Developmental competence of bovine oocytes with increasing concentrations of nano-copper and nano-zinc particles during in vitro maturation

Objective: To evaluate copper and zinc concentrations in plasma and follicular fluid from cattle ovaries, and estimate the impact of rational concentrations of copper and zinc oxide nanoparticles (CuO-NPs & ZnO-NPs) supplemented during in vitro maturation (IVM) against DNA damage of cumulus cells, glutathione content in oocytes and on consequent embryo development. Methods: Oocytes were obtained from 2 to 8 mm follicles by aspiration method for IVM. Replicates of experiments were performed on different days, with a separate batch of cumulus oocytes complex for each day. Results: The DNA damage of oocytes and cumulus cells significantly decreased with supplemental CuO-NPs or ZnO-NPs concentrations of 0.7 and 1.0 μg/mL in the IVM medium compared to medium without CuO-NPs or ZnO-NPs (P < 0.01). Total glutathione concentrations in oocytes and cumulus cells significantly increased following supplementation with both 0.7 and 1.0 μg/mL CuO-NPs or ZnO-NPs in comparison with 0 and 0.4 μg/mL CuO-NPs or ZnO-NPs supplemented groups (P < 0.01). Supplementation of CuO-NPs or ZnO-NPs during IVM medium at any concentration had no significant effect on cleavge rate. Both CuO-NPs and ZnO-NPs significantly increased blastocyst rates when oocytes were matured with 0.7, 1.0 μg/mL CuO-NPs concentrations (P < 0.01). In contrast, addition of 1.5 μg/mL of CuO-NPs or ZnO-NPs to the maturation media resulted in detrimental effects on the developmental competence of bovine oocytes confirming toxicity induced by CuO-NPs and ZnO-NPs in high concentrations. Conclusions: CuO-NPs or ZnO-NPs-treated bovine oocytes during IVM show low level of DNA fragmentation and increased intracellular glutathione content of cumulus cells. In vitro embryo development is improved by supplementation of rational concentrations of CuO-NPs or ZnO-NPs to culture media. Toxicity induced by CuO-NPs and ZnO-NPs is confirmed in high concentrations.

Insects are well recognized as a source of potentially useful compounds for modern medicine. Development of anticancer drugs from natural resources has been performed throughout the world. In the present study, anticancer activity of the hemolymph and fat body of Sarcophaga argyrostoma third larval instars is assayed against human breast cancer cell line (MDA-MB-231 cells). The cytotoxicity of the hemolymph and fat body samples were determined. The results showed that growth of MDA-MB-231 cells was inhibited at different concentrations upon 24 h of exposure. There is no inhibitory activity against Vero cells under these experimental conditions. Protein profile of the hemolymph and fat body were extracted and separated using sodium dodecyl sulphate-polyacrylamide gel electrophoresis (SDS-PAGE). Protein analysis showed differences in number of electrophoretic protein bands with different molecular weights between treated and control larvae. The present work assumes that hemolymph and fat body tissue have cytotoxicity against MDA-MB-231 cells and these results exhibit that larvae from S. argyrostoma could be used as a good source for developing anti-cancer agents and knowledge of these anti-cancer compounds will lead to better control of human disease.

Partial Characterization of Thrombin Inhibitor(s) Derived from Salivary Glands of the Tick, Hyalomma dromedarii, and Related Anti-Cancer Potential

A long-term blood feeder, like the Hyalomma dromedarii tick, requires extended control over all hemostatic defense mechanisms generated by the host during feeding, including blood coagulation. To overcome this, ticks have evolved numerous molecules that target proteases in the blood coagulation cascade. New insights into the role of clotting factors in the development and progression of cancer have identified anticoagulant treatment as a potential therapeutic approach. In this context, the present work assessed the anticoagulation activities of crude and fractionated salivary gland extract (SGE) prepared from semi-fed H. dromedarii females. Additionally, the antitumor effects of the potent anti-thrombin fractions were determined against colon cancer (Caco-2) and normal skin (HFB4) cells. Crude SGE significantly prolonged clotting time in prothrombin time (PT), activated partial thromboplastin time (aPTT) and thrombin time (TT) assays and inhibited thrombin in FII-activity assay. Using anion-exchange chromatography, the fractions that strongly inhibited thrombin (3.A4 and 3.A5) were eluted. Both fractions prolonged the aPTT and TT clotting times and reduced the activity of FII significantly. The protein profiles of both fractions indicated the presence of a single polypeptide band of about 99 kDa. Regarding anti-cancer potential of the tested fractions, Caco-2 cells showed reduced viability with obvious morphological changes, induced apoptosis and a reduced level of vascular endothelial growth factor (VEGF). G2/M cell cycle arrest was observed only in 3.A5-treated cells. No cytotoxic effects were observed in HFB4 cells. These results demonstrated the potential of tick-derived anticoagulants, specifically thrombin inhibitors, as effective tools in colorectal cancer treatment. New Ulva lactuca Algae Based Chitosan Bio-composites for Bioremediation of Cd(II)Ions

The current article covers the production of chitosan(CS)from shells of shrimp waste and its utilization in the preparation of eco-friendly imprinting and non-imprinting composites with Ulva lactuca algae(Alg).These bio-composites namely(Imp-Alg-25wt%CS)and(NImp-Alg-25wt%CS)were used for removal of Cd(II)*** transform infrared(FT-IR)spectra,scanning electron microscope(SEM)equipped with electron dispersive X-ray(EDX),X-ray diffraction(XRD),and elemental analysis measurements were performed to characterize these bio-composites *** highest adsorption of these sorbents towards Cd(II)ions was determined as a function of solutions pH,contact time,Cd(II)ion concentration,beads dose,and *** equilibrium experimental data were treated using various mathematical isotherm and kinetic models to approve the maximum bio-sorption capacities of NImp-Alg-25wt%CS and Imp-Alg-25wt%CS(in mg/g).The results exhibited that Imp-Alg-25wt%CS gave higher removal capacity than NImp-Alg-25wt%CS at the same optimum ***-2nd order dynamic and Langmuir isotherm models were well described in these biosorption ***,the removal behavior of Cd(II)using both bio-composites was spontaneous at room *** reusability of the sorbents,NImp-Alg-25wt%CS and Imp-Alg-25wt%CS,showed three *** addition,comparative study was also conducted for Cd(II)removal onto some reported sorbents.
