A series of macroporous CuO-ZnO-ZrO2 （CZZ） catalysts with different Zn/Zr ratios were successfully prepared by template method and characterized by X-ray diffraction （XRD）, N2 adsorption, reactive N2O adsorption, scanning electron microscopy （SEM）, h2 temperature-pro- grammed reduction （h2-TPR）, and transmission electron microscopy （TEM）. The activity of the catalysts was tested for methanol synthesis from CO2 hydrogenation. It is found that the increase in the Zn/Zr ratio could lead to the sintering of the catalysts, destroying the macroporous structure integrity. The macroporous CZZ catalysts own lower Zn/Zr ratio, exhibiting a better morphology and activity. For comparison, the conventional nonporous CZZ catalysts were also investigated. The results show that the CZZ catalysts with macroporous structure own smaller particles, higher CO2 conversion, and Ch3Oh yield. It reveals that the macroporous structure could inhibit the growth of the par- ticle size, and the special porous structure is favorable for diffusion and penetration of CO2, which could improve the catalytic activities.

[目的]建立稳定的食线虫真菌淡紫紫孢菌遗传转化体系,并获得插入突变体。[方法]介导的方法,以淡紫紫孢菌20-7的分生孢子为受体,将新构建的携带beta tubulin基因的质粒转化进入淡紫紫孢菌的细胞中,通过优化诱导乙酰丁香酮(AS)的浓度、诱导培养时间、农杆菌终浓度OD660值、共培养AS的浓度、共培养时间和共培养温度等因子,建立高效遗传转化体系,获得致病力不同的突变体。[结果]共培养过程中使用萌发孢子是成功建立淡紫紫孢菌遗传转化体系的必要条件;淡紫紫孢菌萌发的孢子与农杆菌EhA105在25℃共振荡培养48 h时,且在共培养阶段当乙酰丁香酮浓度为200μg·mL^(−1)(ph5.5)时转化效率最高,转化效率为1200~3200个转化子/106分生孢子,阳性抗性转化子比率为96%;转化子PCR表明,T-DNA已整合到淡紫紫孢菌的基因组中;Southern杂交验证表明,83.3%的转化子为T-DNA单拷贝插入;成功建立了可靠的淡紫紫孢菌的遗传转化体系,并从20个转化子中筛选到16个致病力变异的突变体。[结论]本研究成功构建了农杆菌介导的、以beta-tubulin基因为选择标记的淡紫紫孢菌高效遗传转化体系,并获得致病力变异的插入突变体,为淡紫紫孢菌的基因功能、致病机制研究及优良菌株选育奠定了基础。

We recently reported that inhibitors against human dihydroorotate dehydrogenase(DhODh)have broad-spectrum antiviral activities including their inhibitory efficacies on SARS-CoV-2 replication in infected cells.however,there are limited data from clinical studies to prove the application of DhODh inhibitors in Coronavirus disease 2019(COVID-19)patients.In the present study,we evaluated Leflunomide,an approved DhODh inhibitor widely used as a modest immune regulator to treat autoimmune diseases,in treating COVID-19 disease with a small-scale of patients.Cases of 10 laboratory-confirmed COVID-19 patients of moderate type with obvious opacity in the lung were included.Five of the patients were treated with Leflunomide,and another five were treated as blank controls without a placebo.All the patients accepted standard supportive treatment for COVID-19.The patients given Leflunomide had a shorter viral shedding time(median of5 days)than the controls(median of 11 days,P=0.046).The patients given Leflunomide also showed a significant reduction in C-reactive protein levels,indicating that immunopathological inflammation was well controlled.No obvious adverse effects were observed in Leflunomide-treated patients,and they all discharged from the hospital faster than controls.This preliminary study on a small-scale compassionate use of Leflunomide provides clues for further understanding of Leflunomide as a potential antiviral drug against COVID-19.

The methylation of lysine 4 of histone h3(h3K4),catalyzed by the histone methyltransferase KMT2/SET1,has been functionally identified in many pathogenic fungi but remains unexplored in nematode-trapping fungi(NTFs).here,we report a regulatory mechanism of an h3K4-specific SET1 orthologue,AoSET1,in the typical nematode-trapping fungus Arthrobotrys oligospora.When the fungus is induced by the nematode,the expression of AoSET1 is up-regulated.Disruption of AoSet1 led to the abolishment of h3K4me.Consequently,the yield of traps and conidia ofΔAoSet1 was significantly lower than that of the WT strain,and the growth rate and pathogenicity were also compromised.Moreover,h3K4 trimethylation was enriched mainly in the promoter of two bZip transcription factor genes(AobZip129 and AobZip350)and ultimately up-regulated the expression level of these two transcription factor genes.In theΔAoSet1 and Aoh3K4A strains,the h3^(K4)me modification level was significantly decreased at the promoter of transcription factor genes AobZip129 and AobZip350.These results suggest that AoSET1-mediated h3KEme serves as an epigenetic marker of the promoter region of the targeted transcription factor genes.Furthermore,we found that AobZip129 negatively regulates the formation of adhesive networks and the pathogenicity of downstream AoPABP1 and AoCPR1.Our findings confirm that the epigenetic regulatory mechanism plays a pivotal role in regulating trap formation and pathogenesis in NTFs,and provide novel insights into the mechanisms of interaction between NTFs and nematodes.

