Objective To uncover the mechanisms underlying the development of colorectal cancer(CRC),we applied bioinformatic analyses to identify key genes and experimentally validated their possible roles in CRC onset and *** We performed Gene Ontology(GO)and Kyoto Encyclopedia of Genes and Genomes(KEGG)pathway analysis on differentially expressed genes(DEGs),constructed a protein-protein interaction(PPI)network to find the top 10 hub genes,and analyzed their expression in colon adenocarcinoma(COAD)and rectum adenocarcinoma(READ).We also studied the correlation between these genes and immune cell infiltration and prognosis and validated the expression of SLC9A2 in CRC tissues and cell lines using qRT-PCR and Western *** experiments were conducted in vitro to investigate the effects of SLC9A2 on tumor growth and *** We found 130 DEGs,with 45 up-regulated and 85 down-regulated in *** analysis indicated that these DEGs were primarily enriched in functions related to the regulation of cellular pH,zymogen granules,and transmembrane transporter *** pathway analysis revealed that the DEGs played pivotal roles in pancreatic secretion,rheumatoid arthritis,and the IL-17 signaling *** identified 10 hub genes:CXCL1,SLC26A3,CXCL2,MMP7,MMP1,SLC9A2,SLC4A4,CLCA1,CLCA4,and *** enrichment analysis showed that these hub genes were predominantly involved in the positive regulation of *** expression analysis revealed that CXCL1,CXCL2,MMP1,and MMP7 were highly expressed in CRC,whereas CLCA1,CLCA4,SLC4A4,SLC9A2,SLC26A3,and ZG16 were expressed at lower *** analysis revealed that 5 key genes were significantly associated with the prognosis of *** mRNA and protein expression levels of SLC9A2 were markedly reduced in CRC tissues and cell ***,SLC9A2 overexpression in SW480 cells led to a notable inhibition of cell proliferation,migration,and *** blotting analysis revealed tha

Structural Dependence Learning Based on Self-attention for Face Alignment

Self-attention aggregates similar feature information to enhance the features. However, the attention covers nonface areas in face alignment, which may be disturbed in challenging cases, such as occlusions, and fails to predict landmarks. In addition, the learned feature similarity variance is not large enough in the experiment. To this end, we propose structural dependence learning based on self-attention for face alignment (SSFA). It limits the self-attention learning to the facial range and adaptively builds the significant landmark structure dependency. Compared with other state-of-the-art methods, SSFA effectively improves the performance on several standard facial landmark detection benchmarks and adapts more in challenging cases.

鲁迅在小说《孤独者》中,借一段叙述者和主人公之间的对话,经由“一粒种子”的好坏,进入对“孩子”的重新审视,从而回应了章太炎有关“善恶种子”的论述。章太炎受佛教唯识学影响而阐释的“俱分进化”说和他对“进化论”的批判思路,在鲁迅的文学作品中激起声声回响,并展演在鲁迅对“业识”与民族根性之遗传的思考中。因而,引入唯识学视野下的“俱分进化”说,将为重释鲁迅的韧性的战斗、决绝的复仇,提供新的视角。当章太炎终将佛学思想融入革命大义,鲁迅却不愿有章一般佛教终末的纵身一跃,也不愿轻易投身无条件革命论。他迥在善恶、罪罚、因果、复仇的深渊前,因此显现极度的紧张性。

RNA-binding proteins(RBPs)-RNA networks have contributed to cancer *** RNAs(circRNAs)are considered as protein recruiters;nevertheless,the patterns of circRNA-protein interactions in colorectal cancer(CRC)are still *** bodies(PBs)formed through liquid-liquid phase separation(LLPS)are membrane-less organelles(MLOs)consisting of RBPs and *** evidence suggests a connection between PBs dynamics and cancer *** the increasingly acknowledged crucial role of RBPs and RNA in the accumulation and maintenance of MLOs,there remains a lack of specific research on the interactions between PBs-related RBPs and circRNAs in ***,we identify that MEX-3 RNA binding family member A(MEX3A),frequently upregulated in CRC tissues,predicts poorer patient *** MEX3A accelerates malignance and inhibits autophagy of CRC ***,MEX3A undergoes intrinsically disordered regions(IDRs)-dependent LLPS in the ***,circMPP6 acts as a scaffold to facilitate the interaction between MEX3A and PBs *** MEX3A/circMPP6 complex modulates PBs dynamic and promotes UPF-mediated phosphodiesterase 5A(PDE5A)mRNA degradation,consequently leading to the aggressive properties of CRC ***,CRC patients exhibiting high MEX3A expression and low PDE5A expression have the poorest overall *** findings reveal a collaboration between MEX3A and circMPP6 in the regulation of mRNA decay through triggering the PBs aggregation,which provides prognostic markers and/or therapeutic targets for CRC.

Currently,human health due to corona virus disease 2019(COVID-19)pandemic has been seriously *** coronavirus severe acute respiratory syndrome coronavirus 2(SARS-CoV-2)spike(S)protein plays a crucial role in virus transmission and several S-based therapeutic approaches have been approved for the treatment of ***,the efficacy is compromised by the SARS-CoV-2 evolvement and *** we report the SARS-CoV-2 S protein receptor-binding domain(RBD)inhibitor licorice-saponin A3(A3)could widely inhibit RBD of SARS-CoV-2 variants,including Beta,Delta,and Omicron BA.1,XBB and ***,A3 could potently inhibit SARS-CoV-2 Omicron virus in Vero E6 cells,with EC50 of 1.016μ*** mechanism was related to binding with Y453 of RBD determined by hydrogen-deuterium exchange mass spectrometry(HDX-MS)analysis combined with quantum mechanics/molecular mechanics(QM/MM)***,phosphoproteomics analysis and multi fluorescent immunohistochemistry(mIHC)respectively indicated that A3 also inhibits host inflammation by directly modulating the JNK and p38 mitogen-activated protein kinase(MAPK)pathways and rebalancing the corresponding immune *** work supports A3 as a promising broad-spectrum small molecule drug candidate for COVID-19.

