Prohibitin(PHB),an evoluti on arily con served mitochondrial inner membra ne protein,is highly expressed in cells that require strong mitoch on drial ***,we dem on strated that the deleti on of Phb in spermatocytes results in impaired mitochondrial *** addition,PHB expression in the mitochondrial sheath of human sperm has a significantly negative correlation with mitochondrial reactive oxygen species levels,but a positive one with mitochondrial membrane potential and sperm *** results suggest that mitochondrial PHB expression plays a role in sperm ***,the mechanism of PHB-mediated regulation of sperm motility remai ns unk ***,we dem on strate for the first time that PHB interacts with protei n kinase B(AKT)and exists in a complex with phospho-PHB(pT258)and phospho-AKT in the mitochondrial sheath of murine sperm,as determined using colocalization and coimmunoprecipitation *** blocking AKT activity using wortmannin(a phosphatidylinositol 3-kinase[PI3K]inhibitor),murine sperm have significantly(P<0.05)decreased levels of phospho-PHB(pT258)and the total and progressive ***,significantly(P<0.05)lower levels of phospho-PI3K P85 subunit a+γ(pY199 and pY46)and phospho-AKT(pS473;pT308)are found in sperm from infertile asthenospermic and oligoasthenospermic men compared with no rmospermic subjects,which suggest a reduced activity of the PI3K/AKT pathway in these in fertile ***,these sperm from infertile subjects also have a significantly(P<0.05)lower level of phospho-PHB(pT258).Collectively,our findings suggest that the interaction of PHB with AKT in the mitochondrial sheath is critical for sperm motility,where PHB phosphorylation(pT258)level and PI3K/AKT activity are key regulatory factors.

Assessment of Serum Vitamin D Levels in Children With Pityriasis Alba:A Retrospective Study

Objective:This study was performed to analyze the correlation between pityriasis alba(PA)and the serum 25-hydroxyvitamin D status in ***:The study population comprised 460 patients with PA and 254 age-and sex-matched healthy *** serum level of 25-hydroxyvitamin D was compared between the two groups by studentt ***:The serum level of 25-hydroxyvitamin D was significantly lower in patients with PA than in healthy controls(P<0.001).There were statistically significant differences in the serum vitamin D status between the PA group and control group(P<0.001);specifically,patients with PA had a significantly higher frequency of vitamin D deficiency(52.83%vs.24.35%,P=0.001)and lower frequency of vitamin D sufficiency(22.83%vs.37.01%,P<0.001)than *** patients showed significantly lower serum level of vitamin D than controls in deficient and sufficient groups(P=0.005 and 0.027,respectively),and in insufficient group,serum vitamin D level was lower in PA patients than in controls,but it showed no statistical difference(P=0.061).Conclusion:Vitamin D may play a role in the pathogenesis of *** investigations are required to establish the relationship between the severity of PA and the vitamin D status and to evaluate the potential for use of vitamin D in the treatment of PA.

Dear Editor,Klebsiella pneumoniae(Kp)has become the most important bacterial pathogen causing high mortality rates in clinical patients due to the continuous evolution to several important variants such as carbapenem-resistant(CR-Kp),hypervirulent(hvKp)and both CR and hv ***(CR-hvKp).The high mortality caused by clinical hvKp is attributed to the non-response to antibiotic treatment.

Antioxidant and hepatoprotective effects of Ajuga nipponensis extract by ultrasonic-assisted extraction

Objective: To investigate suitable condition for extraction of the active components from Ajuga nipponensis(A. nipponensis). Methods: Orthogonal experimental design was used to determine the optimal extraction parameters for ecdysterones and flavonoids. Finally, the hepatoprotective abilities of A. nipponensis extracts were evaluated by CCl_4-induced animal models. Results:Maximum yields of flavonoids(7.87±0.10) mg/g and ecdysterones(0.73±0.02) mg/g could be obtained when the extraction time was 50 min, the extraction temperature was 60 ℃, and the ratio of sample to 70%(v/v) ethanol was 1:20(w/w). The antioxidant property of A. nipponensis was correlated to the concentration of its extracts. At 5 mg/m L, A. nipponensisextract scavenged 84.8% of DPPH radical and had absorbance values of 2.43±0.04 reducing power. Upon CCl_4-induced liver injury, glutamic oxaloacetic transaminase and glutamic pyruvic transaminase decreased significantly after the mice were treated with A. nipponensis. Histological researches also explained that A. nipponensis reduced the extent of liver lesions induced by CCl_4. Conclusions: A. nipponensis exhibited potent antioxidant activity in chemical experimental models and hepatoprotective effect against CCl_4-induced liver damage.