The two mutants idr1-1 and 297-28, which were obtained from the radiation mutation of hD297 and IAPAR9, were used as experimental materials in this study for a 2-year(2012 and 2013) experiment about field drought resistance identification in Beijing, China. Key agronomic traits and water-related physiological indexes were observed and measured, including the leaf anti-dead level(LADL), days to heading, plant height, setting percentage, aboveground biomass, leaf water potential(LWP), net photosynthetic rate(Pn) and transpiration rate. The results showed that the mutant idr1-1 that was under drought stress(DS) conditions for 2 years had the highest LADL grades(1.3 and 2.0) among all the materials, and they were 2–3 grades stronger than the wild-type IAPAR9 with an average that was 21.4% higher for the setting percentage than the wild type. Compared with the IAPAR9 for the 2-year average delay in the days to heading and the reduction rates in the plant height, setting percentage, and aboveground biomass under DS compared with the well-watered(WW) treatment, idr1-1 showed 3.2% less delay and 19.1, 16.4, and 6.1% less reduction, respectively. The idr1-1 in the LWP always exhibited the highest performance among all the materials. The Pn of idr1-1 under severe and mild DS comparing with that under WW was slightly decreased and even slightly increased, respectively, leading to an average reduction rate of only 0.92%, which was 26.93% less than that of IAPAR9. Under the severe DS, idr1-1 still showed the highest value of 16.88 μmol CO2 m–2 s–1 among all the materials and was significantly higher than that of IAPAR9(11.66 μmol CO2 m–2 s–1). Furthermore, only idr1-1 had the increased and the highest transpiration rate values(7.6 and 6.04 mmol h2 O m–2 s–1) under both mild and severe DS compared with the values under WW, when the transpiration rate of all the other materials significantly decreased. Gluconeogenesis responses was assessed during a short starvation period and subsequent refeeding in Siberian sturgeon(Acipenser baerii) previously fed different dietary carbohydrates levels and experienced to a glucose stimuli during early life. The sturgeon larvae were previously fed either a high glucose diet(G) or a low glucose diet(F) from the first feeding to yolk absorption(8 to 12 d post-hatching [dph]). Each group of fish was sub-divided into 2 treatments at 13 dph and was fed either a high-carbohydrate diet(h) or a low carbohydrate diet(L) until 20 wk. In the current study, the fish in 4 groups(GL, FL, Gh and Fh) were experienced to starvation for 21 d following by re-feeding of their corresponding diets for 21 d.Fish were sampled at postprandial 6 and 24 h before starvation(P6 h and P24 h), starvation 7,14 and 21 d(S7, S14 and S21) and 1, 7,14 and 21 d during refeeding(R1, R7, R14 and R21). Plasma samples during refeeding were taken at P6 h at each time point. Glycaemia levels, liver and muscle glycogen contents,activities and mRNA levels of hepatic gluconeogenic enzymes were examined. We found that both dietary carbohydrate levels and early glucose stimuli significantly affected the metabolic responses to starvation and refeeding in Siberian sturgeon(P he liver glycogen and then improved gluconeogenesis when the dietary carbohydrates were abundant, whereas preserved the liver glycogen stores at a stable level and more effectively promoted gluconeogenesis when the dietary carbohydrates are absent to maintain glucose homoeostasis.During refeeding, as most teleostean, Siberian sturgeon failed controlling the activities and mRNA levels of phosphoenolpyruvate carboxykinase cytosolic forms(PEPCK-C), fructose-1,6-bisphosphatase(FBPase),but particularly controlled phosphoenolpyruvate carboxykinase mitochondrial forms(PEPCK-M) activities and mRNA expression of glucose-6-phosphatase(G6 Pase, except in GL group). Background:hemorrhoids are one of the most common conditions that lead to surgery,and until now surgical hemorrhoidectomy has been the major effective treatment.Post-operative pain from hemorrhoidectomy has been experienced by thousands of patients and remains a major inconvenience of the operation.Objective:This study evaluates the clinical efficacy of the pestle needle therapy,an acupoint stimulation method,for relief of post-hemorrhoidectomy pain.Design,setting,participants and interventions:This was a single-center,patient-assessor-blinded and randomized controlled trial with 154 patients receiving Milligan hemorrhoidectomy surgery.Eligible patients were randomly assigned to either a treatment group or a control group at a ratio of 1:1.The treatment group received the pestle needle therapy,with manual stimulation at Yaoshu(DU2),Mingmen(DU4),Changqiang(DU1),Chengshan(BL57),Erbai(EX-UE2)and the perianal points(1,3,5,7,9,and 11 o’clock around the lesion);while the control group received a sham treatment with very light pressure.Three sessions of treatment were performed at 30 min,4 h and 12 h after the surgery,and each lasted for 15 min.Main outcome measures:The primary outcome was post-operative pain measured with the visual analogue scale(VAS)at 12 h after surgery.The secondary outcomes included the VAS scores measured at0.5,2,4,6,8,24 and 48 h after surgery,the analgesic dose,the time and the VAS score of the patients’first defecation after surgery,as well as the hamilton Rating Scale for Anxiety(hAMA)evaluated before discharge.Results:The mean pain score of the treatment group was significantly lower than that of the control group(3.10±1.27 vs 4.82±1.29;Ph after surgery.Compared with the control group,patients in the treatment group needed a smaller dose of analgesic within the first 24 hours after surgery(P=0.002);and their hAMA scores before discharge were lower(4.07±2.40 vs 5.10±2.45,P=0.009).Compared to the treatment group,patients in the control group had a greater time to the first defecat

