Evaluation of media for simultaneous enumeration of total coliform and Escherichia coli in drinking water supplies by membrane filtration techniques

This study evaluated three different dehydrated media for simultaneous detection and enumeration of total coliform （TC） and Escherichia coil in drinking water samples with a standard membrane filtration procedure. The experiment indicated that the differential coliform agar （DCA） medium was the most effective among the tested media in enumerating TC and E. coil, without the need for extensive accompanying confirmation tests. The results for DCA medium were highly reproducible for both TC and E. coil with standard deviation of 6.0 and 6.1, respectively. A high agreement （82%） was found between DCA and m-Endo media on 152 drinking water samples in terms of TC positive. The DCA medium also reduced concealment of background bacteria.

A three-dimensional model of near-surface shear-wave velocity in the deep alluvial basin underlying the metropolitan area of Las Vegas, Nevada （USA）, is being developed for earthquake site response projections. The velocity dataset, which includes 230 measurements, is interpolated across the model using depth-dependent correlations of velocity with sediment type. The sediment-type database contains more than 1 400 well and borehole logs. Sediment sequences reported in logs are assigned to one of four units. A characteristic shear-wave velocity profile is developed for each unit by analyzing closely spaced pairs of velocity profiles and well or borehole logs. The resulting velocity model exhibits reasonable values and patterns, although it does not explicitly honor the measured shear-wave velocity profiles. Site response investigations that applied a preliminary version of the velocity model support a two-zone ground-shaking hazard model for the valley. Areas in which clay predominates in the upper 30 m are predicted to have stronger ground motions than the rest of the basin.

Background: Routine lung function testing requires expensive equipment, or requires maximum expiratory effort. The airflow perturbation device (APD) is a light handheld device, allowing for serial measures of respiratory resistance noninvasively and effortlessly. methods: In a convenience sample of 398 patients undergoing pulmonary function testing, we compared routine spirometric indices (forced expired volume in 1 second (FEV1), peak expiratory flow (PEF)), and airways resistance (Raw-272 patients), to measures of respiratory resistance measured with the APD including inspiratory (IR), expiratory (ER) and averaged (AR) resistance. Results: measures of lung function were significantly correlated (p 0.001). On regression analysis, between 7% - 17% of the variance (R2) for FEV1, PEF, and Raw was explained by APD measurements. Approximately 2/3 of the variance in FEV1 was explained by PEF measurements. Conclusions: APD measurements of lung function correlate with conventional measures. Future studies should be directed at exploring the use of the APD device in serial measures of lung function in patients with lung disease.
