Probiotic-based nanoparticles for targeted microbiota modulation and immune restoration in bacterial pneumonia

While conventional bacterial pneumonia mainly centralizes avoidance of bacterial colonization,it remains unclear how to restore the host immunity for hyperactive immunocompetent primary and immunocompromised secondary bacterial ***,probiotic-based nanoparticles of OASCLR were formed by coating chitosan,hyaluronic acid and ononin on living Lactobacillus *** nanoparticles could effectively kill various clinic common pathogens and antibacterial efficiency was> 99.97%.Importantly,OASCLR could modulate lung microbiota,increasing the overall richness and diversity of microbiota by decreasing pathogens and increasing probiotic and commensal ***,OASCLR could target inflammatory macrophages by the interaction of OASCLR with the macrophage binding site of CD 44 and alleviate overactive immune responses for hyperactive immunocompetent ***,OASCLR could break the state of the macrophage’ s poor phagocytic ability by upregulating the expression of the extracellular matrix assembly,immune activation and fibroblast activation in immunocompromised *** macrophage’ s phagocytic ability was increased from 2.61% to 12.3 %.Our work provides a potential strategy for hyperactive immunocompetent primary and immunocompromised secondary bacterial pneumonia.

Lycium barbarum, a traditional Chinese anti-aging herb, has been shown to protect retinal ganglion cells （RGCs） in a rat chronic ocular hypertension （COH） model. Here, we investigated the expression of endothelin-1 （ET-1）, a strong vasoconstrictor, and its receptors, ETA and ETB, in the COH model and assessed the effects of Lycium barbarum on the ET-1 axis. Elevated intraocular pressure （IOP） was induced in the right eye of SD rats using argon laser photocoagulation. （1） The expression of ET-1, ETA and ETB in normal and COH retinas was studied. （2） Some COH rats were fed daily with Lycium barbarum Polysaccharides （LBP） using 1 mg/kg or phosphate-buffered saline （PBS） for 3 weeks （started 1 week before photocoagulation）. The effects of LBP on the expression of ET-1 and its receptors, ETA and ETB, in COH retina were evaluated. A semi-quantitative analysis of staining intensity was used to evaluate the expression levels of ET-1, ETA and ETB in retinal vasculature. We found that （1） Under COH condition, the immunoreactivity of ET-1 was increased in retina associated with an increase of ETB receptor immunoreactivity and a decrease of ETA receptor immunoreactivity. （2） After feeding COH rats with LBP, the expression of ET-1 was decreased with an increase of ETA expression and a decrease of ETB expression in the retina, especially in RGCs. （3） By comparing the staining intensity in the vasculature of COH retina in LBP-fed group with PBS-fed group, there was a decrease in the expression of ET-1 and ETA and an increase in ETB. In summary, ET-1 expression was up-regulated in the retina in COH model. LBP could decrease the expression of ET-1 and modulate the expression of its receptors, ETA and ETB, under the condition of COH. The neuroprotective effect of LBP on RGCs might be related to its ability to regulate the ET-1-mediated biological effects on RGCs and retinal vasculature.

Both phototherapy via photocatalysts and physical puncture by artificial nanostructures are promising substitutes for antibiotics when treating drug-resistant bacterial infectious ***,the photodynamic therapeutic efficacy of photocatalysts is seriously restricted by the rapid recombination of photogenerated electron-hole ***,the nanostructures of physical puncture are limited to two-dimensional(2D)platforms,and they cannot be fully used ***,this research developed a synergistic system of Ag_(3)PO_(4) nanoparticles(NPs),decorated with black urchin-like defective TiO_(2)(BU-TiO_(2-X)/Ag_(3)PO_(4)).These NPs had a decreased bandgap compared to BU-TiO_(2-X),and BU-TiO_(2)-X/Ag_(3)PO_(4)(3:1)exhibited the lowest bandgap and the highest separation efficiency for photogenerated electron-hole *** combination with BU-TiO_(2-X),the photostability of Ag_(3)PO_(4) improved because the oxygen vacancy of BU-TiO_(2-X) retards the reduction of Ag^(+) in Ag_(3)PO_(4) into Ag^(0),thus reducing its *** addition,the nanospikes on the surface of BU-TiO_(2-X) can,from all directions,physically puncture bacterial cells,thus assisting the hybrid’s photodynamic therapeutic effects,alongside the small amount of Ag^(+) released from Ag_(3)PO_(4).This achieves synergy,endowing the hybrid with high antibacterial efficacy of 99.76±0.15%and 99.85±0.09%against Escherichia coli and Staphylococcus aureus,respectively,after light irradiation for 20 min followed by darkness for 12 *** is anticipated that these findings may bring new insight for developing synergistic treatment strategies against bacterial infectious diseases or pathogenic bacterial polluted environments.

