Topoisomerase I Inhibition Radiosensitizing Hepatocellular Carcinoma by RNF144A-mediated DNA-PKcs Ubiquitination and Natural Killer Cell Cytotoxicity

Background and Aims: Topoisomerase I (TOP1) partici-pates the repair of DNA double-strand breaks (DSBs) upon radiation therapy (RT). RNF144A mediates ubiquitination of catalytic subunit of DNA protein kinase (DNA-PKcs), a critical factor in DSB repair. This study aimed to investigate the natural killer (NK) cell-mediated radiosensitization with TOP1 inhibition and the mechanism by DNA-PKcs/RNF144A. Methods: In vitro synergism with TOP1i or cocultured NK cells and RT were evaluated in human hepatocellular car-cinoma (HCC) cell lines (Huh7/PLC5) by clonogenic surviv-als. Orthotopic xenografts were treated with Lipotecan and/or RT. Protein expression was analyzed by western blotting, immunoprecipitation, subcellular fractionation, and confocal microscopy. Results: Lipotecan/RT had a superior synergis-tic effect to RT on HCC cells. Combined RT/Lipotecan reduced the xenograft size by 7-fold than RT (p<0.05). Lipotecan caused more radiation-induced DNA damage and DNA-PKcs signaling. The expression of major histocompatibility com-plex class I-related chain A and B (MICA/B) on tumor cells is associated with the sensitivity to NK cell-mediated lysis. Cocultured NK and HCC cells with Lipotecan radiosensitized HCC cells/tissues with the expression of MICA/B. RNF144A increased more in Huh7 cells with combined RT/TOP1i, and reduced the prosurvival function of DNA-PKcs. The effect was reversed by inhibiting the ubiquitin/proteasome system. In comparison, RNF144A decreased through nuclear transloca-tion with the cumulated DNA-PKcs and radio-resistance of PLC5 cells. Conclusions: TOP1i reinforces NK cell-activated anti-HCC effect of RT through RNF144A mediated DNA-PKcs ubiquitination. RNF144A provides a reason for differentiating radiosensitization effect between HCC cells.

In this editorial,we comment on the article by Mu et al,published in the recent issue of the World Journal of Gastrointestinal *** pay special attention to the immune tolerance mechanism caused by hepatitis B virus(HBV)infection,the pathogenesis of hepatocellular carcinoma(HCC),and the role of antiviral therapy in treating HCC related to HBV *** infection leads to systemic innate immune tolerance by directly inhibiting pattern recognition receptor recognition and antiviral signaling pathways,as well as by inhibiting the immune functions of macrophages,natural killer cells and dendritic *** addition,HBV leads to an immunosuppressive cascade by expressing inhibitory molecules to induce exhaustion of HBV-specific cluster of differentiation 8+T cells,ultimately leading to long-term viral *** loss of immune cell function caused by HBV infection ultimately leads to ***-term antiviral therapy can improve the prognosis of patients with HCC and prevent tumor recurrence and metastasis.

Translocation of Heterogeneous Flexible Polymers Assisted by Binding Particles

A polymer chain usually contains two or more types of monomeric species from the perspective of polymer chemistry,which poses achallenge to the understanding of structure-property *** is of course true in the field of polymer *** the present work,Iinvestigate the translocation dynamics of heterogeneous flexible polymers composed of two types of monomers labeled A and B through ananopore assisted by binding particles(BPs)by using the coarse-grained Langevin dynamics simulations in two-dimensional ***,multiblock copolymers with different block lengths and monomeric components are considered.I critically examine how thetranslocation dynamics responds to the variations in the block length and the monomeric ***,it is found that the periodicstructure of a multiblock copolymer causes an obvious fingerprint feature in the residence time of individual monomers in which the number ofpeaks is exactly equal to the number of *** findings provide a basic understanding about the sequence-dynamics relationship for theBPs-assisted translocation of heterogeneous flexible polymers.

