OBJECTIVE: To explore the material basis of the difference of efficacy of Dahuang(Radix Et Rhizoma Rhei Palmati)-Taoren(Semen Persicae)(DT) drugs with different proportions. METHODS: Samples of different ratios of Dahuang(Radix et Rhizoma Rhei Palnati, DH) to Taoren(Semen Persicae, TR)(Group A 1∶1, B 2∶3, C 3∶2) were analyzed based on gas chromatography time-of-flight mass spectrometry untargeted metabolomics technique. RESULTS: A total of 240 primary metabolites were detected. Forty-one differential metabolites involved nine differential metabolic pathways, of which four were closely related to the efficacy of DT in the treatment of heat and blood stasis syndrome. These pathways included the biosynthesis of amino acid(phenylalanine tyrosine and tryptophan), flavonoids, unsaturated fatty acids, and the glycolysis/glycogenesis pathway. CONCLUSION: There are significant differences in the efficacy of different ratios of DT drugs, and their optimal ratio for the treatment of heat and blood stasis syndrome should be 1∶1.

Key Physical Factors Affecting Spatial-temporal Variation of Labile Organic Carbon Fractions by Biochar Driven in Mollisols Region of Northeast China

Biochar is widely used to improve soil physical properties and carbon sequestration. However, few studies focuse on the impact of maize stalk biochar on labile organic carbon(LOC) pool and the relationship between physical properties and LOC fractions. A field positioning experiment was performed in Mollisols region of Northeast China to evaluate the influence of maize stalk biochar on the spatial distribution and temporal changes of physical properties and LOC fractions. Maize stalk biochar treatments included C1(1.5 kg·hm^(-2)), C2(3 kg·hm^(-2)), C3(15 kg·hm^(-2)), C4(30 kg·hm^(-2)), and CK(0). The results showed that maize stalk biochar increased soil water contents(SWC) and soil porosity(SP), but reduced bulk density(BD). Maize stalk biochar reduced dissolved organic carbon(DOC) contents in the 0-20 cm soil layer, ranging from 0.25 g·kg^(-1) to 0.31 g·kg^(-1) in harvest period, while increased in the 20-40 cm soil layer. In addition, the application of biochar had a significant impact on the spatial distribution and temporal change of SWC, BD, SP, DOC, hot-water extractable carbon(HWC), acid hydrolyzed organic carbon(AHC Ⅰ, Ⅱ), and readily oxidized organic carbon(ROC). High amounts of maize stalk biochar up-regulated the contents of soil organic carbon SOC, HWC, AHC Ⅰ, AHC Ⅱ, and ROC. In addition, SWC and SP were the key physical factors to affect LOC fractions. In conclusions, maize stalk biochar could improve physical properties, and then influence LOC fractions, and maize stalk biochar could be used as an organic amendment for restoring degraded soils governed by their rates of addition.

Our previous study has demonstrated that lnc_000048 is upregulated in large-artery atherosclerotic stroke and promotes atherosclerosis in ApoE^(-/-)mice.However,little is known about the role of lnc_000048 in classically activated macrophage(M1)polarization.In this study,we established THP-1-derived testing state macrophages(M0),M1 macrophages,and alternately activated macrophages(M2).Real-time fluorescence quantitative PCR was used to verify the expression of marker genes and the expression of lnc_000048 in macrophages.Flow cytometry was used to detect phenotypic proteins(CD11b,CD38,CD80).We generated cell lines with lentivirus-mediated upregulation or downregulation of lnc_000048.Flow cytometry,western blot,and real-time fluorescence quantitative PCR results showed that down-regulation of lnc_000048 reduced M1 macrophage polarization and the inflammation response,while over-expression of lnc_000048 led to the opposite effect.Western blot results indicated that lnc_000048 enhanced the activation of the STAT1 pathway and mediated the M1 macrophage polarization.Moreover,catRAPID prediction,RNA-pull down,and mass spectrometry were used to identify and screen the protein kinase RNA-activated(PKR),then catRAPID and RPIseq were used to predict the binding ability of lnc_000048 to PKR.Immunofluorescence(IF)-RNA fluorescence in situ hybridization(FISH)double labeling was performed to verify the subcellular colocalization of lnc_000048 and PKR in the cytoplasm of M1 macrophage.We speculate that lnc_000048 may form stem-loop structure-specific binding and activate PKR by inducing its phosphorylation,leading to activation of STAT1 phosphorylation and thereby enhancing STAT1 pathway-mediated polarization of THP-1 macrophages to M1 and inflammatory factor expression.Taken together,these results reveal that the lnc_000048/PKR/STAT1 axis plays a crucial role in the polarization of M1 macrophages and may be a novel therapeutic target for atherosclerosis alleviation in stroke.

