AIM:To investigate the association of PNPLA3 polymorphisms with concurrent chronic hepatitis B(CHB) and nonalcoholic fatty liver disease(NAFLD).METHODS:A cohort of Han patients with biopsyproven CHB,with or without NAFLD(CHB group,n = 51;CHB + NAFLD group,n = 57),and normal controls(normal group,n = 47) were recruited from Northern(Tianjin),Central(Shanghai),and Southern(Zhangzhou) *** PNPLA3 polymorphisms were genotyped by gene *** association between PNPLA3 polymorphisms and susceptibility to NAFLD,and clinical characteristics of NAFLD were evaluated on the basis of physical indices,liver function tests,glycolipid metabolism,and histopathologic *** association of PNPLA3 polymorphisms and hepatitis B virus(HBV) load was determined by the serum level of HBV ***:After adjusting for age,sex,and body mass index,we found that four linked single nucleotide polymorphisms(SNPs) of PNPLA3,including the rs738409 G allele(CHB + NAFLD group vs CHB group:odds ratio[OR]= 2.77,95%confidence interval[CI]:1.18-6.54;P = 0.02),rs3747206 T allele(CHB+ NAFLD group vs CHB group:OR = 2.77,95%CI:1.18-6.54;P = 0.02),rs4823173 A allele(CHB +NAFLD group vs CHB group:OR = 2.73,95%CI:1.16-6.44;P= 0.02),and rs2072906 G allele(CHB+ NAFLD group vs CHB group:OR = 3.05,95%CI:1.28-7.26;P = 0.01),conferred high risk to NAFLD in CHB *** patients with both CHB and NAFLD,these genotypes of PNPLA3 polymorphisms were associated with increased susceptibility to nonalcoholic steatohepatitis(NASH)(NAFLD activity score ≥3;P =0.01-0.03) and liver fibrosis(>1 Metavir grading;P =0.01-0.04).As compared to those with C/C and C/G at rs738409,C/C and C/T at rs3747206,G/G and G/A at rs4823173,and A/A and A/G at rs2072906,patients in the CHB + NAFLD group with G/G at rs738409,T/T at rs3747206,A/A at rs4823173,and G/G at rs2072906 showed significantly lower serum levels of HBV DNA(PluSION:Four linked SNPs of PNPLA3(rs738409,rs3747206,rs4823173,and rs2072906) a

Impact of obstructive sleep apnea on pulmonary hypertension in patients with chronic obstructive pulmonary disease

Background:Chronic obstructive pulmonary disease (COPD) and obstructive sleep apnea (OSA) syndrome are highly prevalent respiratory conditions. Their coexistence is referred to as the overlap syndrome. They are both related to pulmonary hypertension (PH) development. This study investigated the effects of OSA on PH in patients with COPD and the associated factors. Methods: Consecutive patients with stable COPD were recruited for an observational cross-sectional study from September 2016 to May 2018 at Peking University Third Hospital. In total, 106 patients with COPD were enrolled and performed home portable monitoring and echocardiography. OSA was defined by an apnea hypopnea index (AHI)>10 events/h. Based on OSA absence or presence, patients were divided into the COPD with OSA and COPD without OSA groups. Factors affecting pulmonary artery pressure (PAP) and PH were identified using univariate analysis and logistic regression models. Results: In the 106 patients with COPD, the mean age was 69.52 years, 91.5% were men, and the mean forced expiratory volume in 1 s (FEV!) percentage of predicted was 56.15%. Fifty-six (52.8%) patients with COPD were diagnosed with OSA, and 24 (22.6%) patients with COPD were diagnosed as PH. Compared with COPD without OSA group, the median PAP in COPD with severe OSA group increased by 5 mmHg (36.00 [26.00-50.00] mmHg vs. 31.00 [24.00-34.00] mmHg, P = 0.036). Spinal cord injuries lead to significant loss of motor, sensory, and autonomic functions, presenting major challenges in neural regeneration. Achieving effective therapeutic concentrations at injury sites has been a slow process, partly due to the difficulty of delivering drugs effectively. Nanoparticles, with their targeted delivery capabilities, biocompatibility, and enhanced bioavailability over conventional drugs, are garnering attention for spinal cord injury treatment. This review explores the current mechanisms and shortcomings of existing treatments, highlighting the benefits and progress of nanoparticle-based approaches. We detail nanoparticle delivery methods for spinal cord injury, including local and intravenous injections, oral delivery, and biomaterial-assisted implantation, alongside strategies such as drug loading and surface modification. The discussion extends to how nanoparticles aid in reducing oxidative stress, dampening inflammation, fostering neural regeneration, and promoting angiogenesis. We summarize the use of various types of nanoparticles for treating spinal cord injuries, including metallic, polymeric, protein-based, inorganic non-metallic, and lipid nanoparticles. We also discuss the challenges faced, such as biosafety, effectiveness in humans, precise dosage control, standardization of production and characterization, immune responses, and targeted delivery in vivo. Additionally, we explore future directions, such as improving biosafety, standardizing manufacturing and characterization processes, and advancing human trials. Nanoparticles have shown considerable progress in targeted delivery and enhancing treatment efficacy for spinal cord injuries, presenting significant potential for clinical use and drug development.

