Face passive failure can severely damage existing structures and underground utilities during shallow shield tunneling,especially in coastal backfill sand.In this work,a series of laboratory model tests were developed and conducted to investigate such failure,for tunnels located at burial depth ratios for which C/D=0.5,0.8,1,and 1.3.Support pressures,the evolution of failure processes,the failure modes,and the distribution of velocity fields were examined through model tests and numerical analyses.The support pressure in the tests first rose rapidly to the elastic limit and then gradually increased to the maximum value in all cases.The maximum support pressure decreased slightly in cases where C/D=0.8,1,and 1.3,but the rebound was insignificant where C/D=0.5.In addition,the configuration of the failure mode with C/D=0.5 showed a wedge-shaped arch,which was determined by the outcropping shear failure.The configuration of failure modes was composed of an arch and the inverted trapezoid when C/D=0.8,1,and 1.3,in which the mode was divided into lower and upper failure zones.

Tetracycline repressor(TetR)family regulators(TFRs)are the largest group of DNA-binding transcription factors and are widely distributed in bacteria and archaea.TFRs play vital roles in controlling the expression of various genes and regulating diverse physiological processes.Recently,a TFR protein Pseudomonas virulence regulator A(PvrA),was identified from Pseudomonas aeruginosa as the transcriptional activator of genes involved in fatty acid utilization and bacterial virulence.Here,we show that PvrA can simultaneously bind to multiple pseudo-palindromic sites and upregulate the expression levels of target genes.Cryo-electron microscopy(cryo-EM)analysis indicates the simultaneous DNA recognition mechanism of PvrA and suggests that the bound DNA fragments consist of a distorted B-DNA double helix.The crystal structure and functional analysis of PvrA reveal a hinge region that secures the correct domain motion for recognition of the promiscuous promoter.Additionally,our results showed that mutations disrupting the regulatory hinge region have differential effects on biofilm formation and pyocyanin biosynthesis,resulting in attenuated bacterial virulence.Collectively,these findings will improve the understanding of the relationship between the structure and function of the TetR family and provide new insights into the mechanism of regulation of P.aeruginosa virulence.

Significant lake expansion has accelerated permafrost degradation on the Qinghai-Tibet Plateau

In recent years, lakes on the Qinghai-Tibet Plateau have become more responsive to climate change. In September 2011, Zonag Lake in Hoh Xil experienced sudden drainage, the water eventually flowed into Yanhu Lake, which caused Yanhu Lake to continue to expand. The potential collapse of Yanhu Lake could directly threaten the operational safety of the adjacent Qinghai-Tibet Highway, Qinghai-Tibet Railway. To explore the implications of expanding lakes on the surrounding permafrost, we selected Hoh Xil Yanhu Lake on the Qinghai-Tibet Plateau to study the effect of lake expansion on permafrost degradation. The permafrost degradation in the Yanhu Lake basin from October 2017 to December 2022 was inverted using Sentinel-1 satellite image data and small baseline subset interferometry synthetic aperture radar(SBAS-In SAR) technology. Additionally, permafrost degradation from February 2007 and February 2010 was analyzed using advanced land observing satellite phased array-type L-band synthetic aperture radar(ALOS PALSAR) satellite images and differential interferometric synthetic aperture radar(D-In SAR) technique. The results showed that the permafrost around Yanhu Lake experienced accelerated degradation. Prior to the expansion of Yanhu Lake, the average annual deformation rate along the line of sight(LOS) direction was 6.7 mm/yr. After the expansion, the rate increased to 20.9 mm/yr. The integration of spatial-temporal distribution maps of surface subsidence, Wudaoliang borehole geothermal data, meteorological data, Yanhu Lake surface area changes, and water level changes supports the assertion that the intensified permafrost degradation could be attributed to lake expansion rather than the rising air temperature. Furthermore, permafrost degradation around Yanhu Lake could impact vital infrastructure such as the adjacent Qinghai-Tibet Highway and Qinghai-Tibet Railway.

