Knockdown of liver-intestine cadherin decreases BGC823 cell invasiveness and metastasis in vivo

AIM:To assess BGC823 gastric cancer(GC) cell metastasis after knockdown of liver-intestine cadherin(CDH17) and the therapeutic value of CDH17-RNAilentivirus in ***:We evaluated primary tumor growth and assessed local infiltration and systemic tumor dissemination using an orthotopic implantation *** therapeutic value of CDH17 knockdown was examined by intratumoral administration of CDH17-RNA interference(RNAi)-lentivirus in an established GC tumor xenograft mouse ***,a comparative proteomic approach was utilized to identify differentially expressed proteins in BGC823 and lenti-CDH17-miRneg cells following CDH17 ***:Metastases in the liver and lung appeared earlier and more frequently in animals with tumors derived from BGC823 or lenti-CDH17-miR-neg cells than in tumors derived from lenti-CDH17-miR-B *** tumor weight and volume in the CDH17-RNAi-lentivirus-treated group were significantly lower than those in the control group(tumor volume:0.89 ± 0.04 cm 3 vs 1.16 ± 0.06 cm 3,P weight:1.15 ± 0.58 g vs 2.09 ± 0.08 g,P < 0.05).Fifteen differentially expressed proteins were identified after CDH17 silencing in BGC823 cells,including a variety of cytoskeletal and chaperone proteins as well as proteins involved in metabolism,immunity/defense,cell proliferation and differentiation,cell cycle,and signal ***:Our data establish a foundation for future studies of the comprehensive protein expression patterns and effects of CDH17 in GC.

Thymoquinone inhibits proliferation in gastric cancer via the STAT3 pathway in vivo and in vitro

AIM: To elucidate the mechanism of thymoquinone(TQ)-induced apoptosis in human gastric cancer cells in vitro and in ***: HGC27, BGC823, and SGC7901 cells were cultured in vitro and treated with TQ(0, 10, 25, 50, 75, 100, 125 μmol/L) for 12 h, 24 h, and 36 h, and then the proliferation inhibitory rates were detected by methylthiazole tetrazolium assay. Apoptosis was observed after Hoechst staining. The protein expressions of signal transducer and activator of transcription(STAT)3, p-STAT3, STAT5, p-STAT5, phospho-janus-activated kinase 2(JAK2), JAK2, p-Src, Src, glyceraldehyde-3-phosphate dehydrogenase, lamin-A, survivin, Cyclin D, Bcl-2, Bax, peroxisome proliferator activated receptor, and caspase-3,7,9 were detected by western blot. Cell cycle and apoptosis weredetermined with flow cytometry. TQ induced dosedependent apoptotic cell death in HGC27 cells was measured by Annexin V-fluorescein isothiocyanate(FITC)/propidium iodide(PI) analysis and Hoechst 33258. RESULTS: TQ inhibited the phosphorylation of STAT3 but not STAT5. TQ-induced downregulation of STAT3 activation was associated with a reduction in JAK2 and c-Src activity. TQ also downregulated the expression of STAT3-regulated genes, such as Bcl-2, cyclin D, survivin, and vascular endothelial growth factor, and activated caspase-3,7,9. Consistent with the in vitro results, TQ was significantly effective as an antitumor agent in a xenograft tumor mouse model. CONCLUSION: This study provides strong evidence that downregulation of the STAT3 signaling pathway mediates TQ-induced apoptosis in gastric cancer.

The Tibetan Plateau vortices(TPVs)are the major rain-producing systems over the Tibetan Plateau(TP).The activities of TPVs are closely related to TP's water source,which supplies fresh water to millions of people in *** of the TPVs can increase understanding about the future of water supply in Asia under global *** this study,the possible activities of TPVs under 1.5℃and 2℃warming scenarios above the pre-industrial level are evaluated through the NCAR CESM(Community Earth System Model)Low-warming(CESM-LW)*** results show that the CESM-LW well reproduces the spatio-temporal characteristics of TPVs in the historical run from 1985 to *** CESM-LW suggests TPVs in warm season(May-September)increase by 15%due to the additional 0.5℃warming by the end of this century(2071—2100).It implies the greater importance of TPVs to the precipitation over the TP in the *** changes of TPVs are closely related to the large-scale circulations *** additional 0.5℃warming strengthens the temperature difference between the TP and its surrounding areas,which results in an enhanced convergence near the TP's surface and divergence in the upper troposphere by about-0.1×10^(-6)and 0.22×10^(6)s^(-1),*** assessment of future TPVs provides a synoptic dynamic perspective on the climate change of precipitation and water resources.

