Comprehensive analysis of the protein phosphatase 2A regulatory subunit B56ε in pan-cancer and its role and mechanism in hepatocellular carcinoma

BACKGROUND B56εis a regulatory subunit of the serine/threonine protein phosphatase 2A,which is abnormally expressed in tumors and regulates various tumor cell functions.At present,the application of B56εin pan-cancer lacks a comprehensive analysis,and its role and mechanism in hepatocellular carcinoma(HCC)are still unclear.The Cancer Genome Atlas,Genotype-Tissue Expression,Gene Expression Profiling Interactive Analysis,and Tumor Immune Estimation Resource databases were used to analyze B56εexpression,prognostic mutations,somatic copy number alterations,and tumor immune characteristics in 33 tumors.The relationships between B56εexpression levels and drug sensitivity,immuno-therapy,immune checkpoints,and human leukocyte antigen(HLA)-related genes were further analyzed.Gene Set Enrichment Analysis(GSEA)was performed to reveal the role of B56εin HCC.The Cell Counting Kit-8,plate cloning,wound healing,and transwell assays were conducted to assess the effects of B56εinterference on the malignant behavior of HCC cells.RESULTS In most tumors,B56εexpression was upregulated,and high B56εexpression was a risk factor for adrenocortical cancer,HCC,pancreatic adenocarcinoma,and pheochromocytoma and paraganglioma(all P<0.05).B56εexpression levels were correlated with a variety of immune cells,such as T helper 17 cells,B cells,and macro-phages.There was a positive correlation between B56εexpression levels with immune checkpoint genes and HLA-related genes(all P<0.05).The expression of B56εwas negatively correlated with the sensitivity of most chemotherapy drugs,but a small number showed a positive correlation(all P<0.05).GSEA analysis showed that B56εexpression was related to the cancer pathway,p53 downstream pathway,and interleukin-mediated signaling in HCC.Knockdown of B56εexpression in HCC cells inhibited the proliferation,migration,and invasion capacity of tumor cells.Core Tip:The expression of protein phosphatase 2A(PP2A)subunit B56εis up-regulated in most tumors,and its high expression is a risk factor for adrenocortical c

Extraction and analysis of saline soil deformation in the Qarhan Salt Lake region (in Qinghai, China) by the sentinel SBAS-InSAR technique

The instability of saline soil foundation affects the safety of artificial construction,and may cause ground collapse,building destruction and road damage.It is fundamental to reduce the potential engineering geological disasters by exploring the dynamic evolution of saline soil.Most of the previous researches of saline soil were conducted by simulated experiments and traditional geodetic surveying methods.Restricted to the limited sampling points,the spatiotemporal evolution characteristics of saline soil were difficult to find out in a large scale.This paper exploited SBAS-InSAR method to extract the deformation of the Qarhan Salt Lake(in Qinghai,China)section along Qinghai-Tibet railway,based on 119 image data acquired by Sentinel-1A from 2015 to 2020.The results showed that the deformation trends varied frequently in this saline mud flat.Between the adjacent areas with different deformation trends along Qinghai-Tibet railway,discontinuities caused by uplift and subsidence were very obvious.Besides,among the areas with brine exploitation or artificial construction,the maximum subsidence rate reached50 mm/yr,and the maximum cumulative subsidence exceeded 320 mm in the latest 5 years.In the saline mud flat closed to the rivers and lakes,the deformation trend was continuous uplift.Nevertheless,there were obvious seasonal deformation characteristics in those areas far away from the water body.Further analysis found out a sharply subsidence caused by the salt collapsibility appeared in rainy seasons.While an uplift trend induced by the salt swelling or frost heave was remarkable in dry seasons.Subsequently,the relationships were analyzed between the time series deformation and external environmental fac-tors.Furthermore,the deformation mechanism of saline soilin the Qarhan Salt Lake region was inter-preted then.In general,this study provides complete spatiotemporal evolution information of saline soil,and demonstrates the deformation characteristics of saline soil in the Qarhan Salt Lake region suc-cessfully.Relate