针对GJ-6型轨道检测系统开启后激光器即进入长时间点亮的单一工作模式,提出了适用于激光摄像同步触发系统做等频脉宽变换及同步协调控制,实现激光频闪最亮发光状态与高速相机等距离空间同步,从而采集钢轨图像的设计方案。因触发时序控制和电信号有效识别是设计方案的关键,因此先进微处理器控制设计和光耦隔离电路、反相整形电路、增益放大电路等硬件电路设计成为系统的关键技术。经试验室与现场试验分析,本设计方案下的激光器与相机同步触发稳定。在120km·h^(-1)速度等级下,与长亮工作模式相比激光器的能耗降低了约94%,延长了激光器的工作寿命,避免了非工作时段外射激光的安全隐患,提高了轨道检测系统的可靠性和安全性。

Fabricating highly efficient and robust oxygen reduction reaction(ORR)electrocatalysts is challenging but desirable for practical Zn-air batteries.As an early transition-metal oxide,zirconium dioxide(ZrO_(2))has emerged as an interesting catalyst owing to its unique characteristics of high stability,anti-toxicity,good catalytic activity,and small oxygen adsorption enthalpies.however,its intrinsically poor electrical conductivity makes it difficult to serve as an ORR electrocatalyst.herein,we report ultrafine N-doped ZrO_(2) nanoparticles embedded in an N-doped porous carbon matrix as an ORR electrocatalyst(N-ZrO_(2)/NC).The N-ZrO_(2)/NC catalyst displays four-electron reduction of oxygen in O.1 M KOh,Upon employment in a Zn-air battery,N-ZrO,/NC presented an exellent activity and long-term durability with a half-wave potential(E,v2)of 0.84 V and a selectivity for the intriguing powerdensity of 185.9 mwcm^(-2).anda high secific capacity of 797.9 mA h gzni,exceeding those of commercial Pt/C(122.1 mw cm^(-2) and 782.5 mA h gzn),This excellent performance is mainly ttributed to the ultrafine ZrO_(2) nanoparticles the conductive carbon substrate,and the modifed electronic band structure of ZrO_(2) after N-doping.Density functional theory calculations demonstrated that N-doping can reduce the band-gap of ZrO_(2) from 3.96 eV to 3.33 eV through the hybridization of the p state of the N atom with the 2p state of the oxygen atom;this provides enhanced electrical conductivity and results in faster electron-transfer kinetics.This work provides a new approach for the design of other enhanced semiconductor and insulator materials.

heterostructure is an effective strategy to facilitate the charge carrier separation and promote the photocatalytic performance.In this paper,uniform SrTiO3 nanocubes were in-situ grown on TiO2 nanowires to construct heterojunctions.The composites were prepared by a facile alkaline hydrothermal method and an in-situ deposition method.The obtained SrTiO3/TiO2 exhibits much better photocatalytic activity than those of pure TiO2 nanowires and commercial TiO2(P25)evaluated by photocatalytic water splitting and decomposition of Rhodamine B(RB).The hydrogen generation rate of SrTiO3/TiO2 nanowires could reach 111.26 mmol·g^−1·h^−1 at room temperature,much better than those of pure TiO2 nanowires(44.18 mmol·g^−1·h^−1)and P25(35.77 mmol·g^−1·h^−1).The RB decomposition rate of SrTiO3/TiO2 is 7.2 times of P25 and 2.4 times of pure TiO2 nanowires.The photocatalytic activity increases initially and then decreases with the rising content of SrTiO3,suggesting an optimum SrTiO3/TiO2 ratio that can further enhance the catalytic activity.The improved photocatalytic activity of SrTiO3/TiO2 is principally attributed to the enhanced charge separation deriving from the SrTiO3/TiO2 heterojunction.