Recent research has demonstrated the impact of physical activity on the prognosis of glioma patients,with evidence suggesting exercise may reduce mortality risks and aid neural *** role of the small ubiquitin-like modifier(SUMO)protein,especially post-exercise,in cancer progression,is gaining attention,as are the potential anti-cancer effects of *** used machine learning to create the exercise and SUMO-related gene signature(ESLRS).This signature shows how physical activity might help improve the outlook for low-grade glioma and other *** demonstrated the prognostic and immunotherapeutic significance of ESLRS markers,specifically highlighting how murine double minute 2(MDM2),a component of the ESLRS,can be targeted by *** underscores the intricate relationship between natural compounds such as nutlin-3 and immune *** comprehensive CRISPR screening,we validated the effects of specific ESLRS genes on low-grade glioma *** also revealed insights into the effectiveness of Nutlin-3a as a potent MDM2 inhibitor through molecular docking and dynamic ***-3a inhibited glioma cell proliferation and activated the p53 *** efficacy decreased with MDM2 overexpression,and this was reversed by Nutlin-3a or *** using a low-grade glioma mouse model highlighted the effect of physical activity on oxidative stress and molecular pathway ***,both physical exercise and Nutlin-3a administration improved physical function in mice bearing tumors derived from MDM2-overexpressing *** results suggest the potential for Nutlin-3a,an MDM2 inhibitor,with physical exercise as a therapeutic approach for glioma *** research also supports the use of natural products for therapy and sheds light on the interaction of exercise,natural products,and immune regulation in cancer treatment.

Wetland and habitat dynamics in the evolving landscape of the Middle Yangtze River Basin

Wetlands play a critical role in the global environment. The Middle Yangtze River Basin(MYRB), known for its abundant wetland resources, has experienced notable changes resulting from the complex interplay of environmental factors. Therefore, we investigated the spatiotemporal characteristics of wetland ecological quality in the MYRB from 2001 to *** the random forest(RF) regression algorithm and patch-generated land-use simulation(PluS) model, we forecasted variations in wetland habitat quality and their determinants under the Shared Socioeconomic Pathway-Representative Concentration Pathway(SSPRCP) framework from 2035 to 2095. The main findings are as follows:(1) The RF algorithm was optimal for land-use and land-cover(luLC) classification in the MYRB from 2001 to 2020,when notable changes were observed in water bodies and buildings. However, the forested area exhibited an increase and decrease of 3.9% and 1.2% under the SSP1-2.6 and SSP5-8.5 scenarios, respectively, whereas farmland exhibited a diminishing trend.(2) Wetlands were primarily concentrated in the central and eastern MYRB, with counties in the southwest exhibiting superior ecological-environmental quality from 2001 to 2020. Notably,wetland coverage revealed significantly high level, significant changes, frequent but relatively minor changes under the SSP1-2.6, SSP2-4.5, and SSP 5-8.5 scenarios, respectively.(3)Regions with lower habitat quality were primarily concentrated in urbanized areas characterized by frequent human activities, indicating a clear degradation in habitat quality across different scenarios. In conclusion, we established a foundational framework for future investigations into the eco-hydrological processes and ecosystem quality of watersheds.

Staged dendrite suppression for high safe and stable lithium-sulfur batteries

The unavoidable dendrite growth and shuttle effect have long been stranglehold challenges limiting the safety and practicality of lithium-sulfur ***,we propose a dual-action strategy to address the lithium dendrite issue in stages by constructing a multifunctional surface-negatively-charged nanodiamond layer with high ductility and robust puncture resistance on polypropylene （PP） *** uniformly loaded compact negative layer can not only significantly enhance electron transmission efficiency and promote uniform lithium deposition,but also reduce the formation of dendrite during early deposition *** importantly,under the strong puncture stress encountered during the deterioration of lithium dendrite growth under limiting current,the high ductility and robust puncture resistance（145.88 MPa） of as-obtained nanodiamond layer can effectively prevent short circuits caused by unavoidable lithium *** Li||Li symmetrical cells assembled with nanodiamond layer modified PP demonstrated a stable cycle of over 1000 h at 2 mA cm-2with a polarization voltage of only 29.3 ***,the negative charged layer serves as a physical barrier blocking lithium polysulfide ions,effectively mitigating capacity *** improved cells achieved a capacity decay of only 0.042%per cycle after 700 cycles at 3 C,demonstrating effective suppression of dendrite growth and capacity attenuation,showing promising prospect.

Efficient large-area flexible organic photovoltaic modules on transparent thin silver electrodes

Large-area flexible organic photovoltaic（OPV） modules are challenging to achieve high power conversion efficiency（PCE）.Electrical shunts or shortages generally occur in large-area OPV modules due to the ultrathin（100–200 nm） and soft active layers. Improving the surface smoothness of bottom transparent electrode will be able to suppress the shunts and shortages. In this work, we report a method to fabricate smooth and flexible transparent electrodes based on thin silver film, and further to fabricate efficient large-area flexible OPV modules. The fabricated thin silver transparent electrodes simultaneously have high conductivity, high optical transmittance, good mechanical flexibility and low surface roughness. Humidity exposure（h.e.）treatment on MoO3effectively facilitates the continuous growth of the thin transparent silver electrode. 6 nm silver film on MoO3（h.e.） on plastic substrate shows a transmittance up to 89%, a sheet resistance of 15 Ω/sq, and a low surface roughness with a rootmean-square value of 0.276 nm. Furthermore, 52-cm2flexible organic solar modules（15 subcells） showed a certified PCE of12.66%. Furthermore, a 1350 cm2flexible component assembled by 25 pieces of 52–55 cm2modules showed certified PCE of12.10% with an open-circuit voltage of 62.85 V.