This study explores the corrosion issues arising from the coupled combustion of coal and biomass and proposes potential solutions.Biomass,as a renewable energy source,offers advantages in energy-saving and carbon reduction.However,the corrosive effects of alkali metal compounds,sulfur(S)and chlorine(Cl)elements in the ash after combustion cannot be underestimated due to the high volatile content of biomass fuels.We investigate the corrosion mechanisms,as well as the transfer of Cl and alkali metal elements during this process.Comparative corrosion analyses are conducted among coal-fired boilers,pure biomass boilers and boilers with coupled combustion.Various biomass types in co-firing are studied to understand different corrosion outcomes.The main factors influencing corrosion include the physicochemical properties of biomass feedstock,furnace temperature and heating surface materials,with the chemical composition and ash content of biomass playing a dominant role.Currently,the methods used for anti-corrosion include water washing pretreatment of biomass feedstock,application of novel alloys and coatings and the development of additives to inhibit fouling,ash deposition and corrosion.Efficient inhibitors are economical and easy to produce.Additionally,biomass can be converted into biomass gasification gas,although challenges related to tar still need to be addressed.

T-box transcription factor T(TBXT;T)is required for mesodermal formation and axial skeletal development.Although it has been extensively studied in various model organisms,human congenital vertebral malformations(CVMs)involving T are not well established.Here,we report a family with 15 CVM patients distributed across 4 generations.All affected individuals carry a heterozygous mutation,T c.596A>G(p.Q199R),which is not found in unaffected family members,indicating co-segregation of the genotype and phenotype.In vitro assays show that T p.Q199R increases the nucleocytoplasmic ratio and enhances its DNA-binding affinity,but reduces its transcriptional activity compared to the wild-type.To determine the pathogenicity of this mutation in vivo,we generated a Q199R knock-in mouse model that recapitulates the human CVM phenotype.Most heterozygous Q199R mice show subtle kinked or shortened tails,while homozygous mice exhibit tail filaments and severe vertebral deformities.Overall,we show that the Q199R mutation in T causes CVM in humans and mice,providing previously unreported evidence supporting the function of T in the genetic etiology of human CVM.

Basic characteristics of key interfaces in Upper Ordovician Wufeng Formation – Lower Silurian Longmaxi Formation in Sichuan Basin and its periphery,SW China

Based on anatomy of key areas and data points and analysis of typical features of shell layer in Guanyinqiao Member, basic characteristics of key interfaces, mainly bentonite layers, in the Upper Ordovician Wufeng Formation-Lower Silurian Longmaxi Formation in the Sichuan Basin and its surrounding areas and the relationship between these key interfaces with the deposition of organic-rich shale have been examined systematically. The Wufeng Formation-Longmaxi Formation has four types of marker beds with interface attributes, namely, the characteristic graptolite belt, Guanyinqiao Member shell layer, section with dense bentonite layers, and concretion section, which can be taken as key interfaces for stratigraphic division and correlation of the graptolite shale. The shell layer in Guanyinqiao Member is the most standard key interface in Wufeng Formation-Longmaxi Formation, and can also be regarded as an important indicator for judging the depositional scale of organic-rich shale in key areas. There are 8 dense bentonite sections of two types mainly occurring in 7 graptolite belts in these formations. They have similar interface characteristics with the shell layer in Guanyinqiao Member in thickness and natural gamma response, and belong to tectonic interfaces(i.e., event deposits). They have three kinds of distribution scales: whole region, large part of the region, and local part, and can be the third, fourth and fifth order sequence interfaces, and have a differential control effect on organic-rich shale deposits. The horizon the characteristic graptolite belt occurs first is the isochronous interface, which is not directly related to the deposition of organic-rich shale. Concretions only appear in local areas, and show poor stability in vertical and horizontal directions, and have no obvious relationship with the deposition of the organic-rich shale.

Deamidation enables pathogenic SMAD6 variants to activate the BMP signaling pathway

The BMP signaling pathway plays a crucial role in regulating early embryonic development and tissue homeostasis. SMAD6 encodes a negative regulator of BMP, and rare variants of SMAD6 are recurrently found in individuals with birth defects. However, we observed that a subset of rare pathogenic variants of SMAD6 consistently exhibited positive regulatory effects instead of the initial negative effects on the BMP signaling pathway. We sought to determine whether these SMAD6 variants have common pathogenic mechanisms. Here, we showed that pathogenic SMAD6 variants accompanying this functional reversal exhibit similar increases in deamidation. Mechanistically, increased deamidation of SMAD6 variants promotes the accumulation of the BMP receptor BMPR1A and the formation of new complexes, both of which lead to BMP signaling pathway activation. Specifically, two residues, N262 and N404, in SMAD6 were identified as the crucial sites of deamidation, which was catalyzed primarily by glutamine-fructose-6-phosphate transaminase 2(GFPT2). Additionally, treatment of cells harboring SMAD6 variants with a deamidase inhibitor restored the inhibitory effect of SMAD6 on the BMP signaling pathway. Conversely,when wild-type SMAD6 was manually simulated to mimic the deamidated state, the reversed function of activating BMP signaling was reproduced. Taken together, these findings show that deamidation of SMAD6 plays a crucial role in the functional reversal of BMP signaling activity, which can be induced by a subset of various SMAD6 variants. Our study reveals a common pathogenic mechanism shared by these variants and provides a potential strategy for preventing birth defects through deamidation regulation, which might prevent the off-target effects of gene editing.