Phenylketonuria in Hong Kong Chinese： a call for hyperphenylalaninemia newborn screening in the Special Administrative Region, China

Hyperphenylalaninemia is one of the commonest inborn errors of metabolism affecting approximately 1 in 15 000 live births. Among Chinese, BH4 deficiency leading to hyperphenylalaninemia is much commoner than in Caucasians. Exact diagnosis is important for the treatment and genetic counseling. In 2000, newborn screening for phenylketonuria is mandatory by law in China throughout the whole country. However, it is not yet included in the newborn screening program of the Hong Kong Special Administrative Region, China. Published data on hyperphenylalaninemia among Hong Kong Chinese are largely lacking. We report a 1-year-old Hong Kong Chinese girl with severe 6-pyruvoyl-tetrahydropterin synthase （PTPS） deficiency. The patient presented with infantile hypotonia and was misdiagnosed as cerebral palsy. She had very mild hyperphenylalaninemia （95 IJmol/L）, significantly high phenylalnine-to-tyrosine ratio （3.1）, and ele~,ated prolactin of 1109 m lU/L. Genetic analysis confirmed a homozygous known disease-causing mutation PTS NM_000317.1： c.259C〉T; NP_000308.1： p.P87S in the proband. In our local experience, while the estimated prevalence of hyperphenylalaninemia due to PTPS deficiency was reported to be 1 in 29 542 live births, not a single case of phenylalanine hydroxylase deficiency has been reported. Furthermore, there is a general lack of awareness of inherited metabolic diseases in the community as well as among the medical professionals. Very often, a low index of clinical suspicion will lead to delay in diagnosis, multiple unnecessary and costly investigations, prolonged morbidity and anxiety to the family affected. We strongly recommend that expanded newborn screening for hyperphenylalaninemia should be implemented for every baby born in the Hong Kong Special Administrative Region, China.

Proactive infection control measures to prevent nosocomial transmission of carbapenem-resistant Enterobacteriaceae in a non-endemic area

Background Identification of hospitalized carbapenem-resistant Enterobacteriaceae （CRE）-positive patient is important in preventing nosocomial *** objective of this study was to illustrate the implementation of proactive infection control measures in preventing nosocomial transmission of CRE in a healthcare region of over 3200 beds in Hong Kong between October 1,2010 and December 31,*** The program included active surveillance culture in patients with history of medical tourism with hospitalization and surgical operation outside Hong Kong within 12 months before admission,and ＂added test＂ as an opportunistic CRE screening in all fecal specimens submitted to the *** investigation and contact tracing were conducted for CRE-positive *** quantitative culture was performed on CRE-positive patients and the duration of fecal carriage of CRE was *** During the study period,a total of 6533 patients were screened for CRE,of which 76 patients were positive （10 from active surveillance culture,65 from ＂added test＂,and 1 secondary case from contact tracing of 223 patients with no nosocomial outbreak）,resulting in an overall rate of CRE fecal carriage of 1.2％.The median time of fecal carriage of CRE was 43 days （range,13-119 days）.Beta-lactam-beta-lactamase-inhibitors,cephalosporins,and fluoroquinolones were associated significantly with high fecal bacterial load when used 90 days before CRE detection,while use of cephalosporins,carbapenems,and fiuoroquinolones after CRE detection are significantly associated with longer duration of *** duration of fecal carriage of CRE also correlates significantly with the initial fecal bacterial load （Pearson correlation：0.53; P=0.02）.Conclusion Proactive infection control measures by enhanced surveillance program identify CRE-positive patients and data obtained are useful for the planning of and resource allocation for CRE control.

Function of chymase in the heart angiotensin Ⅱ formation in transgenic mice

The myosin light chain 2 promoter-human heart chymase (MLC2-hChymase) transgenic mice founded by our laboratory were used as the model to study the function of chymase in the heart angiotension Ⅱ (Ang Ⅱ) formation and heart remodeling. Tissue-specific expression of human heart chymase gene and transcriptional expression of type Ⅰ and type Ⅲ collagens genes were analyzed by RT-PCR. Activities of chymase, ACE and the levels of Ang Ⅱ in heart and plasma were determined with radioimmunoas-say (RIA) kit. Activity of heart matrix metalloproteinase-9 (MMP-9) was detected using gelatin zymography. The cardiac hypertrophic phenotypes were also observed with the physiological and morphological methods. The results in the MLC2-hChymase transgenic mice indicated: ( i ) human heart chymase gene was expressed specially in the heart; (u) heart chymase activity increased markedly in the transgenic mice vs non-transgenic mice (control) (0.27±0.07 U/mg vs. 0.15±0.02 U/mg, P<0.05) with no significant difference in

With the increasing incidence of heavy rainfall events,particularly over the monsoon regions,the highly dense populations are more vulnerable[1].Research initiatives on observation,modeling,and prediction of monsoon heavy rainfall have been promoted actively by World Weather Research Programme's(WWRP)Working Group on Tropical Meteorology Research(WGTMR)of the World Meteorological Organization(WMO)since *** of monsoon-heavy-rainfall workshops were held in Beijing(2011),Petaling Jaya(2012),and New Delhi(2015)to benefit scientists worldwide and forecasters from the National Meteorological and Hydrological *** international Research and Development Project,namely,the Southern China Monsoon Rainfall Experiment(SCMREX)[2]was established in 2013 to coordinate field campaign experiments and to conduct scientific research on presummer(April-June)heavy rainfall processes in southern China.