Channel confluences are universally present in nature. They can be divided into two types： asymmetrical river confluences and symmetric river confluences. The latter is also called as the Y-shaped confluences. Most of previous work has paid more attention to the asymmetrical river confluences, but few studies have been conducted on the Y-shaped confluences. In this article, the effects of bed discordance on the flow patterns at ＂Y＂ shaped open-channel confluences were studied by using a 3-D numerical simulation. It is proved that the model can undertake quantitative assessment of the flow at confluences. The results indicate that there are a lot of differences between the Y-shaped confluence and asymmetrical confluence. The discordant bed height plays an important role at the Y-shaped junction.

Overexpression of Dickkopf-3 induces apoptosis through mitochondrial pathway in human colon cancer

AIM:To investigate the mechanisms of the biological roles of Dickkopf-3(Dkk-3) in cell invasion,survival and apoptosis in colon cancer ***:Three human colon cancer cell lines,i.e.,HT-29,LoVo and SW480,were *** of Dkk-3 induced by pEGFP-N1-Dkk-3-GFP plasmid in LoVo cells was performed using Lipofectamine 2000 *** transcription polymerase chain reaction and Western blotting were performed to determine the mRNA and protein expression levels of Dkk-3,*** proliferation assay,cell cycle analysis,hoechst 33258 assay and Matrigel invasion assay were performed on Dkk-3 overexpressing ***:The mRNA and protein expressions of Dkk-3 in HT-29(mRNA:0.06 ± 0.02,protein:0.06 ± 0.01) and LoVo(mRNA:0.07 ± 0.02,protein:0.07 ± 0.02) cells were significantly lower than that in SW480 cells(mRNA:0.92 ± 0.04,protein:0.69 ± 0.13;all P < 0.05),and the greatest levels of invasiveness wasin LoVo ***-3 overexpression inhibited the proliferation and invasion of LoVo cells and induced cell cycle arrest at G0/G1 phase and subsequent apoptosis,as indicated by increased chromatin condensation and fragments,upregulated Bax and cytochrome c protein,downregulated survivin and Bcl-2 protein,and the activation of caspase-3 and ***,Dkk-3 overexpression reduced the accumulation of cytosolic fraction of β-***:Dkk-3 overexpression induced apoptosis in human colon cancer possibly through the mitochondrial ***-3 may be involved in the Wnt/β-catenin signaling pathways in colon cancer.

Cooperative inhibitory effect of sinomenine combined with5-fluorouracil on esophageal carcinoma

AIM:To investigate the inhibitory effects of sinomenine(SIN)combined with 5-fluorouracil(5-FU)on esophageal carcinoma in vitro and in ***:Esophageal carcinoma(Eca-109)cells were cultured in *** single or combined growth inhibition effects of SIN and 5-FU on the Eca-109 cells were examined by measuring the absorbance of CCK-8dye in living *** 33258 staining and an Annexin V/PI apoptosis kit were used to detect the percentage of cells undergoing *** blotting was used to investigate the essential mechanism underlying SIN and 5-FU-induced *** at 25mg/kg and 5-FU at 12 mg/kg every 3 d,either combined or alone,was injected into nude mice and tumor growth inhibition and side effects of the drug treatment were ***:SIN and 5-FU,both in combination and individually,significantly inhibited the proliferation of Eca-109 cells and induced obvious ***,the combined effects were greater than those of the individual agents(P<0.05).Annexin V/PI staining and Hoechst 33258 staining both indicated that the percentage of apoptotic cells induced by SIN and 5-FU combined or alone were significantly different from the control(P<0.05).The up-regulation of Bax and downregulation of Bcl-2 showed that the essential mechanism of apoptosis induced by SIN and 5-FU occurs via the mitochondrial *** and 5-FU alone significantly inhibited the growth of tumor xenografts in vivo,and the combined inhibition rate was even higher(P<0.05).During the course of chemotherapy,no obvious side effects were observed in the liver or ***:The combined effects of SIN and 5-FU on esophageal carcinoma were superior to those of the individual compounds,and the drug combination did not increase the side effects of chemotherapy.

Tumor suppressor function of ezrin-radixin-moesin-binding phosphoprotein-50 through β-catenin/E-cadher in pathway in human hepatocellular cancer

