Protective Effect of Silent Mating Type Information Regulation 2 Homolog 1 on TGF-β1 Pathway via mTOR in Diabetic Nephropathy

Objective: To demonstrate whether the expression of silent mating type information regulation 2 homolog 1 (SIRT1) affects the level of TGF-β1 and Smad3 in HEK293 cells through regulating mTOR. Methods: First, recombinant plasmids DNA (rSIRT1) and siRNA targeting SIRT1 were constructed which were transfected into Human Embryonic Kidney 293 cell (HEK293) cells, respectively. Then, the generation of intracellular ROS in cells was examined by flow cytometry using the oxidation-sensitive probe. Last, the expressions of TGF-β1, smad3, P53, mTOR, p-mTOR, LC3-I and LC3-II in cells were detected to observe the effect of SIRT1 on TGF-β1 Pathway by western blot analysis. Results: We demonstrated that overexpressing of SIRT1 may decrease TGF-β1 and Smad3 expression in HEK293 cells through regulating mTOR. In addition, the result is the opposite when SIRT1 was silent in HEK293 cells. Conclusions: SIRT1 is closely related to TGF-β1/Smad3 pathway that correlates with the regulation of mTOR and ROS generation and causes diabetic nephropathy. The available evidence implies that SIRT1 has great potential as a clinical target for the prevention and treatment of renal fibrosis in the development of DN.

Special reservoir or fluid has an abnormal response to some certain frequencies, so that seismic decomposition and reconstruction are used to highlight the seismic reflection at certain frequencies useful to identify special geological bodies. Because seismic wavelets are time-varying and spatial-variable in the propagation, synthetic traces based on single wavelet make some weak but useful information lost, and make artifacts form. However, Morlet wavelet aggregation with mathematical analytical expression is able to fully and correctly reflect the variations of wavelet in the propagation of underground medium. The matching pursuit algorithm on the basis of Morlet wavelet improves the calculating efficiency in decomposition and reconstruction greatly. This method is applied to the actual study area to do conjoint analysis of single well and well-tie multi-wavelet decomposition. It is found that frequencies sensitive to interest reservoirs range from 8 to 34 Hz. Reconstructing the wavelets at those special frequencies and analyzing the reconstructed seismic data, it is pointed out that interest reservoirs have abnormal characteristics with respectively strong RMS amplitude in the reconstructed data. Crossplot of gamma value at wells and reconstructed RMS amplitude suggests that anomalies caused by interest reservoirs are well separated from the background anomalies when the reconstructed RMS amplitude is greater than 3650. Quantitative prediction results of interest reservoirs distribution in the study area reveal that interest reservoirs of western and northern study area are distributed annularly and bandedly, while most contiguous sandstone in eastern regions appears sporadically.

The fluid identification of carbonate reservoir is a key factor to hydrocarbon exploration and reservoir development. In order to simulate the seismic response characteristics of the cave in the carbonate reservoir, three sets of models were designed, including the caves varied in width, the caves filled with different solids, and the oil-gas-water model. The numerical simulation technique was used to carry out the forward modeling and the AVO (Amplitude varies with offset) response characteristics of the three groups of models were analyzed. The results show that the AVO characteristics can be observed when the cave reaches a certain extent in the horizontal direction. When the surrounding rock is constant, the absolute value of the intercept of the AVO curve increases with the Vp/Vs decrease. The AVO technology can effectively identify the gas cave. The effect is not obvious to water or oil cave.

Reverse time migration (RTM) is an accurate migration method, which is based on two-way wave equation, eliminates angle limitation, and can be capable for imaging of reverse branch and multiple. In the development of an oilfield in Pearl River Mouth (PRM) Basin, RTM has been applied to solve the problems of fault shadow and structure in distinction and the drilling has proven that the RTM result is reliable. This paper introduces the theory of RTM and emphatically discusses parameter selection. The results of RTM showed that it had advantages in resolving fault shadow and the imaging of steep dip structure, and could be utilized in the oilfield.

