Effectiveness of Wastewater-Based Epidemiology as an Early Warning Tool to Detect SARS-CoV-2 (COVID-19)

Medical diagnostic tests to detect Severe Acute Respiratory Syndrome Coronavirus-2 (SARS-CoV-2) for individuals in the United States were initially limited to people who were traveling or symptomatic to track disease incidence due to the cost of providing testing for all people in a community on a routine basis. As an alternative to randomly sampling large groups of people to track disease incidence at significant cost, wastewater-based epidemiology (WBE) is a well-established and cost-effective technique to passively measure the prevalence of disease in communities without requiring invasive testing. WBE can also be used as a forecasting tool since the virus is shed in individuals prior to developing symptoms that might otherwise prompt testing. This study applied the WBE approach to understand its effectiveness as a possible forecasting tool by monitoring the SARS-CoV-2 levels in raw wastewater sampled from sewer lift stations at a large public university campus setting including dormitories, academic buildings, and athletic facilities. The WBE analysis was conducted by sampling from building-specific lift stations and enumerating target viral copies using RT-qPCR analysis. The WBE results were compared with the 7-day rolling averages of confirmed infected individuals for the following week after the wastewater sample analysis. In most cases, changes in the WBE outcomes were followed by similar trends in the clinical data. The positive predictive value of the applied WBE approach was 86% for the following week of the sample collection. In contrast, positive correlations between the two data with Spearmen correlation (rs) ranged from 0.16 to 0.36. A stronger correlation (rs = 0.18 to 0.51) was observed when WBE results were compared with COVID-19 cases identified on the next day of the sampling events. Synthetic Biology Construct of Ebola Virus in Bacteria Surrogate Is Stable and Safe for Rapid Detection Studies in a BSL-2 Laboratory Setting

Rapid detection of virulent pathogens during an outbreak is critical for public health advisories and control of the disease in a population. While many molecular techniques for point of care and clinical diagnosis abound, the US experience with the COVID-19 testing in the early stages of the pandemic underscores the critical importance of determining the appropriate target gene(s) with in-built controls that reliably detect pathogens with high sensitivity and specificity. Assays and research for diagnostics and therapy could be slowed during an epidemic because access to the required BSL-3 and BSL-4 laboratories are limited. So, during the 2014 West Africa Ebola outbreak, we tested the hypothesis that using synthetic cDNA of Ebolavirus in a bacteria surrogate (fit for all lab settings), would remain unmutated and safe after several generations, serving as an effective positive control in research settings, self test and point-of-care detection platforms. Primers were designed for the detection and quantification of the nucleoprotein (NP) gene of the 2014 Makona Ebola strain (KR781608.1, 733 - 1332 bp). To test the stability of artificially inserted translation arrest in the Orf of the model gene, it was edited to include three STOP codons in the RNA transcript using SNAP GENE. The segment was then spliced into a high copy number plasmid, cloned into One ShotTM TOP10 Escherichia coli (Invitrogen), and tested for stability and safety by periodic subculture, extraction and sequencing. Unlike COVID-19, rapid detection of blood-borne etiologies like Ebola requires optimized protocols for blood matrix. Using real-time PCR and newly designed primer pairs, the EBOV surrogate was detected and enumerated in human blood and regular broth and buffers. Based on aligned sequence analysis, the EBOV synthetic NP gene was stable (>99.9999% similarity coefficient) for at least 3 months. A field experiment was conducted to evaluate the integrated nutrients effect on growth, yield and quality of maize (Zea mays L.) during spring, 2009, at the Agronomic Research Area, University of Agriculture, Faisalabad. The ex-periment was laid out in Randomized Complete Block Design (RCBD) having three replications with following treat-ments: T1 (control), T2(recommended NPK @ 200-120-125 kg ha-1), T3 [single spray of multinutrient (a solution mixture of micronutrients i.e;Zn = 2%, Fe = 1%, B = 1%, Mn = 1%, Cu = 0.2% and macronutrients N = 1%, K2O = 2%, S = 2%) @ 1.25Lha-1], T4 (recommended NPK @ 200-120-125 kg ha-1 + single spray of multinutrient @ 1.25L ha-1), T5 (recom-mended NPK @ 200-120-125 kg ha-1 + two spray of multinutrient @1.25Lha-1) and T6 (recommended NPK @ 200-120-125 kg ha-1+ three spray of multinutrient @ 1.25Lha-1).The recommended dose of NPK in addition with single spray of Multi-nutrients substationally improved all growth parameters, ear characteristics and also enhanced macronutrients use efficiency up to 11.5% which induced significant increase in grain yield as compared to control and also in the treatment where recommended dose of NPK was applied alone. The quality parameter of maize (oil contents) significantly improved by foliar application of multinutrients solution but recommended dose of fertilizer in addition to single spray of Multi-nutrients was economical.
