Intracerebral hemorrhage is often accompanied by oxidative stress induced by reactive oxygen species,which causes abnormal mitochondrial function and secondary reactive oxygen species *** creates a vicious cycle leading to reactive oxygen species accumulation,resulting in progression of the pathological ***,breaking the cycle to inhibit reactive oxygen species accumulation is critical for reducing neuronal death after intracerebral *** previous study found that increased expression of nicotinamide adenine dinucleotide phosphate oxidase 4(NADPH oxidase 4,NOX4)led to neuronal apoptosis and damage to the blood-brain barrier after intracerebral *** purpose of this study was to investigate the role of NOX4 in the circle involving the neuronal tolerance to oxidative stress,mitochondrial reactive oxygen species and modes of neuronal death other than apoptosis after intracerebral *** found that NOX4 knockdown by adeno-associated virus(AAV-NOX4)in rats enhanced neuronal tolerance to oxidative stress,enabling them to better resist the oxidative stress caused by intracerebral *** of NOX4 also reduced the production of reactive oxygen species in the mitochondria,relieved mitochondrial damage,prevented secondary reactive oxygen species accumulation,reduced neuronal pyroptosis and contributed to relieving secondary brain injury after intracerebral hemorrhage in ***,we used a mitochondria-targeted superoxide dismutase mimetic to explore the relationship between reactive oxygen species and *** mitochondria-targeted superoxide dismutase mimetic inhibited the expression of NOX4 and neuronal pyroptosis,which is similar to the effect of *** indicates that NOX4 is likely to be an important target for inhibiting mitochondrial reactive oxygen species production,and NOX4 inhibitors can be used to alleviate oxidative stress response induced by intracerebral hemorrhage.

The evolution of solar magnetic fields is significant for understanding and predicting solar *** our knowledge of solar magnetic fields largely depends on the photospheric magnetic *** this paper,based on the spherical harmonic expansion of the photospheric magnetic field observed by Wilcox Solar Observatory,we analyze the time series of spherical harmonic coefficients and predict Sunspot Number as well as synoptic maps for Solar Cycle *** find that solar maximum years have complex short-period disturbances,and the time series of coefficient g_(7)~0 is nearly in-phase with Sunspot Number,which may be related to solar meridional *** Long Short-Term Memory networks(LSTM),our prediction suggests that the maximum of Solar Cycle 25 is likely to occur in June 2024 with an error of 8 months,the peak sunspot number may be 166.9±22.6,and the next solar minimum may occur around January *** incorporating Empirical Mode Decomposition,we enhance our forecast of synoptic maps truncated to Order 5,validating their relative *** prediction not only addresses a gap in forecasting the global distribution of the solar magnetic field but also holds potential reference value for forthcoming solar observation plans.

Exploitation of oxygen evolution reaction(OER)and urea oxidation reaction(UOR)catalysts with high activity and stability at large current density is a major challenge for energy-saving H_(2) production in water ***,we use the pyridinic-N doping carbon layers coupled with tensile strain of FeNi alloy activated by NiFe_(2)O_(4)(FeNi/NiFe_(2)O_(4)@NC)for efficiently increasing the performance of water and urea *** to the tensile strain effect on FeNi/NiFe_(2)O_(4)@NC,it provides a favorable modulation on the electronic properties of the active center,thus enabling amazing OER(η_(100)=196 mV)and UOR(E_(10)=1.32 V)intrinsic ***,the carbon-coated layers can be used as armor to prevent FeNi alloy from being corroded by the electrolyte for enhancing the OER/UOR stability at large current density,showing high industrial *** work thus provides a simple way to prepare high-efficiency catalyst for activating water and urea oxidation.

Anti-psoriasis molecular targets and active components discovery of Optimized Yinxieling Formula via affinity-purified strategy

