascular cognitive impairment by modulating autophagy-related *** efficacy highlights the potential of traditional Chinese medicine in treating these conditions,though further research is needed to fully understand its

Aerobic degradation of methyl tert-butyl ether by a Proteobacteria strain in a closed culture system

The contamination of methyl ten-butyl ether （MTBE） in underground waters has become a widely concerned problem all over the world. In this study, a novel dosed culture system with oxygen supplied by H2O2 was introduced for MTBE aerobic biodegradation. After 7 d, almost all MTBE was degraded by a pure culture, a member of β-Proteobacteria named as PMI, in a closed system with oxygen supply, while only 40% MTBE was degraded in one without oxygen supply. Dissolved oxygen （DO） levels of the broth in closed systems respectively with and without H2O2 were about 5-6 and 4 mg/L. Higher DO may improve the activity of monooxygemase, which is the key enzyme of metabolic pathway from MTBE to tert-butyl alcohol and finally to CO2, and may result in the increase of the degrading activity of PM1 cell. The purge and trap GC-MS result of the broth in closed systems showed that tea-butyl alcohol, isopronol and acetone were the main intermediate products.

Preparations of TiO_2 film coated on foam nickel substrate by sol-gel processes and its photocatalytic activity for degradation of acetaldehyde

Anatase TiO2 films were successfully prepared on foam nickel substrates by sol-gel technique using tetrabutyl titanate as precursor. The characteristics of the TiO2 films were investigated by XPS, XRD, FE-SEM, TEM and UV-Vis absorption spectra. The photocatalytic activities of TiO2 films were investigated by photocatalytic degradation reactions of gaseous acetaldehyde, an indoor pollutant, under ultraviolet light irradiation. It was found that Ni^2＋ doping into TiO2 films due to the foam nickel substrates resulted in the extension of absorption edges of TiO2 films from UV region to visible light region. The pre-heating for foam nickel substrates resulted in the formation of NiO layer, which prevented effectively the injection of photogenerated electrons from TiO2 films to metal nickel. The TiO2 films displayed high photocatalytic activity for the degradation of acetaldehyde, and were enhanced by calcining the substrates and coating TiO2 films repeatedly. The high activity was mainly attributed to the improvement of the characteristics of substrate surface and the increase of active sites on photocatalyst.

Prediction and evaluation of optimum quenching temperature and microstructure in a 1300 MPa ultra-high-strength Q&P steel

The quenching and partitioning steel is the representative of the third generation of advanced high-strength *** effect of quenching temperature on the microstructure and mechanical property of ferrite-containing quenching and partitioning steel was studied by intercritical annealing quenching and partitioning *** preparing a test steel with a tensile strength of 1300 MPa and total elongation of 19%,it is found that the actual optimum quenching temperature was lower than that calculated according to the constrained carbon *** results indicate that the martensite start temperature of the austenite was overestimated when considering the diffusion of carbon *** grain size which is affected by low temperature and the existence of ferrite during intercritical annealing influenced the optimum quenching temperature.A scheme considering the diffusion of various alloying elements and austenite grain size was proposed and *** this scheme,the optimum quenching temperature of intercritically annealed quenching and partitioning steel with complex microstructures was well predicted.

Effect of vanadium on hydrogen embrittlement susceptibility of high-strength hot-stamped steel

Based on the chemical composition of traditional hot-stamped steel(e.g.,22MnB5 and 30MnB5),Nb and V microalloying elements are added into 30MnB5 steel to meet the requirements of ultra-high strength,excellent ductility and potent resistance to hydrogen embrittlement(HE)at the same *** influence of hot-stamped steel on HE was studied by conducting a hydrogen permeation method and pre-charged hydrogen slow strain rate ***,the experimental steel microstructures and corresponding fracture surfaces are observed and analyzed to characterize HE *** results show that a finer microstructure,a lower apparent diffusion coefficient of hydrogen and a smaller percentage of strength and plasticity reduction are obtained due to the addition of the vanadium element into hot-stamped *** to the V free experimental steel,the steel with 0.14 wt.% V has a large number of dispersive precipitates and more grain boundary areas,which makes hydrogen atoms dispersedly distribute.

For sudden expansion pipes, experiments were carried ＇Out to study the cavitation inception for various enlargement ratios in high speed flows. The flow velocity of the prototype reaches 50 m/s in laboratory. The relationship between the expansion ratio and the incipient cavitation number is obtained. The scale and velocity effects are revealed. It is shown that Keller＇s revised formula should be modified to calculate the incipient cavitation number when the forecasted velocity of the flows in the prototype exceeds the experimental velocity.

Effect of microstructure evolution on Luders strain and tensile properties in an intercritical annealing medium-Mn steel

The influence of microstructural characteristics on Lu¨ders strain and mechanical properties was explored by means of altering thermo-mechanical circumstances in an intercritical annealing(IA)medium-Mn Fe-11Mn-0.09C-0.25Si(wt.%)*** IA of cold-rolled samples with severe plastic deformation,exclusively equiaxed dual phases were obtained because of active recovery and *** equiaxed austenite(gamma E)with a larger size and inadequate chemical concentration was more readily transformed into martensite,and subsequent transformation-induced plasticity(TRIP)effect was triggered actively at relatively higher IA temperature,lessening localized *** addition,grown-in dislocations were prone to multiply and migrate around a broad mean free path for coarser equiaxed ferrite(alpha E)due to weakening dynamic recovery;therefore,it was the ensuing increased mobility of dislocations instead of reserving plentiful initial dislocation density that facilitated the propagation velocity of Luders bands and the accumulation of work *** contrast,the bimodal-grained microstructure with lath-like and equiaxed austenite(gamma L+gamma E)satisfactorily contributed to a smaller yield point elongation(YPE)without compromise of comprehensive mechanical properties on the grounds that austenitic gradient stability gave rise to discontinuous but sustainable TRIP effect and incremental work ***,Luders strain is closely related to the absence of work hardening in the region which yields *** follows that the decreased stability of retained austenite,favorable mobility of dislocations and the bimodal-grained structure all prominently make up for the insufficiency of work hardening,thereof resulting in a limited YPE.

