Docosahexaenoic acid-acylated astaxanthin monoester enhances microglial autophagy for ameliorating amyloid-β load and cognitive deficits in models of Alzheimer’s disease

Autophagy directly regulates the amyloid-peptides （Aβ） clearance， and its dysfunction occurs in the early pathogenesis of Alzheimer’s disease （AD）. We previously reported that docosahexaenoic acid-acylated astaxanthin monoester （AST-DHA） showed neuroprotection against AD pathology. However， its in-depth mechanism and autophagic responses in AD brains are poorly understood. Herein， SH-SY5Y cells overexpressing the APP gene were established to preliminarily evaluate the actions of AST-DHA on reducing Aβ1-42levels and regulating autophagy. In microglial BV2 cells， AST-DHA and free astaxanthin （F-AST） recovered p62 and LC3II/I levels， and restored autophagy flux by rescuing the late phase of microglial autophagy. Notably， autophagic inhibitor bafilomycin A1 blunted the abilities of AST-DHA to reduce Aβ1-42and fibral Aβ， suggesting that AST-DHA probably promoted Aβ clearance in a microglial autophagy-dependent manner. Further studies in APP/PS1 mice verified that dietary AST-DHA and F-AST promoted Aβ phagocytosis via microglial autophagy. Significant decreases of Iba1 and p62 levels were observed around Aβ plaque in the hippocampus and cortex using triple fluorescence staining. Furthermore， AST-DHA exhibited superior performance over F-AST in restoring autophagic dysfunction， ameliorating Aβ burden and cognitive deficit. Our findings suggest a possible mechanism of AST-DHA in improving AD by which it restores microglial autophagy to facilitate cerebral Aβ clearance. It supports the future application of AST-DHA as an autophagic regulator in maintaining brain function.

A novel and reactive oxygen species(ROS)responsive astaxanthin phenylboronic acid derivative(AstaDPBA)was constructed by grafting phenylboronic acid(PBA)onto astaxanthin succinate diester(AstaD),and its chemical structure and physicochemical property were ***-PBA could effectively improve the ROS quenching ability in the lipopolysaccharide(LPS)-induced RAW264.7 cell inflammation ***,the bioactivity of AstaD-PBA was studied by 4 zebrafish ROS-responsive infl ammatory models induced by LPS,copper(Cu^(2+)),high-fat diet,and dextran sodium sulfate(DSS).The results suggest that AstaD-PBA might have high biosafety and the best effect on ulcerative colitis(UC)induced by ***,AstaDPBA significantly alleviated and treated weight loss and colonic shrinkage,inhibited infl ammatory cytokines,and maintained microbiota homeostasis to improve UC in C57BL/6J *** and Oscillibacter were expected to be considered UC marker fl ora according to the Metastats analysis and Pearson correlation Mantel test(P<0.01)of 16S rRNA gene sequencing *** conclusion,AstaD-PBA has been promised to be a functional compound to improve UC and maintain intestinal microbiota homeostasis.

Studies on species diversity of Larix chinensis community ecotone

The communities of Larix chinensis and their ecotone in Qinling Mountains were investigated by sampling belt *** richness,Simpson diversity,Shannon-Weiner diversity,PIE and Pielou evenness indices of vascular plants in their communities and ecotone were *** and composition of the communities in 6 sampling belts within the sites were *** results showed that the edge effect was obvious in the ecotone with a less changing environment(such as soil and terrain)and acute variation of composition and structure,while inconspicuous or even none in the ecotone with worse environmental condition(such as great space fluctuation).It is apparent that the edge effect of community was affected by soil and terrain,but not by microclimate.

Stochastic loss reserving using individual information model with over-dispersed Poisson

For stochastic loss reserving,we propose an individual information model(IIM)which accom-modates not only individual/micro data consisting of incurring times,reporting developments,settlement developments as well as payments of individual claims but also heterogeneity among *** give over-dispersed Poisson assumption about the moments of reporting developments and payments of every individual *** estimation is conducted under quasi-likelihood *** expressions are derived for the expectation and variance of outstanding liabilities,given historical *** utilise conditional mean square error of prediction(MSEP)to measure the accuracy of loss reserving and also theoretically prove that when risk portfolio size is large enough,IIM shows a higher prediction accuracy than individ-ual/micro data model(IDM)in predicting the outstanding liabilities,if the heterogeneity indeed influences claims developments and otherwise IIM is asymptotically equivalent to *** simulations are conducted to investigate the conditional MSEPs for IIM and IDM.A real data analysis is performed basing on real observations in health insurance.