Monosialoganglioside 1 may alleviate neurotoxicity induced by propofol combined with remifentanil in neural stem cells

Monosialoganglioside 1（GM1） is the main ganglioside subtype and has neuroprotective properties in the central nervous system. In this study, we aimed to determine whether GM1 alleviates neurotoxicity induced by moderate and high concentrations of propofol combined with remifentanil in the immature central nervous system. Hippocampal neural stem cells were isolated from newborn Sprague-Dawley rats and treated with remifentanil（5, 10, 20 ng/m L） and propofol（1.0, 2.5, 5.0 μg/m L）, and/or GM1（12.5, 25, 50 μg/m L）. GM1 reversed combined propofol and remifentanil-induced decreases in the percentage of 5-bromodeoxyuridine（＋） cells and also reversed the increase in apoptotic cell percentage during neural stem cell proliferation and differentiation. However, GM1 with combined propofol and remifentanil did not affect β-tubulin（＋） or glial fibrillary acidic protein（＋） cell percentage during neural stem cell differentiation. In conclusion, we show that GM1 alleviates the damaging effects of propofol combined with remifentanil at moderate and high exposure concentrations in neural stem cells in vitro, and exerts protective effects on the immature central nervous system.

Delayed bone defect repairs lead to severe health and socioeconomic impacts on patients. Hence, there are increasing demands for medical interventions to promote bone defect healing. Recombinant proteins such as BMP-2 have been recognized as one of the powerful osteogenic substances that promote mesenchymal stem cells (MSCs) to osteoblast differentiation and are widely applied clinically for bone defect repairs. However, recent reports show that BMP-2 treatment has been associated with clinical adverse side effects such as ectopic bone formation, osteolysis and stimulation of inflammation. Here, we have identified one new osteogenic protein, named ‘HKUOT-S2’ protein, from Dioscorea opposita Thunb. Using the bone defect model, we have shown that the HKUOT-S2 protein can accelerate bone defect repair by activating the mTOR signaling axis of MSCs-derived osteoblasts and increasing osteoblastic biomineralization. The HKUOT-S2 protein can also modulate the transcriptomic changes of macrophages, stem cells, and osteoblasts, thereby enhancing the crosstalk between the polarized macrophages and MSCs-osteoblast differentiation to facilitate osteogenesis. Furthermore, this protein had no toxic effects in vivo. We have also identified HKUOT-S2 peptide sequence TKSSLPGQTK as a functional osteogenic unit that can promote osteoblast differentiation in vitro. The HKUOT-S2 protein with robust osteogenic activity could be a potential alternative osteoanabolic agent for promoting osteogenesis and bone defect repairs. We believe that the HKUOT-S2 protein may potentially be applied clinically as a new class of osteogenic agent for bone defect healing.

Controlling the corrosion rate is critical for practical applications of Mg-based alloys. In this work, we constructed a protective coating of hybrid polycaprolactone(H-PCL)/indocyanine green(ICG) on AZ31 Mg alloy,whose degradation rate was controlled by 808-nm nearinfrared(NIR) light irradiation. The corrosion behaviors of H-PCL/ICG coated Mg alloys were systematically investigated by potentiodynamic polarization tests, electrochemical impedance spectroscopy(EIS) and hydrogen evolution experiments. The results disclosed that the H-PCL/ICG composite coating could effectively protect Mg alloy from corroding without NIR light irradiation. In contrast, under 808-nm NIR light irradiation, the corrosion resistance of this composite coating was decreased significantly, i.e., the corrosion current density(i_(corr))increasedfrom(8.81 ± 1.068) 9 10^(-8) to(1.22 ± 0.545) 9 10^(-6) A·cm^(-2). This is because the component of ICG in the coating was excited to produce heat locally, which triggered the glass transition temperature(T_(g)) of H-PCL in the coating, resulting in the motion of the molecular chain segment. Consequently, the electrolytes penetrated the coating and corroded the Mg substrate. In vitro biological experiment indicated that the synthesized coating exhibited good ***, these findings will provide a new strategy for designing novel photoresponsive coatings to remotely adjust the degradation rate of biodegradable metals for biomedical applications.

How Not to Miss a Case of Malaria in Emergency Department in Malaria Non-Endemic Areas? Practical Approach &Experiences in Hong Kong

Human malaria is a life-threatening mosquito-borne protozoan parasitic infection in human involving female anopheline mosquitoes as vector for transmission. It is caused by Plasmodium species, most commonly, P. vivax, P. ovale, P. malariae and P. falciparum, and rarely P. knowlesi. Malaria remains a significant global health issue and is a medical emergency. It is also an important cause of morbidity and mortality in endemic areas, particularly in at-risk groups. In Hong Kong, where malaria is non-endemic, more than 20 cases of malaria per year have been notified in recent years. We still have chances encountering patients with malaria presented to public or private emergency departments. High index of clinical suspicious is utmost important for not missing a case of malaria. A practical approach for prompt identification of patients with severe malaria is essential, followed by appropriate initiation of appropriate effective antimalarial treatment within 24 to 48 hours of symptoms onset after blood taken for thick and thin smears for diagnosis. Vigilance with increased awareness of not falling into common diagnostic traps has to be alerted. The risk of missing any case of malaria presenting to emergency department could be largely minimized.