AIM:To determine the effect and molecular mechanism of ezrin-radixin-moesin-binding phosphoprotein-50(EBP50) in hepatocellular carcinoma(HCC).METHODS:Three human HCC cell lines,i.e.,SMMC7721,HepG2 and Hep3B,were *** transfected the Pbk-CMV-HA-EBP50 plasmid into SMMC7721 cells with Lipofectamine 2000 to overexpress *** blotting were performed to determine the effects of the plasmid on EBP50 expression and to detect the expression of β-catenin and E-cadherin before and after the transfection of the plasmid into SMMC7721 *** vitro cell proliferation was assessed with a Cell Counting Kit-8(CCK-8) *** cycle distribution was assessed with flow *** and migration ability of before and after the transfection were determined with a transwell *** apoptosis was demonstrated with Annexin *** effect of EBP50 overexpressing on tumor growth in vivo was performed with a xenograft tumor model in nude ***:The transfection efficiency was confirmed with Western blotting(1.36 ± 0.07 vs 0.81 ± 0.09,P < 0.01).The CCK8 assay demonstrated that the growth of cells overexpressing EBP50 was significantly lower than control cells(P < 0.01).Cell cycle distribution showed there was a G0/G1 cell cycle arrest in cells overexpressing EBP50(61.3% ± 3.1% vs 54.0% ± 2.4%,P < 0.05).The transwell assay showed that cell invasion and migration were significantly inhibited in cells overexpressing EBP50 compared with control cells(5.8 ± 0.8 vs 21.6 ± 1.3,P < 0.01).Annexin V-FITC revealed that apoptosis was significantly increased in cells overexpressing EBP50 compared with control cells(14.8% ± 2.7% vs 3.4% ± 1.3%,P < 0.05).The expression of β-catenin was downregulated and E-cadherin was upregulated in cells overexpressing EBP50 compared with control cells(0.28 ± 0.07 vs 0.56 ± 0.12,P < 0.05;0.55 ± 0.08 vs 0.39 ± 0.07,P < 0.05).In vivo tumor growth assay confirmed that up-regulation of EBP50 could obviously slow the growth of HCC derived from SMM

Schistosoma japonicum attenuates dextran sodium sulfateinduced colitis in mice via reduction of endoplasmic reticulum stress

AIM To elucidate the impact of Schistosoma(S.) japonicum infection on inflammatory bowel disease by studying the effects of exposure to S. japonicum cercariae on dextran sodium sulfate(DSS)-induced colitis. METHODS Infection was percutaneously established with 20 ± 2 cercariae of S. japonicum, and colitis was induced by administration of 3% DSS at 4 wk post infection. weight change, colon length, histological score(HS) and disease activity index(DAI) were evaluated. Inflammatory cytokines, such as IL-2, IL-10 and IFN-γ, were tested by a cytometric bead array and real-time quantitative polymerase chain reaction(RT-PCR). Protein and m RNA levels of IRE1α, IRE1β, GRP78, CHOP, P65, P-P65, P-IκBα and IκBα in colon tissues were examined by Western blot and RT-PCR, respectively. Terminal deoxynucleotidyl transferase-mediated d UTP nick-end labeling positive cells, cleaved-caspase 3 expression and Bcl2/Bax were investigated to assess the apoptosis in colon *** Mice infected with S. japonicum cercariae were less susceptible to DSS. Mice infected with S. japonicum cercariae and treated with DSS showed decreased weight loss, longer colon, and lower HS and DAI compared with mice treated with DSS alone. A substantial decrease in Th1/Th2/Th17 response was observed after infection with S. japonicum. Endoplasmic reticulum(ER) stress and the nuclear factor-kappa B(NF-κB) pathway were reduced in mice infected with S. japonicum cercariae and treated with DSS, along with ameliorated celluar apoptosis, in contrast to mice treated with DSS alone. CONCLUSION Exposure to S. japonicum attenuated inflammatory response in a DSS-induced colitis model. In addition to the Th1/Th2/Th17 pathway and NF-κB pathway, ER stress was shown to be involved in mitigating inflammation and decreasing apoptosis. Thus, ER stress is a new aspect in elucidating the relationship between helminth infection and inflammatory bowel disease(IBD), which may offer new therapeutic methods for IBD.

A Case Study of the Impacts of Dust Aerosols on Surface Atmospheric Variables and Energy Budgets in a Semi-Arid Region of China

The authors present a case study investigating the impacts of dust aerosols on surface atmospheric variables and energy budgets in a semi-arid region of China. Enhanced observational meteorological data, radiative fluxes, near-surface heat fluxes, and concentrations of dust aerosols were collected from Tongyu station, one of the reference sites of the International Coordinated Energy and Water Cycle Observations Project (CEOP), during a typical dust storm event in June 2006. A comprehensive analysis of these data show that in this semi-arid area, higher wind velocities and a continuously reduced air pressure were identified during the dust storm period. Dust storm events are usually associated with low relative humidity weather conditions, which result in low latent heat flux values. Dust aerosols suspended in the air decrease the net radiation, mainly by reducing the direct solar radiation reaching the land surface. This reduction in net radiation results in a decrease in soil temperatures at a depth of 2 cm. The combination of increased air temperature and decreased soil temperature strengthens the energy exchange of the atmosphere-earth system, increasing the surface sensible heat flux. After the dust storm event, the atmosphere was dominated by higher pressures and was relatively wet and cold. Net radiation and latent heat flux show an evident increase, while the surface sensible heat flux shows a clear decrease.
