Curcumin Inhibits Lipopolysaccharide-lnduced Mucin 5AC Hypersecretion and Airway Inflammation via Nuclear Factor Erythroid 2-Related Factor 2

Background： Excess mucus production is an important pathophysiological feature of chronic inflammatory airway diseases. Effective therapies are currently lacking. The aim of the study was to evaluate the effects ofcurcumin （CUR） on lipopolysaccharide （LPS）-induced mucus secretion and inflammation, and explored the underlying mechanism in vivo and in vitro. Methods： For the in vitro study, human bronchial epithelial （NCI-H292） cells were pretreated with CUR or vehicle for 30 min, and then exposed to LPS for 24 h. Next, nuclear factor erythroid 2-related factor 2 （Nrf2） was knocked down with Nrf2 small interfering RNA （siRNA） to confirm the specific role of Nrf2 in mucin regulation of CUR in NCI-H292 cells. In vivo, C57BL/6 mice were randomly assigned to three groups （n = 7 for each group）： control group, LPS group, and LPS ＋ CUR group. Mice in LPS and LPS ＋ CUR group were injected with saline or CUR （50 mg/kg） intraperitoneally 2 h before intratracheal instillation with LPS （ 100 μg/ml） for 7 days. Cell lysate and lung tissue were obtained to measured Mucin 5AC （MUC5AC） and Nrf2 mRNA and protein expression by a real-time polymerase chain reaction and Western blotting. Bronchoalveolar lavage fluid （BALF） was collected to enumerate total cells and neutrophils. HistopathologicaI changes of the lung were observed. Data were analyzed by one-way analysis of variance. Student＇s t-test was used when two groups were compared. Results： CUR significantly decreased the expression ofMUC5AC mRNA and protein in NCI-H292 cells exposed to LPS. This effect was dose dependent （2.424 ± 0.318 vs. 7.169 ± 1.785, t = 4.534, and 1.060 ± 0.197 vs. 2.340 ± 0.209, t = 7.716; both P 〈 0.05, respectively） and accompanied by increased mRNA and protein expression of Nrf2 （1.952 ± 0.340 vs. 1.142 ± 0.176, t = -3.661, and 2.010 ± 0.209 vs. 1.089 ±0. 132, t = -6.453; both P 〈 0.05, respectively）. Objective: : To explore the value of a combined computed tomography(CT) and magnetic resonance imaging(MRI) in evaluating profound sensorineural deafness patients before cochlear implant(CI) surgery.Methods: A retrospective analysis of 1012 cases of profound sensorineural deafness that received CI was performed.Results: A total of 96 cases were diagnosed with inner ear abnormalities including large vestibular aqueduct syndrome(LVAS, n ? 61), Michel deformity(n ? 3), cochlear incomplete partition I(n ? 2), cochlear incomplete partition II(n ? 6), cochlear hypoplasia with vestibular malformation(n ? 3), cochlear ossification(n ? 3), bilateral internal auditory canal obstruction(n ? 5) and internal auditory canal stenosis(n ? 2).Conclusion: High resolution CT(HRCT) can display bony structures while MRI can image the membranous labyrinth in preoperative evaluation for cochlear implantation. The combination of these two modalities provides reliable anatomical information regarding the bony and membranous labyrinths, as well as the auditory nerve.

The global spread of severe acute respiratory syndrome coronavirus 2 has resulted in a significant number of individuals developing pulmonary fibrosis(PF),an irreversible lung injury.This condition can manifest within a short inter-val following the onset of pneumonia symptoms,sometimes even within a few days.While lung transplantation is a potentially lifesaving procedure,its limited availability,high costs,intricate surgeries,and risk of immunological rejection present significant drawbacks.The optimal timing of medication administration for coronavirus disease 2019(COVID-19)-induced PF remains controversial.Despite this,it is crucial to explore pharmacotherapy interventions,involving early and preventative treatment as well as pharmacotherapy options for advanced-stage PF.Additionally,studies have demonstrated disparities in anti-fibrotic treatment based on race and gender factors.Genetic mutations may also impact therapeutic efficacy.Enhancing research efforts on pharmacotherapy interventions,while considering relevant pharmacological factors and optimizing the timing and dosage of medication administration,will lead to enhanced,personalized,and fair treatment for individuals impacted by COVID-19-related PF.These measures are crucial in lessening the burden of the disease on healthcare systems and improving patients'quality of life.