Serpentinisation is Required for the Magnetization of the Martian Crust

The remnant magnetism in the crust of Martian southern highland is associated with the magnetic sources at an average depth of~32 *** this work,we investigate the magnetization of Martian crust via 1-D parameterized model for the stagnant-lid mantle *** to our model,the magnetization of Martian crust is likely to take place in the top-down manner during 4.1-3.7 *** reproduce the average depth of magnetic sources below the southern highland,magnetite and Mg-ferrite are anticipated to be the magnetic carriers in the Martian crust,implying the serpentinisation *** magnetite is the only magnetic carrier in the Martian crust,the early climate must be warm enough to maintain a surface temperature of 300 K during 4.1-3.7 Ga at *** a warm climate is more likely to be a regional phenomenon associated with the serpentinisation in the crust of the southern highland or the hot ejecta of Borialis impact depositing on the southern hemisphere.

Effect of Wavefront Properties on Numerical Aperture of Fresnel Hologram in Incoherent Holographic Microscopy

Fresnel incoherent correlation holographic （FINCH） microscopy is a recently developed new technique, which employs a spatial light modulator （SLM） as the beam splitter. Light originating from the same object point is split into two beams by the wavefront-division phase mask displayed on SLM, and a Fresnel-zone plate-like hologram is formed by the interference of the two beams. The numerical aperture NAH of the recorded Fresnel hologram is introduced as a comprehensive parameter for evaluating the imaging characteristics in the FINCH scheme. The effect of wavefront properties on NAH of the hologram is investigated theoretically in the sense of optimizing imaging resolution. The variation trends of NAH are described and an optimal NAH is achieved by implementing an appropriate phase mask for a given recording distance between SLM and CCD; thus optimized imaging resolution and signal-to-noise ratio are demonstrated experimentally.

Penetration and lubrication evaluation of vegetable oil with nanographite particles for broaching process

With increasing environmental concerns,the substitution of mineral oil-based cutting fluid has become an urgent *** vegetable soybean oil as base fluid,nanofluid cutting fluids(NFCFs)were prepared by adding different weight concentrations of nanographite particles(NGPs),and their penetration and lubrication performances were studied.A novel simulated tool-chip slit with micrometersized geometry was manufactured to evaluate and quantify the penetration rate of the NFCFs by image analysis ***,a large number of comparative experiments on the closed-type broaching machine were carried out to compare the performance of the proposed NFCFs and a commercial cutting fluid in terms of cutting force,workpiece surface roughness,and metal *** is found that there is an optimal NGP concentration in NFCF for practical cutting *** the concentration of NGP is 0.4 wt%,the broaching process lubrication exhibits an ideal mixed lubricate state,resulting in minimal friction resistance,and thus,both the cutting force and chip curling angle reach their corresponding best ***,the proposed NGP-based vegetable-oil cutting fluid exhibits excellent environmentfriendliness and low-cost consumption in the minimal quantity lubrication(MQL)method;this demonstrates its potential for replacing the traditional broaching cutting fluid.

Background:Non-alcoholic fatty liver disease(NAFLD)is a liver disorder characterized by the accumulation and degeneration of fat in the liver cells,a condition that may further deteriorate and lead to cirrhosis and liver *** studies showed that metabolic dysfunction can promote NAFLD *** Zhugan Decoction(LGZGD)has therapeutic effects on *** mechanism of LGZGD still remains *** study was to examine the impact of LGZGD on the metabolic processes involved in the development of ***:A mice model of NAFLD was treated with *** therapeutic potential of LGZGD was evaluated by assessing the activity of transaminases,lipids levels of blood,and pathological changes in the liver of the mice model of ***,this study also evaluated the influence of LGZGD on liver inflammation and oxidative ***:The results of untargeted metabolomics analysis showed that LGZGD reduced the disordered lipid metabolism in NAFLD *** improved the oxidative stress and also reduced the levels of pro-inflammatory cytokines in the *** metabolomics analysis of liver samples revealed that LGZGD treatment improved metabolic disorders,including alanine,aspartate,glutamate,glycerophospholipid metabolism,and citrate *** RT-qPCR and Western blot results showed that LGZGD could regulate the expression of key enzymes in the metabolic pathway of the citrate cycle,including ATP-citrate lyase(ACLY),alanine-glyoxylate aminotransferase-2(AGXT2),phosphatidylethanolamine N-methyltransferase(PEMT),and succinate dehydrogenase(SDH).Conclusion:We found that LGZGD can treat NAFLD by reducing inflammatory responses,inhibiting oxidative stress,regulating alanine,aspartate,glutamate,and glycerophospholipid metabolism,and citrate cycle pathways.

