Effect of 5-Aza-2’-deoxycytidine on immune-associated proteins in exosomes from hepatoma

AIM: To study the effect of 5-Aza-2’-deoxycytidine (5-Aza-CdR) on heat shock protein 70 (HSP70), human leucocyte antigen-Ⅰ (HLA-Ⅰ) and NY-ESO-1 proteins in exosomes produced by hepatoma cells, Hepg2 and Hep3B. METHODS: Exosomes derived from Hepg2 and Hep3B cells treated with or without 5-aza-CdR were isolated and purified by ultrafiltration centrifugation and sucrose gradient ultracentrifugation. The number of exosomes was counted under electron microscope. Concentration of proteins in exosomes was measured by bicinchoninic acid protein assay. Expression of HSP70, HLA-Ⅰ and NY-ESO-1 proteins in exosomes was detected by Western blotting and immunoelectron microscopy. mRNA expression of p53 gene was detected by reverse transcription polymerase chain ***: The mRNA expression of p53 gene was increased in both hepatoma cell lines after treatment with 5-Aza-CdR. The number of exosomes and the concentration of total proteins in exosomes were increased signifi cantly after treatment with 5-aza-CdR (P g showed that the HSP70, HLA-Ⅰ and NY-ESO-1 proteins were increased in exosomes produced by both hepatoma cell lines. CONCLUSION: 5-aza-CdR, an inhibitor of DNA methyltransferase, can increase exosomes produced by hepatoma cells and immune-associated protein component of exosomes, which may be mediated by p53 gene upregulation and 5-Aza-CdR demethylation.

Practical implementations of rechargeable lithium(Li)metal batteries have long been plagued by multiple problems of Li anode,such as Li dendrite growth,large volume change,low Coulombic ***,we report a protein-enabled film that can provide effective protection for Li *** protective film with an integrated design of high flexibility,strong adhesion and high Li-ion transference number(0.80)is fabricated by incorporating denatured zein(corn protein)with polyethylene oxide(PEO)acting as an age nt for sustaining the denatured protein chains against refolding via the intermolecular interactions between ***,a conformable zein-enabled protective film(zein@PEO)with simultaneous en hancement in flexibility,modulus and adhesion strength is gen erated to offer both functi ons of self-adapting and anion-anchoring *** results show that the zein@PEO film is able to accommodate the volume change,reduce the side reactions,and homogenize the ion *** from these significant properties/fu nctions,the Li/Cu cell with the zein@PEO film delivers prolonged cycle life for over 500 hours with stable *** with LiMn_(2)O_(4) cathode,the capacity,cycle stability and rate performance of the cell are remarkably improved as well,demonstrati ng the effectiveness in stabilizing Li metal batteries.

Microblade assemblages are among the most common prehistoric archaeological materials found on the Tibetan Plateau(TP)and are thought to indicate large scale migration to and settlement of the *** microblade sites,however,have been systematically excavated,especially in the remotest,highest-elevation regions of the *** timing of the large-scale arrival,spread,and permanent settlement of people on the TP therefore remains *** this paper,we report on a recently excavated site,Locality 3 of the Nwya Devu Site(ND3),located at 4600 meters above sea level(masl),near the shore of Ngoin Lake,on the interior *** analyses reveal a fairly typical microblade technological orientation and two types of microblade cores:wedge-shaped and semi-conical,which are similar to those found throughout North *** Optically Stimulated Luminescence(OSL)dating and AMS^(14)C dating,the age of ND3 ranges from 11 to 10 *** date range indicates ND3 is the oldest microblade site yet recorded in the remote,high-elevation regions of the TP and thus provides important information about when and how hunter-gatherers using microblades began exploiting the higher altitudes of the *** together,studies at ND3 and throughout the TP suggest that a microblade adaptation is associated with the first prolonged human occupation of the plateau and that microblades played a significant role in mediating the risks and facilitating the mobility necessary to permanently inhabit the TP.

