The binding energy spectra and the momentum distributions of the valence orbitals of iso-butane, also known as methylpropane (CH_(3)CH(CH_(3))CH_(3)), are studied by using a high resolution binary (e, 2e) electron momentum spectrometer, at an impact energy of 1200eV plus the binding energy (8-32eV) with symmetric non-coplanar kinematics. Binding energy spectra of the valence shell are obtained at a coincidence energy resolution of 0.95 eV in full width at halfmaximum. The experimental momentum profiles of the inner valence orbitals are obtained and compared with the theoretical momentum distributions calculated by using Hartree-Fock and density functional theory methods with the basis sets 6-31G and 6-311++G**. The pole strengths of the inner orbitals are estimated and the final correlation effects are discussed.

Asparagus setaceus is a popular ornamental plant cultivated in tropical and subtropical regions ***,we constructed a chromosome-scale reference genome of *** to facilitate the investigation of its genome characteristics and *** a combination of Nanopore long reads,Illumina short reads,10×Genomics linked reads,and Hi-C data,we generated a high-quality genome assembly of *** covering 710.15 Mb,accounting for 98.63%of the estimated genome size.A total of 96.85%of the sequences were anchored to ten superscaffolds corresponding to the ten *** genome of *** was predicted to contain 28,410 genes,25,649(90.28%)of which were functionally annotated.A total of 65.59%of the genome was occupied by repetitive sequences,among which long terminal repeats were predominant(42.51%of the whole genome).Evolutionary analysis revealed an estimated divergence time of *** from its close relative *** of~9.66 million years ago,and *** underwent two rounds of whole-genome *** addition,762 specific gene families,96 positively selected genes,and 76 resistance(R)genes were detected and functionally predicted in *** findings provide new knowledge about the characteristics and evolution of the *** genome,and will facilitate comparative genetic and genomic research on the genus Asparagus.

Starch is the most important component in endosperm of sorghum ***,two types of starch are present:amylose(AM)and amylopectin(AP).The levels of AM and AP contents play a significant role in the appearance,structure,and quality of sorghum grains and in marketing *** the present study,a panel of 634 sorghum(Sorghum bicolor(L.)Moench)accessions were evaluated for starch,AM,and AP contents of grain,which included a mini core collection of 242 accessions from the International Crops Research Institute for the Semi-Arid Tropics(ICRISAT)in India,and 252 landraces and 140 cultivars from *** average starch content was 67.64%and the average AM and AP contents were 20.19 and 79.81%,*** developed a total of 260000 high-confidence single nucleotide polymorphism(SNP)markers in the panel of 634 accessions of *** using specific locus amplified fragment sequencing(SLAF-seq).We performed genome-wide association studies(GWAS)of starch,AM,and AM/AP of grain and SNP markers based on a mixed linear model(MLM).In total,70 significant association signals were detected for starch,AM,and AM/AP ratio of grain with P<4.452×10^-7,of which 10 SNPs were identified with significant starch,51 SNPs were associated with AM,and nine SNPs were associated with the AM/AP *** Gene Ontology(GO)analysis identified 12 candidate genes at five QTLs associated with starch metabolism within the 200-kb intervals,located on chromosomes 1,5,6,and *** these genes,Sobic.006G036500.1 encodes peptidyl-prolyl cis-trans-isomerase CYP38 responsible for hexose monophosphate shunt(HMS)and Sobic.009G071800 encodes 6-phospho-fructokinase(PFK),which is involved in the embden-meyerhof pathway(EMP).Kompetitive allele specific PCR(KASP)markers were developed to validate the GWAS *** C allele is correlated with a high starch content,while the T allele is linked with a low level of starch content,and provides reliable haplotypes for MAS in sorghum quality improvement.

The heterotrimeric GTP-binding proteins（G-proteins） in eukaryotes consisted of α, β and γ subunits and are important in molecular signaling by interacting with G-protein-coupled receptors（GPCR）, on which to transduce signaling into the cytoplast through appropriate downstream effectors. However, downstream effectors regulated by the G-proteins in plants are currently not well defined. In this study, the transcripts of AGB1, a G protein β subunit gene in Arabidopsis were found to be down-regulated by cold and heat, but up-regulated by high salt stress treatment. AGB1 mutant（agb1-2） was more sensitive to high salt stress than wild-type（WT）. Compared with WT, the cotyledon greening rates, fresh weight, root length, seedling germination rates and survival rates decreased more rapidly in agb1-2 along with increasing concentrations of Na Cl in normal（MS） medium. Physiological characteristic analysis showed that compared to WT, the contents of chlorophyll, relative proline accumulation and peroxidase（POD） were reduced, whereas the malonaldehyde（MDA） content and concentration ratio of Na＋/K＋ were increased in agb1-2 under salt stress condition. Further studies on the expression of several stress inducible genes associated with above physiological processes were investigated, and the results revealed that the expressions of genes related to proline biosynthesis, oxidative stress response, Na＋ homeostasis, stress- and ABAresponses were lower in agb1-2 than in WT, suggesting that those genes are possible downstream genes of AGB1 and that their changed expression plays an important role in determining phenotypic and physiologic traits in agb1-2. Taken together, these findings indicate that AGB1 positively regulates salt tolerance in Arabidopsis through its modulation of genes transcription related to proline biosynthesis, oxidative stress, ion homeostasis, stress- and ABA-responses.