Stochastic heterogeneous microstructures are widely applied in structural and functional materials,playing a crucial role in determining their performance.X-ray tomography and focused ion beam serial sectioning are frequently used methods to reconstruct three-dimensional(3D)microstructures,yet are demanding techniques and are ***,a highthroughput multi-stage 3D reconstruction method via distance correlation functions is developed using a single representatively large-sized 2D micrograph for stochastic microstructures,and verified by X-ray micro-tomography datasets of isotropic and anisotropic solid oxide fuel cell *** method provides an economic,easy-to-use and high-throughput approach for reconstructing stochastic heterogeneous microstructures for energy conversion and storage devices,and can readily be extended to other materials.

Numerical simulation on the pressure wave in a 30 mm electrothermal-chemical gun with the discharge rod plasma generator

An axisymmetric two-dimensional(2D)internal ballistic model including the transient burning rate law is used to simulate the 30mm electrothermal-chemical(ETC)launch with the discharge rod plasma generator(DRPG).The relationship between the pressure wave and the initial parameters,such as input electric power,discharging timing sequence,loading density and propellant web thickness,is researched through the change of initial parameters in the *** the condition of synchronous discharging,the maximum of the pressure wave can be controlled while the ratio of the input electric energy to the propellant chemical energy(electric energy ratio)is less than *** the electric energy ratio is larger than 0.11,the maximum of the pressure wave increases rapidly with the electric energy *** the increasing of the electric energy ratio,the change of the first negative amplitude value can be *** the condition of timing sequence discharging,the allowed input electric energy ratio to control the pressure wave is proportional to the current pulse *** the high electric energy ratio,the maximum of the pressure wave is inverse proportional to the current pulse *** pressure wave increases with the increasing of the loading *** the allowed electric energy ratio to control the pressure wave and the variation trend of the first negative amplitude wave value doesn't *** the discharging of the DRPG,the influence of changing propellant web thickness in ETC launch can be ignored.

Changing patterns of urban-rural nutrient flows in China:driving forces and options

Nutrient recycling has been practiced for thousands of years in China to maintain food production without environmental pollution. In the past three decades, however, the traditional nutrient recycling systems have been replaced with waste treatment systems, which have resulted in rapid and severe environmental pollution. By analyzing the primary driving forces of the changing nutrient flows(technology, labor costs, food supplies, fertilizer demands, environmental quality, human health, and public awareness), this paper argues that technology fundamentally motivated the nutrient-recycling strategy to address the malnutrition problem in traditional societies but has constrained the reconstruction of nutrient recycling systems in modern cities. With the availability of synthetic fertilizers in modern society, the lack of interdisciplinary views in policy making for nutrient management is the root cause of today's environmental situation. Ongoing fast urbanization has concentrated more nutrients in urban areas, creating the need for a national nutrient management plan to coordinate multiple ministries and fix the uncoupled nutrient cycling between urban and rural systems. Rebuilding the traditional nutrient-recycling systems is an environmentally and economically effective solution. There are three fundamental technological barriers to reconstructing the nutrient recycling systems, as follows: userfriendly toilets, the separation of sewage pipelines, and easy-to-use organic fertilizers made from human manure or other organic waste. Overcoming these barriers requires building institutional mechanisms,developing the necessary infrastructure, creating research funding, and providing open experimental platforms for technological development.