To efficiently develop deep shale gas in southern Sichuan Basin,under the guidance of“extreme utilization”theory,a basic idea and solutions for deep shale gas development are put forward and applied in practice.In view of multiple influencing factors of shale gas development,low single-well production and marginal profit of wells in this region,the basic idea is to establish“transparent geological body”of the block in concern,evaluate the factors affecting shale gas development through integrated geological-engineering research and optimize the shale gas development of wells in their whole life cycle to balance the relationship between production objectives and development costs.The solutions are as follows:(1)calculate the gold target index and pinpoint the location of horizontal well drilling target,and shale reservoirs are depicted accurately by geophysical and other means to build underground transparent geological body;(2)optimize the drilling and completion process,improve the adaptability of key tools by cooling,reducing density and optimizing the performance of drilling fluid,the“man-made gas reservoir”is built by comprehensively considering the characteristics of in-situ stress and fractures after the development well is drilled;(3)through efficient management,establishment of learning curve and optimization of drainage and production regime,the development quality and efficiency of the well are improved across its whole life cycle,to fulfil“extreme utilization”development of shale gas.The practice shows that the estimated ultimate recovery of single wells in southern Sichuan Basin increase by 10%-20%than last year.

Autism spectrum disorders(ASD) are highly heterogeneous pediatric developmental disorders with estimated heritability more than 70%. Although the genetic factors in ASD are mainly unknown, a large number of gene mutations have been found, especially in genes involved in neurogenesis. The Neurexin-Neuroligin-Shank(NRXN-NLGN-SHANK) pathway plays a key role in the formation, maturation and maintenance of synapses, consistent with the hypothesis of neurodevelopmental abnormality in ASD. Presynaptic NRXNs interact with postsynaptic NLGNs in excitatory glutamatergic synapses. SHANK proteins function as core components of the postsynaptic density(PSD) by interacting with multiple proteins. Recently, deletions and point mutations of the SHANK1 gene have been detected in ASD individuals, indicating the involvement of SHANK1 in ASD. This review focuses on the function of SHANK1 protein, Shank1 mouse models, and the molecular genetics of the SHANK1 gene in human ASD.

Microfacies types and distribution of epicontinental shale: A case study of the Wufeng–Longmaxi shale in southern Sichuan Basin, China

For black shales,laminae and bedding are hard to identify,grain size is difficult to measure,and trace fossils do not exist.Taking the Ordovician Wufeng–Silurian Longmaxi shale in southern Sichuan Basin,China,as an example,the types,characteristics and models of microfacies in epicontinental shale are analyzed by means of full-scale observation of large thin sections,argon-ion polishing field emission-scanning electron microscopy(FE-SEM),and kerogen microscopy.The epicontinental sea develops delta,tidal flat and shelf facies,with black shale found in microfacies such as the underwater distributary channel and interdistributary bay under delta front facies,the calcareous and clayey flats under intertidal flat facies,the calcareous and clayey shelfs under shallow shelf facies,the deep slope,deep plain and deep depression under deep shelf facies,and the overflow under gravity flow facies.Basinward,silty lamina decreases and clayey lamina increases,the grain size changes from coarse silt to fine mud,the silica content increases from about 20%to above 55%,the carbonate and clay minerals content decreases from above 40%to around 10%,and the kerogen type changes from type II2 to type II1 and type I.Provenance and topography dominate the types and distribution of shale microfacies.The underwater distributary channel,interdistributary bay,clayey flat,clayey shelf,and overflow microfacies are developed in areas with sufficient sediment supply.The calcareous flat and calcareous shelf are developed in areas with insufficient sediment supply.The deep shelf shale area is divided into deep slope,deep plain,and deep depression microfacies as a result of three breaks.The formation of epicontinental shale with different microfacies is closely related to the tectonic setting,paleoclimate,and sea level rise.The relatively active tectonic setting increases the supply of terrigenous clasts,forming muddy water fine-grained sediment.The warm and humid paleoclimate is conducive to the enrichment of organic matter.The rapid sea level rise
