Nanoengineered sonosensitive platelets for synergistically augmented sonodynamic tumor therapy by glutamine deprivation and cascading thrombosis

Ultrasound(US)-activated sonodynamic therapy(SDT)stands for a distinct antitumor modality because of its attractive characteristics including intriguing noninvasiveness,desirable safety,and high tissue penetration depth,which,unfortunately,suffers from compromised therapeutic efficacy due to cancer cell-inherent adaptive mechanisms,such as glutathione(GSH)neutralization response to reactive oxygen species(ROS),and glutamine addictive properties of *** this work,we developed a biological sonosensitive platelet(PLT)pharmacytes for favoring US/GSH-responsive combinational therapeutic of glutamine deprivation and augmented *** amino acid transporter SLC6A14 blockade agentα-methyl-DL-tryptophan(α-MT)-loaded and MnO_(2)-coated porphyrinic metal-organic framework(MOF)nanoparticles were encapsulated in the PLTs through the physical adsorption of electrostatic attraction and the intrinsic endocytosis of *** the sonosensitive PLT pharmacytes reached tumor sites through their natural tendencies to TME,US stimulated the PLTs-loaded porphyrinic MOF to generate ROS,resulting in morphological changes of the PLTs and the release of ***,intracellular high concentration of GSH and extracellular spatio-temporal controlled US irradiation programmatically triggered the release ofα-MT,which enabled the synergistically amplified SDT by inducing amino acid starvation,inhibiting mTOR,and mediating *** addition,US stimulation achieved the targeted activation of PLTs at tumor vascular site,which evolved from circulating PLTs to dendritic PLTs,effectively blocking the blood supply of tumors through thrombus formation,and revealing the encouraging potential to facilitate tumor therapeutics.

It is essential to explore high efficient catalysts for nitrogen reduction in ammonia *** on the first-principles calculation,we find that B/g-C_(2)N can serve as high performance photocatalyst in N_(2)fixation,where single boron atom is anchored on the g-C_(2)N to form B/g-C_(2)*** the introduction of B atom to g-C_(2)N,the energy gap reduces from 2.45 eV to 1.21 eV and shows strong absorption in the visible light *** addition,N_(2)can be efficiently reduced on B/g-C_(2)N through the enzymatic mechanism with low onset potential of 0.07 V and rate-determining barrier of 0.50 ***"acceptance-donation"interaction between B/g-C_(2)N and N_(2)plays a key role to active N_(2),and the BN_(2)moiety of B/g-C_(2)N acts as active and transportation *** activity originates from the strong interaction between 1π1π*orbitals of N_(2)and molecular orbitals of B/g-C_(2)N,the ionization of 1πorbital and the filling of 1π*orbital can increase the N≡N bond length greatly,making the activation of N_(2).Overall,this work demonstrates that B/g-C_(2)N is a promising photocatalyst for N_(2)fixation.

Increasing importance has been placed on particle shape implementation within discrete element mod elling （DEM） in order to more accurately reflect the nonspherical behaviour of the bulk material being handled. As computational resources grow, complex particle shapes are increasingly being modelled as the associated simulation times become more realistic to provide timely solutions. The objective of this research is to assess particle shape descriptors through a digital image segmentation technique, and to further implement particle shape parameters into generation of corresponding irregular shaped DEM particles. Separated and lumped particle images were analysed and reconstructed through the devel opment of two distinct methodologies. Subsequently, various particle shape descriptors were obtained using combinations of image segmentation algorithms, including mathematical morphology processing, thresholding, edge detection, region growing, region splitting and region merging. DEM particles were subsequently created using particle shape results obtained above. Shape parameters of DEM particles were then examined and validated against the real particle shape parameters.

A mathematical model of friction coefficient was proposed for the roll force calculation of hot-rolled strips. The online numerical solving method of the roll force calculation formula based on the proposed friction model was developed and illustrated by the practical calculation case. Then, the friction coefficient during hot strip rolling was estimated from the measured roll force by force model inversion. And then, the expression of friction model was pro posed by analyzing the calculation process of stress state coefficient, and the model parameters were determined by the shared parameter multi-model nonlinear optimization method. Finally, the industrial experiments demonstrated the feasibility and effectiveness of the related models. The accuracy of the new roll force model based on the built friction model was much higher than that of the traditional Sims model, and it could be applied in the online hot rolling process control.

Sensitive detection of cancer biomarker microRNAs （miRNAs） is of vital importance for cancer diagnosis and treatment. Nonetheless, the detection sensitivity in the existing miRNA bioassays is severely limited by the structural characteristics of miRNA （including small length and high sequence homology） because most of these methods are based on target amplification. Herein, we report a novel approach to sensitive and specific detection of low-abundance miRNA via a unique strategy of nanoprobe dissolution-enhanced fluorescence amplification, in which a capture probe featuring molecular beacon structure is designed. By means of this strategy, miRNA-21 was detected in a linear range from 10 fM to 100 pM with a detection limit as low as 1.38 fM. High selectivity of the newly developed biosensor was demonstrated by the good discrimination against a target with a single-base mismatch. Furthermore, this assay was used for the detection of miRNA-21 added into fetal bovine serum samples with the recovery in the range of 90.2%--108% and coefficients of variation below 10.1%, indicating its promising applications to RNA immunoassays and early cancer diagnosis.

Experiment and Simulation for Rolling of Diamond–Cu Composites

We demonstrate an innovative preparation approach of diamond/Cu composites by powder-in-tube technique and rolling. A small copper tube was loaded with Ti- and Cu-coated diamond particles, mad then the diamond particles were combined with Cu matrix by composite rolling. The morphology and element distribution of the interface between diamond and Cu were determined by scanning electron microscopy and energy-dispersive spectrometer. Finite element method （FEM） simulation was used to analyze the rolling process associated with experiment by DEFORM-3D. The final experimental results showed that homogeneous distribution of diamond particles could be observed in the center layer of the composites. According to the contrast experiments, the sample, whose diamond particle size is 0.12-0.15 mm and thickness of pre-rolling is 1.2 mm, showed relatively complete morphologies and homogeneous distribution. Experimental results indicated a poor efficacy of excessive rolling reduction. The thermal conductivity of the composites is about 453 W （m K）-1 by theoretical calculation. For FEM simulation, roiling strain and temperature field of the composites were simulated by DEFORM-3D. Simulation results were interpreted, and numerical results verified the reliability of the model. The simulation predicted that the local area of large strain, indicative of the strain along the thickness direction, could be intensified by adding diamond particles.

White mold of pea caused by Sclerotinia sclerotiorum is a common disease in ***,we discovered that the diverse Sclerotinia species could cause white mold on pea plants in Chongqing and Sichuan of China during recent disease ***,the objective of this study was to confirm the causal agents from diseased pea *** obtained isolates of white mold from Chongqing and Sichuan were identified by morphological characters and molecular characterization to determine the pathogen species,and their pathogenicity was confirmed on pea through completing Koch’s *** isolates of Sclerotinia-like were obtained from diseased plants or *** on morphological characteristics and molecular characterization,30 isolates were identified to three species,six isolates as ***,seven as ***,and 17 as *** pathogenicity tests on pea cultivars Zhongwan 4 and Longwan 1,all 30 isolates caused typical symptoms of white mold on the inoculated plants,and the inoculated pathogens were re-isolated from the diseased *** study confirmed that white mold of pea was caused by three Sclerotinia species,***,*** and *** in Chongqing and *** is the first report that *** and *** cause white mold of pea in Southwest China.