路面病害检测是保障人民交通安全的重要方式,为了提高路面病害检测的准确率,实现及时、精准的路面病害检测,提出了一种重参数化YOLOv8路面病害检测算法。在主干网络引入CNX2f模块,提高网络对路面病害特征的提取能力,有效解决路面病害特征与背景环境特征易混淆问题;引入RepConv和DBB重参数化模块,增强多尺度特征融合能力,解决路面病害尺度差异较大问题;改进头部采用共享参数结构,并引入RBB重参数模块,解决头部参数冗余问题,并提高特征提取能力;引入SPPF_Avg模块,解决路面病害特征丢失问题,丰富多尺度特征表达。实验结果表明,改进后的路面病害检测网络精度为73.3%、召回率为62.3%、mAP为69.3%,较YOLOv8网络分别提高了2.6、3.0、2.8个百分点,提高了模型的检测效果。

The tree ring has been regarded as an emerging archive to reconstruct historical atmospheric mercury(Hg)trends,but with the large knowledge gaps in the reliability.In this study,we comprehensively evaluated the Hg source,radial translocation and age effect of Masson pine(Pinus massoniana)tree ring at Mt.Jinyun in Chongqing,to assess the suitability of such tree ring as the archive of atmospheric Hg.Results showed that distinct variabilities among Masson pine tree-ring Hg concentration profiles.The Hg concentration significantly increased along with stem height(P<0.05),indicating the Hg in tree rings mainly derived from foliage uptake atmospheric Hg.We found a distinct age effect that the tree ring of young trees had the higher Hg concentration.Besides,we used the advection-diffusion model to demonstrate how Hg concentration shifted by the advection or/and diffusion in tree rings.The modeling results showed that the advection induced radial translocation during the young growth period of tree was a plausible mechanism to result in the tree-ring Hg record largely different from the trend of anthropogenic Hg emissions in Chongqing.We finally suggest that in further Hg dendrochemistry,better discarding the tree-ring Hg profile of the young growth period to reduce impacts of the radial translocation and age effect.

Anammox is widely considered as the most cost-effective and sustainable process for nitrogen removal.However,how to achieve the enrichment of anammox biomass remains a challenge for its large-scale application,especially in mainstream wastewater treatment.In this study,the feasibility of enrichment of anammox biomass was explored through the realization of partial denitrification and the addition of bio-carriers.By using ordinary activated sludge,a sequencing batch reactor(SBR)followed by an up-fow anaerobic sludge bed(UASB)was operated at 25±2℃ for 214 days.The long-term operation was divided into five phases,in which SBR and UASB were started-up in Phases I and II,respectively.By eliminating oxygen and adjusting the infow ratios in Phases III-V,advanced nitrogen removal was achieved with the effuent total nitrogen being 4.7 mg/L and the nitrogen removal efficiency being 90.5%in Phase V.Both in-situ and ex-situ activity tests demonstrated the occurrence of partial denitrification and anammox.Moreover,16S rRNA high-throughput sequencing analysis revealed that Candidatus Brocadia was enriched from below the detection limit to in biofilms(0.4%in SBR,2.2%in UASB)and the foc sludge(0.2%in SBR,1.3%in UASB),while Thauera was mainly detected in the foc sludge(8.1%in SBR,8.8%in UASB),which might play a key role in partial denitrification.Overall,this study provides a novel strategy to enrich anammox biomass driven by rapid achievement of partial denitrification through the addition of bio-carriers,which will improve large-scale application of anammox processes in mainstream wastewater treatment.

Performance and mechanism of La-Fe metal-organic framework as a highly efficient adsorbent for fluoride removal from mine water

Water fluoride pollution has caused non-negligible harm to the environment and humans,and thus it is crucial to find a suitable treatment technology.In this study,La-Fe@PTA adsorbent was synthesized for the defluoridation of mine water.The results showed that the optimum conditions for defluoridation by La-Fe@PTA were p H close to 7.0,the initial F-concentration of 10 mg/L,the dosage of 0.5 g/L and the adsorption time of 240 min.Compared with SO_4^(2-),Cl^(-),NO_(3)^(-),Ca^(2+)and Mg^(2+),CO_(3)^(2-)and HCO_(3)^(-)presented severer inhibition on fluoride uptake by La-Fe@PTA.The adsorption process fits well with the pseudo-second-order kinetic model and Freundlich model,and the maximum adsorption capacity of Langmuir model was 95 mg/g.Fixed-bed adsorption results indicated that fluoride in practical fluorinated mine water could be effectively removed from 3.6 mg/L to less than 1.5 mg/L within130 bed volume(BV)by using 1.5 g La-Fe@PTA.Furthermore,the adsorbent still had good adsorption capacity after regeneration,which confirms the great application potential of La-Fe@PTA as a fluoride ion adsorbent.The mechanism analysis showed that La-Fe@PTA adsorption of fluorine ions is a physicochemical reaction driven by electrostatic attraction and ion exchange.