Study of Pyrolysis Characteristics and Kinetic Analysis of Shenmu Coal at a High Heating Rate Using TG-FTIR

Coal pyrolysis is a fundamental reaction in the thermal processing and utilization of *** the behavior and kinetics of coal pyrolysis is crucial for optimizing,designing,and developing a composite riser for the staged pyrolysis gasification process of pulverized *** this study,the non-isothermal pyrolysis behavior and kinetics of coal were examined at different heating rates(30,50,100,300,500,700,and 900℃/min)using thermogravimetry(TG)coupled with Fourier-transform infrared *** of the TG/derivative TG(TG/DTG)curves indicated that coal pyrolysis mainly occurred between 300℃ and 700℃.Higher heating rates led to more volatiles being released from the coal,and a higher temperature was required to achieve rapid *** analysis showed that both the model-free methods(Friedman,Flynn-Wall-Ozawa,and Kissinger-Akahira-Sunose)and the model-based method(Coats-Redfern)effectively describe the coal pyrolysis *** change in the Ea values between the two kinetic models was consistent throughout the pyrolysis process,and the most probable mechanism was the F2 model(secondary chemical reaction).In addition,the heating rate did not change the overall reaction order of the pyrolysis process;however,a higher heating rate resulted in a decrease in the Ea value during the initial pyrolysis stage.

目的通过光镜及电镜下观察感染后肠易激综合征（IBS）患者结肠黏膜细胞超微结构的变化，探讨这些改变在IBS病理生理机制中的可能作用和临床意义。方法经结肠镜钳取30例感染后IBS患者和10例健康者的乙状结肠黏膜标本，每例2块。1块标本在光镜下观察、计算每10个高倍视野下染色阳性表达细胞的平均数；1块标本采用石蜡连续切片及原位包埋法，透射电镜观察肥大细胞的形态变化及其相邻组织结构，并应用病理图像分析软件进行分析。结果感染后IBS患者标本在光镜下可见结肠黏膜肥大细胞明显增多，存在显著变异，大量肥大细胞表现为活跃的脱颗粒状态，而电镜下可见患者结肠黏膜上皮细胞膜完整，膜与膜之间桥粒样结构连接，细胞间隙增宽（1例于增宽处见肠道细菌）。肠黏膜微绒毛分布尚规整，但密度不均，长短不一，多处微绒毛断裂，线粒体嵴突消失，粗面内质网发达。在黏膜固有层中见较多肥大细胞、浆细胞等神经内分泌细胞紧密相靠，其内可见大量高密度内分泌颗粒，伴有脱颗粒后的空洞，呈现功能活跃和分泌旺盛状态。结论感染后IBS患者结肠黏膜细胞超微结构发生改变，提示IBS并非一种单纯的消化系统功能性疾病，其发病具有分子生物学基础。

Detector performance of the Gamma-ray Transient Monitor onboard DRO-A satellite

The Gamma-ray Transient Monitor(GTM) is an all-sky monitor onboard the Distant Retrograde Orbit-A(DRO-A) satellite with the scientific objective of detecting gamma-ray transients ranging from 20 ke V to 1 MeV. The GTM was equipped with five Gamma-ray Transient Probe(GTP) detector modules utilizing a NaI(Tl) scintillator coupled with a SiPM array. To reduce the SiPM noise, GTP uses a dedicated dual-channel coincident readout design. In this work, we first studied the impact of different coincidence times on the detection efficiency and ultimately selected a 0.5 μs time coincidence window for offline data processing. To test the performance of the GTPs and validate the Monte-Carlo-simulated energy response, we conducted comprehensive ground calibration tests using the Hard X-ray Calibration Facility(HXCF) and radioactive sources, including the energy response, detection efficiency, spatial response, bias-voltage response, and temperature dependence. We extensively present the ground calibration results and validate the design and mass model of the GTP detector, thus providing the foundation for in-flight observations and scientific data analysis.

目的探讨窄带成像技术(NBI)在早期食管癌及其癌前病变诊断中的临床应用价值。方法在白光和NBI模式下观察食管黏膜,记录病变的大小、范围,同时进行NBl分级。最后应用1.2%碘液进行全食管染色,记录碘染色阳性部位大小、范围,并进行碘染色分级。对所有NBI阳性及碘染色阳性部位均取活检。以病理结果作为诊断金标准,将其他检查结果与之作对照。结果①应用白光、NBI模式及碘染色检查60例患者中共发现86个病变。其中白光模式下,高年资和低年资内镜医师对病变检出率分别为79.1%(68/86)和65.1%(56/86);应用NBI模式及碘染色后两位医师对病变的检出率相同,NBI模式均为86.0%(74/86),碘染色均为90.7%。②高级别黏膜内瘤变均主要表现为NBIⅠ级(79.1%)和碘染色Ⅰ级(84.7%),低级别黏膜内瘤变主要表现为碘染色Ⅱ/Ⅲ级(78.0%)。结论与白光模式相比,NBI模式与碘染色均可增强病变的可识别性,能清楚显示病变轮廓和形态。能提高平坦型微小黏膜病变的检出率,同时也有效提高活检的准确性。操作方便,易于推广应用。NBI在早期食管癌及癌前病变诊断有一定的临床应用价值。