Chain Dynamics Heterogeneity in Plasticized Poly(vinyl butyral)(PVB) as Elucidated by Solid-State NMR

The chain dynamics heterogeneity of the poly(vinyl butyral)(PVB) plasticized by triethylene glycol bis(2-ethylhexa noate)(TEG-EH) was investigated by various solid-state NMR techniques.The plasticized PVB shows two domains in distinct molecular dynamics differences,namely,rigid and soft domains,where the latter is the plasticizer-rich domain.The time domain low field NMR was first used to investigate the dynamics heterogeneity of the plasticized PVB,and the results show the decreasing activated energy of components in the soft domain of plasticized PVB(E_a=20.2 kJ/mol) as compared with that of the pristine one(E_a=24.3 kJ/mol).Detailed dynamics heterogeneity was obtained by high-field NMR with site-specific features.The quadrupole-echo ~2H-NMR was adopted to elucidate the dynamics heterogeneity of the vinyl alcohol(VA) units,where only the hydroxyl group of VA is deuterated.The ~1H-^(13)C WISE NMR spectra show that there is not much difference in the mobility of the VB unit in PVB with and without plasticizer,whereas the glass transition temperature differed by approximately 53℃.This is further supported by Torchia's T_1 relaxation measurements.The origin of such an unusual phenomenon is attributed to the critical role of the remaining VA(~22%) in the soft domain,where the VA units locally aggregate through hydrogen bonding.Also,the existence of a mobility gradient in the VB unit has been demonstrated.Moreover,the mobility difference for VB with different stereo-geometry(meso or racemic conformation) is observed for the first time.This indicates the importance of modulating the ratio of meso over racemic VB for controlling the macroscopic perfo rmance of PVB.

In engineering application,there is only one adaptive weights estimated by most of traditional early warning radars for adaptive interference suppression in a pulse reputation interval(PRI).Therefore,if the training samples used to calculate the weight vector does not contain the jamming,then the jamming cannot be removed by adaptive spatial filtering.If the weight vector is constantly updated in the range dimension,the training data may contain target echo signals,resulting in signal cancellation effect.To cope with the situation that the training samples are contaminated by target signal,an iterative training sample selection method based on non-homogeneous detector(NHD)is proposed in this paper for updating the weight vector in entire range dimension.The principle is presented,and the validity is proven by simulation results.

High-pressure minerals and new lunar mineral changesite-(Y) in Chang’e-5 regolith

Forty-five years after the Apollo and Luna missions, China’s Chang’e-5 (CE-5) mission collected ∼1.73 kg of new lunar materials from one ofthe youngest basalt units on the Moon. The CE-5 lunar samples provide opportunities to address some key scientific questions related to theMoon, including the discovery of high-pressure silica polymorphs (seifertite and stishovite) and a new lunar mineral, changesite-(Y). Seifertitewas found to be coexist with stishovite in a silica fragment from CE-5 lunar regolith. This is the first confirmed seifertite in returned lunarsamples. Seifertite has two space group symmetries (Pnc2 and Pbcn) and formed from an α-cristobalite-like phase during “cold” compressionduring a shock event. The aftershock heating process changes some seifertite to stishovite. Thus, this silica fragment records different stagesof an impact process, and the peak shock pressure is estimated to be ∼11 to 40 GPa, which is much lower than the pressure condition forcoexistence of seifertite and stishovite on the phase diagram. Changesite-(Y), with ideal formula (Ca_(8)Y)◻Fe^(2+)(PO_(4))_(7) (where ◻ denotes avacancy) is the first new lunar mineral to be discovered in CE-5 regolith samples. This newly identified phosphate mineral is in the form ofcolumnar crystals and was found in CE-5 basalt fragments. It contains high concentrations of Y and rare earth elements (REE), reaching upto ∼14 wt. % (Y,REE)2O3. The occurrence of changesite-(Y) marks the late-stage fractional crystallization processes of CE-5 basalts combinedwith silicate liquid immiscibility. These new findings demonstrate the significance of studies on high-pressure minerals in lunar materials andthe special nature of lunar magmatic evolution.

Regression Method for Rail Fastener Tightness Based on Center-Line Projection

In the railway system,fasteners have the functions of damping,maintaining the track distance,and adjusting the track level.Therefore,routine maintenance and inspection of fasteners are important to ensure the safe operation of track lines.Currently,assessment methods for fastener tightness include manual observation,acoustic wave detection,and image detection.There are limitations such as low accuracy and efficiency,easy interference and misjudgment,and a lack of accurate,stable,and fast detection methods.Aiming at the small deformation characteristics and large elastic change of fasteners from full loosening to full tightening,this study proposes high-precision surface-structured light technology for fastener detection and fastener deformation feature extraction based on the center-line projection distance and a fastener tightness regression method based on neural networks.First,the method uses a 3D camera to obtain a fastener point cloud and then segments the elastic rod area based on the iterative closest point algorithm registration.Principal component analysis is used to calculate the normal vector of the segmented elastic rod surface and extract the point on the centerline of the elastic rod.The point is projected onto the upper surface of the bolt to calculate the projection distance.Subsequently,the mapping relationship between the projection distance sequence and fastener tightness is established,and the influence of each parameter on the fastener tightness prediction is analyzed.Finally,by setting up a fastener detection scene in the track experimental base,collecting data,and completing the algorithm verification,the results showed that the deviation between the fastener tightness regression value obtained after the algorithm processing and the actual measured value RMSE was 0.2196 mm,which significantly improved the effect compared with other tightness detection methods,and realized an effective fastener tightness regression.

