益舒软肝丸逆转肝硬化的机制

目的:探讨益舒软肝丸对肝硬化模型的逆转及其作用机制。方法:实验于2004-10/2006-03在三峡大学医学院天然药物实验室完成。①选取雄性Wistar大鼠48只,随机数字表法分为正常对照组8只、模型对照组12只、益舒软肝丸0.4g/kg组10只、益舒软肝丸0.8g/kg组9只、益舒软肝丸1.6g/kg组9只。益舒软肝丸由蛴螬、虻虫、冬虫夏草、水蛭、鳖甲、桃仁等中药组成,宜昌县人民医院药剂科制备,实验中用双蒸水配成混悬液。②除正常对照组外,其余各组均采用CCL4复合因素法建立肝硬化模型。造模后,益舒软肝丸0.4,0.8,1.6g/kg组分别给予对应剂量的益舒软肝丸混悬液灌胃,1次/d,连续灌胃12周;并持续背部皮下注射CCL40.3mL/100g,1次/周,共12周。正常对照组、模型对照组同时间点灌服生理盐水1mL/只。③第12周末禁食12h后,大鼠麻醉后开胸,心脏采血,分离血清,-20℃保存。取左中叶相同部位制作病理石蜡切片,苏木精-伊红染色观察组织形态学变化,苦味酸酸性品红染色观察纤维增生程度,免疫组织化学检测转化生长因子β1和基质金属蛋白酶抑制因子1阳性表达总面积。余下肝脏置于塑料薄膜袋,迅速封口置于-80℃冰箱,制备肝组织匀浆,检测羟脯胺酸、丙二醛含量及超氧化物歧化酶水平。结果:正常对照组8只大鼠无死亡。造模后,模型对照组死亡4只,益舒软肝丸0.4,0.8,1.6g/kg组各死亡3,2,2只。①肝功能检测结果:与模型对照组比较,益舒软肝丸0.4,0.8,1.6g/kg组均可明显降低血清丙氨酸氨基转移酶、天冬氨酸氨基转移酶水平[(3.66±1.00),(2.72±0.39),(2.98±0.84),(2.82±0.23)nkat/L;(11.00±2.18),(7.80±1.32),(9.56±3.09),(9.92±2.48)nkat/L;P0.05)。②肝纤维化相关指标检测结果:与模型对照组比较,益舒软肝丸0.4,0.8,1.6g/kg组均可明显降低透明质酸、层粘连蛋白、PⅢNP胶原水平[(132.67±40.31),(99.67±30.79),(96.63±36.04),(92.30±27.95)μg/L;(72.40±13.36),(60.87±12.30),(56.87±17.41),(59.63±6.29)μg/L;(41.34±6.40),(35.87±5.84),(31.57±11.13),(39.01±8.23)μg/L;P均0.05)。④肝组织形态学观察结果:正常对照组肝小叶结构清晰完整,未见纤维组织异常增生。模型对照组肝小叶结构破坏,肝细胞索排列紊乱,纤维组织大量异常增生,形成假小叶。益舒软肝丸各剂量组肝小叶结构基本完整,有少量胶原纤维增生,未见假小叶结构。⑤肝组织免疫组化半定量分析结果:与模型对照组比较,益舒软肝丸0.4,0.8,1.6g/kg组转化生长因子β1和基质金属蛋白酶抑制因子1阳性总面积均有不同程度减少[(6.33±1.16),(6.28±0.88),(5.11±0.92),(5.83±0.96)μm2;(624.82±237.11),(282.48±139.28),(80.75±28.28),(246.29±155.65)μm2;P<0.01或0.05]。结论:益舒软肝丸具有明显的抗肝硬化作用,其作用机制可能与以下因素有关:①改善肝功能。②降低丙二醛水平,减轻肝细胞损伤,逆转肝硬化病理形态结构。③下调肝硬化组织转化生长因子β1和基质金属蛋白酶抑制因子1的表达,抑制细胞外基质的合成和胶原增生、沉积。

目的:观察益舒软肝丸对肝硬化大鼠肝功能及肝组织形态学的影响。方法:实验于2004-10/2006-04在三峡大学中药药理国家二级实验室完成。①实验分组:80只大鼠随机分正常对照组8只,模型组72只。在造模过程中死亡30只,造模6周末随机取2只检测造模成功后,剩余40只大鼠随机分为模型对照组12只、益舒软肝丸0.4g/kg组10只、益舒软肝丸0.8g/kg组9只、益舒软肝丸1.6g/kg组9只。②实验方法:采用四氯化碳油剂、高脂低蛋白食物复合因素造肝硬化模型。模型对照组给予生理盐水(1mL/只)灌服,治疗组给予益舒软肝丸的混悬液灌胃,1次/d,连续灌胃12周。③实验评估:观察动物的饮食、活动、毛色、死亡等情况。于实验末称动物体质量及全肝、脾脏湿质量,计算脾指数和肝脏指数。取部分血清用自动生化分析仪检测肝功能相关指标。另取部分血清按放射免疫试剂盒方法检测血清透明质酸、层黏连蛋白和Ⅲ型前胶原水平。取肝左叶,制备切片分别用于苏木精-伊红染色和VG染色,观察组织学变化。结果:①大体观察结果:正常对照组大鼠无死亡,营养状况良好。模型对照组大鼠毛质粗糙,消瘦,精神萎靡,动作迟钝,易激惹,个别老鼠趾甲易出血,死亡4只;益舒软肝丸0.4g/kg组死亡3只,益舒软肝丸0.8g/kg组死亡2只,益舒软肝丸1.6g/kg组死亡2只。②体质量和肝脏、脾脏脏器系数:模型对照组大鼠体质量明显低于正常对照组(P<0.01),用药各组大鼠体质量明显回升(P<0.05);模型对照组肝脏、脾脏脏器系数高于正常对照组(P<0.01);用药各组肝脏、脾脏脏器系数低于模型对照组(P<0.01,P<0.05),益舒软肝丸0.8g/kg组与正常对照组最接近。③肝功能各项指标:用药各组血清丙氨酸氨基转移酶、天冬氨酸氨基转移酶水平低于模型对照组,胆碱酯酶水平高于模型对照组(P<0.01,P<0.05)。用药各组血清透明质酸、层黏连蛋白、Ⅲ型前胶原水平低于模型对照组(P<0.05);其中益舒软肝丸0.8g/kg组降低血清透明质酸、层黏连蛋白、Ⅲ型前胶原水平作用最明显。用药各组羟脯氨酸含量也低于模型对照组(P<0.01,P<0.05)。④组织学变化:与模型对照组大鼠比,用药组组织切片显示汇管区有少量纤维组织增生,但未见假小叶结构。结论:采用复合因素造模法成功构建了大鼠肝硬化模型。益舒软肝丸有很好的逆转肝硬化的作用。

