Peri-centrosomal localization of small interfering RNAs in C. elegans

The centrosome is the microtubule-organizing center and a crucial part of cell division. Centrosomal RNAs(cnRNAs) have been reported to enable precise spatiotemporal control of gene expression during cell division in many species. Whether and how cnRNAs exist in C. elegans are unclear. Here, using the nuclear RNAi Argonaute protein NRDE-3 as a reporter, we observed potential peri-centrosome localized small interfering(si)RNAs in C. elegans. NRDE-3 was previously shown to associate with pre-mRNAs and pre-rRNAs via a process involving the presence of complementary siRNAs. we generated a GfP-NRDE-3 knock-in transgene through CRISPR/Cas9 technology and observed that NRDE-3 formed peri-centrosomal foci neighboring the tubulin protein TBB-2, other centriole proteins and pericentriolar material(PCM)components in C. elegans embryos. The peri-centrosomal accumulation of NRDE-3 depends on RNA-dependent RNA polymerase(RdRP)-synthesized 22G siRNAs and the PAZ domain of NRDE-3, which is essential for siRNA binding. Mutation of eri-1, ergo-1, or drh-3 significantly increased the percentage of pericentrosome-enriched NRDE-3. At the metaphase of the cell cycle, NRDE-3 was enriched in both the peri-centrosomal region and the spindle. Moreover, the integrity of centriole proteins and pericentriolar material(PCM) components is also required for the peri-centrosomal accumulation of NRDE-3. Therefore, we concluded that siRNAs could accumulate in the pericentrosomal region in C. elegans and suggested that the peri-centrosomal region may also be a platform for RNAi-mediated gene regulation.

Influence of hydrogen attack on mechanical and fatigue properties of 1020, 15CrMo and 2.25Cr-1Mo steels has been studied, which were exposed to gaseous hydrogen at pressure of 18MPa and 300-480℃ for up to 480h. The results of fatigue test showed that hydrogen attack (HA) resulted in an obvious change in fatigue properties of 1020 and 15CrMo steels, however, 2.25Cr-1Mo steel showed a lower susceptibility to hydrogen attack due to the addition of Cr and Mo. fatigue crack behaviour for 1020 steel is attributed to hydrogen attack damage and the crack closure induced by the rough cracking surface, which lead to a valley on △kth～HA% plot. The change in the fatigue behaviour of 15CrMo steel is possibly caused by severe surface decarburization.

Merging bound states in the continuum in an open acoustic resonator

Bound states in the continuum （BICs） are perfectly localized resonances despite embedding in the continuum ***,an isolated BIC is very sensitive to the structure ***,we report merging acoustic BICs in a single open resonator,robust against the structure *** find that both symmetry-protected BIC and friedrich-Wintgen BIC are sustained in a single coupled waveguide-resonator *** varying the height and length of the resonator,these two BICs move toward each other and merge into a single one at a critical *** to an individual BIC,the merged BIC is robust against fabrication error because its Q-factor is proportional to ΔL-4,whereΔL embodies the structure *** essence of this extraordinary phenomenon is perfectly explained by the two-and three-level approximations of the effective nonHermitian ***,we present direct experimental demonstrations of the moving and merging of BICs in a coupled 3D waveguide-resonator,which are evidenced by the vanishing of the linewidth of fano resonance in the transmission *** results may find exciting applications in designing high-quality acoustic sources,sensors and filters.

Trans-scale mechanics: looking for the missing links between continuum and micro/nanoscopic reality

Problems involving coupled multiple space and time scales offer a real challenge for conventional frame-works of either particle or continuum mechanics. In this paper, four cases studies （shear band formation in bulk metallic glasses, spallation resulting from stress wave, interaction between a probe tip and sample, the simulation of nanoindentation with molecular statistical thermodynamics） are provided to illustrate the three levels of trans-scale problems （problems due to various physical mechanisms at macro-level, problems due to micro-structural evolution at macro/micro-level, problems due to the coupling of atoms/ molecules and a finite size body at micro/nano-level） and their formulations. Accordingly, non-equilibrium statistical mechanics, coupled trans-scale equations and simultaneous solutions, and trans-scale algorithms based on atomic/molecular interaction are suggested as the three possible modes of trans-scale mechanics.