A silylated melamine sponge(SMS)was prepared by two simple steps,namely,immersion and dehydration of a melamine sponge coated with *** silylated structure of SMS was characterized by FT-IR(Fourier-transform infrared)spectroscopy,SEM(Scanning electron microscopy)and in terms of water contact *** oil-water absorption and separation capacities were measured by FT-IR and UV-visible *** experimental results have shown that oligomeric silanol covalently bonds by Si-N onto the surface of melamine sponge *** has shown superhydrophobicity with a water contact angle exceeding 150°±1°,a better separation efﬁciency with regard to diesel oil(by 99.31%(wt/wt%)in oil-water mixture and even up to 99.99%(wt/wt%)for diesel oil in its saturated aqueous ***,SMS inherited the intrinsicﬂame retardancy of the melamine *** general,SMS has shown superhydrophobicity,high porosity,excellent selectivity,remarkable recyclability,and better absorption capacity for various oils and organic solvents,and a high separation efﬁciency for oil in saturated aqueous solutions.

在VTI介质中,由于引入了各向异性参数使得多分量多参数地震反演问题的非线性程度显著增加,因此采用传统的权重加权法构建单目标函数进行反演得到的反演结果往往并不理想.本文以反射率法为基础,结合快速非支配排序遗传算法研究了一种VTI介质的多分量叠前联合反演方法.该方法以反射率法为正演方程,应用互相关原理构建PP波和PSV波的多目标函数,进而采用快速非支配排序遗传算法全局寻优获得VTI介质的厚度、纵横波速度、密度和各向异性参数等多个参数.在正演的过程中,反射率法可以考虑几何扩散、吸收衰减、透射损失、多次波以及纵横波旅行时不匹配等地震波传播效应,更能精确地描述地震波在地下地层中的真实传播情况;在反演的过程中,快速非支配排序遗传算法可以在不引入权重系数的条件下同时优化多个目标函数,获得联合反演问题的Pareto最优解,既不添加权重系数影响又充分利用多分量地震数据.模型测试结果验证了该反演方法的有效性和可行性.

Application of Multi-Attribute Crossplots to Tight Gas in X Area, Western China

The gas-bearing reservoir in X area is mainly the tight sandstone reservoir characterized by low porosity and permeability, frequently lateral variation and poor connectivity of single sand. The previous research results reveal that the general seismic attributes analysis cannot meet the requirement of fluid identification. This is because the relationship between seismic attributes and their implication is uncertain and ambiguous, which decreases the precision of both reservoir prediction and fluid identification. To overcome the problem, multi-attribute crossplot technology is proposed from the mathematical statistical point of view rather than the correspondence between the seismic attributes and their geological implication. In this method, the wells which have the same statistical law are classified firstly, and then all the interest wells are retained while the wells beyond the statistical law are eliminated, and the seismic attributes sensitive to the same types of eliminated wells are optimized and used to generate crossplots. The nonzero area of their crossplots results just predicts the potential distribution. The discontinuity of subsurface geological conditions results in the non-continuous shape and the seismic bin lead to the mosaic form. The optimization of sensitive attributes relative to the same types of wells is independent from each other, and thus the order of attributes in crossplots does not affect the final prediction results. This method is based on the statistical theory and suitable for the areas such as the study area abundant of lots of well data. Application to X area proves the effectiveness of this method and predicts plane distribution about different types of gas production. Due to the effect of faults and other geological factors, the partition prediction results using multi-attribute crossplots reach 95% of coincidence which is obviously and far higher than the results of the whole area. The predicted extraordinary properties of carbon nanotubes(CNTs)from theoretical calculations have great potential for many ***,reliable experimental determination of intrinsic properties at the single-tube level is currently a matter of concern,and many challenges remain because of the unhandled and nanoscale size of individual ***,we demonstrated a prototype to detect the intrinsic thermal conductivity of the single-wall carbon nanotube(SWCNT)and verify the significant non-resonant optical absorption behavior on tiny nanotubes by integrating the nanotube and ice into a new core-shell *** particular,a reversible optical visualization method based on the individual suspended ultra-long SWCNT was first developed by wrapping a nanotube with ice in the cryogenic air *** light-induced thermal effect on the hybrid core-shell structure was used tomelt the ice shell,which subsequently acted as a temperature sensor to verify the intrinsic thermal conductivity of the core-like *** interestingly,we successfully determined for the first time the thermal response phenomenon of the tiny absorption cross section in SWCNT in the vertical-polarization configuration and the significant non-resonant absorption behavior in the parallel-polarization *** investigations will provide a better understanding for the unique optical behaviors of CNT and enable the detection of intrinsic properties of various one-dimensional nanostructures such as nanotubes,nanowires,and nanoribbons.
