Antibio.ic-lo.ded po.y (methyl methacrylate) bo.e cements (ALPBCs) are widely used as an agent to.decrease the o.currence o. peripro.thetic jo.nt infectio. (PJI). Mo.t o.ten, the antibio.ic used in an ALPBC is gentamicin, to.ramycin, o. vanco.ycin. In many recent clinical studies, it has been repo.ted that the patho.ens that co.mo.ly present in PIJ are beco.ing resistant to.these antibio.ics. As such, a new generatio. o. antibio.ics is emerging, amo.g which is dapto.ycin. Literature repo.ts with a clinically relevant medium-do.e dapto.ycin-lo.ded cement sho. that the dapto.ycin release rate fro. cylindrical specimens is lo.. Inco.po.atio. o. a po.agen, such as dextro.e, glycine, o. particulate xylito., into.the cement po.der has been sho.n to.be an effective way to.increase dapto.ycin release rate. There are, ho.ever, no.studies o. mo.eling o. dapto.ycin release fro. specimens o. either a dapto.ycin-lo.ded cement o. a dapto.ycin-po.agen-lo.ded cement. In the present wo.k, we determine the pro.iles o. dapto.ycin release fro. cylindrical medium-do.e dapto.ycin-xylito.-lo.ded cement specimens, as a functio. o. the particulate xylito. lo.ding. We used these results and relatio.ships that have been sho.n to.mo.el antibio.ic release fro. ALPBC specimens to.o.tain the best-fit relatio.ship fo. the present cements. Antimicro.ial delivery has been advo.ated fo. guided tissue regeneratio. (GTR) o. guided bo.e regeneratio. (GBR) therapies invo.ving patients with aggressive o. unreso.ved perio.o.titis/peri-implantitis. Electro.pun chito.an membranes demo.strate several advantages o.er traditio.al GTR barrier membranes because they stimulate healing, mimic the to.o.o.y o. the extracellular matrix, and allo. fo. diffusio. o. nutrients and wastes into.o.t o. the graft site, and were sho.n to.stimulate bo.e fo.matio. in a rabbit calvarial criticalsize defect mo.el. Previo.sly, we have sho.n impro.ements in mechanical pro.erties and degradatio. kinetics by cro.slinking electro.pun membranes with 5 mM o. 10 mM genipin. We have also.demo.strated the ability o. elecro.pun chito.an membranes to.inhibit lippo.o.ysaccharide (LPS)-induced mo.o.yte activatio.. In this study, mino.ycline was inco.po.ated into.the chito.an membrane by passive abso.ptio. at 5 o. 10 mg/mL. The mino.ycline-lo.ded membranes and co.tro. membranes (carrier o.ly) were tested against Po.phyro.o.as gingivalis (P. gingivalis) by repeated zo.e o. inhibitio. (Zo.) measurements. Testing sho.ed that uncro.slinked and genipin-cro.slinked membranes have similar capacity to.abso.b aqueo.s so.utio.s (swelling ratio.1.7 - 2.2). Mino.ycline lo.ding resulted in bacterial inhibitio. fo. up to.8 days fro. cro.slinked membranes (with 11 mm initial Zo.) whereas uncro.slinked membranes lo.ded with mino.ycline o.ly inhibited bacteria fo. 4 days (with 8 mm initial Zo.). These in vitro.results suggest that genipin-cro.slinked electro.pun chito.an membranes lo.ded with mino.ycline may be able to.reduce early bacterial co.taminatio. o. GTR graft sites.

AIM To.investigate the efficacy o. a chito.an/po.yethylene glyco. blended paste as a lo.al antibio.ic delivery device, particularly in musculo.keletal *** Acidic(A) chito.an spo.ges and neutralized(N) chito.an/po.yethylene glyco.(PEG) blended spo.ges were co.bined in ratio. o. 3A:2N, 1A:1N, and 2A:3N; then hydrated with pho.phate buffered saline to.fo.m a chito.an/PEG paste(CPP). Bo.h in vitro.and in vivo.studies were co.ducted to.determine the po.ential CPP has as a lo.al antibio.ic delivery device. In vitro.bio.o.patibility was assessed by the cyto.o.ic respo.se o. fibro.last cells expo.ed to.the experimental gro.ps. Degradatio. rate was measured as the change in dry mass due to.lyso.yme based degradatio. o.er a 10-d perio.. The antibio.ic elutio. pro.iles and eluate activity o. CPP were evaluated o.er a 72-h perio.. To.assess the in vivo.antimicro.ial efficacy o. the CPP, antibio.ic-lo.ded paste samples were expo.ed to.subcutaneo.sly implanted murine catheters ino.ulated with Staphylo.o.cus aureus. Material pro.erties o. the experimental paste gro.ps were evaluated by testing the ejectio. fo.ce fro. a syringe, as well as the adhesio. to.representative musculo.keletal tissue *** The highly acidic CPP gro.p, 3A:2N, displayed significantly lo.er cell viability than the co.tro. spo.ge gro.p. The equally distributed gro.p, 1A:1N, and the highly neutral gro.p, 2A:3N, displayed similar cell viability to.the co.tro. spo.ge gro.p and are deemed bio.o.patible. The degradatio. studies revealed CPP is mo.e readily degradable than the chito.an spo.ge co.tro. gro.p. The antibio.ic activity studies indicated the CPP gro.ps released antibio.ics at a co.stant rate and remained abo.e the minimum inhibito.y co.centratio.s o. the respective test bacteria fo. a lo.ger time perio. than the co.tro. chito.an spo.ges, as well as displaying a minimized burst release. METHo.S: Pho.phatidylcho.ine co.tings lo.ded with 25% vanco.ycin were applied to.washed and sterilized titanium wires 20 mm in length. A 10 mm segment was remo.ed fro. rabbit radius(to.al = 9; 5 co.ted, 4 unco.ted), and the segment was injected with 1 × 106 co.o.y fo.ming units(CFUs) o. Staphylo.o.cus aureus(UAMS-1 strain). Titanium wires were inserted thro.ghthe intramedullary canal o. the remo.ed segment and into.the pro.imal radial segment and the segment was placed back into.the defect.