The introduction of spinel phase to form the layered-spinel structure(LSS)is an effective way to improve the electrochemical performance of Li-and Mn-rich layered oxides(LMR).But is this structure universal for all LMR systems?In this work,different Mn/Ni ratio systems with the LSS are discussed in detail.It is found that,high discharge capacity(200.8 mA h g^(-1) at 1C rate;1C=250 mA h g^(-1))as well as high capacity-retention(94%at 1C rate after 100 cycles)can be achieved by forming the LSS for low-Ni system(Mn/Ni=5.0).However,the capacity retention decreases severely in the high-Ni system(Mn/Ni=3.5,2.6).For example,when the ratio of Mn/Ni is 3.5,the capacity-retention of the layered-spinel sample was only 65.8%,compared to the 83%of the original LMR sample.The Ex-situ XRD,XPS,and HRTEM results demonstrate that the introduction of spinel phase in high-Ni system accelerates the transition and collapse of the crystal structure.This work provides guidance for optimizing the proportions of elements and the design of structures for the LMR.

Tuning redox activity through delithiation induced protective layer and Fe-O coordination for Li-rich cathode with improved voltage and cycle performance

Li-rich layered transition metal oxides are one of the most promising cathode materials for their high energy density.However,the cathodes usually suffer from severe potential dropping and capacity fading during cycling,which are associated with the surface oxygen release and accompanied by cation densification and structural collapse.Herein,an integrative approach of simultaneous constructing uniform 3d Fe-ion doping in the transition metal layer and Li-rich Li_(5)FeO_(4) shell to grab the oxygen and prevent interfacial side reactions is proposed.The introduction of Fe induces higher redox potential and stronger 3 d Fe-O_(2)p covalent bond,triggering reversible anionic redox via a reductive coupling mechanism.And the delithiated product of Li-rich Li_(5)FeO_(4) not only acts as a protective layer alleviating the side reactions but also enhances the surface kinetic property.With the benefit of promoted reversibility of oxygen redox and enhanced surface stability,the cathode exhibits high reversible capacity and superior cycle performance.Density function theory calculation indicates that the O_(2)p non-bonding state in the cathode incorporated with Fe sits at a lower energy band,resulting in higher energy storage voltage and improved oxygen stability.Consequently,the modified cathode exhibits a discharge specific capacity of 307 m A h g^(-1)(1 C=250 m A g^(-1)),coulombic efficiency of 82.09%in the initial cycle at 0.1 C and 88.34%capacity retention after 100 cycles at 1 C.The work illustrates a strategy that could simultaneously enhance oxygen redox reversibility and interface stability by constructing lattice bond coordination and delithiation induced protective layer to develop Li-rich materials with high reversible capacity and long lifespan.

Effect of microscopic pore structures on ultrasonic velocity in tight sandstone with different fluid saturation

Microcosmic details of pore structure are the essential factors affecting the elastic properties of tight sandstone reservoirs,while the relationships in between are still incompletely clear due to the fact that quantitative or semi-quantitative experiments are hard to achieve.Here,three sets of tight sandstone samples from the Junggar Basin are selected elaborately based on casting thin sections,XRD detection,and petro-physical measurement,and each set is characterized by a single varied microcosmic factor(pore connectedness,pore type,and grain size)of the pore structure.An ultrasonic pulse transmission technique is conducted to study the response of elastic properties to the varied microcosmic details of pore structure in the situation of different pore fluid(gas,brine,and oil)saturation and confining pressure.Observations show samples with less connectedness,inter-granular dominant pores,and smaller grain size showed greater velocities in normal conditions.Vpis more sensitive to the variations of pore type,while Vsis more sensitive to the variations of grain size.Samples with better connectedness at fluid saturation(oil or brine)show greater sensitivity to the confining pressure than those with gas saturation with a growth rate of 6.9%-11.9%,and the sensitivity is more likely controlled by connectedness.The pore types(inter-granular or intra-granular)can be distinguished by the sensitivity of velocities to the variation of pore fluid at high confining pressure(>60 MPa).The samples with small grain sizes tend to be more sensitive to the variations of confining pressure.With this knowledge,we can semi-quantitatively distinguish the complex pore structures with different fluids by the variation of elastic properties,which can help improve the precision of seismic reservoir prediction for tight sandstone reservoirs.

