High drug loading and pH-responsive nanomedicines driven by dynamic boronate covalent chemistry for potent cancer immunotherapy

Tumor cells undergoing immunogenic cell death (ICD) have emerged as an in situ therapeutic vaccine helping to activate a persistent anti-tumor response. Several chemotherapeutic agents have been demonstrated to induce ICD, however accompanied with severe adverse effects in the clinic, weakening its immune responses. Herein, to elicit an intensive ICD while minimizing the systemic toxicity, we introduce a tumor targeting peptide modified bortezomib (BTZ) loading nanomedicine (i-NPBTZ) for the efficient delivery and controlled release of BTZ in tumors. This system is constructed by conjugating BTZ to PEGylated polyphenols via a pH-sensitive covalent boronate-phenol bond that allows them to self-assemble into nanovesicles in neutral condition with high drug loading efficiency. Once accumulated in acidic environment, BTZ-phenolic network is disassembled and thereby accelerates the release of BTZ from nanocarriers. The released BTZ selectively kill tumor cells with a concomitant evocation of tumor-specific cytotoxic T cells by triggering ICD in vivo. This can finally lead to an extended tumor ablation and prevention of distant metastasis in a syngeneic tumor mouse model, while reducing the systemic toxicity of BTZ. In general, our system offers a novel concept with clinical potential to exploit ICD for potentiating tumor immunotherapy and also provides an excellent example of the application of polymer-drug interaction for efficient drug delivery and controllable release.

To meet the growing demand for socioeconomic development,a large amount of groundwater is extracted from confined aquifers *** North China Plain has experienced considerable groundwater depletion and subsidence during the past six *** this study,we use Sentinel-1A/B SAR images from 2015 to 2020 to map the ground subsidence of the Tianjin–Langfang *** subsiding zones centered at Guangyang,Wuqing–Bazhou,and Jinghai are identified with maximum subsidence rates of 98.1,121.8,and 104.7 mm/*** and long-term signals are separated from time series subsidence and hydraulic measurements using continuous wavelet transform to retrieve aquifer *** long-term subsidence,which fits well with an exponential decaying model,remarkably slows down in our study *** elastic skeletal storage coefficients range between 0.52×10−3 and 9.66×10−*** then retrieve the spatial–temporal variations of total groundwater storage,recoverable groundwater storage,and irreversible groundwater *** storage depletion rates are apparently reducing,which benefits from the operation of the South-to-North Water Transfer Project and local groundwater management practices.

Comparative analysis of the ecological fitness and transcriptome between two genotypes of the brown planthopper Nilaparvata lugens

Shear stress-induced delamination method for the mass production of Ti_(3)C_(2)Tx MXene nanosheets

MXene nanosheets are considered advantageous for functional materials,but current delamination methods to prepare MXene nanosheets have many limitations including high cost,small production scale,low efficiency,and deteriorated structure integrity of obtained ***,we propose a simple,efficient,and scalable shear stress-induced delamination(SSID)strategy to boost the production of single-/few-layered Ti_(3)C_(2)T_(x) MXene *** dynamics simulation indicates that the pan mill-type grinding discs create a strong hydrodynamic flow field,which exerts gigantic shear stress to substantially delaminate the multilayered MXene stacks into homogeneously dispersed MXene ***,shear stress generated from vigorous water flow has limited fragmentation effect,ensuring large lateral size and good structure integrity to the obtained MXene nanosheets as evidenced by the morphological and structural *** to conventional delamination methods,this novel SSID strategy exhibits great advantages in terms of efficiency,scalability and the properties of resultant MXene nanosheets,which opens up great opportunity for the scalable production and commercialization of high-performance MXene-based materials.

An experiment on multibubble sonoluminescence spectra in sodium chloride solution

We investigated experimentally the spectra of MBSL in sodium chloride water solution with krypton as dissolved gas. We observed and compared the spectra of hydroxyl ion at 310 nm and that of sodium atom at 589 nm. It has been found that under the same experimental condition, the intensity of sodium atom spectra is obviously higher than that of the hydroxyl ion spectra, and is more sensitive to the experimental condition. The krypton content, the concentration of sodium chloride solution, and the driving sound pressure obviously affect the spectra intensity in certain range.

Tumor microenvironment has been widely utilized for advanced drug delivery in recent years,among which hypoxia-responsive drug delivery systems have become the research *** hypoxia-responsive micelles or polymersomes have been successfully developed,a type of hypoxia-degradable nanogel has rarely been reported and the advantages of hypoxia-degradable nanogel over other kinds of degradable nanogels in tumor drug delivery remain ***,we reported the synthesis of a novel hypoxia-responsive crosslinker and the fabrication of a hypoxia-degradable zwitterionic poly(phosphorylcholine)-based(HPMPC)nanogel for tumor drug *** obtained HPMPC nanogel showed ultra-long blood circulation and desirable immune compatibility,which leads to high and long-lasting accumulation in tumor ***,HPMPC nanogel could rapidly degrade into oligomers of low molecule weight owing to the degradation of azo bond in hypoxic environment,which leads to the effective release of the loaded ***,HPMPC nanogel showed superior tumor inhibition effect both in vitro and in vivo compared to the reduction-responsive phosphorylcholine-based nanogel,owing to the more complete drug ***,the drug-loaded HPMPC nanogel exhibits a pronounced tumor inhibition effect in a humanized subcutaneous liver cancer model with negligible side effects,which showed great potential as nanocarrier for advanced tumor drug delivery.

A long-term record(1995-2019)of the dynamics of land desertification in the middle reaches of Yarlung Zangbo River basin derived from Landsat data

Widespread desertification in the middle part of the Yarlung Zangbo River(YZR)basin is threatening the sustain-able development of this *** capture this process,a method was proposed for large-scale desertification monitoring by using Landsat images from 1995 to *** method used an integrated classification method combined with a hierarchical decision tree and nearest neighbor *** spatio-temporal dynamics of the desertification pattern were analyzed to assist in the detection of possible driving *** validation samples collected from Google Earth high-resolution images and field investigations,the overall accuracy of the classification in 2019 was 92.3%with a Kappa coefficient of *** major results were:(1)total sandy land area in 2019 was 734.1 km^(2),which accounted for 3.7%of the study area,prominently distributed along the wide river valleys and inlets of tributaries with a strip and discontinuous *** land tends to be distributed in the southern aspect regions with lower elevations and that are closer to rivers;(2)sandy land areas showed two temporal stages:a gradual increase of 102.4 km^(2)from 1995 to 2015 and a large decrease of 106.8 km^(2)from 2015 to 2019;(3)newly increased sandy land was distributed in the YZR Valley,while the revegetation on sandy land occurred mainly in the Lhasa River basin and some regions in the YZR Valley;and(4)increased sandy land area of 142.1 km^(2)was mainly distributed in the southern band of the two ***,revegetation on sandy land was more effective on the northern banks of the river *** findings provide guidance for implementing vegetation recovery on sandy lands and provide important insights for maintaining sustainable development.

Allergic asthma is an inflammatory disease of the airways characterized by recurrent episodes of wheezing,bronchoconstriction,and airway hyperresponsiveness(AHR)[1].In recent years,allergic asthma has become a heated public health problem because of the highly increased incidence and prevalence[2].The main pathogenesis of asthma is airway inflammation and *** is caused by an imbalance of Th1/Th2 cells with predominance to Th2,whereas airway inflammation is caused by increased levels of Th2 cytokine[3].Major effects of pathological changes in the respiratory include eosinophil infiltration,bronchial epithelial cell damage,obstruction of the mucus plug,and thickening of the submucosal muscle layer[4,5].