Comparison of Soil Nitrogen Availability Indices under Two Temperate forest Types

To evaluate the validity of different indices in estimating soil readily mineralizable N, soil microbial biomass （Nmic）, soil active N （SAN）, soluble organic N （SON）, net N mineralization rate （NNR） and gross N mineralization rate （GNR） in mineral soils （0-10 cm） from six forest stands located in central Germany were determined and compared with two sampling times： April and November. Additionally, soil density fractionation was conducted for incubated soils （with addition of ^15NH4-N and glucose, 40 days） to observe the sink of added ^15N in different soil fractions. The study showed that Nmic and NNR in most stands differed significantly （P 〈 0.05） between the two sampling times, but not GNR, SAN and SON. In November, no close relationships were found between GNR and other N indices, or between Nrnic, SON, and SAN and forest type. However, in April, GNR was significantly correlated （P 〈 0.05） with Nmic, SAN, and NNR along with Nmlc under beech being significantly higher （P 〈 0.05） than under conifers. furthermore, density fractionation revealed that the light fraction （Lf, 0.063-2 mm, 〉 1.7 gcm^-3） was not correlated with the other N indices. In contrast, results from the incubation study proved that more 15N was incorporated into the heavy fraction （Hf 〈 0.063 ram, 〉 1.7 g cm^-3） than into Lf, indicaing that more labile N existed in Hf than in Lf. These findings suggested that attention should be paid to the differences existing in N status between agricultural and forest soils.

Biofuel-driven trigeneration systems for non-residential building applications:A holistic assessment from the energy,environmental and economic perspectives

In the metropolises,it is unlikely to use merely solar and wind energy to pursue zero carbon building ***,it would become possible if biofuel-driven trigeneration systems(BDTS)are *** is thus essential to assess the application opportunity of BDTS in a holistic *** this study,BDTS offered definite primary energy saving of up to 15%and carbon emissions reduction of at least 86%in different types of non-residential buildings as compared to the conventional *** 24/7 operation for the hotel and hospital buildings,the corresponding BDTS could even achieve zero carbon *** the BDTS primed with compression-ignition internal combustion engine were not economically viable even in running cost due to the high local biodiesel price *** BDTS primed with spark-ignition engine and fueled by biogas,however,would have economic merit when carbon price was considered for the conventional systems that fully utilize fossil *** of carbon tax and social cost could have the payback ceilings of 8 years and 2 years respectively for most of building ***,the results could reflect the application potential of BDTS for non-residential buildings,leading the pathway to carbon neutrality for sustainable sub-tropical cities.

Short-time Oxidation of Alloy 690 in High-temperature and High-pressure Steam and Water

The oxidation behavior of alloy 690 exposed to high-temperature and high-pressure steam and water at 280℃ for 1 h was investigated by scanning electron microscopy （SEM）, atomic force microscopy （AfM） and X-ray photoelectron spectroscopy （XPS）. In high-temperature and high-pressure steam, the oxide film is composed of an outermost Ni-rich hydroxides layer, an intermediate layer of hydroxides and oxides enriched in Cr, an inner oxide layer. The film formed in high-temperature water is similar to that in steam, except for missing the Ni-rich hydroxides layer. Samples with different surface finishes （electropolished, mechanically polished, ground, and as-received） were prepared for comparison. A general increase of the oxide thickness with the degree of surface roughness is observed. The equivalent oxide thicknesses lie in the range of 100-200 nm for the as-received samples, 150-250 nm for the samples ground to 400# and 10-20 nm for the samples ground to 1500#, mechanically polished, and electropolished.

Chemical Chaperones Increasing Expression Level of Soluble Single-chain fv Antibody(scfv2f3)

Glutathione peroxidase（GPX） is a sclenoenzyme that protects the biomembrane and other cellular components against oxidative damage. The selenium-containing single chain fv fragment of monoelonal antibody 2173 （Se scfv2f3） is a kind of GPX mimic and it has a wide clinical applications because of its high activity and low antigenicity. Se-sefv2f3 is generated by the chemical modification of the single chain fv fragment of monoelonal antibody 2f3 （ scfv2f3 ）, which can be expressed in E. coll. In this article, the effect of chemical chaperones, such as glycerol, glucose, and β-cyclodextrin added to the culture medium, on the expression of soluble scfv2f3 was investigated. The expression level was evaluated by the determination of soluble scfv2f3 contents in the whole cell lysates. The results suggest that both glycerol and β-cyclodextrin greatly increase the expression level of soluble scfv2f3, and β-cyclodextrin is found to be more effective compared with glycerol. Glucose has a slight effect on the expression level of soluble scfv2f3. This is the first example, wherein β-cyclodextrin has been used as a chemical chaperone during the cell culture to improve the expression level of recombinant proteins. In addition, chemical chaperones are found to decrease the toxic effect of IPTG on cells.