南京临时政府与清政府的财政命运极其相似,皆受财政地方主义的羁制,陷入“中央孤悬”的境况。在南京临时政府的财政收入中,以临时性的非税收收入为主,缺少稳定的税收来源。税收本应由各地解送,但革命省份在接收原财政体系之后,并未积极向中央解送应缴款项协助纾困救急,反而还希望获得中央财政的拨款,甚至截留成为常态。这当然与各省的政治态度及财政赤字有关,但仍受清末以来财政地方主义的直接影响。中央允许地方就地筹款,外销和摊派范围不断扩大,原属中央财政的正额赋税钱粮之征收权、分配权和使用权部分被转至地方。在武昌首义爆发后,大部分省份对清政府的催解谕令置之不理;在宣布易帜独立之后,各省对临时政府的催款命令同样消极应付。财政地方主义的延续,对政权更迭及革命本身产生了重要影响。

Acute aortic dissection is one of the most life-threatening cardiovascular diseases,with a high mortality rate.Its prevalence ranges from 0.2%to 0.8%in humans,resulting in a significant number of deaths due to being missed in manual examinations.More importantly,the aortic diameter—a critical indicator for surgical selection—significantly influences the outcomes of surgeries post-diagnosis.Therefore,it is an urgent yet challenging mission to develop an automatic aortic dissection diagnostic system that can recognize and classify the aortic dissection type and measure the aortic diameter.This paper offers a dual-functional deep learning system called aortic dissections diagnosis-aiding system(DDAsys)that enables both accurate classification of aortic dissection and precise diameter measurement of the aorta.To this end,we created a dataset containing 61190 computed tomography angiography(CTA)images from 279 patients from the Division of Cardiovascular Surgery at Tongji Hospital,Wuhan,China.The dataset provides a slice-level summary of difficult-to-identify features,which helps to improve the accuracy of both recognition and classification.Our system achieves a recognition F1 score of 0.984,an average classification F1 score of 0.935,and the respective measurement precisions for ascending and descending aortic diameters are 0.994 mm and 0.767 mm root mean square error(RMSE).The high consistency(88.6%)between the recommended surgical treatments and the actual corresponding surgeries verifies the capability of our system to aid clinicians in developing a more prompt,precise,and consistent treatment strategy.

In flowering plants(angiosperms),fertilization of the egg cell by one sperm cell produces an embryo,whereas fusion of a second sperm cell with the central cell generates the endosperm.In most angiosperms like Arabidopsis,a pollen grain contains two isomorphic sperm cells required for this double fertilization process.A long-standing unsolved question is whether the two fertilization events have any preference.A tool to address this question is the usage of the cyclin-dependent kinase a1(cdka;1)mutant pollen,which produces a single sperm-like cell(SLC).Here,we first adopt a complementation-based fluorescence-labeling method to successfully separate and collect cdka;1 mutant pollen containing a single SLC.Single-cell RNA-sequencing analysis revealed that cdka;1 SLCs show a gene expression profile highly similar to that of sperm cells and not to the generative cell,precursor of the two sperm cells.Pollination assays using a limited number of cdka;1 mutant pollen revealed that in 98.2%of the ovules,single fertilization of the egg cell occurred.Pollination of pistils with excessive cdka;1 mutant pollen allowed the delivery of a second SLC via fertilization recovery,which fertilized the central cell,resulting in 20.7%double-fertilized ovules.This indicates that cdka;1 SLCs are able to fertilize both the egg and the central cell.Taken together,our findings have answered a long-standing question and support that preferential fertilization of the egg cell is evident in Arabidopsis.
