Targeting cancer stem cell pathways for cancer therapy

Since cancer stem cells(CSCs)were first identified in leukemia in 1994,they have been considered promising therapeutic targets for cancer *** cells have self-renewal capacity and differentiation potential and contribute to multiple tumor malignancies,such as recurrence,metastasis,heterogeneity,multidrug resistance,and radiation *** biological activities of CSCs are regulated by several pluripotent transcription factors,such as OCT4,Sox2,Nanog,KLF4,and *** addition,many intracellular signaling pathways,such as Wnt,NF-κB(nuclear factor-κB),Notch,Hedgehog,JAK-STAT(Janus kinase/signal transducers and activators of transcription),PI3K/AKT/mTOR(phosphoinositide 3-kinase/AKT/mammalian target of rapamycin),TGF(transforming growth factor)/SMAD,and PPAR(peroxisome proliferator-activated receptor),as well as extracellular factors,such as vascular niches,hypoxia,tumor-associated macrophages,cancer-associated fibroblasts,cancer-associated mesenchymal stem cells,extracellular matrix,and exosomes,have been shown to be very important regulators of ***,vaccines,antibodies,and CAR-T(chimeric antigen receptor T cell)cells have been developed to specifically target CSCs,and some of these factors are already undergoing clinical *** review summarizes the characterization and identification of CSCs,depicts major factors and pathways that regulate CSC development,and discusses potential targeted therapy for CSCs.

Here,we report a Pd/PdO_(x) sensing material that achieves 1-s detection of 4% H_(2) gas(i.e.,the lower explosive limit concentration for H_(2))at room temperature in *** Pd/PdO_(x) material is a network of interconnected nanoscopic domains of Pd,PdO,and PdO_(2).Upon exposure to 4% H_(2),PdO and PdO_(2) in the Pd/PdO_(x) are immediately reduced to metallic Pd,generating over a>90% drop in electrical *** mechanistic study reveals that the Pd/PdO_(2) interface in Pd/PdOx is responsible for the ultrafast PdO_(x) *** Pd at the Pd/PdO_(2) interface enables fast H_(2) dissociation to adsorbed H atoms,significantly lowering the PdO2 reduction *** addition,control experiments suggest that the interconnectivity of Pd,PdO,and PdO2 in our Pd/PdO_(x) sensing material further facilitates the reduction of PdO,which would otherwise not *** 1-s response time of Pd/PdO_(x) under ambient conditions makes it an excellent alarm for the timely detection of hydrogen gas leaks.

We analyze finite volume schemes of arbitrary order r for the one-dimensional singu- larly perturbed convection-diffusion problem on the Shishkin mesh. We show that the error under the energy norm decays as （N-11n（N 4- 1））r, where 2N is the number of subinter- vals of the primal partition. Furthermore, at the nodal points, the error in function value approximation super-converges with order （N-11n（N ＋ 1））2r, while at the Gauss points, the derivative error super-converges with order （N-11n（N ＋ 1））r＋1. All the above conver- gence and superconvergence properties are independent of the perturbation parameter e. Numerical results are presented to support our theoretical findings.

This work is devoted to practical stability of a class of regime-switching diffusions. First, the notion of practical stability is introduced. Then, sufficient conditions for practical stability and practical instability in probability and in pth mean are provided using a Lyapunov function argument. In addition, easily verifiable conditions on drift and diffusion coefficients are also given. Moreover, examples are supplied for demonstration purposes.

The signal transduction network in regulating lipid metabolism is a hot topic of biomedical research. Recent research endeavors reveal that intracellular stress signaling from a cellular organelle called endoplasmic reticulum (ER) is critically involved in lipid homeostasis and the development of metabolic disease. The ER is a site where newly-synthesized proteins are folded and assembled into their three-dimensional structures, modified and transported to their precise cellular destinations. A wide range of biochemical, physiological and pathological stimuli can interrupt the protein folding process in the ER and cause accumulation of unfolded or misfolded proteins in the ER lumen, a condition referred to as ER stress. To cope with this stress condition, the ER has evolved highly-specifi c signaling pathways collectively termed Unfolded Protein Response (UPR) or ER stress response. The UPR regulates transcriptionaland translational programs, affecting broad aspects of cellular metabolism and cell fate. Lipogenesis, the metabolic process of de novo lipid biosynthesis, occurs primarily in the liver where metabolic signals regulate expression of key enzymes in glycolytic and lipogenic pathways. Recent studies suggest that the UPR plays crucial roles in modulating lipogenesis under metabolic conditions. Here we address some of recent representative evidence regarding the role of the UPR in lipogenesis.

Inflammatory conditions increase the risk of cancer. Strong evidences showed that inflammation contributes to breast cancer and prostate cancer in different ways such as inflammationinduced DNA or RNA damage, overexpression cytokines, chemokines etc. Recent studies have begun to unravel molecular pathways linking inflammation and cancer. Some possible mechanisms by which inflammation can contribute to carcinogenesis have been found. These mechanisms by which inflammation contributes to cancer give broader views of cancer development. These insights are fostering new antiinflammatory therapeutic approaches to cancer development.

目的:通过网络药理学方法探寻非甾体抗炎药(nonsteroidal anti-inflammatory drugs,NSAIDs)作为抗炎止痛药对呼吸道合胞病毒(RSV)的潜在作用机制,通过网络药理学研究筛选抗RSV的最有影响力的NSAIDs药物。方法:将美国食品和药物管理局(FDA)批准的NSAIDs(19种活性药物和一种前药)用于本研究。通过Swiss Target Prediction和PubChem数据库收集NSAIDs的靶点,在GeneCards数据库中检索疾病RSV靶标。利用韦恩网绘制NSAIDs靶点蛋白与RSV靶点蛋白的交集,在STRING数据库对药物-疾病交集靶蛋白进行蛋白互作分析,在Cytoscape 3.9.0中对该网络进行美化。在DAVID数据库中对交集靶蛋白进行京都基因和基因组百科全书(KEGG)通路富集分析,利用微生信在线绘图工具绘制KEGG通路富集分析的气泡图,通过Cytoscape 3.9.0进行通路-靶点-成分网络分析,最后通过分子对接(AutoDock Vina)确定NSAIDs对靶蛋白的结合亲和力。结果:共搜集到421个NSAIDs靶点,391个与RSV相关的靶点,39个重叠的靶蛋白,基因组富集分析显示了37条抗RSV的信号通路,探索出MAPK1、AKT1和MAP2K1是与B细胞受体信号通路相关性最大的靶蛋白。结论:三种NSAIDs(舒林酸、罗非昔布、双氯芬酸)可能通过使MAPK1、AKT1和MAP2K1等相关靶蛋白失活来阻断B细胞受体信号通路,从而缓解RSV引起的支气管炎和肺炎。