The Three-layer Structure and Systems of Petroleum Migration and Accumulation of Continental Basins

The concept of the three-layer structure of continental basins is presented based on the characteristics of layered structure of basins. The reservoir could be classified into accumulation system assemblage, accumulation system, accumulation assemblage and reservoir. This paper discusses the characteristics of hydrocarbon accumulation system assemblages of the Zhanhua Depression, which include four kinds of genetic patterns：（1） buried-hill hydrocarbon accumulation system assemblage; （2） self-sourced accumulation system assemblage from the upper interval of member 4 to member 1 of Shahejie formation; （3） transition accumulation system assemblage from member 1 of the Shahejie formation to Dongying formation and （4） externally sourccd accumulation system assemblage in the late Tertiary. The hydrocarbon-source transport network layer consisted of faults and unconformities, which connected with the reservoir layer.

亚热带次生常绿阔叶林普遍存在生长滞缓、固碳功能和经济价值低下等问题,迫切需要通过人工抚育提高其生态系统功能。对植物功能性状及其多样性的深入研究,能够找寻植物共存与群落稳定发展的内在机制,为提升森林生态系统功能的实践提供基础。为了探究人工抚育对亚热带次生常绿阔叶林质量提升的影响,以杭州午潮山国家森林公园内的次生常绿阔叶林为研究对象,实施了不同强度的抚育措施,通过群落植被复查,分析群落植物功能性状以及功能多样性对不同强度人工抚育的响应。结果发现:抚育2年后,群落最大潜在树高与叶片氮、磷、钾含量显著上升,而叶片干物质含量和叶片叶绿素含量均有显著下降,比叶面积有下降趋势,两种强度的抚育之间差异不显著;叶片干物质含量、叶绿素含量与叶片氮含量间的相关性由不显著变为了显著负相关,而叶片干物质含量与叶片叶绿素含量,叶片磷、钾含量与叶片氮含量,叶片磷含量与叶片钾含量间的相关性由不显著变为显著正相关;在20%强度抚育下,功能丰富度指数均值从对照的0.52显著下降为0.16,功能均匀度指数与功能分离度指数的均值则分别上升到0.61和0.67,显著高于对照组的0.51和0.56;在10%强度抚育下,功能丰富度有所下降,功能均匀度与功能分离度均有所上升,但差异不显著。研究表明,人工抚育在一定程度上改变了群落水平的植物功能性状与功能多样性,提高了群落植物利用资源的能力和效率;而适度的抚育强度更能解放资源空间,对群落产生的影响更加明显。该结果为亚热带次生常绿阔叶林的经营与管理提供依据与参考。

目的:探究生物钟基因大脑和肌肉芳香烃受体核转运蛋白样蛋白1(BMAL1)过表达对脓毒症小鼠心肌损伤中线粒体自噬途径的影响。方法:将40只小鼠随机分为模型对照组、模型组、阴性对照(NC)组、BMAL1组,每组10只。超声心动图检测小鼠心功能指标左室短轴缩短率(LVFS)、左室射血分数(LVEF)、左室舒张末期内径(LVIDd)、左室收缩末期内径(LVIDs),ELISA检测血清肌酸激酶同工酶(CK-MB)、肌钙蛋白I(cTnI)水平,HE染色观察心肌组织病理变化,TUNEL染色检测心肌细胞凋亡,透射电镜观察心肌线粒体超微结构变化,Western blotting测定心肌组织中微管相关蛋白1轻链3(LC3)、BH3域自噬蛋白(Beclin1)、PTEN诱导激酶(Pink1)、E3泛素-连接酶活性帕金森病蛋白2(Parkin)、电压依赖性阴离子通道蛋白1(VDAC1)表达水平。结果:与模型组和NC组比较,BMAL1组小鼠LVFS、LVEF升高(P<0.05),LVIDd、LVIDs减小(P<0.05),血清中CK-MB、cTnI降低(P<0.05),心肌组织病变得到改善,TUNEL阳性细胞率减少(P<0.05),线粒体损伤减轻,心肌组织中LC3Ⅱ/LC3Ⅰ值升高(P<0.05),Beclin1、Pink1、Parkin、VDAC1蛋白表达上调(P<0.05)。结论:BMAL1过表达能够明显改善脓毒症小鼠心功能,减轻心肌损伤,该作用可能与其促进线粒体自噬有关。