Gene expression and MR diffusion-weighted imaging after chemoembolization in rabbit liver VX-2 tumor model

AIM: To investigate the dynamic characteristics and the correlation between PCNA, Bax, nm23, E-cadherin expression and apparent diffusion coefficient (ADC) on MR diffusion-weighted imaging (DWI) after chemoembolization in rabbit liver VX-2 tumor model. METHODS: Forty New Zealand rabbit liver VX-2 tumor models were included in the study. DWI was carried out periodically after chemoembolization. All VX-2 tumor samples in each group were examined by histopathology and Strept Avidin-Biotin Complex (SABC) immunohistochemical staining. RESULTS: The PCNA expression index in VX-2 tumors was higher than in the normal parenchyma around the tumor (P < 0.001). Nm23, Bax or E-caderin expression index in VX-2 tumors were lower than in the normal parenchyma around the tumor (all P < 0.001). PCNAand nm23 expression in the VX-2 tumor periphery first increased and then decreased (P < 0.001 and P = 0.03, respectively), while the expression of Bax and E-cadherin before and after chemoembolization was insignificant. When b-value was 100 s/mm2, there was a linear correlation between PCNA expression and ADC in the area of VX-2 tumor periphery (P < 0.001), and PCNA expression in VX-2 tumor periphery influenced the ADC. CONCLUSION: The potential of VX-2 tumor infiltrating and metastasizing decreases, while its ability to proliferate increases for a short time after chemoembolization. To some degree, the ADC value indirectly reflects the proliferation of VX-2 tumor cells.

AIM: To investigate dynamic characteristics and pathological mechanism of signal in rabbit VX-2 tumor model on diffusion-weighted imaging (DWI) after chemoembolization. METHODS: Forty New Zealand rabbits were included in the study and forty-seven rabbit VX-2 tumor models were raised by implanting directly and intrahepatically after abdominal cavity opened. Forty VX-2 tumor models from them were divided into four groups. DWI was performed periodically and respectively for each group after chemoembolization. All VX-2 tumor samples of each group were studied by pathology. The distinction of VX-2 tumors on DWI was assessed by their apparent diffusion coefficient (ADC) values. The statistical significance between different time groups, different area groups or different b-value groups was calculated by using SPSS12.0 software. RESULTS: Under b-value of 100 s/mm2, ADC values were lowest at 16 h after chemoembolization in area of VX-2 tumor periphery, central, and normal liver parenchyma around tumor, but turned to increase with further elongation of chemoembolization treatment. The distinction of ADC between different time groups was significant respectively (F = 7.325, P < 0.001; F = 2.496, P < 0.048; F = 6.856, P < 0.001). Cellular edema in the area of VX-2 tumor periphery or normal liver parenchyma around tumor, increased quickly in sixteen h after chemoembolization but, from the 16th h to the 48th h, cellular edema in the area of normal liver parenchyma around tumor decreased gradually and that in the area of VX-2 tumor periphery decreased lightly at, and then increased continually. After chemoembolization, Cellular necrosis in the area of VX-2 tumor periphery was more significantly high than that before chemoembolization. The areas of dead cells in VX-2 tumors manifested low signal and high ADC value, while the areas of viable cells manifested high signal and low ADC value. AIM： To investigate dynamical and image pathological characteristics of the liver on magnetic resonance （MR） diffusion-weighted imaging （DWI） in the rabbit VX-2 tumor model. METHODS： Forty New Zealand rabbits were included in the study and VX-2 tumor piece was implanted intrahepatically. Fifteen animals received two intrahepatic implantations while 25 had one intrahepatical implantation. DWI, T1- and T2-weighted of magnetic resonance imaging （MRI） were carried out on the 7th and the 14th d after implantation and DWI was conducted, respectively on the 21th d. Ten VX-2 tumor samples were studied pathologically. RESULTS： The rate of lump detected by DWI, TlWI and T2WI was 78.7%, 10.7% and 53.5% （X^2 =32.61, P 〈 0.001） on the 7th d after implantation and 95.8%, 54.3% and 82.9% （X^2 = 21.50, P 〈 0.001） on the 14th d. The signal of most VX-2 tumors on DWI was uniform and it was equal diffusion coefficient （ADC）. did not decrease on the 7th on the map of apparent The signal of VX tumors d after implantation, most of them slowly growing during the week following implantation without significant cell dying within the tumor. VX-2 tumors grew increasingly within 14 d after implantation but the signal of most VX-2 tumors on DWI or on the map of ADC was uniform or uneven and ADC of VX tumors decreased obscurely or slightly because tumor necrosis was still not obvious. On the 21th d after implantation, the signal of most VX-2 tumors on DWI or on the map of ADC was uneven because tumor necrosis was evident and ADC of VX-2 tumor necrotic areas decreased. The areas of viable cells in VX-2 tumors manifested a high signal on DWI and a low signal on the map of ADC. The areas of dead cells or necrosis in VX-2 tumors manifested low signals on DWI and low, equal or high signals on the map of ADC but they manifested high signals on DWI and on the map of ADC at the same time when the areas of necrotic tumor became liquefied or cystic.