The expanding cost of ordinary fuel in urban zones requires the investigation of other vitality *** exploitation of agricultural wastes for biogas production could protect our environment and can also solve emerging energy crises *** chrysosporium,Trichoderma virdie and Trichoderma harzianum were used to ferment rice straw and cattle dung for production of compost and *** yield was determined using displacement technique,using acidified water(1 N H_(2)SO_(4))to prevent solubilization of carbon dioxide contained in the *** results showed that the highest rate of biogas and methane were recorded with treated rice straw mixed with cattle dung where the amount of production from the total gas was 51.84 of biogas and 37.86 of methane per Liter(L)/Kilogram(Kg)of fermented ***,the results showed that the consumed biogas and methane related to volatile solids gave the highest amount with untreated rice straw(86.32 and 56.72 L/Kg respectively)followed by treated rice straw and untreated and treated mixtures of rice straw and cattle dung *** results also showed that the highest bacterial count was obtained from fungi treated rice straw mixed with cattle dung(125×10^(3)cells/g)followed by treated rice straw(32×10^(3)cells/g).This study confirms that ***,*** and *** significantly speed up the process of aerobic and anaerobic fermentation of rice straw in respect to biogas and methane production.

Ni-based anodes of SOFCs are susceptible to coking, which greatly limits practical application of direct methane-based fuels. An indirect internal reformer is an effective way to convert methane-based fuels into syngas before they reach anode. In this work, catalytic activity of a redox-stable perovskite La0.7Sr0.3Cr0.8Fe0.2O3-δ(LSCrFO) for methane conversion was evaluated. The catalyst was fabricated as an anodic protective layer to improve coking resistance of a Ni cermet anode. Using wet CH4 as a fuel, the LSCrFO-modified cell showed excellent power output and good coking resistance with peak power density of 1.59 W cm-2 at 800℃. The cell demonstrated good durability lasting for at least 100 h. While the bare cell without the protective layer showed poor durability with the cell voltage fast dropped from 0.75 V to 0.4 V within 30 min. Under wet coal bed methane (CBM) operation, obvious performance degradation within 35 h (1.7 mV h^-1) was observed due to the influence of heavy carbon compounds in CBM. The pre-and post-mortem microstructures and carbon analysis of the anode surface and catalyst surface were further conducted.

A glasshouse experiment was conducted using a root-bag technique to study the root exudates, rhizosphere Zn fractions, and Zn concentrations and accumulations of two ryegrass cultivars （Lolium perenne L. cvs. Airs and Tede） at different soil Zn levels （0, 2, 4, 8, and 16 mmol kg^-1 soil）. Results indicated that plant growth of the two cultivars was not advérsely affected at soil Zn level ≤ 8 mmol kg^-1. Plants accumulated more Zn as soil Zn levels increased, and Zn concentrations of shoots were about 540 μg g^-1 in Aris and 583.9 μg g^-1 in Tede in response to 16 mmol Zn kg^-1 soil. Zn ratios of shoots to roots across the soil Zn levels were higher in Tede than in Airs, corresponding with higher rhizosphere available Zn fractions （exchangeable, bound to manganese oxides, and bound to organic matter） in Airs than in Tede. Low-molecular-weight （LMW） organic acids （oxalic, tartaric, malic, and succinic acids） and amino acids （proline, threonine, glutamic acid, and aspartic acid, etc.） were detected in root exudates, and the concentrations of LMW organic acids and amino acids increased with addition of 4 mmol Zn kg^-1 soil compared with zero Zn addition. Higher rhizosphere concentrations of oxalic acid, glutamic acid, alanine, phenylalanine, leucine, and proline in Tede than in Airs likely resulted in increased Zn uptake from the soil by Tede than by Airs. The results suggested that genotypic differences in Zn accumulations were mainly because of different root exudates and rhizosphere Zn fractions.