Traditional expert-designed branching rules in branch-and-bound(B&B) are static, often failing to adapt to diverse and evolving problem instances. Crafting these rules is labor-intensive, and may not scale well with complex *** the frequent need to solve varied combinatorial optimization problems, leveraging statistical learning to auto-tune B&B algorithms for specific problem classes becomes attractive. This paper proposes a graph pointer network model to learn the branch rules. Graph features, global features and historical features are designated to represent the solver state. The graph neural network processes graph features, while the pointer mechanism assimilates the global and historical features to finally determine the variable on which to branch. The model is trained to imitate the expert strong branching rule by a tailored top-k Kullback-Leibler divergence loss function. Experiments on a series of benchmark problems demonstrate that the proposed approach significantly outperforms the widely used expert-designed branching rules. It also outperforms state-of-the-art machine-learning-based branch-and-bound methods in terms of solving speed and search tree size on all the test instances. In addition, the model can generalize to unseen instances and scale to larger instances.

Mitochondriopathy inspired adenosine triphosphate(ATP) depletions have been recognized as a powerful way for controlling tumor growth. Nevertheless, selective sequestration or exhaustion of ATP under complex biological environments remains a prodigious challenge. Harnessing the advantages of in vivo self-assembled nanomaterials, we designed an Intracellular ATP Sequestration(IAS) system to specifically construct nanofibrous nanostructures on the surface of tumor nuclei with exposed ATP binding sites,leading to highly efficient suppression of bladder cancer by induction of mitochondriopathy-like ***, the reported transformable nucleopeptide(NLS-FF-T) self-assembled into nuclear-targeted nanoparticles with ATP binding sites encapsulated inside under aqueous conditions. By interaction with KPNA2, the NLS-FF-T transformed into a nanofibrous-based ATP trapper on the surface of tumor nuclei,which prevented the production of intracellular energy. As a result, multiple bladder tumor cell lines(T24,EJ and RT-112) revealed that the half-maximal inhibitory concentration(IC50) of NLS-FF-T was reduced by approximately 4-fold when compared to NLS-T. Following intravenous administration, NLS-FF-T was found to be dose-dependently accumulated at the tumor site of T24 xenograft mice. More significantly, this IAS system exhibited an extremely antitumor efficacy according to the deterioration of T24 tumors and simultaneously prolonged the overall survival of T24 orthotopic xenograft mice. Together, our findings clearly demonstrated the therapeutic advantages of intracellular ATP sequestration-induced mitochondriopathy-like damages, which provides a potential treatment strategy for malignancies.

Experimental study and simulation on the creep characteristics of machineharvested seed cotton

In view of the unclear understanding of the basic scientific problems such as the rheological mechanism of seed cotton,especially the lack of research on the creep characteristics of seed cotton,the machine-harvested seed cotton in the Xinjiang region was taken as the research object to find out the compression creep *** universal material testing machine was used to carry out a one-factor creep test,taking moisture content,feed quality,compression times,and trash content as test factors and instantaneous elastic modulus,hysteretic elastic modulus,viscosity coefficient,and delay time as test *** ANOVA and correlation were analyzed by SPSS,and the creep process of the seed cotton was simulated by *** show that moisture content significantly affects the instantaneous elastic modulus,hysteretic elastic modulus,and viscosity coefficient(p<0.01).In addition,each value of which decreases with the increase in moisture *** quality significantly affects the hysteretic elastic modulus and viscosity coefficient(p<0.05).Moreover,the hysteretic elastic modulus and viscosity coefficient increase with the increase in feed *** compression times significantly influence the instantaneous elastic modulus,hysteretic elastic modulus,and viscosity coefficient(p<0.01),each value of which increases with the increase of compression ***,the trash content significantly influenced the hysteretic elastic modulus and viscosity coefficient(p<0.05).The absolute error between the simulated and experimental values ek is within−0.011-0.030 mm,and the relative errorφk is less than 7%.The experimental results can provide theoretical and data support for the study of rheological characteristics of machine-harvested seed cotton,the design of seed cotton packing devices,and the molding quality of cotton bale(mold).