Central precocious puberty(CPP) refers to a human syndrome of early puberty initiation with characteristic increase in hypothalamic production and release of gonadotropin-releasing hormone(Gn RH). Previously, loss-of-function mutations in human MKRN3, encoding a putative E3 ubiquitin ligase, were found to contribute to about 30% of cases of familial CPP. MKRN3 was thereby suggested to serve as a ‘brake’ of mammalian puberty onset, but the underlying mechanisms remain as yet ***, we report that genetic ablation of Mkrn3 did accelerate mouse puberty onset with increased production of hypothalamic Gn RH1. MKRN3 interacts with and ubiquitinates MBD3, which epigenetically silences GNRH1 through disrupting the MBD3 binding to the GNRH1 promoter and recruitment of DNA demethylase TET2. Our findings have thus delineated a molecular mechanism through which the MKRN3–MBD3 axis controls the epigenetic switch in the onset of mammalian puberty.

Efficient separation of palladium(Pd(Ⅱ))from high-level liquid waste(HLLW)not only helps the safe disposal of nuclear waste,but also is a vital importan supplement to Pd(Ⅱ)***,16 kinds of nove Amberlite XAD resins-based adsorbents were prepared by vacuum impregnation method for the separation of Pd(Ⅱ)from simulated HLLW,and the MMBT/XAD7HP prepared using 4-methyl-2-mercaptobenzothiazole(MMBT)as ligand and Amberlite XAD7HP resin(XAD7HP)as the carrier was selected as the optimal research *** analysis experiments proved the successful preparation of MMBT/XAD7HP,which exhibited ultrahigh adsorption selectivity for Pd(Ⅱ)(distribution coefficient(K_d)≥12584.5 ml·g^(-1),separation factor(SF_(Pd/M))≥2818.4)and superior adsorption capacity(Q≥130.1 mg·g^(-1)).The adsorption kinetics and adsorption isotherms for Pd(Ⅱ)matched with pseudo-secondorder(PSO)kinetics model and Langmuir model,indicating that the adsorption mode for Pd(Ⅱ)by MMBT/XAD7 HP was monolayer chemical *** dynamic column experiments showed that MMBT/XAD7 HP achieved efficiently separation of Pd(Ⅱ)from simulated HLLW,and the recovery rate was nearly 100% The adsorption mechanism was analyzed by Fourier transform infrared spectroscopy(FTIR),X-ray photoelectron spectroscopy(XPS)and density functional theory(DFT),and the ultrahigh selectivity of MMBT/XAD7 HP is attributed to the preferred affinity of the soft-N,S donor atoms in MMBT for Pd(Ⅱ).MMBT has the shortest frontline molecular orbital energy band gap(HOMO-LUMO)among the four ligands,which makes it easier to become an electron donor to attract Pd^(2+)to participate in ***_(3)^(-)participates in the adsorption to keep charge *** summary,MMBT/XAD7 HP has broad application prospects for the treatment of Pd(Ⅱ)in HLLW.

This study was designed to explore the RNA interference technique in inhibition of the expression of the mouse fibrinogen like protein 2(mfgl2),which has been reported to be involved in the development a variety of diseases including fulminant viral hepatitis.A plasmid named p-mfgl2shRNA,complementary to the sequence of mfgl2 was constructed,while another short hairpin RNA(shRNA) which was a mutated form of the mfgl2shRNA sequences was used as a control.A plasmid named pEGFP-mfgl2 expressing the mfgl2-EGFP fusion protein was also constructed for the screening of the effect of p-mfgl2shRNA on mfgl2 *** cotransfection of p-mfgl2shRNA and pEGFP-mfgl2 or pcDNA3.1-mfgl2 expression construct into CHO cells or HeLa cells,the inhibition of mfgl2 expression by mfgl2shRNA was analyzed by direct observation through fluorescent microscopy,FACS,RT-PCR and immunohistochemistry *** experiments showed the significant inhibitory effect of p-mfgl2shRNA on mfgl2 expression at 48h post-transfection in both CHO and Hela cell lines with the inhibitory efficiency as high as 80.1%.The study demonstrated that the construct of p-mfgl2shRNA successfully interfered with the mfgl2 expression in vitro.

Pedogenic Carbonate and Soil Dehydrogenase Activity in Response to Soil Organic Matter in Artemisia ordosica Community

Little attention has been paid to the role of soil organic matter （OM） in the formation of pedogenic carbonate in desert soils. The relationships among soil OM, soil dehydrogenase activity （DHA）, and soil CaCO3 in a plant community dominated by Artemisia ordosica, located on the eastern boundary of Tcngger Desert in the Alxa League, Inner Mongolia, China, were studied to understand whether OM was directly involved in the formation of pedogenic carbonate. The results showed that DHA and CuCO3 positively correlated with OM content, and DHA, OM, and CaCO3 were correlated with each other in their spatial distribution, indicating that abundant OM content contributed to the formation of CaCO3. Therefore, the formation of pedogenic CaCO3 was a biotic process in the plant community dominated by A. ordosica.
