Comparing invasive effects of five foodborne bacterial pathogens in human embryonic intestine 407 cells and human ileocecum HCt-8 cells

Objective: to refine the infectious doses of enteric bacterial pathogens in animal assays and vaccine clinical trials by studying the invasion kinetics of five bacterial pathogens with human intestinal ***: Utilizing in vitro cultured cell invasion assays with gentamicin-killing step,the invasive effects were analyzed in foodborne pathogens including Salmonella,Shigella, Yersinia, Escherichia coli(E. coli) O157 and opportunistic pathogens Citrobacter in human embryonic intestine 407 cells and ileocecum HCt-8 cells at multiplicities of infection(MOIs) of 0.04–4 000.00 E. coli HS served as a noninvasive ***: the study results showed that the bacterial invasive efficiency and the average number of internalized bacteria per host cell changed with different starting MOIs. Higher starting MOIs did not always produce more bacterial internalization. the bacterial invasion effects varied with different bacterial strains and host cell lines. E. coli O157:H7 did invade human ileocecum HCt-8 ***: this study shows that these bacteria possess different invasive patterns at various starting MOIs and also in different cell lines. the results could help to figure out the appropriate infectious doses of the bacteria in animal assays and in vaccine clinical trials. the bacterial invasion kinetics is also valuable in evaluating the safety and efficacy of live attenuated bacterial vaccines.

this study identifies the atmospheric circulation features that are favorable for the occurrence of low-level turbulence at Hong kong International Airport [below 1600 feet （around 500 m）]. By using LIDAR data at the airport, turbulence and nonturbulence cases are selected. It is found that the occurrence of turbulence is significantly related to the strength of the southerly wind at 850 hPa over the South China coast. On the other hand, the east-west wihd at this height demonstrates a weak relation to the occurrence. this suggests that turbulence is generated by flow passing Lantau Island from the south. the southerly wind also transports moisture from the South China Sea to Hong kong, reducing local stability. this is favorable for the development of strong turbulence. It is also noted that the strong southerly wind during the occurrence of low-level turbulence is contributed by an anomalous zonal gradient of geopotential in the lower troposphere over the South China Sea. this gradient is caused by the combination of variations at different timescales. these are the passage of synoptic extratropical cyclones and anticyclones and the intraseasonal variation in the western North Pacific subtropical high. the seasonal variation in geopotential east of the tibetan Plateau leads to a seasonal change in meridional wind, by which the frequency of low-level turbulence is maximized in spring and minimized in autumn.

the use of sensors for monitoring livestock has opened up new possibilities for the management of livestock in extensive grazing *** work presented in this paper aimed to develop a model for predicting the metabolisable energy intake(MEI)of sheep by using temperature,pitch angle,roll angle,distance,speed,and grazing time data obtained directly from wearable sensors on the sheep.A Deep Belief Network(DBN)algorithm was used to predict MEI,which to our knowledge,has not been attempted *** results demonstrated that the DBN method could predict the MEI for sheep using sensor data *** mean square error(MSE)values of 4.46 and 20.65 have been achieved using the DBN model for training and testing datasets,*** also evaluated the influential sensor data variables,i.e.,distance and pitch angle,for predicting the *** study demonstrates that the application of machine learning techniques directly to on-animal sensor data presents a substantial opportunity to interpret biological interactions in grazing systems directly from sensor *** expect that further development and refinement of this technology will catalyse a step-change in extensive livestock management,as wearable sensors become widely used by livestock producers.

Long-term nucleos(t)ide analogue therapy in chronic hepatitis B virus(HBV)infection is effective in suppressing viral replication and reducing liver-related complications. However, HBV-related liver events can still occur in different patient sub-groups. there is emerging evidence that, similar to chronic hepatitis C virus infection, metabolic risk factors may play a role in the disease process of chronic HBV. While the mechanistic nature of metabolic-HBV interactions remains uncertain, studies in different HBV-infected populations have demonstrated that hepatic steatosis, increased body-mass index, diabetes, or a combination of different metabolic risk factors are associated with an increased risk of hepatocellular carcinoma and cirrhosis. the impact of metabolic risk factors is especially prominent in patients with quiescent virological activity,including on-treatment patients with effective viral suppression. As the proportion of on-treatment chronic HBV patients increases worldwide,longitudinal studies determining the relative risks of different metabolic parameters with respect to clinical outcomes are needed. Future studies should also determine if metabolic-directed interventions can improve disease outcomes in chronic HBV.

Objective to investigate sex hormone deficiency related osteoporosis and efficacy of different therapies. Methods Orchiectomized and ovariectomized rat models are used to investigate sex hormone deficiency related osteoporosis and efficacy of different therapies. A rat vertebral body can be longitudinally divided into central portion, which contain more trabecular bone, and para-endplate portions which contain more compact bone. In matured male and female Wistar and Sprague-Dawley rat lumbar spines, we investigated baseline bone mineral density （BMD） characteristics and the differential segmental responses in bone loss within the lumbar vertebral body post gonadal surgery with clinical multidetector computed tomography. Results Para-endplate sections had a higher BMD than central sections. the cephalad para-endplate sections had a higher BMD than the caudad para-endplate sections. Eight weeks after gonadal removal, there was more bone loss in central sections than para-endplate sections. the relative difference of bone loss between para-endplate and central sections was more apparent in male rats than in female rats. there was more bone loss in caudad sections than cephalad sections; this lead to a further increase of BMD difference between caudad para-endplate sections and cephalad para-endplate sections post gonadal surgery. Conclusion the approach described in this study provided a consistent way to study BMD change within predominantly compact bone portion and trabecular bone portion of the vertebral body.

Although lanthanide double-decker complexes with hetero-macrocyclic ligands as functional luminescent and magnetic materials have promising properties,their inferior water solubility has negated their biomedical ***,four water-soluble homoleptic lanthanide(Ln=Gd,Er,Yb and La)sandwiches with diethylene-glycoldisubstituted porphyrins(DD)are reported,with their structures proven by both quantum chemical calculations and scanning tunneling *** findings demonstrate that the near-infrared emission intensity and singlet oxygen(1O2)quantum yields of YbDD and GdDD in aqueous media are higher than those of the reported capped lanthanide monoporphyrinato analogues,YbN and GdN;the brightness and luminescence lifetime in water of YbDD are greater than those of *** work provides a new dimension for the future design and development of molecular theranostics-based water-soluble double-decker lanthanide bisporphyrinates.