Psoriasis,a prevalent inherited skin condition,involves an inflammatory response as a key pathogenic *** Optimized Yinxieling Formula(OYF),rooted in traditional Chinese medicine,is extensively utilized in clinical settings to treat *** previous studies have demonstrated OYF’s significant anti-inflammatory effects in psoriasis,its potential molecular targets and active components remain *** study aimed to unveil the anti-psoriasis molecular targets and active components of *** findings indicated that OYF extract markedly reduced the production of several inflammatory mediators,including IL-23,nitric oxide,TNF-α,and IL-1β,in LPS-induced RAW264.7 *** synthesized OYF extract-crosslinked beads to isolate pharmacological targets from RAW264.7 lysates using an affinity purification strategy,known as Target *** enriched target proteins were subsequently identified via LC-MS/MS,followed by bioinformatics analysis to map the psoriasis-associated pathway-gene *** identified a total of 76 potential target proteins,which were highly associated with mRNA transcription *** particular,pathway-gene network analysis revealed that the IL-23 inflammatory pathway was involved in the anti-psoriasis effect of OYF *** further utilized a target protein-based affinity capture strategy,combined with LC-MS and SPR analysis,to globally screen OYF’s active components,focusing on the mRNA transcription regulator,fused in sarcoma(FUS).This process led to the identification of umbelliferone,vanillic acid,protocatechuic acid,gentisic acid,and echinacoside as key compounds targeting FUS to inhibit IL-23 ***,we formulated a compound cocktail(CpdC),which significantly reduced psoriasis area and severity index(PASI)scores and the expressions of IL-23 and Ki67 in an imiquimod(IMQ)-induced psoriasis mouse ***,our study elucidates the primary molecular targets and active components of OYF,offeri

Objective To explore the genotyping characteristics of human fecal Escherichia coli(E. coli) and the relationships between antibiotic resistance genes(ARGs) and multidrug resistance(MDR) of E. coli in Miyun District, Beijing, an area with high incidence of infectious diarrheal cases but no related *** Over a period of 3 years, 94 E. coli strains were isolated from fecal samples collected from Miyun District Hospital, a surveillance hospital of the National Pathogen Identification Network. The antibiotic susceptibility of the isolates was determined by the broth microdilution method. ARGs,multilocus sequence typing(MLST), and polymorphism trees were analyzed using whole-genome sequencing data(WGS).Results This study revealed that 68.09% of the isolates had MDR, prevalent and distributed in different clades, with a relatively high rate and low pathogenicity. There was no difference in MDR between the diarrheal(49/70) and healthy groups(15/24).Conclusion We developed a random forest(RF) prediction model of TEM.1 + baeR + mphA + mphB +QnrS1 + AAC.3-IId to identify MDR status, highlighting its potential for early resistance identification. The causes of MDR are likely mobile units transmitting the ARGs. In the future, we will continue to strengthen the monitoring of ARGs and MDR, and increase the number of strains to further verify the accuracy of the MDR markers.

Animal body size variation is of particular interest in evolutionary biology,but the genetic basis remains largely *** studies have shown the presence of two parallel evolutionary genetic clusters within the fish genus Epinephelus with evident divergence in body size,providing an excellent opportunity to investigate the genetic basis of body size variation in ***,we performed phylotranscriptomic analysis and reconstructed the phylogeny of 13 epinephelids originating from the South China *** genetic clades with an estimated divergence time of approximately 15.4 million years ago were correlated with large and small body size,respectively.A total of 180 rapidly evolving genes and two positively selected genes were identified between the two *** enrichment analyses of these candidate genes revealed distinct enrichment categories between the two *** pathways and genes may play important roles in body size variation in groupers through complex regulatory *** on our results,we speculate that the ancestors of the two divergent groups of groupers may have adapted to different environments through habitat selection,leading to genetic variations in metabolic patterns,organ development,and lifespan,resulting in body size divergence between the two locally adapted *** findings provide important insights into the genetic mechanisms underlying body size variation in groupers and species differentiation.

Progress in the applications of flux analysis of metabolic networks

The metabolic network has become a hot topic in the area of system biology and flux-based analysis plays a very important role in understanding the characteristics of organism metabolic networks. We review mainly the static methods for analyzing metabolic networks such as flux balance analysis (FBA), minimization of metabolic adjustment (MOMA), regulatory on / off minimization (ROOM), and dynamic flux balance analysis with linear quadratic regulator (DFBA-LQR). Then several kinds of commonly used software for flux analysis are introduced briefly and compared with each other. Finally, we highlight the applications of metabolic network flux analysis, especially its usage combined with other biological characteristics and its usage for drug design. The idea of combining the analysis of metabolic networks and other biochemical data has been gradually promoted and used in several aspects such as the combination of metabolic flux and the regulation of gene expression, the influence of protein evolution caused by metabolic flux, the relationship between metabolic flux and the topological characteristics, the optimization of metabolic engineering. More comprehensive and accurate properties of metabolic networks will be obtained by integrating metabolic flux analysis, network topological characteristics and dynamic modeling.