通过1年期现场暴露试验、腐蚀速率分析、锈层形貌观察、锈层组成分析及电化学测试等试验方法,研究了Q235碳钢在达州典型大气环境下的腐蚀行为。结果表明,位于工业大气环境的达州市通川区,Q235碳钢的腐蚀速率为23.78μm/a,而位于乡村大气环境的达州市万源市,Q235碳钢的腐蚀速率仅为10.33μm/a。Q235碳钢表面腐蚀产物主要组成为γ-FeOOH、α-FeOOH和Fe 3 O 4。电化学结果表明,在达州重工业环境下的碳钢腐蚀较为严重,腐蚀产物层电阻和电荷转移电阻均高于达州乡村环境,说明其表面锈层能有效保护基体,减缓基体的进一步腐蚀。

为了探究羟基亚乙基二膦酸(HEDP)镀铜工艺的工程化应用的可行性,分别采用HEDP镀铜与氰化(CN)镀铜方法在A3钢表面制备铜镀层,对比2种工艺制备铜镀层的宏微观形貌、沉积速率、分散能力及深镀能力,同时对HEDP铜镀层的结合力及氢脆性能进行表征。结果表明:HEDP铜镀层结晶细致、晶粒呈紧密的沟壑起伏分布;氰化铜镀层呈大小不一的颗粒状结构。当电流密度为1.0 A/dm^(2)时,HEDP铜镀层的沉积速率达到0.19μm/min,较CN镀铜工艺0.14μm/min的沉积速率提高了35.7%。HEDP镀铜与CN镀铜工艺的分散能力、深镀能力相当。HEDP铜镀层在钢铁基体表面的结合力良好,氢脆性能合格。

Effect of Incoming Vortex on Secondary Flows in Turbine Cascades with Planar and Non-Axisymmetric Endwall

Non-Axisymmetric Endwall Profiling(NAEP) is commonly utilized in turbines to eliminate secondary ***,most of the NAEP methods consider a single-blade row environment without incorporating the effect of the stage *** paper aims to investigate the influence mechanism of the incoming vortex on the endwall secondary flow structures of NAEP in a highly loaded turbine *** model the incoming vortex in a stage environment,this study considers a half-delta wing as the vortex generator at the upstream of the turbine *** NAEP is then carried out for a highly loaded turbine cascade with an in-house numerical optimization design platform subject to no incoming *** simulation is also carried out under the influence of the incoming vortex for the turbine cascades with both planar and non-axisymmetric *** paper furthers investigated the pitchwise effect of the incoming vortex on the near endwall secondary *** results indicate that the NAEP effectively improves the endwall secondary flow of the turbine cascade,where the total pressure loss coefficient and the secondary kinetic energy(SKE) are reduced by 7.3%,and 45.7%,*** is further seen that with the incoming vortex,the NAEP achieves a considerable control effect on the endwall secondary flow of the turbine *** incoming vortex,the NAEP can still achieve considerable control effect on the endwall secondary flow of the turbine cascade;the averaged reductions of loss coefficient and SKE are 7.8% and 14.2%,*** some pitchwise locations,incoming vortex can suppress the convection of cross-passage flow toward the suction corner greatly and reduce the loss coefficient of the baseline *** incoming vortex at 4/7 pitch impinged right at the blade leading edge,leading to the generation of low-momentum fluid,which increased the size and the strength of the horseshoe *** all the pitchwise locations,NAEP can suppress the seco