Cellular senescence-driven transcriptional reprogramming of the MAFB/NOTCH3 axis activates the PI3K/AKT pathway and promotes osteosarcoma progression

Osteosarcoma is the most common primary malignancy of bones and primarily occurs in adolescents and young adults.However,a second smaller peak of osteosarcoma incidence was reported in the elderly aged more than 60.Elderly patients with osteosarcoma exhibit different characteristics compared to young patients,which usually results in a poor prognosis.The mechanism underlying osteosarcoma development in elderly patients is intriguing and of significant value in clinical applications.Senescent cells can accelerate tumor progression by metabolic reprogramming.Recent research has shown that methylmalonic acid(MMA)was significantly up-regulated in the serum of older individuals and played a central role in the development of aggressive characteristics.We found that the significant accumulation of MMA in elderly patients imparted proliferative potential to osteosarcoma cells.The expression of MAFB was excessively up-regulated in osteosarcoma specimens and was further enhanced in response to MMA accumulation as the patient aged.Specifically,we first confirmed a novel molecular mechanism between cellular senescence and cancer,in which the MMA-driven transcriptional reprogramming of the MAFB-NOTCH3 axis accelerated osteosarcoma progression via the activation of PI3K-AKT pathways.Moreover,the down-regulation of the MAFB-NOTCH3 axis increased the sensitivity and effect of AKT inhibitors in osteosarcoma through significant inhibition of AKT phosphorylation.In conclusion,we confirmed that MAFB is a novel age-dependent biomarker for osteosarcoma,and targeting the MAFB-NOTCH3 axis in combination with AKT inhibition can serve as a novel therapeutic strategy for elderly patients with osteosarcoma in experimental and clinical trials.

Bioinspired Stretchable MXene Deformation-Insensitive Hydrogel Temperature Sensors for Plant and Skin Electronics

Temperature sensing is of high value in the wearable healthcare,robotics/prosthesis,and noncontact physiological monitoring.However,the common mechanic deformation,including pressing,bending,and stretching,usually causes undesirable feature size changes to the inner conductive network distribution of temperature sensors,which seriously influences the accuracy.Here,inspired by the transient receptor potential mechanism of biological thermoreceptors that could work precisely under various skin contortions,we propose an MXene/Clay/poly(N-isopropylacrylamide)(PNIPAM)(MCP)hydrogel with high stretchability,spike response,and deformation insensitivity.The dynamic spike response is triggered by the inner conductive network transformation from the 3-dimensional structure to the 2-dimensional surface after water being discharged at the threshold temperature.The water discharge is solely determined by the thermosensitivity of PNIPAM,which is free from mechanical deformation,so the MCP hydrogels can perform precise threshold temperature(32℃)sensing under various deformation conditions,i.e.,pressing and 15%stretching.As a proof of concept,we demonstrated the applications in plant electronics for the real-time surface temperature monitoring and skin electronics for communicating between human and machines.Our research opens venues for the accurate temperature-threshold sensation on the complicated surface and mechanical conditions.

Three dimensional velocity model and its tectonic implications at China Seismic Experimental Site,eastern margin of the Tibetan Plateau

The China Seismic Experimental Site(CSES)is located at the intersection of the Tibetan Plateau,South China Block,and Indian Plate and has complex geological settings and intense crustal deformation,making it one of the most seismically active areas in Chinese mainland.A high-resolution,three-dimensional(3D)crust-mantle velocity structure is crucial for understanding seismotectonic environments,lithospheric deformation mechanisms,and deep dynamic processes.We first constructed a high-vertical-resolution 3D initial velocity model using the joint inversion of receiver functions and surface waves and then obtained a 3D P-and S-wave velocity model(CSES-VM1.0)with the highest lateral resolution of 0.25°for the CSES using double-difference tomography.Owing to the limitations of the Sn observation data,the resolution of the S-wave velocity model in the lower crust and upper mantle was reduced,making it closer to the initial model provided by joint inversion.A comparison with explosive-source seismic data showed that the synthetic P-wave first-arrival travel times of the new model were closer to the observations than those of the previous velocity models.The velocity cross-sections across the source areas of the 2022 Lushan MS6.1 and Ludian MS6.8 earthquakes reveal that the former earthquake occurred near a weak contact zone between the Tibetan Plateau and Sichuan Basin,and the rupture of the latter earthquake occurred in a granitic area,with the northern end blocked by rigid high-velocity bodies.A clear high-velocity anomaly zone is distributed along the western margin of the Yangtze Block,revealing the spatial distribution of Neoproterozoic intermediate-basic intrusions.This high-velocity zone significantly controls the morphology of fault zones and influences the rupture processes of major earthquakes.Two northeast-southwest and north-south trending high-velocity anomalies were found near Panzhihua,potentially related to Neoproterozoic and Middle-Late Permian intermediate-basic intrusions.The imaging results revealed t
